Immunity-related genes and gene families in Anopheles gambiae   总被引:2,自引:0,他引:2  
We have identified 242 Anopheles gambiae genes from 18 gene families implicated in innate immunity and have detected marked diversification relative to Drosophila melanogaster. Immune-related gene families involved in recognition, signal modulation, and effector systems show a marked deficit of orthologs and excessive gene expansions, possibly reflecting selection pressures from different pathogens encountered in these insects' very different life-styles. In contrast, the multifunctional Toll signal transduction pathway is substantially conserved, presumably because of counterselection for developmental stability. Representative expression profiles confirm that sequence diversification is accompanied by specific responses to different immune challenges. Alternative RNA splicing may also contribute to expansion of the immune repertoire.  相似文献

The dry season in tropical regions urges livestock to feed on nutritionally very poor diets. It has not been explored how tropical sheep—assumed grazers—and tropical goats—intermediate browsers—cope with a high-fibre tannin-rich diet. This study was designed to determine the effects of a high-fibre and tannin-rich diet on the flow of important microminerals iron (Fe), zinc (Zn) and copper (Cu) throughout the digestive tract of tropical sheep and goats. The feeding trial was set up with twelve adult male animals, six sheep with mean body weight (BW) of 30.3 ± 1.6 kg and six goats with mean BW of 26.4 ± 2.2 kg. The feed consisted of 36% leaves of Millettia ferruginea, 61% hay and 3% concentrate and was offered at 3% of BW (all on dry matter (DM) basis). The total faecal collection was carried out for 7 consecutive days. At the end of the experimental period, the animals were slaughtered to collect liver and digesta samples from the gastrointestinal tract. Feed, digesta and faecal samples underwent analysis of Fe, Zn, and Cu and total tannins (TT). Goats had significantly higher reticulum Cu concentrations expressed on DM as compared to sheep. Faecal Cu concentrations were higher for goats compared to sheep. Reticulum and colon digesta Zn levels were higher in goats than sheep. Abomasum and colon Fe levels were higher in sheep than goats when expressed on DM. These results suggest differences in feed intake, micromineral absorption, secretion and excretion between sheep and goats, pointing to a divergent mineral metabolism as an adaptation to the challenge of a dry season diet having very low nutritive value.  相似文献

The isotopic composition of lead pollution in the environment provides information as to the source of emission. The 206Pb/207Pb ratio for Canadian industrial emissions (1.153±0.005) is significantly less than U.S. industrial emissions (1.213±0.008) making it possible to determine the relative contributions of emissions from both countries within Eastern North America by means of a simple isotopic mixing model. Profundal sediments in lakes chronicle contaminant inputs and are therefore useful monitors of environmental pollution. Surface sediment from 32 sediment cores across Quebec and Ontario, Canada were analyzed for 206Pb/207Pb to ascertain the relative proportions of Pb emission from Canada and the U.S.A. Data show that U.S. contributions to the total lead burden in surficial sediments across much of southern Quebec and Ontario are often in excess of 50%. Local sources were particularly important in the Eastern Township region of Quebec which lies 200 km south east of the city of Montreal. The results are discussed in relation to the major sources of industrial lead emissions in North America.  相似文献

Tomato products and especially concentrated tomato paste are important sources of antioxidants in the Mediterranean diet. Tomato fruit contain well-known antioxidants such as vitamin C, carotenoids, flavonoids, and hydroxycinnamic acids. The industrial processing of this fruit into tomato paste involves several treatments that potentially affect the final profile of antioxidants and other metabolites in the commercial product. Here we have used both biochemical and metabolomic techniques to assess the effect of each separate step in the industrial production chain starting from fresh fruit to the final tomato paste. Material was collected from five independent tomato paste production events spread over two successive years. Samples comprised the intact ripe fruits and semifinished products after fruit-breaking, separation of the pulp from skin and seeds, evaporation, and finally after canning and pasteurization. The effect of each processing step was determined by different types of analysis. First, the total antioxidant capacity and total phenolic content were determined by commonly used spectrophotometric methods. Second, individual antioxidants in the extracts were identified and compared using an HPLC with online antioxidant detection. Third, in each sample the levels of the major individual antioxidants present, i.e., vitamin C, phenolic compounds (such as rutin and chlorogenic acid), tocopherols, and carotenoids, were quantified. Fourth, an untargeted metabolomic approach using LC-QTOF-MS was used to identify those production steps that have the largest impact on the overall metabolic profile in the final paste as compared to the original fruits. This multifaceted approach has revealed that each processing step induces specific alterations in the metabolic profile, as determined by the different analysis procedures, and that in particular the fruit-breaking step and the removal of seed and skin significantly affect the levels of antioxidants and many other metabolites present in commercial tomato paste.  相似文献

Strawberry contains high levels of micronutrients and phytochemical compounds. These exhibit functional roles in plant growth and metabolism and are also essential for the nutritional and organoleptic qualities of the fruit. The aim of the present work was to better characterize the phytochemical and antioxidant profiles of the fruit of nine different genotypes of strawberry, by measuring the total flavonoid, anthocyanin, vitamin C, and folate contents. Cultivar effects on the total antioxidant capacities of strawberries were also tested. In addition, the individual contribution of the main antioxidant compounds was assessed by HPLC separation coupled to an online postcolumn antioxidant detection system. This study showed the important role played by the genetic background on the chemical and antioxidant profiles of strawberry fruits. Significant differences were found between genotypes for the total antioxidant capacity and for all tested classes of compounds. The HPLC analyses confirmed qualitative and quantitative variability in the antioxidant profiles. These studies show that differences exist among cultivars, applicable in dietary studies in human subjects.  相似文献

Methods for introduction of an improved cultivar of Ber tree (Ziziphus mauritiana Lam.) also called Jujube and its management on farms through intercropping with African eggplant (Solanum aethiopicum L.) and sorghum [Sorghum bicolor (L.) Moench] were tested in Sanankoroba, Mali. An experimental plantation was established on-farm with seedlings of the local variety of Ber half of which were grafted in situ with an improved cultivar of Ber called Seb. Crop production, Ber growth and fruit production were assessed over two cropping seasons. Both the improved and local varieties of Ber had no detrimental effect on either eggplant or sorghum, both in terms of yield and nutritional quality. In fact a beneficial effect of trees was found on the performance of both crops (yield and dry matter production) which suggests complementarities in resource use. The high level of fruit production by the improved variety of Ber observed on farms under rain-fed conditions may be a source of additional income and diversification of diet for rural communities in West Africa. Therefore, farmers’ adoption of growing improved Ber varieties in association with food crops may help considerably in improving food security and alleviating poverty in the region.  相似文献

Xanthomonas campestris pv. musacearum, the causal agent of Xanthomonas wilt of banana (XW), does not infect or cause symptom development in all physically attached shoots in an infected mat. Incomplete/partial systemicity and latent infections often occur. The single diseased stem removal (SDSR, the removal of only symptomatic plants) technique depends on these observations. The SDSR technique, as an alternative or complementary practice to complete mat uprooting (CMU) for XW control, was evaluated at eight XW pilot sites in eastern Democratic Republic of Congo as a novel control option. This technique is low-cost, simple and easily applicable. Within one month, XW plant incidence at the experimental sites declined to below 10%, while within three and 10 months declined to below 2% and 1%, respectively. Restoration of banana plots was observed even in plots that initially had over 80% plant disease incidence. CMU removes a larger portion of the inoculum in a field but is very tedious, time consuming and costly in terms of labour and lost production, due to the premature cutting of symptomless plants that potentially could bear a bunch. CMU can potentially prevent further spread when XW appears for the first time on a farm or location. The choice of CMU relative to SDSR also depends largely on farming objectives. CMU can be carried out in intensive and market-oriented production systems, whose ultimate target is eradication, for example, in South-Western Uganda. In contrast, SDSR is more appealing to subsistence-oriented production, such as in eastern DR Congo, Burundi or central Uganda, whose target is more oriented towards management/control. SDSR can be suggested where access to clean planting material is difficult, thus could be recommended to a very large percentage of small-scale farmers in the currently affected banana-based production systems in east and central Africa.  相似文献

OBJECTIVE: To evaluate canarypox-vectored equine influenza virus (EIV) vaccines expressing hemagglutinins of A/equine/Kentucky/94 (vCP1529) and A2/equine/Ohio /03 (vCP2242) for induction of antibody responses against canine influenza virus (CIV) in dogs. ANIMALS: 35 dogs. PROCEDURES: Dogs were randomly allocated into 4 groups; group 1 (n = 8) and group 2 (9) were inoculated SC on days 0 and 28 with 1.0 mL (approx 10(5.7) TCID(50)) of vCP1529 and vCP2242, respectively. Dogs in group 3 (n = 9) were inoculated twice with 0.25 mL (approx 10(5.7) TCID(50)) of vCP2242 via the transdermal route. The 9 dogs of group 4 were control animals. All dogs were examined for adverse reactions. Sera, collected on days -1, 7, 13, 21, 28, 35, and 42, were tested by hemagglutination inhibition (HI) and virus neutralization (VN) assays for antibodies against CIV antigens A/Canine/FL/43/04-PR and A/Canine/NY/115809/05, respectively. RESULTS: Inoculations were tolerated well. The HI and VN antibodies were detected by 7 days after primary inoculation. Most dogs of groups 1 and 2 and all dogs of group 3 had detectable antibodies by 14 days after initial inoculation. The second inoculation induced an anamnestic response, yielding geometric mean HI titers of 139, 276, and 1,505 and VN titers of 335, 937, and 3,288 by day 42 (14 days after booster inoculation) in groups 1, 2, and 3, respectively. CONCLUSIONS AND CLINICAL RELEVANCE: Canarypox-vectored EIV vaccines induce biologically important antibodies and may substantially impact CIV transmission within a community and be of great value in protecting dogs against CIV-induced disease.  相似文献
