Soil microbial biomass N is commonly determined through fumigation-extraction (FE), and a conversion factor (KEN) is necessary to convert extractable N to actual soil biomass N. Estimation of KEN has been constrained by various uncertainties including potential microbial immobilisation. We developed a mass-balance approach to quantify changes in microbial N storage during nutrient-amended incubation, in which microbial uptake is determined as the residual in a ‘mass-balance’ based on soil-water N before and after amended incubation. The approach was applied to three sandy soils of southwestern Australia, to determine microbial N immobilisation during 5-day incubation in response to supply of 2.323 mg C g−1, 100 μg N g−1 and 20 μg P g−1. The net N immobilisation was estimated to be 95-114 μg N g−1 in the three soils, equivalent to 82.7-85.1% of soil-water N following the amendment. Such estimation for microbial uptake does not depend on fumigation and KEN conversion, but for comparison purposes we estimated ‘nominal’ KEN values (0.11-0.14) for the three soils, which were comparable to previously reported KEN from soils receiving C and N amendment. The accuracy of our approach depends on the mass-balance equation and the integrated measurement errors of the multiple N pools, and was assessed practically through recoveries of added-N when microbial uptake can be minimised. Near-satisfactory recoveries were achieved under such conditions. Our mass-balance approach provides information not only about changes in the microbial biomass nitrogen storage, but also major N-pools and their fluxes in regulating soil N concentrations under substrate and nutrient amended conditions.  相似文献

Pathology of experimentally-induced, acute toxoplasmosis in macropods   总被引:1,自引:0,他引:1  
SUMMARY Thirteen Tammar wallabies (Macropus eugenii) were dosed orally with 500, 1000 or 10 000 oocysts of Toxoplasma gondii, as part of a vaccination trial. Eleven animals died of acute toxoplasmosis 9 to 15 days after challenge. The lesions were similar in all animals, consisting of foci of necrosis and inflammation in the intestines, lymphoid tissue, adrenal cortex, heart, skeletal muscle and brain, and severe generalised pulmonary congestion and oedema. Free and intracellular tachyzoites of Toxoplasma were associated with the lesions. The remaining 2 animals had shown no signs of disease when euthanased four months after challenge. Small, focal, non-suppurative inflammatory lesions were seen in brain, heart and skeletal muscle of these animals and chronic Toxoplasma infection was confirmed by mouse inoculation.  相似文献

Genetic traits within Ornithopus sativus Brot. make it suitable for breeding as a forage legume in agriculture. The breeding system of O. sativus must be understood to stabilize valuable traits once developed. Hybridization between white‐flowered and pink‐flowered genotypes of O. sativus revealed the white flower colour to be inherited recessively to pink, while early flowering showed genetic dominance over late flowering (d = 0.5). The pink flower colour was always associated with red pigmentation on seedling hypocotyls, which provided a pragmatic aid in the selection process. Pollination of O. sativus is largely autogamous, but examination of hypocotyl colour of seedlings from white‐flowered trap plants placed among pink‐flowered plants in a sward indicated a xenogamic cross‐pollination rate of 25% is possible. This significant level of cross‐pollination revealed opportunities for breeding desirable traits within the species, while providing a warning that shifts in population structure could occur in response to selection pressure.  相似文献

The terminal navigation of the Near Earth Asteroid Rendezvous (NEAR) spacecraft during its close flyby of asteroid 253 Mathilde involved coordinated efforts to determine the heliocentric orbits of the spacecraft and Mathilde and then to determine the relative trajectory of the spacecraft with respect to Mathilde. The gravitational perturbation of Mathilde on the passing spacecraft was apparent in the spacecraft tracking data. As a result of the accurate targeting achieved, these data could be used to determine Mathilde's mass as 1.033 (+/- 0.044) x 10(20) grams. Coupled with a volume estimate provided by the NEAR imaging team, this mass suggests a low bulk density for Mathilde of 1.3 grams per cubic centimeter.  相似文献

Phytophthora cinnamomi is a necrotrophic pathogen of woody perennials and devastates many biomes worldwide. A controlled perlite–hydroponic system with no other hyphae‐producing organisms as contaminants present allowed rapid assessment of ten annual and herbaceous perennial plant species most of which have a wide distribution within the jarrah (Eucalyptus marginata) forest in Western Australia where this pathogen has been introduced. As some annuals and herbaceous perennials have recently been reported as symptomatic and asymptomatic hosts, laboratory screening of some of the field‐tested annuals and herbaceous perennials and additional species was used to further evaluate their role in the pathogen's disease cycle. Nine of the species challenged with the pathogen were asymptomatic, with none developing root lesions; however, Trachymene pilosa died. The pathogen produced thick‐walled chlamydospores and stromata in the asymptomatic roots. Furthermore, haustoria were observed in the roots, indicating that the pathogen was growing as a biotroph in these hosts.  相似文献

Resistant annual and herbaceous perennial plant species were identified as key hosts which allow Phytophthora cinnamomi to persist on severely impacted black gravel sites within the Eucalyptus marginata (jarrah) forest of southwest Western Australia. Of the annual and herbaceous perennial plant species present on black gravel sites, 15 out of 19 species were found to be hosts of P. cinnamomi, and 10 of these were symptomless hosts. In particular, the native annual Trachymene pilosa and the two native herbaceous perennials Stylidium diuroides and Chamaescilla corymbosa were commonly found to be hosts of the pathogen. Species from 12 new genera including three from new families (Crassulaceae, Droseraceae and Primulaceae) are reported for the first time to be hosts of P. cinnamomi. The species from which P. cinnamomi was recovered were the native species: Chamaescilla corymbosa, Crassula closiana, Drosera erythrorhiza, Hydrocotyle callicarpa, Levenhookia pusilla, Paracaleana nigrita, Podotheca angustifolia, Pterochaeta paniculata, Rytidosperma caespitosum, Siloxerus multiflorus, Stylidium diuroides and Trachymene pilosa, and the introduced annual weeds Hypochaeris glabra, Lysimachia arvensis and Pentameris airoides.  相似文献

Worldwide, forests and woodlands have shown progressive declines in health as a result of global environmental changes in combination with local anthropogenic drivers. This study examined the incidence and progression of a canker disease of marri (Corymbia calophylla) caused by the endemic fungal pathogen Quambalaria coyrecup at three paired forest and anthropogenically disturbed sites in the southwest of Western Australia over 13 years. At the time of plot establishment in 2001, cankers were present on trees at all six sites with 22.7% of the assessed trees cankered. By 2014, cankers had led to the death of 6.7% of the trees, and an additional 10.0% of the trees developed cankers during this time. A further 2.3% of trees died due to causes other than canker, resulting in a final figure of 65.0% of trees remaining alive and free of cankers for the duration of the survey period. Canker incidence was significantly greater on trees present at anthropogenically disturbed sites (along roadsides and in paddocks) than forest trees (35.3% increasing to 50.7%, 10.0% increasing to 14.7%, respectively). Trunk diameter at breast height, tree height and crown ratings were not correlated with canker presence. This long‐term study provides evidence of the increasing severity of this canker pathogen and the impact it is having on the survival of marri.  相似文献

The concentrations of the hydrogen radicals OH and HO2 in the middle and upper troposphere were measured simultaneously with those of NO, O3, CO, H2O, CH4, non-methane hydrocarbons, and with the ultraviolet and visible radiation field. The data allow a direct examination of the processes that produce O3 in this region of the atmosphere. Comparison of the measured concentrations of OH and HO2 with calculations based on their production from water vapor, ozone, and methane demonstrate that these sources are insufficient to explain the observed radical concentrations in the upper troposphere. The photolysis of carbonyl and peroxide compounds transported to this region from the lower troposphere may provide the source of HOx required to sustain the measured abundances of these radical species. The mechanism by which NO affects the production of O3 is also illustrated by the measurements. In the upper tropospheric air masses sampled, the production rate for ozone (determined from the measured concentrations of HO2 and NO) is calculated to be about 1 part per billion by volume each day. This production rate is faster than previously thought and implies that anthropogenic activities that add NO to the upper troposphere, such as biomass burning and aviation, will lead to production of more O3 than expected.  相似文献

Subpolar North Atlantic proxy records document millennial-scale climate variations 500,000 to 340,000 years ago. The cycles have an approximately constant pacing that is similar to that documented for the last glacial cycle. These findings suggest that such climate variations are inherent to the late Pleistocene, regardless of glacial state. Sea surface temperature during the warm peak of Marine Isotope Stage 11 (MIS 11) varied by 0.5 degrees to 1 degrees C, less than the 4 degrees to 4.5 degrees C estimated during times of ice growth and the 3 degrees C estimated for glacial maxima. Coherent deep ocean circulation changes were associated with glacial oscillations in sea surface temperature.  相似文献
