Even relatively short exposure to high altitude canproduce changes in the metabolic capacity of muscle. This study examined the changes in citrate synthase (CS). β-hydroxyacyl-CoA-dehydrogenase (HOAD), lactate dehydrogenase (LDH) and total protein (TP) activity of skeletal muscle in equids after acute high altitude exposure (3800 m). The middle gluteal muscles of trained equids (one Quarter Horse, one Shetland pony, and four Arabians) were sampled before altitude exposure (225 m) and after nine days of sub-maximal exercise at altitude (3800 m). Muscle biopsies were take from a location 10 cm dorsocaudal to the tuber coxae at an angle of 45°. The insertion site was standardized to a depth of 8 cm. Tissue homogenates were assayed for TP and maximal activity of CS, HOAD, and LDH. All samples were run in duplicates and comparisons performed using a paired Student's t-test (significance set at P<0.05). Altitude did not change CS, HOAD or TP. CS for pre-altitude and altitude averaged 31.2±2.9 and 32.6±4.4 μmoles/g/min, respectively (P=0.51). HOAD values averaged 17.9 ±2.1 and 18.2±2.9 μmoles/g/min, respectively (P=0.85). Altitude acclimatization decreased LDH activity. Pre-altitude and altitude LDH averaged 725.4 ±43.4 and 672.7±51.5 μmoles/g/min, respectively (P=0.04). The decrease in LDH is consistent with decreases in skeletal muscle observed in other mammals at high altitude and suggests that muscles do not become more glycolyctic in altitude hypoxia.  相似文献

A 3-yr study (2009–2011) was conducted to examine the spatial and temporal dynamics of stink bugs in three commercial farmscapes. Study locations were replicated in South Carolina and Georgia, in an agriculturally diverse region known as the southeastern coastal plain. Crops included wheat, Triticum aestivum (L.), corn, Zea mays (L.), soybean, Glycine max (L.), cotton, Gossypium hirsutum (L.), and peanut, Arachis hypogaea (L.). Farmscapes were sampled weekly using whole-plant examinations for corn, with all other crops sampled using sweep nets. The predominant pest species of phytophagous stink bugs were the brown stink bug, Euschistus servus (Say), the green stink bug, Chinavia hilaris (Say), and the southern green stink bug, Nezara viridula (L.). Chi-square tests indicated a departure from a normal distribution in 77% of analyses of the variance to mean ratio, with 37% of slopes of Taylor’s power law and 30% of coefficient β of Iwao’s patchiness regression significantly greater than one, indicating aggregated distributions. Spatial Analyses by Distance IndicEs (SADIE) indicated aggregated patterns of stink bugs in 18% of year-end totals and 42% of weekly counts, with 80% of adults and nymphs positively associated using the SADIE association tool. Maximum stink bug densities in each crop occurred when the plants were producing fruit. Stink bugs exhibited greater densities in crops adjacent to soybean in Barnwell and Lee Counties compared with crops adjacent to corn or fallow areas. The diversity of crops and relatively small size of fields in the Southeast leads to colonization of patches within a farmscape. The ecological and management implications of the spatial and temporal distribution of stink bugs within farmscapes are discussed.  相似文献

A 5‐year‐old Arabian stallion was managed for breeding with fresh/extended semen during a period of 8 months with a resulting per cycle pregnancy rate of 26.3%. The stallion was in good health and no abnormalities of the reproductive tract were observed. Evaluation of several ejaculates revealed that sperm production and semen quality were mostly unchanged during the period of evaluation, that sperm production was normal and that semen quality was extremely poor. The most prevalent sperm defects were abnormal heads and mid‐pieces. Most abnormal heads were microcephalic and/or tapered and considerable variation in sperm head dimensions within and among ejaculates was observed. A unique defect characterized by swollen/roughened mid‐piece caused by accumulation of cytoplasmic‐like material and abnormal mitochondrial sheath was observed. Nuclear vacuoles, acrosome defects, and teratoids were also prevalent and most sperm presented multiple abnormalities. The absence of any clear cause or any signs of testicular degeneration, combined with normal sperm production, and constant abnormal sperm production suggest an inherent, congenital disturbance of spermatogenesis as the cause of teratospermia in this case.  相似文献

Forest floor carbon stocks, which include different components of litter, hemic and sapric materials, have not been empirically quantified in tropical montane forest, although they influence soil carbon (C) pools. To date, the contribution of arbuscular mycorrhizae in C sequestration potentials in tropical montane forests have not been clearly investigated. This study determined the amount of C stocks in the different decomposing layers of forest floor, mainly litter, hemic and sapric materials. The abundance of arbuscular mycorrhizal root colonisation differed among forest floor fractions. Forest floor was measured for depth, area density, dry mass and carbon fraction separately in Sungai Kial Forest Reserve, Pahang, Malaysia to calculate C stocks. Percentages of root colonisation in the hemic and sapric materials were investigated. The results showed that forest floor C stocks were significantly higher in hemic (5 Mg C ha?1) and sapric (7.7 Mg C ha?1) compared with the litter fragments (1.5 Mg C ha?1). Mycorrhizal root colonisation was significantly higher (75%) in the toeslope compared with the summit area in the hemic materials. Segregation of forest floor layers provided greater accuracy in forest floor C stocks reporting.  相似文献

Decreasing dietary N inputs into beef cattle feeding operations could potentially decrease environmental concerns relating to air and water quality. Previous studies with sheep suggest that oscillating dietary CP concentrations may improve N use efficiency and thereby decrease dietary N requirements. Therefore, two studies were conducted to determine the effects of oscillating dietary CP concentrations on performance, acid-base balance, and manure characteristics of steers fed high-concentrate diets. Steers were fed to a constant backfat thickness in both studies. In the first trial, 92 steers (mean BW = 408 +/- 2.8 kg; four pens/treatment) were fed the following diets: 1) constant 12% CP, 2) constant 14% CP, and 3) 10 and 14% CP oscillated at 2-d intervals. Steer performance and carcass characteristics were measured. In the second trial, 27 steers were individually fed the same three experimental dietary regimens (nine steers/treatment). Animal performance, arterial acid-base balance, plasma metabolites, and fecal characteristics were measured. In both trials, steers fed the 14% CP diet tended (P < 0.10) to have greater ADG and gain:feed than steers fed the 12% CP diet. Steers fed the oscillating CP regimen had intermediate performance. In Trial 1, steers fed the 14% CP diet tended (P = 0.09) to have smaller longissimus area and higher quality grades than steers fed the oscillating CP regimen. Protein retentions (g/d) calculated from NRC (2000) equations were greater (P = 0.04) for steers fed the 14% CP diet than steers fed the 12% CP diet. Steers fed the oscillating CP regimen tended (P = 0.08) to have greater calculated protein retention (g/d) than steers fed the 12% CP diet. Steers fed the 14% CP diet had greater (P < 0.05) calculated urinary N excretion than steers fed the 12% CP or oscillating CP regimens. Venous plasma concentrations of urea N were greater (P < 0.001) in steers fed the 14% CP diet than in steers fed the 12% CP diet; steers fed the oscillating CP regimen were intermediate but fluctuated over days. Based on arterial blood gas concentrations, acid-base balance was not significantly affected by dietary CP regimen. Results of these trials suggest that the CP requirement of steers in these studies was greater than 12% of the diet DM, and/or that the degradable CP requirement was greater than 6.3% of diet DM. However, the effects of oscillating dietary CP were minimal.  相似文献

Eight crossbred steers (avg wt, 350 kg) with duodenal and ileal cannulas were used in two experimental periods in a single-reversal design to determine the effects of monensin on site and extent of apparent absorption of Mg and Ca. Steers were fed a cottonseed hull:concentrate diet (30:70) with or without 25 mg monensin/kg. The diet contained .20% Mg and .38% Ca. Each diet contained .25% Cr2O3 as a digesta marker. During each period, steers were fed the control diet for 10 d before allotment to treatments. Each period consisted of 17 d for diet adjustment and 6 d for collection. During the collection phase, fecal samples were obtained beginning on d 1 and continuing through d 6 at 12-h intervals progressing 2 h each day. Apparent Mg availability (grams apparently absorbed expressed as a percentage of grams of intake) increased (P less than .05) from 18.4 to 32.5% when monensin was fed. Feeding monensin increased (P less than .05) the apparent Mg availability and amount of Mg absorbed in the preintestinal region (47.7 vs 29.8% and 8.5 vs 5.1 g/d, respectively). Larger quantities (P less than .05) of Mg were secreted into the small intestine when monensin was fed. Monensin did not alter (P less than .05) apparent Ca availability or absorption in any segment of the digestive tract. These data indicate that monensin increased Mg and did not change Ca availability in the preintestinal region.  相似文献

During the first part of a study, cats were inoculated with Cryptococcus neoformans via the following routes: intradermal, intranasal, IV, and intracisternal. Only use of the IV route of inoculation consistently induced disseminated cryptococcosis. In the second part of the study, disseminated cryptococcosis was experimentally induced in cats via IV inoculation of C neoformans. One month after inoculation, 3 cats were treated with ketoconazole (10 mg/kg of body weight/d) and 3 cats were treated with itraconazole (10 mg/kg/d) for 3 months. One of the ketoconazole-treated and 2 of the itraconazole-treated cats also had cryptococcosis of the CNS when treatment was begun. During treatment, serum cryptococcal antigen titer progressively decreased in all cats. Abnormalities in CBC values or the serum biochemical profile were not found in any cat during treatment. However, all ketoconazole-treated cats became anorectic and lost weight. Side effects were not seen in itraconazole-treated cats. During the 3-month posttreatment observation period, all cats remained healthy. At necropsy, histologic evidence of cryptococcosis was not found in the 3 ketoconazole-treated cats or in 2 of the itraconazole-treated cats. In the third itraconazole-treated cat, cryptococcal organisms were found in the kidneys.  相似文献

Four commercial anaerobic systems (CASs) were evaluated for usefulness in identification of Eubacterium suis. Twelve strains were evaluated in each system in triplicate, and results were interpreted independently by 5 individuals. Statistically significant differences (P less than 0.01) due to strain variation and reader interpretation accounted for discrepancies encountered. The reactivity, repeatability, and unique profiles generated made both CAS-1 and CAS-2 suitable adjuncts for identification of E. suis when colony morphology and Gram reaction were considered. Limited reactivity in CAS-3 limited its use as an aid in identification. Variability in test observations and the large number of numerical profiles generated precluded use of CAS-4.  相似文献

Ten abomasally cannulated crossbred wether lambs (avg wt, 33 kg) were used in a replicated 5 x 5 latin square design to determine the site and extent of apparent absorption of Mg when fed different sources of Mg. Lambs were fed twice daily 220 g of chopped mixed grass hay and 180 g of a corn-based supplement (control; .13% mg, DM basis), or the control diet supplemented with Mg (.26% Mg, DM basis) from MgO, magnesium citrate (MgC), smectite-vermiculite (Mg-Mica) or MgOH. Lambs were maintained in metabolism stalls during each of the five experimental periods. Each period consisted of a 7-d dietary adjustment followed by a 3-d collection of abomasal samples, feces and urine. Abomasal contents were sampled four times daily during the 3-d collection period. The diet contained .5% chromium oxide as a digestion marker. Apparent absorption of Mg was .17, .55, .85, .78 and .82 g/d for lambs fed the control, MgO, MgC, Mg-Mica and MgOH diets, respectively. Apparent absorption of Mg (g/d) was similar (P greater than .05) in the lambs fed the supplemented diets and greater (P less than .05) than in those fed the control diet. Preintestinal absorption of Mg was .21, .57, 1.08, .14 and .92 g/d when the control, MgO, MgC, Mg-Mica and MgOH diets were fed. Lambs fed the control and Mg-Mica diets absorbed similar (P greater than .05) quantities of Mg in the preintestinal region and less (P less than .05) than lambs fed the MgO, MgC and MgOH diets.(ABSTRACT TRUNCATED AT 250 WORDS)  相似文献
