Poliovirus virion RNA contains a single covalently bound sequence of polyadenylic acid which is approximately 49 nucleotides long. A single, slightly longer polyadenylic acid sequence is contained in Eastern Equine Encephalitis virus RNA. Since these viruses are otherwise dissimilar these sequences may play a common role in viral replication, possibly in translation of the viral RNA.  相似文献

Hepcidin is a peptide hormone secreted by the liver in response to iron loading and inflammation. Decreased hepcidin leads to tissue iron overload, whereas hepcidin overproduction leads to hypoferremia and the anemia of inflammation. Ferroportin is an iron exporter present on the surface of absorptive enterocytes, macrophages, hepatocytes, and placental cells. Here we report that hepcidin bound to ferroportin in tissue culture cells. After binding, ferroportin was internalized and degraded, leading to decreased export of cellular iron. The posttranslational regulation of ferroportin by hepcidin may thus complete a homeostatic loop: Iron regulates the secretion of hepcidin, which in turn controls the concentration of ferroportin on the cell surface.  相似文献

Hemoproteins are critical for the function and integrity of aerobic cells. However, free heme is toxic. Therefore, cells must balance heme synthesis with its use. We previously demonstrated that the feline leukemia virus, subgroup C, receptor (FLVCR) exports cytoplasmic heme. Here, we show that FLVCR-null mice lack definitive erythropoiesis, have craniofacial and limb deformities resembling those of patients with Diamond-Blackfan anemia, and die in midgestation. Mice with FLVCR that is deleted neonatally develop a severe macrocytic anemia with proerythroblast maturation arrest, which suggests that erythroid precursors export excess heme to ensure survival. We further demonstrate that FLVCR mediates heme export from macrophages that ingest senescent red cells and regulates hepatic iron. Thus, the trafficking of heme, and not just elemental iron, facilitates erythropoiesis and systemic iron balance.  相似文献

When African violet leaf explants are cultured in vitro, buds and shoots develop directly from the upper leaf surfaces. Three developmentally different African violet chimeras were cultured, and in each case adventitious shoots that developed into plants had the parent chimera pattern. Wound-healing tuber tissues from two potato cultivars, one resistant (cv. Reddale) and one susceptible (cv. Kennebec) to Verticillium wilt caused by the fungusVerticillium dahliae were treated with elicitor preparations of either an autoclaved mycelial extract of the fungus or with the fungal component arachidonic acid (AA). Wound-healing tissues were analyzed for hypersensitive cell browning (HCB) and alterations in lipoxygenase (LOX), polyphenoloxidase (PPO), and peroxidase (PER) activities, because all three enzymes are thought to be involved in the hypersensitive response and plant defense. Healing Reddale tissues that were treated with the elicitors exhibited a much more rapid and intense HCB than did the healing Kennebec tissues. By 96 h, Reddale tissues treated with either elicitor preparation appeared dark brown and necrotic. Treated Kennebec tissues appeared only slightly darker than the controls after 96 h. LOX, PPO and PER activities in treated Reddale tissues were significantly less than that found in the control after 48 h, and by 96 h PPO and PER activities were nearly zero. In contrast, LOX activity in AA-treated Kennebec disks was higher than in the control at 96 h, while the activities of PPO and PER had decreased to about 60% of the control at the same time period. Our results show that HCB is strongly correlated withV. dahliae and AA elicitor treatments in the resistant vs. the susceptible cultivar, while the overall role of the three enzymes in cell browning and death remain unclear.  相似文献

Cats are among the most popular pets in the U.S., and the majority of these animals are kept indoors where litter boxes containing some type of absorbent litter material are needed. Dried distillers grains (DDGs) are a major co-product of the ethanol industry, and are primarily sold as animal feed. We have been studying value-added uses for DDGs by extracting valuable phytochemicals from them with a variety of organic solvents. The objective of this research was to determine if the extracted DDGs could be formulated as cat litter. Extracted DDGs absorbed significantly more water (termed hydration capacity) than unextracted DDGs, although sorting the extracted DDGs by particle size had no effect on hydration capacity. Through the addition of glycerol as a dust retardant and guar gum as a clumping agent, a formulation was obtained with desirable physical properties. The addition of copper sulfate to this formulation significantly reduced the release of a volatile odor compound that is chemically similar to the odor compound produced by the decomposition of cat urine. From these results it appears that extracted DDGs have potential as commercial cat litter.  相似文献
