Cloning and Sequence Analysis of PTTG1 Gene in Luchuan Pig

This experiment was aimed to clone PTTG1 gene CDS sequence of Luchuan pig,and was analyzed by bioinformatics methods.A pair of special primers was designed according to predicted sequence of porcine PTTG1 in GenBank.The coding sequence of PTTG1 in Luchuan pig was amplified by RT-PCR,its gene sequence characteristics and protein structure was systemically analyzed by bioinformatics techniques.The results showed that the cloned PTTG1 fragment included a 609 bp CDS (coding 202 amino acids).The sequence multi-aligned results showed that Luchuan pig shared 90.15%,87.85%,87.52%,87.03%,76.03%,74.38%,55.74% and 44.48% of similar nucleotide sequence with that of Bos,Pan troglodytes,Homos,Macaca,Rattus,Mus,Gallus and Danio retio,respectively.The protein structure analysis results showed that the protein attributed hydrophilic protein without signal peptide,localized in cell cytoplasm and had 16 phosphorylation sites.The phylogentic tree of amino acid indicated that PTTG1 was highly conserved in the process of evolution of different species.The cloning and analysis of PTTG1 gene provided an important foundation for further study biological function of porcine PTTG1 during early embryonic development.     

都兰县基本草原划定试点工作及做法

根据《农牧厅关于基本草原划定试点工作实施方案》的要求,都兰县2009～2010年开展基本草原划定试点工作,完成了野外调查、核实,内业整理、制图,政府公告、基本草原保护标志设立等工作。对试点工作加以总结,以期对全省基本草原划定工作提供借鉴。     

星点木组培芽扦插技术研究

为了探索星点木的大规模繁殖技术,以星点木四年生植株当年生枝条、半年生组培苗枝条和组培试管芽条为试材,研究了不同激素种类、浓度及不同插穗来源对星点木试管外扦插生根的影响。结果表明:星点木试管外扦插生根率较高,生长激素能够很好地促进生根,50mg·L-1 IBA处理后生根综合指标较好,生根率可达95%以上,根系效果指数达0.549。半年生组培苗枝条较适合作为插穗来源。     

不同欧李品种(系)光合与水分利用特性比较

以9种不同欧李品种(系)为试材,使用GFS3000光合测定系统测定了自然条件下叶片的光合和水分利用特性。结果表明:9种欧李品种(系)的净光合速率(Pn)、叶片气孔导度(Gs)、胞间CO2浓度(Ci)、蒸腾速率(Tr)、蒸腾比率(TR)、水分利用效率(WUE)均存在差异;在新疆生产建设兵团第十四师二二四团的生长环境下,Pn与Gs、Tr、WUE呈极显著正相关关系,而与Ci、TR呈极显著负相关关系;Ci与TR呈极显著正相关相关关系,与WUE呈极显著负相关关系;Gs与Tr呈极显著正相关关系,与TR呈显著负相关关系;Pn最高的"宏宇Ⅵ号"其WUE较高,但不是最高;WUE最高的"宏宇Ⅰ号"其Pn较高,但不是最高;"01-01号"的Pn和WUE均最低,其它品种(系)的Pn和WUE均处于中等水平,不同的欧李品种(系)通过不同方式协调自身的光合和水分利用来适应生长的环境。     

防护条件下梭梭幼苗生长及养分吸收特性

以新疆古尔班通古特沙漠南缘管件防护与传统移栽2种方式移栽的梭梭幼苗为实验材料,通过野外监测与室内分析,探讨了管件防护条件下梭梭幼苗的生长及养分吸收特性。结果表明:整个生育期内,管件防护梭梭幼苗的各生长指标与传统移栽苗相比,增幅最大的为茎粗,其次为株高。年周期内管件防护梭梭幼苗的生物量呈递增趋势,单株生物量累积可达45.25 g·株~(-1);净增加27.10 g·株~(-1),是传统移栽苗的1.38倍(P0.05)。管件防护梭梭幼苗体内的氮素、磷素、钾素含量呈先升高后降低趋势,在同化枝生长期达到峰值,分别为11.376 g·kg~(-1)、1.066 g·kg~(-1)和23.340 g·kg~(-1),较传统移栽苗高24%、15%和8%,差异显著;而其梭梭幼苗氮素、磷素、钾素的总积累量却呈递增趋势,年净增积累量分别为0.297 g·株~(-1)、0.027 g·株~(-1)和0.560 g·株~(-1),是传统移栽苗的2.23倍、1.72倍和1.65倍;各时期梭梭幼苗体内的氮素、磷素、钾素含量及总积累量均高于传统移栽苗。     

抗TY、抗线虫樱桃番茄新品种长丰88的选育

番茄新品种长丰88是以CT15为母本,CT101-3为父本配制的抗TY、抗线虫杂交一代良种,属无限生长红果类型。生长势强,第7片叶着生第1花穗,每隔2片叶着生一花穗,中早熟。坐果率高、连续结果能力强,春季每穗平均结果 20个,产量59 850 kg/hm~2;果实圆形,无绿色果肩,表面光滑,外形美观。果实硬度大,耐贮运,货架寿命长,单果质量20 g左右,大小均匀。可溶性固形物含量8.9%,VC含量32.43 mg/100 g,总糖3.44%,总酸0.236%。抗病性强,抗番茄黄化曲叶病毒病,根结线虫病发病率为0,灰霉病和早疫病均未见发生。

Preparation of Adjuvant Diluent for Live Vaccine against Classical Swine Fever and Evaluation of its Immune Effects

The aim of this study was to develop a novel oil-in-water (O/W) emulsion as adjuvant diluents (AD) for live vaccine against classical swine fever (CSF) that could effectively enhance the immune effect of vaccine.The AD was prepared by high-pressure homogenization technique.Formulations and preparation parameters were optimized with response surface design.Its stability, particle size, polydispersity (PDI) and Zeta potential were characterized.The humoral immune response and cellular immune response of the AD were evaluated with BALB/c mice by intramuscular injection.The particle size of the AD prepared by optimized formulation and parameters was 100.4 nm, PDI was 0.147, and Zeta potential was —28.7 mV.The experiment results showed that the AD had good stability.The AD was inoculated combined with live vaccine against CSF into BALB/c mice by intramuscular injection.The results showed that the live vaccine against CSF specific immune responses could be evoked in mice by co-inoculation with AD and vaccine.The cellular immune response levels in co-inoculated groups were significantly higher than control group (P<0.05), with obvious phenomena of higher levels of IFN-γ, IL-6 and IL-4 in serum.The result revealed that cellular immune capability significantly improved with the AD.The results strongly revealed that cellular immune capability significantly improved by introducing AD for effective immune-adjuvant for live vaccine against CSF.     

广西油茶桑寄生发病情况调查及清除成效分析

对广西巴马、那坡、田阳、三江、龙胜等5个油茶主产区油茶桑寄生发病情况进行了调查,并对清除桑寄生的成效进行了分析,结果表明:5个县区均发现了不同程度的桑寄生,其中田阳的桑寄生情况最为严重(80.00%),其次是巴马(63.33%)和那坡(50.00%),三江(20.00%)和龙胜县(13.33%)桑寄生比率较低;5枝以下桑寄生枝,对植株生长发育不明显,但寄生枝的数量超过5个时,植株冠幅、叶绿素相对含量、花芽数量和座果数降低明显;清除桑寄生后2年,植株的冠幅、结果枝的数量、叶绿素相对含量、花芽数量以及座果数、单果重均显著增加,清除桑寄生后2年,可增加收入4 796元/hm~2。     

常春藤茎段初代培养研究

利用正交设计表研究常春藤茎段诱导无菌苗的初代培养,试验结果经过方差分析及邓肯氏新复极差法显示:常春藤茎段腋芽萌发的最佳激素组合为:BA1.0mg/L+KT0.5mg/L+NAA0.1mg/L。
