This study examined the influence of swimbladder inflation (SBI) failure on mortality (Experiment 1), lordotic deformity (Experiment 2) and growth (Experiment 1, 2) in Pacific bluefin tuna, Thunnus orientalis, postflexion larvae and juveniles by producing larvae lacking inflated swimbladders. Experiment 1 was conducted for postflexion larvae to juveniles (from 18 to 30 days‐post‐hatch; dph). Mortality was not significantly different between the fish with (WIS) and without (WOIS) inflated swimbladders. Standard length (SL) and body weight (BW) were significantly smaller in WOIS than WIS. Moreover, the SBI was found in WOIS after postflexion stage. In Experiment 2, two examination trials were conducted on SBI, vertebral deformity and growth for juvenile stage. Lordotic deformities were found neither in the WOIS nor in the WIS. Although SL and BW were significantly smaller in WOIS than WIS at 22 dph, after 37 dph no significant differences were found between them. The results of Experiment 1 and 2 indicate that SBI failure causes growth retardation until juveniles of 30 dph; however, it does not cause growth retardation in juveniles after 37 dph and mortality of postflexion larvae and juveniles. This study was undertaken to examine pre- and postimplantation developmental potency of cryopreserved embryos that had undergone in vitro growth (IVG), maturation (IVM) and fertilization (IVF) of oocytes from the preantral follicle stage. An oocyte culture system for IVG and IVM was used in oocyte-granulosa cell complexes (OGCs) derived from preantral follicles in 12-day-old mice. The rate of oocyte maturation was improved by the addition of gonadotropins (FSH/LH) and cytokines (IGF-I/SCF) to culture medium for IVG. During culture for IVG, estradiol-17β and progesterone concentrations increased progressively to the latter period of culture. This culture system enabled IVG, IVM, IVF and pre- and postimplantation development. From 90 cryopreserved 2-cell stage embryos transferred into recipients after warming, 10 live pups were produced. Cryopreservation of embryos by vitrification at the 2-cell stage showed no harmful effect on development to the blastocyst stage or on the cell numbers of the inner cell mass (ICM) and trophectoderm (TE). This study demonstrated that embryos derived from oocytes grown in vitro have tolerance for vitrification and competence to develop to term after warming. This IVG-IVM-IVF technology combined with embryo cryopreservation might be useful for assisted reproduction in mice. 相似文献

A method of measuring moisture content of wood is introduced. This method uses a vibrational technique. A shaker driven by swept sine signals was used for the accurate generation of vibration. A cylindrical wood sample was used to verify the method of measuring the moisture content. Resonance frequencies of the wood sample decreased as its moisture content increased. An experiment was performed by measuring the vibration of a tree to explore the feasibility of applying the method to the measurement of the moisture content of trees. In tomato plants, salt stress has been induced to improve the quality of fruit. In general, plants under salt stress produce reactive oxygen species (ROS). Plants have their own ROS scavenging systems (antioxidant systems). In tomato plants, salt-induced changes in antioxidant systems have been examined extensively in leaves and roots; however, detailed information about salt-stressed fruits is not available. We examined the salt-induced changes in the antioxidant systems of the pericarp (containing epidermis) and pulp (containing seeds, placenta, and locule) during fruit ripening. Salt treatments were applied by adding 100 mM NaCl to the nutrient solution. In the pericarp and pulp, lipid peroxidation and hydrogen peroxide content were not increased by salt stress during ripening, indicating the absence of salt-induced oxidative stress. In the pericarp, the activities of superoxide dismutase (EC 1.15.1.1) and ascorbate (ASA)–glutathione (GSH) cycle-related enzymes increased with salt stress at the turning stage. Thus, at the turning stage, the antioxidant system may contribute to the enzymatic reaction involved ASA–GSH cycle. However, during the red and over-ripe fruit stages, salt stress produces little effect on antioxidant enzymes. In addition, the concentrations of antioxidants, such as the reduced form of ASA and GSH, increased during ripening in the control fruit, but those in the salt-stressed fruit remained unchanged. Therefore, the antioxidant system may contribute to the nonenzymatic reactions such as ASA and GSH taking place during the red and over-ripe fruit stages. In contrast, in the pulp, salt stress produces little effect on antioxidants and antioxidant enzymes. These results indicate that the salt-stressed fruit has protection mechanisms against salt-induced oxidative stress during ripening in both the pericarp and pulp. Abstract: The question of whether the ovulation and spawning time in chub mackerel Scomber japonicus is entrained by a circadian rhythm was raised by our previous experiments. Further questions were also raised about whether the time course of human chorionic gonadotropin (hCG)-induced final oocyte maturation (FOM) and ovulation reflected the natural time course induced by endogeneous pituitary gonadotropin (GtH). To address these questions, hCG and gonadotropin-releasing hormone analog (GnRHa) were administered at two 'opposite' times, 14:00 and 02:00 hours, and the time courses of FOM and ovulation were compared. When hCG was injected, ovulation occurred 33 h post-injection in both groups, regardless of the timing of the hCG injection. The timing of ovulation in chub mackerel depends on the timing of hCG injection, but apparently not on circadian rhythms. When GnRHa was injected, ovulation began at 36 h post-injection of GnRHa, regardless of the timing of injection. These results indicate that the time course of FOM and ovulation in the chub mackerel followed a similar pattern whether stimulated by hCG injection or spontaneous luteinizing hormone (LH) surge because GnRHa induces the secretion of endogenous GtH (primarily LH) from the fish pituitary. Thus, it is concluded that the time course of hCG-induced FOM and ovulation in chub mackerel follows the natural time course induced by endogenous pituitary LH. 相似文献

ABSTRACT: In bluefin tuna aquaculture, collision of juveniles with the tank or net walls is a major cause of high mortality. This problem may be related to color sensibility of the visual mechanisms of this species. As a first step in understanding of color vision of Pacific bluefin tuna Thunnus orientalis , we applied a molecular technique and histology to study cone cell distribution in the retina of juvenile fish. We isolated three cone opsin genes encoding one blue-sensitive ( SWS2 ) and two green-sensitive ( RH2 ) visual pigments. In situ hybridization revealed that SWS2 mRNA is localized in the single-cone photoreceptors. The localization of the two RH2 signals in distinct cone cells was not determined, probably because of the high homology between the two RH2 genes. Single-cone photoreceptors appeared frequently in the ventral–temporal retina in approximately 80-mm fish and in the temporal retina in approximately 230-mm fish. These cone distributions may define a visual field with best color contrast vision in front and above the fish with a short wavelength (blue) reflecting target (sensed by single cones), and may be enhanced against the longer wavelength (green) background when fish see a target below them (sensed by double cones). 相似文献

Three photoperiods (12L:12D, 16L:8D and 24L:0D) were used to investigate the growth performance and stress response in red sea bream, Pagrus major (body weight 200–400 g). Fish were fed a commercial diet to apparent satiation, two times a day for 8 weeks. Fish exposed to a 24L:0D photoperiod showed a significantly higher weight gain (%) than those exposed to other photoperiods (P<0.05). The best specific growth rate and feed conversion efficiency were achieved at 24L:0D and 16L:8D, without significant differences among them. Although fish exposed to 16L:8D showed a significantly higher plasma level of cortisol than those exposed to other photoperiods in the third week, there was no major variation in the cortisol level among the treatments either at the sixth week or at the end of this study. There were no significant differences either in the haematocrit level or the plasma levels of glucose, total cholesterol and total protein among the treatments during this study. The results revealed that the growth performance of red sea bream reared from 200 to 400 g can be stimulated significantly using a continuous (24L:0D) photoperiod without any measurable significant stress response in fish. 相似文献

The growth of juvenile chub mackerel Scomber japonicus collected in the western North Pacific Ocean in 2007 and 2009 was examined based on the evidence of otolith daily increment formation in captive specimens. There was a significant difference in the relationship between known age and number of increments in the frontal and sagittal planes. Repeated markings on the otolith using Alizarin complexone and the coefficient of variation in number of increments suggest that the increments in the frontal plane of the otolith are more suitable for age estimation than those in the long and short axes of the sagittal plane. The increments in the frontal plane formed daily, and the first ring was usually deposited 3 days after hatch. Age of wild juveniles ranged from 24 to 211 days after hatch based on the frontal plane method. The estimated hatching periods of specimens ranged from February to June, but the April-hatched specimens were collected throughout the sampling periods of 2007 and 2009. The Gompertz growth model showed a difference in growth pattern in specimens between 2007 and 2009. The juveniles in 2009 appeared to grow more quickly than those in 2007 until summer, but thereafter the 2009 specimens seemed to grow more slowly. 相似文献
