In attempts to induce differentiation of lymphoid cells from hematopoietic stem cells in vitro, the effects of allogeneic effect factor on the growth of murine bone marrow cultures were studied. Allogeneic effect factor is a soluble mediator derived from mixed secondary murine leukocyte cultures. For several weeks it supported the growth of bone marrow cultures, as indicated by the maintenance of stem cell activity, cellular proliferation, and heterogeneity. Another lymphokine, T cell growth factor, did not, Pre-T lymphocytes could be detected in these cultures for several weeks.  相似文献

Heterologous prime/boost regimens have the potential for raising high levels of immune responses. Here we report that DNA priming followed by a recombinant modified vaccinia Ankara (rMVA) booster controlled a highly pathogenic immunodeficiency virus challenge in a rhesus macaque model. Both the DNA and rMVA components of the vaccine expressed multiple immunodeficiency virus proteins. Two DNA inoculations at 0 and 8 weeks and a single rMVA booster at 24 weeks effectively controlled an intrarectal challenge administered 7 months after the booster. These findings provide hope that a relatively simple multiprotein DNA/MVA vaccine can help to control the acquired immune deficiency syndrome epidemic.  相似文献

The number of synapses in the molecular layer of the rat cerebellum is reduced by early hypo-and hyperthyroidism within 30 days. Hypothyroidism retards synaptogenesis after 10 days, while hyperthyroidism accelerates synaptogenesis initially, but by 21 days the number of synapses is reduced. The sensitivity of developing synapses to thyroid hormone may permit analysis of the events triggering synaptogenesis.  相似文献

CASE HISTORY: A feral cat captured in the Manawatu region of New Zealand was treated for worms and fleas, and kept confined in a metabolic cage. It showed good appetite and weight gain but had intermittent watery, yellow diarrhoea.

CLINICAL FINDINGS: Clinical examination under sedation was unremarkable and routine blood tests showed no significant abnormalities. The cat was negative for feline immunodeficiency virus (FIV) and feline leukaemia virus (FeLV). Different canned cat foods did not alter the course of the diarrhoea, and the cat was euthanised 6 months after capture. At necropsy, two sections of adult Spirometra tapeworms were found in the jejunum and typical Spirometra eggs were found in colonic contents. Molecular identification of the parasite was undertaken, using the cytochrome- c oxidase subunit-1 gene (cox1) sequence.

DIAGNOSIS: Chronic intermittent diarrhoea associated with Spirometra erinacei / S. erinaceieuropaei infection.

CLINICAL RELEVANCE: Spirometra has not been reported in New Zealand before but has been associated with gastrointestinal disease in cats in other parts of the world. It requires speciestargeted treatment to be eliminated effectively, and is zoonotic. Diagnosis could be diffi cult for clinicians who are not familiar with the parasite and its life cycle.  相似文献

ObjectiveTo compare anaesthesia induced with either alfaxalone or ketamine in horses following premedication with xylazine and guaifenesin.Study designRandomized blinded cross-over experimental study.AnimalsSix adult horses, five Standardbreds and one Thoroughbred; two mares and four geldings.MethodsEach horse received, on separate occasions, induction of anaesthesia with either ketamine 2.2 mg kg?1 or alfaxalone 1 mg kg?1. Premedication was with xylazine 0.5 mg kg?1 and guaifenesin 35 mg kg?1. Incidence of tremors/shaking after induction, recovery and ataxia on recovery were scored. Time to recovery was recorded. Partial pressure of arterial blood oxygen (PaO2) and carbon dioxide (PaO2), arterial blood pressures, heart rate (HR) and respiratory rates were recorded before premedication and at intervals during anaesthesia. Data were analyzed using Wilcoxon matched pairs signed rank test and are expressed as median (range).ResultsThere was no difference in the quality of recovery or in ataxia scores. Horses receiving alfaxalone exhibited a higher incidence of tremors/shaking on induction compared with those receiving ketamine (five and one of six horses respectively). Horses recovered to standing similarly [28 (24–47) minutes for alfaxalone; 22 (18–35) for ketamine] but took longer to recover adequately to return to the paddock after alfaxalone [44 (38–67) minutes] compared with ketamine [35 (30–47)]. There was no statistical difference between treatments in effect on HR, PaO2 or PaCO2 although for both regimens, PaO2 decreased with respect to before premedication values. There was no difference between treatments in effect on blood pressure.Conclusions and clinical relevanceBoth alfaxalone and ketamine were effective at inducing anaesthesia, although at induction there were more muscle tremors after alfaxalone. As there were no differences between treatments in relation to cardiopulmonary responses or quality of recovery, and only minor differences in recovery times, both agents appear suitable for this purpose following the premedication regimen used in this study.  相似文献

Plasma samples from dogs with infections of Dirofilaria immitis were assessed using ELISA and Western blotting techniques. These results were then assessed in relation to age and sex of the host, and to the numbers of microfilariae and adult filariae. Dogs with microfilariae tended to have lower levels of infection. In infected dogs, mean ELISA titres increased directly with the degree of infection. Dogs that were either young or mildly infected, showed a preferential antibody reactivity to antigens in the high molecular weight regions of the immunoblots. With increasing age and/or the extent of infection, an antibody response to antigens in the low molecular weight regions was apparent.  相似文献
