An enzyme-linked immunosorbent assay (ELISA) was developed and compared with 2 reference diagnostic tests (indirect immunofluorescence [IF] and complement fixation) to detect myxoma virus-specific antibodies in sera from 50 rabbits experimentally vaccinated with an attenuated strain of myxoma virus or with a Shope fibroma virus. The ELISA was highly specific (100% specificity) and sensitive (100%, 21 days after homologous vaccination). In a comparison of the ELISA with the IF test in 128 wild rabbits from France, discrepant results were obtained in only 11 (8.6%) animals, which were positive with the ELISA and negative with the IF test. The higher sensitivity and the good specificity of the ELISA was confirmed in a serologic survey of 118 rabbits from 2 Kerguelen (Indian Ocean) islands, where the prevalence of myxomatosis varied considerably. The ELISA is an alternative serologic test for diagnosis, vaccine evaluation, and seroepidemiologic surveys of myxomatosis. 相似文献

The aim of this paper is to provide explanations of how dietary phospholipid (PL) globally improves fish larval development, including growth and survival, digestive functions and skeletal development, and to propose optimal PL levels and sources in fish larval diets. Dietary incorporation of 8–12% PL related to dry matter (d.m.) promotes growth and enhanced survival in various species. Marine source PL, incorporating highly unsaturated fatty acids, was most efficient than soybean lecithin. This beneficial effect was explained by an enhancement in digestive functions, assessed by digestive enzyme activities and histomorphology. Nevertheless, 1.5–2.5% highly unsaturated fatty acids related to diet d.m. supplied by PL improved growth, survival and skeletal development, while 5% induced different skeletal deformities. The high incidence of deformities was associated with the down-regulation of genes involved in development, such as RXRα, RARα, RARβ and BMP-4, observed in the early stages in larvae fed a high highly unsaturated fatty acids level. 相似文献

ABSTRACT We recovered a spontaneous albino strain from ascospores of Ceratocystis resinifera, a sapstain fungus that grows deeply and rapidly in freshly felled conifer trees. This albino strain, named Kasper, was tested for its ability to prevent discoloration of spruce sapwood caused by wild-type sapstain fungi and compared with Cartapip 97, a commercially available biological control agent of sapstain in lodgepole pine and red pine logs. In a laboratory trial, Kasper reduced sapstain of white spruce logs as much as 94.4% and was more efficient than Cartapip 97. In field trials conducted in an area north of Québec City, Kasper reduced sapstain of black spruce as much as 80%. In three of four field trials, Kasper was significantly more efficient than Cartapip 97 in reducing sapstain development. The exception was encountered in a 2003 trial conducted in a sawmill yard where Kasper did not reduce sapstain. In a field trial conducted in western Canada, at Aleza Lake forest near Prince George, Kasper almost totally prevented the development of sapstain, even after 24 weeks. These results suggest albino strains derived from C. resinifera might be an additional source of potential biocontrol agents against sapstain. 相似文献

Biotransformation of the insecticide lindane by the white rot basidiomycete Phanerochaete chrysosporium has been investigated in liquid cultures. Some polar metabolites and carbon dioxide were produced from the pesticide. Among the metabolites identified were tetrachlorocyclohexene, tetrachlorocyclohexene epoxide and tetrachlorocyclohexenol. When used as a substrate, tetrachlorocyclohexene was also converted by the fungus to tetrachlorocyclohexenol, polar metabolites and carbon dioxide. Three incubation conditions leading to low and high peroxidase production were assayed. Data from these experiments, as well as in-vitro incubations with purified enzymes, ruled out any involvement of the peroxidases in lindane biotransformation and mineralization. Moreover, 1-aminobenzotriazole (a P450 inactivator) drastically reduced pesticide metabolism. Conversely, phenobarbital (a P450 inducer) did not significantly increase lindane breakdown. 相似文献

This work describes the ontogeny of the digestive tract in thick lipped grey mullet (Chelon labrosus) larvae reared until day 36 post-hatching with the semi-extensive technology in mesocosms. Diet was constituted by live preys, rotifers, Artemia and wild zooplankton, then compound diet was added from day 20 (p. h.). Linear growth, weight growth and digestive enzymes specific activities were studied during larval ontogeny. Pancreatic enzymes (trypsin and amylase) and intestinal enzymes (leucine-alanine peptidase “Leu-ala”, aminopeptidase N “AN” and alkaline phosphatase “AP”) were assayed in larvae sampled throughout the rearing trial to evaluate gastrointestinal maturation along the development.The trypsin specific activities were very high during the first two weeks and then declined as observed in marine fish species. A following increase in trypsin specific activity from day 20 was attributed notably to ingestion of particle compound diet. In contrast to the pattern generally described in fish larvae, amylase specific activity showed a continuous increase. This could be attributed to the fact that C. labrosus is an omnivorous species and suggests that the fish might be able to use efficiently diets containing higher levels of starch or other carbohydrates since the end of larval development.Relative expression of intestinal brush border membrane enzymes (AP and AN) and cytosolic enzyme (Leu-ala), showed an abrupt increase of both AP/leu-ala and AN/leu-ala ratios at day 8 (p. h.), indicating that maturation of intestinal tract in C. labrosus larvae is particularly precocious. In situ changes in pectin fractions for thermally processed carrots were related to textural changes. The texture of pretreated and subsequently thermally processed carrot disks was determined. Alcohol insoluble residue (AIR) was extracted from the pretreated and thermally processed tissues. The AIR was characterized in terms of the degree of methylation (DM) and changes in pectin fractions. Distinct differences in texture and DM were observed during thermal processing. Pretreatment conditions that induced a significant decrease in DM showed better textures. Demethoxylation caused interconversion of pectin fractions, water soluble pectin (WSP) changing into water insoluble pectin [chelator (CSP) and alkali (NSP) soluble pectin]. This process was reversed during cooking accompanied by remarkable alterations in molecular weight (MW) distribution patterns. The WSP depicted polydisperse MW distribution patterns, strongly dependent on the pretreatment condition. Confirmatory results of interconversions of pectin fractions (WSP, NSP) were demonstrated by the MW distribution patterns and neutral sugar profiles. The effect of feeding foliage from Afzelia africana, Pterocarpus erinaceus or Khaya senegalensis on growth performance was evaluated using 32 West African Djallonké rams of about 8 months of age and with a mean initial
body weight (BW) of 16.1 kg. The animals were randomly assigned to four groups of 8 animals and the experiment lasted for
13 weeks. All the animals received the same amount of hay from Andropogon gayanus and maize bran (200 g/day each) and dried foliage ad libitum. The control group was fed cottonseed cake in restricted amounts. The animals were able to consume higher amounts of A. africana than of P. erinaceus and K. senegalensis. There was no significant difference in growth rate between sheep offered A. africana or P. erinaceus, 62.9 and 58.8 g/d, respectively, but sheep offered K. senegalensis had a lower average daily gain, 48 g/day (P < 0.05) due to lower consumption of both energy and crude protein. Animals in
the control group had the highest growth rate, 95.8 g/day. Sheep offered P. erinaceus and K. senegalensis had similar carcass characteristics and dressing percentage but lower fasted BW, empty BW, carcass weight and dressing percentage

Three groups of newly hatched spotted wolffish (Anarhichas minor) were held at three different temperatures in order to determine relationships between metabolic, digestive and growth response in rapidly developing larvae. Growth rates were successfully modulated by temperature (5, 8 and 12 °C). Activity levels of trypsin and lactate dehydrogenase (LDH) were positively linked to specific growth rates at all temperatures. Trypsin showed a positive compensation (higher activity at lower temperature) whereas glycolytic enzymes (pyruvate kinase and Lactate dehydrogenase) and aspartate aminotransferase (AST) showed a negative compensation (lower activity at lower temperature). Citrate synthase was not affected by growth rate, indicating that the level of aerobic capacity was adequate in sustaining the high energetic needs associated with rapid growth early in the life of the spotted wolffish. In light of our results, it is suggested that protein digestion, as demonstrated by the activity profile of trypsin in relation to growth rate and temperature, is likely a key growth‐limiting agent during the early‐life stages of wolffishes. Our results are discussed in comparison with A. lupus, a closely related species displaying different temperature preferences and growth capacities. 相似文献
