Premature farrowings caused by feeding cottonseed meal

Increasing the level of cottonseed meal (CSM) in sow diets from less than 5% to 10% increased the incidence of premature farrowings (gestation length less than 111 days) from 1.1% to 2.7% (p less than 0.001) and reduced the mean gestation length from 114.07 +/- 1.53 to 113.70 +/- 1.59 days (p less than 0.0001). Survival of piglets born prematurely was poor. After removal of CSM from the diet there was a residual effect lasting several weeks before the gestation length returned to normal. Experimental feeding of diets containing 20% and 40% CSM to small groups of sows caused significant shortening of gestation length and 3 of 26 sows fed 40% CSM farrowed prematurely. The mechanism by which CSM causes this effect has yet to be determined.     

Photosensitisation and crystal-associated cholangiohepatopathy in sheep grazing Brachiaria decumbens

Two outbreaks of photosensitivity and jaundice in sheep grazing the grass, Brachiaria decumbens. In West Java in 1983 and 1983 and 1990 are described. Aggregates of acicular clefts and crystals were present in bile canaliculi, bile ducts, Kupffer cells, hepatocytes and in kidney tubules. The number of crystals increased with the length of exposure to the plant and sheep that died had the largest accumulations. These crystalline structures have been reported before in this syndrome, but are similar to those seen in Tribulus terrestris and Panicum spp toxicities.     

Identification of the etiologic agent of epizootic bovine abortion in field-collected Ornithodoros coriaceus Koch ticks

Epizootic bovine abortion (EBA), or foothill abortion as it has often been termed, is a tick-borne disease of pregnant cattle recognized in California, Nevada and Oregon. The primary objective of this study was to better define the relationship of a novel deltaproteobacterium, the putative etiological agent of EBA (aoEBA), with the Pajaroello tick (Ornithodoros coriaceus Koch), the recognized vector of EBA. Three developmental stages of O. coriaceus (larva, nymph, and adult) were collected from five locations in California, Nevada and Oregon. A polymerase chain reaction (PCR), developed for detection of aoEBA, was applied to DNA extracted from ticks. Southern blotting of the PCR products increased the number of ticks determined to be carrying the bacteria by seven-fold, suggesting the majority of infected ticks carry relatively low numbers of the pathogen. An effort was made to determine if an artificial blood meal would stimulate replication of the bacterial pathogen, thereby increasing the frequency in which aoEBA could be identified; no statistically significant effect was evident. The number of ticks determined to be carrying aoEBA varied with geographic location and ranged from 5 to 20%. aoEBA was found in both adults (12% of the males and 12% of the females) and nymphs (13%) but not larvae. Comparative analysis of dissected ticks provided strong evidence that the salivary gland was the most common location of aoEBA in field-collected ticks. No significant correlations were identified between the frequency of infection and tick weight, suggesting that increasing tick age and increased number of blood meals did not increase infectivity.     

Observation of individual DNA molecules undergoing gel electrophoresis

Individual DNA molecules undergoing agarose gel electrophoresis were viewed with the aid of a fluorescence microscope. Molecular shape and orientation were studied in both steady and pulsed electric fields. It was observed that (i) DNA macromolecules advanced lengthwise through the gel in an extended configuration, (ii) the molecules alternately contracted and lengthened as they moved, (iii) the molecules often became hooked around obstacles in a U-shape for extended periods, and (iv) the molecules displayed elasticity as they extended from both ends at once. A computer model has been developed that simulates the migration of the molecules in a rotating-field gel electrophoresis experiment.     

Desflurane vapor pressure depression

Due to its high vapor pressure and low boiling point, desflurane requires a specially designed, electronically controlled, temperature and pressure compensated vaporizer to regulate agent delivery to the anesthetic circuit. However, if the vapor pressure and boiling point were decreased, desflurane could be used in any conventional variable bypass vaporizer. Raoult's Law states that the vapor pressure of a liquid is proportional to its molar fraction in a solution. Accordingly, propylene glycol was used as a solvent for desflurane, and the physical characteristics of this mixture were evaluated at various molar concentrations and temperatures. Desflurane boiling point increased and vapor pressure decreased as a nonlinear function of dilution, but these changes were less than predicted by Raoult's Law. Using a circle system with a breathing bag attached at the patient end and a mechanical ventilator to simulate respiration, an in‐circuit, nonprecision vaporizer containing 40% desflurane and 60% propylene glycol achieved a 11.5 ± 1.0% (mean ± SD) circuit desflurane concentration with a 5.2 ± 0.4 (0 = off, 10 = maximum) vaporizer setting. This experiment was repeated with a dog attached to the breathing circuit under spontaneous ventilation with a fresh gas flow of 0.5 L min^–1. Anesthesia was maintained for over two hours at a mean vaporizer setting of 6.2 ± 0.4, yielding mean inspired and end‐tidal desflurane concentrations of 8.7 ± 0.5% and 7.9 ± 0.7%, respectively. Within 5 minutes after cessation of anesthesia, the dog was awake, extubated and standing. In clinical practice, propylene glycol may not prove an ideal solvent for desflurane due to its instability in solution and substantial positive deviation from Raoult's Law. However, rather than alter the vaporizer to suit physical properties of anesthetic agents, this study demonstrates that it may also be possible to alter anesthetic agents to suit physical properties of the vaporizer.     

Humoral immune responses to phocine herpesvirus-1 in Pacific harbor seals (Phoca vitulina richardsii) during an outbreak of clinical disease

Infection with phocine herpesvirus type-1 (PHV-1) has been associated with morbidity and high mortality in neonatal harbor seals (Phoca vitulina). A PHV-1 specific indirect enzyme linked immunosorbent assay (ELISA) was developed to sequentially measure the serological status of 106 harbor seal neonates admitted to a Pacific coast rehabilitation center (total number of sera tested was 371). Early in the season (February-April), the majority of pups had low serum levels of PHV-1 specific antibody. A dramatic increase in PHV-1 specific antibody, involving the majority of hospitalized pups, was observed during a 4-week period in May. This coincided with a high incidence of PHV-1 associated adrenal lesions and mortality. Although there was overall agreement between the timing of seroconversion to PHV-1 and histological evidence of PHV-1 infection, 82.4% of individual pups with adrenalitis had no evidence of a humoral response to PHV-1 at the time of their death. This suggests either a rapid disease course, or an inability to develop a humoral response in some neonatal seals.
