Staphylococcus aureus is one of the main etiological agents of mastitis in ruminants. In the present retrospective study, we evaluated the potential interest of a previously described automated multiple loci Variable Number of Tandem Repeats (VNTR) Assay (MLVA) comprising 16 loci as a first line tool to investigate the population structure of S. aureus from mastitis. We determined the genetic diversity of S. aureus strains from cases of clinical and subclinical mastitis in dairy cattle (n = 118, of which 16 were methicillin-resistant), sheep (n = 18) and goats (n = 16). The 152 strains could be subdivided into 115 MLVA genotypes (including 14 genotypes for the ovine strains and 15 genotypes for the caprine strains). This corresponds to a discriminatory index (D) value of 0.9936. Comparison with published MLVA data obtained using the same protocol applied to strains from diverse human and animal origins revealed a low number (8.5%) of human-related MLVA genotypes among the present collection. Eighteen percent of the S. aureus mastitis collection belonged to clonal complexes apparently not associated with other pathological conditions. Some of them displayed a relatively low level of diversity in agreement with a restricted ecological niche. These findings provide arguments suggesting that specific S. aureus lineages particularly adapted to ruminant mammary glands have emerged and that MLVA is a convenient tool to provide a broad overview of the population, owing to the availability via internet of databases compiling published MLVA genotypes.

Mutations in the human neurotrypsin gene are associated with autosomal recessive mental retardation. To further understand the pathophysiological consequences of the lack of this serine protease, we studied Tequila (Teq), the Drosophila neurotrypsin ortholog, using associative memory as a behavioral readout. We found that teq inactivation resulted in a long-term memory (LTM)-specific defect. After LTM conditioning of wild-type flies, teq expression transiently increased in the mushroom bodies. Moreover, specific inhibition of teq expression in adult mushroom bodies resulted in a reversible LTM defect. Hence, the Teq pathway is essential for information processing in Drosophila. 相似文献
The water dynamics and retrogradation kinetics behavior of gelatinized wheat starch by either ultrahigh pressure (UHP) processing or heat are investigated. Wheat starch completely gelatinized in the condition of 90, 000 psi at 25 degrees C for 30 min (pressurized gel) or 100 degrees C for 30 min (heated gel). The physical properties of the wheat starches were characterized in terms of proton relaxation times (T2 times) measured using time-domain nuclear magnetic resonance spectroscopy and evaluated using commercially available continuous distribution modeling software. Different T2 distributions in both micro- and millisecond ranges between pressurized and heated wheat starch gels suggest distinctively different water dynamics between pressurized and heated wheat starch gels. Smaller water self-diffusion coefficients were observed for pressurized wheat starch gels and are indicative of more restricted translational proton mobility than is observed with heated wheat starch gels. The physical characteristics associated with changes taking place during retrogradation were evaluated using melting curves obtained with differential scanning calorimetry. Less retrogradation was observed in pressurized wheat starch, and it may be related to a smaller quantity of freezable water in pressurized wheat starch. Starches comprise a major constituent of many foods proposed for commercial potential using UHP, and the present results furnish insight into the effect of UHP on starch gelatinization and the mechanism of retrogradation during storage. 相似文献
The effect of two different preslaughter procedures (limited or 15-min intense muscular activity) on muscle trout proteins was investigated. Muscle was sampled 45 min and 24 h post-mortem, proteins were separated using two-dimensional electrophoresis, and spots of interest were tentatively identified by MALDI-TOF spectrometry. Twenty-nine and 4 spots were differentially represented between the two groups of fish at 45 min and 24 h post-mortem, respectively. Spots that could be identified corresponded mainly to proteins involved in energy-producing pathways (triosephosphate isomerase, enolase, pyruvate dehydrogenase) or to structural proteins (desmin, cap-Z, myosin heavy chain fragment). Persistent under-representation of desmin, a key cytoskeletal protein, in fish submitted to intense muscular activity suggests that such a preslaughter treatment can have an effect on post-mortem muscle integrity. 相似文献
Thermal denaturation and aggregation abilities of salmon myofibrils and myosin were studied measuring turbidity, intrinsic fluorescence, 8-anilino-1-naphthalene sulfonic acid binding, and 1-ethyl-3-(3-dimethylaminopropyl) carbodiimide cross-linking. The thermal behaviors of protein preparation from white and red muscles were compared, and the relationship with thermal gelation properties is discussed. The low gelation ability of salmon muscle proteins was related to a limited extent of protein denaturation and aggregation upon heating. These properties seemed to be carried by myosin molecules as a similar behavior was observed for both myofibrils and myosin preparations. The higher thermal stability observed for red muscle proteins with higher transition temperatures in rheological profiles was related to a shift to higher temperature in denaturation and aggregation processes. The extent of denaturation and aggregation was very similar for both muscle types as was the final rigidity of the gels formed. 相似文献
Dispersal capacity is a life-history trait that may have profound consequences for earthworm populations: it influences population dynamics, species persistence and distribution and community structure. It also determines the level of gene flow between populations and affects processes such as local adaptation, speciation and the evolution of life-history traits. It may play a great role in soil functioning by determining the spatial distribution of ecosystem engineers such as earthworms. Dispersal is an evolutionary outcome of the behaviour in response to the ecological constraints of the species. Hence different dispersal behaviour is expected from the different ecological types of earthworms. Nevertheless the dispersal behaviour of earthworms has been little documented.In this work we test a series of basic mechanisms that are fundamental and complementary to understand earthworms dispersal behaviour. We focus on the dispersal triggered by environmental conditions, a fundamental process usually termed “conditional dependent dispersal”. We show experimentally in mesocosms that in one week: 1) earthworm dispersal can be triggered by low habitat quality, either through soil quality or the presence/absence of litter. 2) Earthworms can be subject to positive density dependent dispersal, that is the rate of dispersal increases when density increases; and 3) earthworm dispersal can be reduced by the pre-use of the soil by conspecific individuals that are no longer present.Our results suggest that earthworms may be more mobile than expected from previous estimations, and that they present high capacities of habitat selection. In the light of our findings we elaborate a behavioural scenario of earthworm foraging, and propose several priority working directions. 相似文献
Xenobiotic chemicals that are present in aquatic environments are typically concentrated on suspended particles and sedimentary materials where they represent a source of chronic contamination to benthic and pelagic organisms. Laboratory and field studies of bioaccumulation, toxicity, and food-chain transfer of chemicals bound to sediment have shown the potential ecological impacts that may result from the contamination of sediments. In this review, the chemical and physical processes that determine the environmental fate of sediment-sorbed organic chemicals are outlined, and their relationship to bioavailability is discussed. Methods currently used to predict the fate of pollutants in the aquatic environment are of limited use for the assessment of sediment-sorbed chemical bioavailability because they are based on the compound's behavior in the water column. Furthermore, an organism's morphology and ecological niche can profoundly affect its ability to accumulate xenobiotic chemicals from sediments and these must be considered before the populations at greatest risk can be identified. 相似文献
Species distribution modelling is a common tool in conservation biology but two main criticisms remain: (1) the use of simplistic variables that do not account for species movements and/or connectivity and (2) poor consideration of multi-scale processes driving species distributions.
Objectives
We aimed to determine if including multi-scale and fine-scale movement processes in SDM predictors would improve accuracy of SDM for low-mobility amphibian species compared with species-level analysis.
Methods
We tested and compared different SDMs for nine amphibian species with four different sets of predictors: (1) simple distance-based predictors; (2) single-scale compositional predictors; (3) multi-scale compositional predictors with a priori selection of scale based on knowledge of species mobility and scale-of-effect; and (4) multi-scale compositional predictors calculated using a friction-based functional grain to account for resource accessibility with landscape resistance to movement.
Results
Using friction-based functional grain predictors produced slight to moderate improvements of SDM performance at large scale. The multi-scale approach, with a priori scale selection, led to ambiguous results depending on the species studied, in particular for generalist species.
Conclusion
We underline the potential of using a friction-based functional grain to improve SDM predictions for species-level analysis.
Changes in Collembola richness and diversity along a land-use intensity gradient were studied in eight European countries (Portugal, Spain, France, Switzerland, Hungary, UK, Ireland and Finland). In each country a set of six 1 km2 land-use units (LUUs) were selected forming a gradient ranging from natural forest to agricultural dominated landscapes, passing through mixed-use ones. In addition to data on Collembola, detailed information regarding landscape diversity and structure was collected for each LUU. A total of 47,774 individuals were identified from 281 species. Collembola reacted not only to changes in the diversity of the landscape, but also to the composition of that diversity and the area occupied by each land-use type at each LUU. Although species richness patterns were not concordant among the different countries, the total number of species per LUU (landscape richness) was generally higher in natural forests and mixed-used landscapes, and lower in agricultural dominated landscapes. Moreover, high richness and diversity of Collembola at each LUU were associated with a diverse landscape structure, both in terms of number of patches and patch richness. Despite this comparable species richness between mixed-use landscapes and those dominated by natural forests, average species richness on forested areas (local richness) decreased along the gradient, showing that forest patches on mixed-use landscapes support a lower richness than in landscapes dominated by forest. This aspect is important when addressing the role of native forests in structuring biodiversity in disturbed and fragmented landscapes. Although a diverse landscape can support a high biodiversity, the results suggest that intensive fragmentation should be avoided with the risk of collapsing local species richness with the consequent result for regional biodiversity. 相似文献
