Maize is the major staple food in southern Africa with human consumption averaging 91 kg capita?1 year?1, and normal maize is nutritionally deficient in two essential amino acids: tryptophan and lysine. Despite the development of quality protein maize (QPM) with high tryptophan and lysine, stunting and kwashiorkor remain high in sub-Saharan Africa due to lack of high yielding and adapted QPM varieties. This study aimed at evaluating a new generation of QPM varieties for yield and related agronomic traits. Before the QPM varieties were validated on-farm, they were simultaneously selected on-station under five different management conditions. In the 2014/2015 season, 10 elite QPM varieties were selected from on-station trials based on high grain yield and stability, and were compared with the best commercial check varieties on-farm. During the 2015/2016 season, some poorly performing QPM varieties were dropped while new ones were added, resulting in 12 elite QPM varieties being evaluated on-farm. Analysis of variance for the 2014/2015 season showed non-significant hybrid × management condition interaction. Mean grain yields across management conditions ranged from 1.5 to 4 t ha?1 and were higher under mild stress (2.3–5.5 t ha?1) compared to random stress conditions (1.1–2.9 t ha?1). Broad sense heritability estimates were low to moderate (11–69%), and thus could still permit effective selection of better genotypes. Yield advantage ranged from 12 to 25% across the 2 years, suggesting effective genetic gains in QPM breeding. QPM hybrids CZH132044Q, CZH142238Q and CZH142236Q were stable and high yielding. Soil and root samples were collected from major tomato growing areas of Ethiopia during the 2012/2013 growing season to identify root-knot nematode problems. DNA-based and isozyme techniques revealed that Meloidogyne incognita and M. javanica were the predominant Meloidogyne species across the sampled areas. The aggressiveness of different populations of these species was assessed on tomato cultivars Marmande and Moneymaker. The two most aggressive populations of each species were selected and further tested on 33 tomato genotypes. The resistance screening and mechanism of resistance was performed after inoculation with 100 freshly hatched (<24 h) second-stage juveniles (J2). Eight weeks after inoculation the number of egg masses produced on each cultivar was assessed. For the resistance mechanism study, J2 penetration and their subsequent development inside the tomato roots were examined at 1, 2, 4 and 6 weeks after inoculation. On both cultivars Marmande and Moneymaker all M. incognita and M. javanica populations formed a high number of egg masses indicating highly aggressive behaviour. Populations from ‘Jittu’ and ‘Babile’ for M. incognita and ‘Jittu’ and ‘Koka’ for M. javanica were selected as most aggressive. None of the 33 tomato genotypes were immune for these M. incognita and M. javanica populations. However, several tomato genotypes were found to have a significant effect on the number of egg masses produced indicating possible resistance. For M. javanica populations there were more plants from cultivars or breeding lines on which no egg masses were found compared to M. incognita populations. The lowest number of egg masses for both populations of M. incognita was produced on cultivars Bridget40, Galilea, and Irma while for M. javanica it was on Assila, Eden, Galilea, Tisey, CLN-2366A, CLN-2366B and CLN-2366C. Tomato genotypes, time (weeks after inoculation) and their interaction were significant sources of variation for J2 penetration and their subsequent development inside the tomato roots. Differential penetration was found in breeding lines such as CLN-2366A, CLN-2366B and CLN-2366C, but many of the selected tomato genotypes resistance for the tested M. incognita and M. javanica populations were expressed by delayed nematode development. Lack of quantitative observations of extent, frequency, and severity of large historical fires constrains awareness of departure of contemporary conditions from those that demonstrated resistance and resilience to frequent fire and recurring drought.
Objectives
Compare historical and contemporary fire and forest conditions for a dry forest landscape with few barriers to fire spread.
Methods
Quantify differences in (1) historical (1700–1918) and contemporary (1985–2015) fire extent, fire rotation, and stand-replacing fire and (2) historical (1914–1924) and contemporary (2012) forest structure and composition. Data include 85,750-ha tree-ring reconstruction of fire frequency and extent; >?375,000-ha timber inventory following >?78,900-ha fires in 1918; and remotely-sensed maps of contemporary fire effects and forest conditions.
Results
Historically, fires?>?20,000 ha occurred every 9.5 years; fire rotation was 14.9 years; seven fires?>?40,469 ha occurred during extreme drought (PDSI <?? 4.0); and stand-replacing fire occurred primarily in lodgepole (Pinus contorta var. murrayana). In contemporary fires, only 5% of the ecoregion burned in 30 years, and stand-replacing fire occurred primarily in ponderosa (Pinus ponderosa) and mixed-conifer. Historically, density of conifers?>?15 cm dbh exceeded 120 trees/ha on?<?5% of the area compared to 95% currently.
Conclusions
Frequent, large, low-severity fires historically maintained open-canopy ponderosa and mixed-conifer forests in which large fire- and drought-tolerant trees were prevalent. Stand-replacing patches in ponderosa and mixed-conifer were rare, even in fires >?40,469 ha (minimum size of contemporary “megafires”) during extreme drought. In this frequent-fire landscape, mixed-severity fire historically influenced lodgepole and adjacent forests. Lack of large, frequent, low-severity fires degrades contemporary forest ecosystems.
Large mounds amended with plant residues are a common cultivation method for sweetpotato (Ipomoea batatas) in the western parts of the Papua New Guinea highlands, with a suggested benefit of elevated root zone temperature (RZT) from decomposition. This study aimed to evaluate whether RZT affects the formation and growth of sweetpotato storage roots. Experiments evaluated the effects of a constant elevated RZT on the Beauregard cultivar and of a short‐term increase in RZT on the Beauregard and Northern Star cultivars. Plants were grown in 18‐L pots divided into a control compartment (constant RZT at 20°C) and a treated compartment (RZT at 30, 40 or 50°C). Constant elevated RZT was maintained until harvest, while short‐term increase in RZT occurred in weeks 2 and 3 only. Constant elevated RZT did not affect the total number of storage roots, but RZT of 30°C and above reduced the dry‐biomass of storage roots. The short‐term increase in RZT did not alter the total number or dry‐biomass of storage roots in Beauregard, while both of these attributes were reduced at RZT of 40°C or above in Northern Star. Elevated RZT inhibited dry‐biomass accumulation of storage root, with a more pronounced impact at constant elevated RZT. These results demonstrate that any benefits of the mound culture on sweetpotato yield production do not arise through the influence of temperature elevation. 相似文献
Managers are faced with numerous methods for delineating wildlife movement corridors, and often must make decisions with limited data. Delineated corridors should be robust to different data and models.
Objectives
We present a multi-method approach for delineating and validating wildlife corridors using multiple data sources, which can be used conserve landscape connectivity. We used this approach to delineate and validate migration corridors for wildebeest (Connochaetes taurinus) in the Tarangire Ecosystem of northern Tanzania.
Methods
We used two types of locational data (distance sampling detections and GPS collar locations), and three modeling methods (negative binomial regression, logistic regression, and Maxent), to generate resource selection functions (RSFs) and define resistance surfaces. We compared two corridor detection algorithms (cost-distance and circuit theory), to delineate corridors. We validated corridors by comparing random and wildebeest locations that fell within corridors, and cross-validated by data type.
Results
Both data types produced similar RSFs. Wildebeest consistently selected migration habitat in flatter terrain farther from human settlements. Validation indicated three of the combinations of data type, modeling, and corridor detection algorithms (detection data with Maxent modeling, GPS collar data with logistic regression modeling, and GPS collar data with Maxent modeling, all using cost-distance) far outperformed the other seven. We merged the predictive corridors from these three data-method combinations to reveal habitat with highest probability of use.
Conclusions
The use of multiple methods ensures that planning is able to prioritize conservation of migration corridors based on all available information.
CRISPR-Cas technology has raised considerable interest among plant scientists, both in basic science and in plant breeding. Presently, the generation of random mutations at a predetermined site of the genome is well mastered, just like the targeted insertion of transgenes, although both remain restricted to species or genotypes amenable for plant transformation. On the other hand, true genome editing, i.e. the deliberate replacement of one or several nucleotides of the genome in a predetermined fashion, is limited to some rather particular examples that generally concern genes allowing positive selection, for example tolerance to herbicides. Therefore, further technological developments are necessary to fully exploit the potential of genome editing in enlarging the gene pool beyond the natural variability available in a given species. In principle, the technology can be applied to any quality related, agronomical or ecological trait, under the condition of upstream knowledge on the genes to be targeted and the precise modifications necessary to improve alleles. Published proof of concepts concern a wide range of agronomical traits, the most frequent being disease resistance, herbicide tolerance and the biochemical composition of harvested products. The regulatory status of the plants obtained by CRISPR-Cas technology raises numerous questions, in particular with regard to the plants that carry in their genomes the punctual modifications caused by the presence of the Cas9 nuclease but not the nuclease itself. Without clarification by the competent authorities, CRISPR-Cas technology would continue to be a powerful tool in functional genomics, but its potential in plant breeding would remain untapped. 相似文献
Using a 37‐year recruitment time series, we uncovered a field pattern revealing a strong, inverse relationship between bloater Coregonus hoyi recruitment success and slimy sculpin Cottus cognatus biomass in Lake Michigan (United States), one of the largest freshwater lakes of the world. Given that slimy sculpins (and deepwater sculpin Myoxocephalus thompsonii) are known egg predators that spatiotemporally overlap with incubating bloater eggs, we used recently published data on sculpin diets and daily ration to model annual bloater egg consumption by sculpins for the 1973–2010 year‐classes. Although several strong year‐classes were produced in the late 1980s when the proportion of eggs consumed by slimy sculpins was extremely low (i.e., <0.001) and several weak year‐classes were produced when the proportion of bloater eggs consumed was at its highest (i.e., >0.10–1.0), egg predation failed to explain why recruitment was weak for the 1995–2005 year‐classes when the proportion consumed was also low (i.e., <0.02). We concluded that egg predation by slimy and deepwater sculpins could have limited bloater recruitment in some years, but that some undetermined factor was more important in many other years. Given that slimy sculpin densities are influenced by piscivorous lake trout Salvelinus namaycush, the restoration of which in Lake Michigan has lagged behind those in lakes Superior and Huron, our study highlights the importance of an ecosystem perspective when considering population dynamics of fishes. 相似文献
