对我国蔬菜温室的评价及新型日光温室的研发

针对目前我国温室发展中的问题，研发了内保温组装式日光温室，介绍了内保温组装式日光温室的开发进展及应用情况。     

Effects of pirenzepine on expression of MMP-2 and TIMP-2 in stroma of sclera in chick myopic eyes

AIM:To observe the effect of intravitreal injection of pirenzepine on form-deprivation myopia in chicks. METHODS:Forty-one day old chicks were randomly divided into 4 groups:normal control group, form deprivation group, vehicle control group and pirenzepine injection group. The right eyes of all chicks were used as experimental eyes. The deprived eyes in vehicle control group and pirenzepine injection group received daily intravitreal injection of vehicle control solution(0.01 mol/L PBS) and pirenzepine(1%in PBS), respectively. Optical examinations such as refraction, axial length and equatorial diameter were made at the end of the 5th day. The mRNA and protein levels of matrix metalloproteinase 2(MMP-2) and tissue inhibitor of metalloproteinase-2(TIMP-2), and the activity of MMP-2 were detected by RT-PCR, Western blotting and zymography analysis,respectively. RESULTS:Refraction, axial length and equatorial diameter of the eyes in pirenzepine injection group were significantly lower than those in form deprivation group and vehicle control group(P<0.05), but those were higher(P<0.05) and the eyes were relatively myopic as compared with normal control group. The mRNA expression, protein le vels and activity of MMP-2 in pirenzepine group were significantly higher than those in normal control group(P<0.01), and were significantly lower than those in form deprivation group and vehicle control group(P<0.01, P<0.01). The mRNA and protein levels of TIMP-2 in pirenzepine group were significantly lower than those in normal control group(P<0.01), and was significantly higher than those in form deprivation group and vehicle control group(P<0.01, P<0.01). CONCLUSION:Intravitreal injection of pirenzepine may partly prevent form-deprivation myopia by modulating the expression of MMP-2 and TIMP-2 in the fibrous layer of sclera.     

Sampling and Investigating Zearalenone Contamination of Feeds and Feedstuffs of Pig Farms in Beijing Area

本研究采用免疫亲和柱高效液相色谱法测定了北京地区猪场饲料原料及全价饲料中玉米赤霉烯酮(ZEN)的含量,以了解北京地区饲料中ZEN的污染情况.试验抽样采集北京市昌平区、大兴区、延庆区、平谷区、顺义区5个区县15个猪场131份饲料样[55份玉米、豆粕、麸皮、干酒糟及其可溶物( DDGS)原料,76份猪全价饲料]进行ZEN含量的测定.结果表明:玉米、豆粕、麸皮、DDGS和全价饲料中ZEN的检出率分别为100.00％、54.45％、100.00％、100.00％和58.88％,超标率分别为0.00％、0.00％、0.00％、41.18％和0.00％,平均含量分别为109.08、9.19、14.92、882.68和58.88 μg/kg.结果提示,不同饲料原料中ZEN含量存在差异,DDGS中ZEN平均含量超标,猪全价饲料及玉米、豆粕等饲料原料中ZEN平均含量均未超标.     

OsGA20ox2不同长度RNAi片段对水稻株高等农艺性状的遗传效应

利用OsGA20ox2基因序列构建不同长度的RNAi片段并导入水稻，获得不同高度的矮化植株。将这些矮化植株与野生型植株回交获得B₁F₂群体，卡方检测表明B₁F₂群体矮秆植株数和高秆植株数符合3∶1比例，表现为矮秆显性的遗传规律。对矮化植株的F₅和B₁F₂群体株高、各茎节间长度和一些主要农艺性状方差分析显示, OsGA20ox2基因的RNAi能显著缩短株高和各节间长度(P<0.05)，RNAi干扰片段越长，使植株株高和节间长度缩短程度越大，可使株高降低24~42 cm，矮化22%~39%。在同一长度的RNAi干扰片段下，倒一节节间长度平均缩短与倒二节节间长度平均缩短非常相近，倒三节和倒四节节间长度平均缩短非常相近，总的缩短程度是倒四节>倒三节>倒二节>倒一节。这种近基部节间长度缩短幅度和比例较大的特点，利于提高水稻的抗倒伏能力，同时上部节间缩短幅度和比例较小，有效地保持合理株高，不使生物产量明显降低，有利于水稻的稳产和高产。OsGA20ox2基因的RNAi不影响如千粒重、结实率、穗长等其他主要农艺性状或影响很小。     

25%霜霉威·甲霜灵可湿性粉剂防治黄瓜霜霉病药效试验

研究不同剂量25%霜霉威.甲霜灵可湿性粉剂对黄瓜霜霉病防治效果。结果表明:469.5～703.5g/hm2可湿性粉剂对温室黄瓜霜霉病的防治效果达80.80%～82.90%,该药剂对黄瓜安全,可以在农业生产中与其它药剂交替使用。     

牛GHR0基因编码区生物信息学分析

GHR基因作为产奶性状的候选基因之一,已引起人们越来越多的关注.本文应用生物信息学方法比较分析了人、牛、猪、绵羊等14个物种的GHR基因编码区(CDS)序列,并对其遗传多样性、氨基酸序列的理化性质、信号肽、蛋白质二级结构和结构功能域等进行了预测分析.结果表明,从14个物种的30条序列检测到1218个多态位点,共生成22种单倍型,物种间GHR基因序列编码区存在较丰富的遗传多样性.GHR蛋白等电点均低于7,呈酸性；大多GHR蛋白N端存在信号肽,均有跨膜结构域；蛋白质二级结构的主要结构元件是无规则卷曲.     

小花忍冬硬枝扦插影响因素分析

小花忍冬是观赏性和抗逆性兼具的优良树种之一。为了解决其种子繁殖子代分化变异大的问题,结合小花忍冬特性,通过不同基质、激素及不同浓度的处理,对其进行了硬枝扦插试验。结果表明：基质种类、激素种类、激素浓度都直接影响其扦插成活率,激素种类直接影响地下根系的生长量。其中,使用珍珠岩︰沙子=1︰2、NAA、100 mg/L组合,...     

花生组培苗嫁接技术的研究

为解决花生组培再生苗及转基因苗驯化移栽成活率低的难题,本试验以花生实生苗作为砧木,对花生嫁接技术进行了研究。试验结果表明:超净台内无菌嫁接效果较好,以再生苗或实生苗为接穗进行无菌嫁接,其成活率均达90%以上,明显高于室内嫁接成活率(71%～72%);无菌嫁接时12～15d苗龄的砧木效果最好;将无菌嫁接苗在培养基中培养3～5d后进行驯化移栽为最佳时期;不同花生基因型嫁接苗成活率在87%～95%;嫁接苗移栽田间后,其成活率高达94%,且100%结果。     

高职院校生物技术专业顶岗实习的实践与思考

顶岗实习是高职院校生物技术专业人才培养中一个重要的实践性环节。笔者对郑州牧业工程高等专科学校生物技术及应用专业进行顶岗实习的实践过程、所取得的成效及存在的问题等进行深入思考,提出了一些看法和认识。     

PPARγ agonist pioglitazone attenuates cortical neuron lesion and gliosis in rat brain of post-traumatic injury

AIM: To investigate the neuroprotective effect of pioglitazone (Pio), a potent agonist of peroxisome proliferator-activated receptor gamma (PPARγ), on the traumatic brain injury (TBI) in rats. METHODS: SD rats were randomly divided into 4 groups: sham group, vehicle+TBI group, Pio+TBI group and Pio+T0070907+TBI group. TBI was induced by the method of controlled cortical impact (CCI) injury. Neutral red staining technique was used to determine the cortical lesion volume. NeuN, GFAP and OX-42 were measured by immunohistochemical technique to evaluate the morphology of neurons, activation and infiltration of astrocytes and microglia at the edge of cortical lesion. RESULTS: CCI injury in rat elicited activation and proliferation of the astrocytes and microglia. The glial scar wall formation at the edge of cortical lesion, which was accompanied by the loss of neurons, was observed. Pio significantly reduced the cortical lesion volume, the activation and infiltration of the astrocytes and microglia, and the loss of pyramidal neurons at the edge of cortical lesion. T0070907, an antagonist of PPARγ, reversed the effects of Pio. CONCLUSION: Pioglitazone exerts a neuroprotective efficacy, attenuates the loss of neurons and cortical lesion volume following CCI injury by inhibiting the activation and infiltration of astrocytes and microglia, especially glial scar formation.
