AIM: To characterise New Zealand's livestock biosecurity databases, and investigate their compatibility and capacity to provide a single integrated data source for quantitative outbreak analysis.

METHODS: Contemporary snapshots of the data in three national livestock biosecurity databases, AgriBase, FarmsOnLine (FOL) and the National Animal Identification and Tracing Scheme (NAIT), were obtained on 16 September, 1 September and 30 April 2014, respectively, and loaded into a relational database. A frequency table of animal numbers per farm was calculated for the AgriBase and FOL datasets. A two dimensional kernel density estimate was calculated for farms reporting the presence of cattle, pigs, deer, and small ruminants in each database and the ratio of farm densities for AgriBase versus FOL calculated. The extent to which records in the three databases could be matched and linked was quantified, and the level of agreement amongst them for the presence of different species on properties assessed using Cohen's kappa statistic.

RESULTS: AgriBase contained fewer records than FOL, but recorded animal numbers present on each farm, whereas FOL contained more records, but captured only presence/absence of animals. The ratio of farm densities in AgriBase relative to FOL for pigs and deer was reasonably homogeneous across New Zealand, with AgriBase having a farm density approximately 80% of FOL. For cattle and small ruminants, there was considerable heterogeneity, with AgriBase showing a density of cattle farms in the Central Otago region that was 20% of FOL, and a density of small ruminant farms in the central West Coast area that was twice that of FOL. Only 37% of records in FOL could be linked to AgriBase, but the level of agreement for the presence of different species between these databases was substantial (kappa >0.6). Both NAIT and FOL shared common farm identifiers which could be used to georeference animal movements, and there was a fair to substantial agreement (kappa 0.32–0.69) between these databases for the presence of cattle and deer on properties.

CONCLUSIONS: The three databases broadly agreed with each other, but important differences existed in both species composition and spatial coverage which raises concern over their accuracy. Importantly, they cannot be reliably linked together to provide a single picture of New Zealand's livestock industry, limiting the ability to use advanced quantitative techniques to provide effective decision support during disease outbreaks. We recommend that a single integrated database be developed, with alignment of resources and legislation for its upkeep.  相似文献

The use of vesicles co‐incubated with plasmids showed to improve the efficiency of cytoplasmic injection of transgenes in cattle. Here, this technique was tested as a simplified alternative for transgenes delivery in porcine zygotes. To this aim, cytoplasmic injection of the plasmid alone was compared to the injection with plasmids co‐incubated with vesicles both in diploid parthenogenic and IVF zygotes. The plasmid pcx‐egfp was injected circular (CP) at 3, 30 and 300 ng/μl and linear (LP) at 30 ng/μl. The experimental groups using parthenogenetic zygotes were as follows: CP naked at 3 ng/μl (N = 105), 30 ng/μl (N = 95) and 300 ng/μl (N = 65); Sham (N = 105); control not injected (N = 223); LP naked at 30 ng/μl (N = 78); LP vesicles (N = 115) and Sham vesicles (N = 59). For IVF zygotes: LP naked (N = 44) LP vesicles (N = 94), Sham (N = 59) and control (N = 79). Cleavage, blastocyst and GFP+ rates were analysed by Fisher's test (p < 0.05). The parthenogenic CP naked group showed lower cleavage respect to control (p < 0.05). The highest concentration of plasmids to allow development to blastocyst stage was 30 ng/μl. There were no differences in DNA fragmentation between groups. The parthenogenic LP naked group resulted in high GFP rates (46%) and also allowed the production of GFP blastocysts (33%). The cytoplasmic injection with LP vesicles into parthenogenic zygotes allowed 100% GFP blastocysts. Injected IVF showed higher cleavage rates than control (p < 0.05). In IVF zygotes, only the use of vesicles produced GFP blastocysts. The use of vesicles co‐incubated with plasmids improves the transgene expression efficiency for cytoplasmic injection in porcine zygotes and constitutes a simple technique for easy delivery of plasmids.  相似文献

A hydraulically integrated serial turbidostat algal reactor (HISTAR) was mathematically modeled and designed for large-scale production of microalgae. HISTAR, which combines turbidostats with a series of continuous-flow stirred-tank reactors (CFSTRs), was designed with contaminant mitigation as a primary design parameter. The concept of contaminant mitigation in the series of CFSTRs is discussed and three mathematical models presented to predict contaminant washout within the CFSTRs. Model simulations indicated that the hydraulic regime within each CFSTR is crucial to the contaminant mitigation process in the series of CFSTRs. Experiments on the HISTAR system with a liquid volume of about 8500 l, culturing Chaetoceros muelleri (CHAET 10), demonstrated that 0.8 million live rotifers, intentionally added to the first CFSTR, were washed out of the six CFSTRs at a local dilution rate of 4 day−1. In another experiment, Thalassiosira sp. cultured at a dilution rate of 0.667 day−1 fell victim to a few rotifers that inadvertently entered the CFSTRs. The validity of the washout concepts was demonstrated further when the ≈300 million live rotifers in the CFSTRs were completely washed out by simply increasing the dilution rate to 4 day−1.  相似文献

The purpose of this study was to develop a mathematical model to assist in the determination of design numbers for color destruction by ozone as a function of feed rate. A mass balance model was developed that takes into account methods of introduction and removal of color, including removal by ozone. Because direct measurement of color mass or concentration is difficult, a representative unit was developed during this study called the color mass equivalent (CME). The CME represents a direct measurement of absorbance at 436 nm, which is a unitless measurement, multiplied by the system volume. The CME is directly proportional to the true concentration of color causing compounds at that wavelength. Once the model was developed, two studies were run to test the model. A 1,500-L recirculating fish system was set up with a 57-L bead filter and six 1-L fluidized sand beds. Approximately 45 kg of channel catfish Ictalurus puncfatus were placed in the system and fed 0.25 kg of 32% protein feed per day (0.55% body wt.). Once the system reached stable conditions, the system was flushed with clean water and samples were taken to estimate the accumulation rate of color in the system. After 5 wk, an ozone unit was activated and samples were taken to determine the destruction rate for color. Once an accumulation rate had been determined, a second study was performed to corroborate this value. The same ozone unit was installed on a 5,000-L recirculating fish system being fed 0.49 kg per day with a 171-L bead filter. Samples were taken to determine the destruction rate for color. The accumulation rate for color was 12.6 CME/kg feed and the destruction rates were 1.7 CME/g O3 in the first study and 0.82 CMWg O3 in the second study. This calculates to a range of 7 to 15 g O3/kg feed to remove the color produced by the feed.  相似文献

A hydraulically integrated, serial turbidostat algal reactor (HISTAR) was mathematically modeled and developed for the mass production of microalgae. HISTAR, which hydraulically links precisely controlled turbidostats with continuous-flow stirred-tank reactors (CFSTRs) into a single production technology, was conceptualized emphasizing contaminant mitigation at the design stage to maintain system stability. This paper discusses the conceptual basis for the development of HISTAR and presents a theoretical overview of the CFSTRs. Mathematical modeling, using first-order algal growth kinetics combined with CFSTR reactor kinetics, was used to investigate the effects of system dilution rate, Ds; net algal specific growth rate, Ua; number of reactors, N; input algal biomass, Xi; and contaminant concentration, Cn, on algal productivity, as well as local dilution rate, Dn, on suspended contaminant washout. A double antibody sandwich ELISA developed by ID-DLO, Lelystad to detect Corynebacterium pseudotuberculosis infection was used on 329 sheep from four pedigree Suffolk flocks in which clinical cases of caseous lymphadenitis (CLA) had occurred. At subsequent necropsy, typical CLA lesions were seen in 133 sheep, and the diagnosis was confirmed on culture. Lesions were most commonly seen in lungs (n = 46), parotid lymph nodes (n = 44), prescapular lymph nodes (n = 38) and mediastinal lymph nodes (n = 31). The sensitivity of the ELISA test for detecting culture-positive sheep was 0.88, while the specificity of the test was 0.55. The antibody ELISA detected 87.5 per cent of sheep that had CLA lesions restricted to internal organs only. It was concluded that the ELISA test has a valuable role in detecting sheep with both clinical and subclinical CLA.  相似文献

Endometrial expression of oestrogen receptor‐α (ERα), progesterone receptor (PR) and cyclooxigenase‐2 (COX‐2) was evaluated in non‐pregnant and pregnant llamas during the period when luteolysis/maternal recognition of pregnancy is expected to occur. Females (n = 28) were divided into two groups: non‐pregnant llamas were induced to ovulate with a Buserelin injection, and endometrial biopsies were obtained on day 8 (n = 5) or 12 (n = 5) post‐induction of ovulation. Animals of the pregnant group (n = 18) were mated with a fertile male. Pregnancy was confirmed by the visualization of the embryo collected by transcervical flushing in 5 of 9 animals on day 8 post‐mating and by progesterone profile on day 12 post‐mating in 4 of 9 animals, when endometrial biopsies were obtained. An immunohistochemical technique was used to evaluate receptors population and COX‐2 expression. Pregnant llamas showed a higher percentage of positive cells and stronger intensity for ERα than for non‐pregnant llamas in stroma on day 8 and in the luminal epithelium on day 12 post‐induction of ovulation, while a deep decrease in endometrial PR population was reported in pregnant llamas on that day in luminal and glandular epithelia and stroma. In the luminal epithelium, COX‐2 expression was lower in pregnant than in non‐pregnant animals. Briefly, the increase of ERα in pregnant llamas gives further support to the hypothesis that oestrogens are involved in the mechanism of maternal recognition of pregnancy. Endometrial PR decrease in pregnant llamas might be a necessary event to allow the expression of proteins involved in conceptus attachment, a mechanism widely accepted in other species. Moreover, embryo seems to attenuate maternal PGF2α secretion during early pregnancy by decreasing the endometrial expression of COX‐2 in the luminal epithelium of pregnant llamas.  相似文献

Accurate simulation of agricultural management effects on N loss in tile drainage is vitally important for understanding hypoxia in the Gulf of Mexico. An experimental study was initiated in 1978 at Nashua, Iowa of the USA to study long-term effects of tillage, crop rotation, and N management practices on subsurface drainage flow and associated N losses. The Root Zone Water Quality Model (RZWQM) was applied to evaluate various management effects in several previous studies. In this study, the simulation results were further analyzed for management effects (tillage, crop rotation, and controlled drainage) on crop production and N loss in drain flow. RZWQM simulated the observed increase in N concentration in drain flow with increasing tillage intensity from NT (no-till) to RT (ridge till) to CP (chisel plow) and to MP (moldboard plow). It also adequately simulated tillage effects on yearly drain flow and yearly N loss in drain flow. However, the model failed to simulate lower corn and soybean yields under NT than under MP, CP, and RT. On the other hand, RZWQM adequately simulated lower yearly drain flow and lower flow-weighted N concentration in drain flow under CS (corn-soybean) and SC (soybean-corn) than under CC (continuous corn). The model adequately simulated higher corn yield under CS and SC than under CC. Applying the newly suggested N management practice for the Midwest of controlled drainage, the model simulated a 30% reduction in drain flow and a 29% decrease in N losses in drain flow under controlled drainage (CD) compared to free drainage (FD). With most of the simulations in reasonably close agreement with observations, we concluded that RZWQM is a promising tool for quantifying the relative effects of tillage, crop rotation, and controlled drainage on N loss in drainage flow. Further improvements on simulated management effects on crop yield and N mineralization are needed, however.  相似文献
