Inbred strains of rodents have become indispensable for a wide range of biological studies. It has generally been accepted that genetic uniformity is unlikely to be achieved before 20 generations of brother x sister matings discouraging attempts to inbreed larger mammals. Nevertheless, pigs, homozygous for the swine MHC haplotype SLA b/b, have been inbred at the Babraham Institute for almost thirty years and used for immunological studies. Since the herd had not been studied at the DNA level, DNA profiling at multiple hypervariable loci was performed and surprisingly little genetic polymorphism and extremely high inter-individual resemblance were observed reminiscent of that observed in inbred strains of mice. 相似文献

In situ digestion kinetics of neutral detergent insoluble nitrogen (NDIN) from alfalfa (Medicago sativa L.) harvested at one-tenth bloom and eastern gamagrass (Tripsacum dactyloides L.) harvested at the boot (GGB), anthesis (GGA), and physiological maturity (GGM) stages of growth were determined with nonlinear regression techniques. Whole-plant tissue and associated leaf and stem fractions were incubated in the ventral rumen simultaneously. On a wholeplant basis, potential extents of degradation were particularly high (> or =904 g/kg NDIN) for GGB and GGA, relative to those of GGM and alfalfa (772 and 658 g/kg NDIN, respectively). For all plant parts, degradation rates of NDIN were faster (P<.05) for alfalfa than for all gamagrass forages. Degradation rate of NDIN did not differ (P>.05) across maturities for any gamagrass tissue type. These results indicate 1) that phenological development and lignification do not limit the rate of NDIN degradation in gamagrass forages but do markedly limit the potential extent of NDIN availability and 2) that most of the NDIN in these forages is potentially available in the rumen and can contribute to the ruminal N supply. Our secondary objective was to compare estimates of N escaping ruminal degradation that were determined on the basis of NDIN degradation kinetics (NDIN method) with those determined traditionally, on the basis of total residual N. The NDIN method mathematically eliminates all neutral detergent soluble N from consideration as part of the pool of dietary N potentially escaping the rumen intact. Estimates of rumen escape nitrogen determined on the basis of degradation rates of NDIN were consistently less than corresponding estimates that were determined on the basis of total residual N. When ruminal escape N that was determined with the NDIN method was regressed on corresponding estimates with the total residual N method, the slopes of the regression lines were .53 and .66 for assumed passage rates of .02 and .06 h(-1), respectively. For the forages evaluated in this study, these results indicate that neutral detergent soluble N may make important contributions to the pool of N escaping ruminal degradation. 相似文献

Two experiments were completed to compare the supplemental effects of yeast Saccharomyces cerevisiae and sub-therapeutic antibiotics in high-fibre and low-protein diets for broiler chicks. In experiment 1, yeast was added at 1.5, 3.0 and 6.0 g kg-1 while penicillin, tylosin or neoterramycin were added at 150 mg kg-1 into different batches of a high-fibre diet containing 250 g kg-1 palm kernel meal. Saccharomyces cerevisiae and antibiotics increased BW, BWG, FCR, feed intake, carcass and breast weights above the control. Small intestine weight was reduced by antibiotics, while yeast caused a lower deposition of abdominal fat. Liver weights of different treated broilers were similar. The performance indices were superior on penicillin and 3.0 g kg-1 yeast compared to other supplements evaluated. In the second experiment, 1.5 and 3.0 g kg-1 of yeast, and 0.75 mg kg-1 of procaine penicillin, zinc bacitracin and tylosin were separately added to a 180 g kg-1 crude protein diet. Performance of the chicks with additives was compared with unsupplemented negative and positive controls containing 180 and 210 g kg-1 crude protein respectively. Broilers fed with supplements had superior BW, BWG, FCR and feed intake compared to the negative control, but carcass data were similar. Dietary penicillin increased BW, BWG, feed intake more than other supplements, but yeast stimulated higher FCR. Data from both studies indicated that Saccharomyces cerevisiae comparatively increased growth and carcass weight and reduce abdominal fat, therefore, it can serve as a natural substitute for antibiotics in broiler diet. 相似文献

A retrospective case series of 26 cats with diffuse retinal degeneration is presented. The most common presenting complaints included bumping into objects, dilated pupils, and reluctance to jump. Ophthalmic examination findings were consistent with those reported in dogs with progressive retinal atrophy. Breed predilection of the Siamese cat was observed. Cats with primary retinal degeneration presented late in the clinical course of their disease, when vision loss was severe. Early symptoms such as night blindness and secondary ocular complications (i.e., cataract and retinal detachment), reported in dogs with progressive retinal degeneration, were not observed in this study. In order to develop procedures to label the main bovine leucocyte populations in paraffin embedded sections, the immunoreactivity of 25 monoclonal antibodies (mAbs) to different leucocyte antigens was assessed with formal dichromate (FD5) and 10% formalin fixation, a battery of antigen retrieval (AR) methods, and the biotin-tyramide amplification system. All the leucocyte populations investigated (CD2+, CD4+, CD8+, WC1+ T lymphocytes, B cells and macrophages) were strongly and specifically detectable under an appropriate combination of mAb, AR method and signal amplification system. CD4 and CD8 required the most stringent conditions and could only be demonstrated in FD5 fixed sections. For detection of CD2, WC1+ T lymphocytes, B cells and macrophages, all the mAbs produced immunoreactivity in FD5 or formalin fixed tissues. The need to check a range of different AR methods is stressed, as the method of choice varied for each individual mAb. The incorporation of the signal amplification system was necessary to observe a strong signal and the complete distribution of CD4, CD8 and B cells. Fixation by FD5 proved to be better than formalin for the preservation of surface antigens but it was inferior for the detection of markers which were found to show cytoplasmic immunoreactivity, such as the macrophage marker MAC387 or the B cell markers BAQ155 or IL-A59. 相似文献

OBJECTIVE: To characterize variables used to monitor rabbits during inhalation anesthesia. ANIMALS: 8 male New Zealand White rabbits. PROCEDURE: Rabbits were similarly anesthetized with halothane (HAL) or isoflurane (ISO) in a crossover study; half received HAL followed by ISO, and the protocol was reversed for the remaining rabbits. After induction, minimum alveolar concentration (MAC) was determined for each agent, using the tail-clamp method, and variables were recorded at 0.8, 1.0, 1.5, and 2.0 MAC (order randomized). RESULTS: Mean +/- SEM MAC was 1.42 +/- 0.05 and 2.07 +/- 0.09% for HAL and ISO, respectively. Directly measured auricular mean arterial blood pressure was 52.8 +/- 5.6 and 54.8 +/- 6.1 mm Hg at 0.8 MAC for HAL and ISO, respectively, and decreased from these values in a parallel dose-dependent manner. Respiratory frequency remained constant (range, 69 to 78 breaths/min) over the range of HAL doses but incrementally decreased from a mean of 53 (at 0.8 MAC) to 32 breaths/min (at 2.0 MAC) for ISO. The PaCO2 was similar at 0.8 MAC for HAL and ISO and progressively increased with increasing doses of both agents; PaCO2 at 2.0 MAC for ISO was significantly greater than that at 2.0 MAC for HAL (79.8 +/- 13.7 vs 54.9 +/- 4.0 mm Hg, respectively). Eyelid aperture consistently increased in a dose-dependent manner for both anesthetics. CONCLUSIONS: Arterial blood pressure, PaCO2, and eyelid aperture consistently and predictably changed in rabbits in response to changes in anesthetic doses. This study investigates serum immunoglobulin (SIg) levels and lymphocyte subpopulations in normal dogs in response to putative immunosuppressive doses of prednisone and/or azathioprine. The objectives were to quantify SIg levels and lymphocyte subpopulations, including Thy-1+, CD4+, CD8+ and B cells, in normal dogs both before and after the administration of prednisone and/or azathioprine at 2 mg/kg, PO, each. Eighteen beagles were divided into 3 groups of 6 dogs each. Blood samples for radial immunodiffusion assay of IgG, IgM and IgA, complete blood count (CBC)and flow cytometry were collected prior to the administration of any drugs and again after 14 d of azathioprine, prednisone or azathioprine and prednisone. Peripheral blood mononuclear cells were isolated using density centrifugation and were incubated with monoclonal antibodies reacting with CD4+, CD8+, Thy-1+ and membrane immunoglobulin. Lymphocyte subsets were quantified using flow cytometry. Azathioprine-treated dogs had no significant changes in SIg levels or lymphocyte subpopulations. Prednisone-treated dogs had significant (P < 0.05) decreases in all SIg levels, all lymphocyte subpopulations and erythrocyte numbers, and had an increase in neutrophil counts. Prednisone and azathioprine-treated dogs had significant (P < 0.05) decreases in serum IgG levels and Thy-1+ and CD8+ lymphocyte subpopulations, with an increase in the CD4:CD8. These dogs also had a significant decrease in erythrocyte number and a significant increase in the monocyte count. These findings suggest that azathioprine and prednisone in combination or prednisone alone may be useful for the treatment of T cell-mediated diseases since decreased circulating T cell levels were demonstrated following treatment. The combination of drugs or azathioprine alone may not be appropriate for treatment of acute or autoantibody-mediated immune disease, because SIg levels were minimally affected by treatment. 相似文献

Myeloperoxidase (MPO) is a specific enzyme of neutrophil azurophilic granules with a strong oxidative activity. Thanks to a radioimmunoassay of equine myeloperoxidase, the authors have observed a significantly higher plasma level of MPO in horses operated for strangulation obstruction of the large intestine (n = 6) than in horses suffering from a non-strangulating displacement of the large intestine (n = 9). For the 2 groups, 3 phases were distinguished: reception (P1), intensive care (P2) and terminal phase (P3). The mean peak values of MPO for these phases were 121.6 ng/mL (P1), 168.6 ng/mL (P2), and 107.0 ng/mL (P3) for the non-strangulating group, and 242.6 ng/mL (P1); 426.0 ng/mL (P2), and 379.5 ng/mL (P3) for the strangulation group. The variations of the mean peak values of plasma MPO were significantly different between the 2 groups and between the different phases. A significant increase of the least square means of MPO was observed between P1 and P2. A significant decrease of the least square means of the number of circulating leukocytes was observed between P1 and P3. Polymorphonuclear neutrophil activation could play a major role in the pathogenesis of acute abdominal disease and endotoxic shock. 相似文献

Pregnancy rates, foal-heat conception rates, early and late embryo loss, polyovulations and delivery loss were compared on a Russian breeding farm between Don mares with a low percentage of Thoroughbred (TB) blood and those with more than 25% TB blood, called Budenny mares. The overall pregnancy rate in the two groups was not significantly different; 85.5% and 85.6%, respectively. There was a significant difference in the foal heat conception rate, with the Don mares at 54.9% and the Budenny mares at 50.3%. Embryo loss before Day 100 was only 4.9% for the Don mares, but was 9.1% for the Budenny mares. Embryo loss after Day 100 was similar in the two groups; 10.2% for the Don mares and 9.0% for the Budenny mares. The loss of foals in the process of delivery was 3.3% for the Don mares and 5.0% for the Budenny mares. Estrous cycles with polyovulations was 8.7% for the Don mares and 13.1% for the Budenny mares. 相似文献
