Effect of mutant β-catenin gene on the proliferation of hepatocytes

AIM:The β-catenin is a key molecule in the Wnt signal pathway, which plays a critical role in normal development and tumorigenesis. However, the mechanisms of the β-catenin on the cell growth control are still not completely defined. The aim of this study was to test the hypothesis that the mutant β-catenin may regulate the hepatocyte proliferation. METHODS: The immortalized murine hepatocyte cell line, AML12, was used for this study. A plasmid that contain mutant β-catenin S33Y was transfected into the AML12 cells and a stable cell line AML12S33Y was established. The cell growth property of this cell line and the parental cell were compared by flow cytometry analysis and direct cell count. The cells were also tested for the ability to form soft agar colonies, and the ability to form tumors in the severe immune deficient mice (SCID). RESULTS:1. The mutant β-catenin containing cell line AML12S33Y has higher proliferating index compared with the parental AML12 cells (P<0.01), suggesting that mutant β-catenin promotes cell growth. 2. The mutant β-catenin cells formed small colonies in soft agar after 4 weeks of culture, but did not generate tumor in SCID mice. CONCLUSION:The mutant β-catenin promotes liver cell growth.     

Effect of antifibrotic herb serum on rat hepatic stellate cell proliferation and collagen synthesis

AIM: To investigate the effect of Chinese herbs, Ganxianfang(GXF), on rat hepatic stellate cells (HSC) proliferation and collagen synthesis. METHODS: Two types of herb serum, portal venous serum and circumferential venous serum, were prepared from rats infused intragastrically with 16, 8, 4 times adult dose of GXF decoction. HSC isolated from rat liver were processed with the above sera in vitro. Then we mensurated the radioactivity of HSC admixed with [[³H]H]proline and [[³H]H]thymine to judge the effect on proliferation and collagen synthesis of HSC. RESULTS: Both two types of serum collected 0.5, 1, 2 h after intragastrical infusion inhibited HSC proliferation (P＜0.05), and the serum collected 1 h after intragastrical infusion had the strongest effect (P＜0.05). Portal serum decreasea collagen synthesis (P＜0.05), but circumferential serum had no effect (P＞0.05). CONCLUSION: Inhibition of HSC proliferation and decrease of collagen synthesis may contribute to the GXF antifibrotic action.     

ITS—RFLP分析进行昆虫病原线虫斯氏属和异小杆属的分类鉴定

本研究通过PCR扩增和六种限制性内切酶（AluⅠ，HinfⅠ，MboⅠ，RsaⅠ，HaeⅢ和PvuⅡ）酶切，对国内害虫防治上常用的几种昆虫病原线虫，包括斯氏属S.car-pocapsae,S.feltiae和S.glaseri以及异小杆属H.bacteriophora,H.zealandica,H.indicus和H.megidis等8个品系rDNA-ITS进行分析。建立起可以区分各线虫种的标准RFLP图谱。该方法快速简便，稳定可靠，需要的样品量少。可以用于新鲜的，或冻存的样品，甚至分析单条的线虫，不仅可进行昆虫病原线虫的快速分类鉴定。而且进一步可以应用于线虫田间释放的辅助监测。实际田间感染率的测定和线虫毒力的比较。     

山西菜田捕食性食蚜蝇优势种及其对菜蚜的控制效果

对山西菜田捕食性食蚜蝇优势种及对菜蚜的控制效果进行了研究，菜田常见捕食性食蚜蝇有黑带食蚜蝇，大灰食蚜蝇，斜斑鼓额食蚜蝇，野食蚜蝇，四条小食蚜蝇，短翅细腹食蚜蝇6种，以黑带食蚜蝇种群数量最大，平均占全年发生总量的36.3%，通过对食蚜蝇产卵量，卵化率，幼虫期捕食量和羽化率测试，筛选出的黑带食蚜蝇，大灰食蚜蝇，斜斑鼓额食蚜蝇幼虫对菜蚜均有较强的捕食能力，其中黑带食蚜蝇为山西菜田菜蚜捕食性食蚜蝇优势种，用黑带食蚜蝇三龄幼虫对菜蚜进行防治试验，结果以1：180防效最佳，72h后控制效果达93.5%。     

滑菇优良品种筛选试验初报

本试验对 6个不同滑菇品种进行了菌丝体生长和制种观察 ;在栽培试验过程中进行了多点小试和中试 ,初选出Ph0 8号和Ph0 6号两个品种。Ph0 8的单盘产量比CK1高出 2 5 8% ,比CK2 高出 35 3% ;Ph0 6的单盘产量比CK1高出 2 1 5 % ,比CK2 高出 30 6 %。它们均属早生品种 ,适合辽宁地区的气候特点 ,在幼菇期菌盖呈半球形、桔红色、附着的粘液多 ,菌柄比对照短 ,菇体健壮 ,符合商品收购和产品加工的要求     

Metallothionein inhibits homocysteine-induced proliferation of rat vascular smooth muscle cells

AIM:To investigate the effect of metallothionein(MT) on proliferation of rat vascular smooth muscle cells (VSMCs) stimulated by homocysteine and its mechanism. METHODS:VSMCs proliferation was measured by [³-H]-TdR incorporation, mitogen-activated protein kinase(MAPK)activity were determined by immunoprecipitation method, the intracellular contents of MT and malondialdehyde (MDA)were assayed by -hemoglobin saturation method and TBA reaction, respectively, and lactate dehydrogenase (LDH) leakage was measured by NADH oxidation. RESULTS:Hcy(10^-6-10^-4 mmol/L) stimulated [³-H]-TdR incorporation by the VSMCs in a concentration-dependent manner. Compared with control, [³-H]-TdR incorporation in VSMCs treated with 0.1 mmol/L Hcy was increased by 4.2 fold (P＜0.01). Meanwhile, Hcy enhanced MAPK activity, MDA formation and LDH release (P＜0.01)in a concentration-dependent manner. Treatment of VSMCs with MT alone did not change above parameters, compared with control. However, MT (10^-6-10^-4 mol/L)attenuated significantly Hcy-stimulated proliferation of VSMCs (P＜0.01)in a concentration-dependent manner. And MT inhibited obviously Hcy-induced activation of MAPK activity, MDA formation and LDH release. Preincubation of VSMCs with 0.5 mmol/L ZnCl₂ for 6 h induced an increase cellular MT content by 5.7-fold (P＜0.01). The MT-overexpressed VSMCs resisted Hcy-stimulating action on MAPK activity, MDA formation and LDH leakage (P＜0.01). CONCLUSION:These results show that MT has an inhibitory effect on Hcy-induced VSMCs proliferation, and that MT could inhibit Hcy-stimulated MAPK activity and lipid peroxidation.     

Study on the electrophysiological characteristics of ion channels of stem cell derived cardiomyocytes of mouse

AIM: To study the electrophysiological characteristics of ion channels of stem cell derived cardiomyocytes(SCDC) of mouse. METHODS: Embryonic stem cells of D₃ line(ES-D₃) were cultured on the MEF feeder layer with BRL conditioned medium, and fetal mouse heart cells(FMHC)were cultured in vitro. Then ES-D₃ cells were induced to differentiate into many kinds of cells. SCDC were harvested on day 12 after differentiation initiating and identified by electro-microscope and immunocytochemistry. SCDC and FMHC were prepared for the patch-clamp research. Sodium and calcium currents together were elicited and compared between SCDC and FMHC. RESULTS: The current characteristics of sodium and calcium channels of SCDC were very similar to FMHC. CONCLUSION: The functional expression of ion channels occurred during ES-D₃ cells differentiation and the electrophysiological characteristics of sodium and calcium channels of SCDC are very similar to FMHC.     

香菇杂交新菌株1363号的选育研究

采用单孢杂交的方法培育出香菇新菌株 136 3号。它具有产量高 ,质量好 ,遗传性状稳定 ,抗逆性状强等优点。     

金耳的液体发酵研究

研究表明，金耳(Tremella aurantialba)液体发酵最适培养基组成为玉米粉2％，蔗糖3％，酵母膏0．1％，KH2PO40．1％，MgSO40．05％；培养基最佳初始pH、250mL三角瓶装量、接种量、种子菌龄分别是3．5、80mL、10％和120h；用10L发酵罐发酵时，最佳培养条件是种子种龄144h、28℃、接种量10％、通风量2．5L／min。     

酸益(SI)在断奶仔猪日粮中的应用比较

选用杂种仔猪杜×(长×川白)24头,于28±2日龄断奶,以玉米-豆粕型日粮饲喂,进行为期30d的饲养试验,研究日粮中添加0.1%复合酸化剂-酸益SI、0.1%复合酸化剂FD和降低1.5%乳清粉用量对断奶仔猪生产性能、日粮酸和力及肠道大肠杆菌数的影响.结果表明,添加复合酸化剂-酸益(SI)可明显提高仔猪日增重,改善料肉比,大幅度降低腹泻率,降低乳清粉用量与饲料成本.其料肉比、日增重与使用进口产品FD差异不显著(P＞0.05).
