Newcastle disease (ND) is a highly contagious avian disease and one of the major causes of economic losses in the poultry industry. The emergence of virulent NDV genotypes and repeated outbreaks of NDV in vaccinated chickens have raised the need for fundamental studies on the virus–host interactions. In this study, the profiles of B and T lymphocytes and macrophages and differential expression of 26 immune-related genes in the spleen of specific-pathogen-free (SPF) chickens, infected with either the velogenic genotype VII NDV strain IBS002 or the genotype VIII NDV strain AF2240, were evaluated. A significant reduction in T lymphocyte population and an increase in the infiltration of IgM+ B cells and KUL01+ macrophages were detected in the infected spleens at 1, 3 and 4 days post-infection (dpi) (P < 0.05). The gene expression profiles showed an up-regulation of CCLi3, CXCLi1, CXCLi2 (IL-8), IFN-γ, IL-12α, IL-18, IL-1β, IL-6, iNOS, TLR7, MHCI, IL-17F and TNFSF13B (P < 0.05). However, these two genotypes showed different cytokine expression patterns and viral load. IBS002 showed higher viral load than AF2240 in spleen at 3 and 4 dpi and caused a more rapid up-regulation of CXCLi2, IFN-γ, IL-12α, IL-18, IL-1β, iNOS and IL-10 at 3 dpi. Meanwhile, the expression levels of CCLI3, CXCLi1, IFN-γ, IL-12α, IL-1β and iNOS genes were significantly higher in AF2240 at 4 dpi. In addition, the expression levels of IL-10 were significantly higher in the IBS002-infected chickens at 3 and 4 dpi. Hence, infection with velogenic genotype VII and VIII NDV induced different viral load and production of cytokines and chemokines associated with inflammatory reactions.  相似文献

Growth factors play critical role in cell proliferation, regulate tissue differentiation and modulate organogenesis. Several growth factors have been identified in the testes of various mammalian species in last few years. In present investigation, the objective was to determine the expression of epidermal growth factor (EGF) and the epidermal growth factor receptor (EGFR) in yak testicular tissue by relative quantitative real time polymerase chain reaction (RT‐PCR), Western blot (WB) and immunohistochemistry (IHC) from mRNA and protein levels. The testicular tissues were collected from male yak at 6 and 24 months old. Results of RT‐PCR and WB showed that the expression quantity of EGF and EGFR at 24 months of age was higher than at 6 months, and the increase rate of EGFR on mRNA and protein levels was higher than the increase rate EGF during post‐natal testes development. Positive staining for EGF and EGFR was very low and mainly localized to Leydig cells testes at 6 months of age with immunohistochemistry, and seminiferous tubules were not observed. At 24 month of age, both the EGF and EGFR could be detected in Leydig cells, peritubular myoid cells, sertoli cells and germ cells of the yak testes. However, EGF and EGFR were localized to preferential adluminal compartment and basal compartment in the seminiferous tubules, respectively. In conclusion, the findings in present studies suggest that EGF and EGFR as important paracrine and/or autocrine regulators in yak testes development and spermatogenesis.  相似文献

The finite element method was used to analyse the cutting process of a sandy loam soil with medium-deep subsoiler, used as a compaction sensor, aiming to calculate the subsoiler draught for various combinations of dry bulk density, moisture content and tillage depth. The finite element results showed that draught increased with depth and dry bulk density, whereas it decreased with moisture content. A multiple linear regression analysis was performed to establish a formula for relating subsoiler draught (dependent variable) with the three independent variables. The regression equation developed was simple and had a high determination coefficient close to 0.95. An equation for prediction of dry bulk density as a function of moisture content, depth and draught was derived from the regression equation developed. This equation was used to calculate dry bulk density, for measured depth, moisture content and draught at nine different points along a single line in a meadow field of a silty clay loam soil. The predicted dry bulk density indicated that there was a considerable variation in the degree of compaction throughout the measured line. However, the on-line depth control and measurement of moisture content still need to be integrated with the on-line measurement of draught, to govern the model suitability for performing on-line detection of the spatial distribution of soil compaction, assessed as dry bulk density.  相似文献

The adsorption of four dyestuffs onto bagasse pith has been studied. A method has been presented for the prediction of concentration decay vs time. The model is based on external mass transfer and pore diffusion and enables the external transport coefficients and the effective diffusivities to be determined. Constant mass transport. coefficients were obtained for each dye-pith system to correlate the effects of varying the initial dye concentration and pith mass. The external mass transfer coefficients are 1.4 × 10?2, 1.24 × 10?2, 1.5 × 10?3, and 2.5 × 10?3 cm s?1 and the effective diffusivities are 6.5 × 10?6, 2.6 × 10?6, 4.5 × 10?7, and 5.7 × 10?7 cm2 s?1 for Basic Blue 69, Basic Red 22, Acid Blue 25, and Acid Red 114, respectively.  相似文献

Thirty seven diverse genotypes of Vigna mungo and three of V. radiataresembling to V. mungo for seed characters were studied to determine the extent of genetic variation based on morphological characters. In addition, 4black seeded genotypes in each species and8 green seeded V. radiata genotypes were included for SDS-PAGE analysis. Seed proteins were analyzed through slab type SDS-PAGE. High variance was observed for plant height, days to flowering, days to maturity, number of branches/plant, number of pods/plant, pod length, seeds per pod,biomass yield/plant, grain yield/plant and harvest index (%) during 2 consecutive years. First four components of PCA with eigenvalues >1 contributed 78.7 and79.1% of the total variance amongst 40genotypes during 1998 and 1999. The populations with high PC1 values were expected to be high yielding characterised by earlyness, high seed weight and harvest index, all traits strictly related to reproductive phase. Four clusters of genotypes were observed during both the years and genetic diversity was in association for both the years. Based on SDS-PAGE, specific bands were suggested to be used for identifying Vigna radiatafrom mixed germplasm with Vigna mungo. The SDS-PAGE proved to be a powerful tool for differentiating Vigna radiata and Vigna mungo, whereas a low level inter-specific genetic diversity was observed and no clear differentiation was observed both for agronomic characteristics and for geographical origin. Various feeding studies have been conducted with the different species of animals to evaluate the possible transfer of transgenic DNA (tDNA) from genetically modified (GM) feed into the animal tissues. However, the conclusions drawn from most of such studies are sometimes controversial. Thus, in the present study, an attempt has been made to evaluate the fate of tDNA in rabbits raised on GM cotton-based diet through PCR analysis of the DNA extracted specifically from blood, liver, kidney, heart and intestine (jejunum). A total of 48 rabbits were fed a mixed diet consisting variable proportions of transgenic cottonseeds meal (i.e. 0% w/w, 20% w/w, 30% w/w and 40% w/w) for 180 days. The presence of transgenic DNA fragments (Cry1Ac, Cry2A and CP4 EPSPS) or plant endogenous gene (Sad1) was traced in those specific tissues and organs. The presence of β-actin (ACTB) was also monitored as an internal control. Neither the transgenic fragments (459 bp of Cry1Ac gene, 167 bp of Cry2A gene and111 bp of CP4 EPSPS gene) nor cotton endogenous reference gene (155 bp of Sad1) could be detected in any of the DNA samples extracted from the rabbit's tissues in both control and transgenic groups. However, 155 bp fragment of the rabbit's reference gene (ACTB) was recovered in all the DNA samples extracted from rabbit tissues. The results obtained from this study revealed that both plant endogenous and transgenic DNA fragments have same fate in rabbit's tissues and were efficiently degraded in the gastrointestinal tract (GIT).  相似文献

Boron (B) is one of the essential micronutrients having a specific role, particularly during reproductive phase, in rice. In a previous experiment on aerobic rice, panicle sterility was noted as one of the major challenges. This experiment was conducted to evaluate the influence of soil-applied B on tillering, panicle sterility, water relations, and grain enrichment in fine-grain aromatic rice cultivars ‘Super Basmati’ and ‘Shaheen Basmati’. Boron was soil applied at 0.50, 0.75, 1, 1.25, and 1.50 kg ha?1 while the control treatment did not receive B. Rate of leaf emergence and elongation and tiller appearance were significantly improved by B application. Likewise, B application also improved the leaf chlorophyll contents and water relations in both rice cultivars. Substantial improvement in kernel yield and yield contributing traits was also observed by B application owing to decrease in panicle sterility. A linear increase in leaf and kernel B contents was observed with increase in B application rate. However, the range for an optimum B application rate is very narrow and increase of B application beyond 1 kg ha?1 was toxic. In conclusion, soil application of B is an effective way to decrease panicle sterility and increase the kernel yield and grain B enrichment in rice.  相似文献

The production performance of genetically improved farmed tilapia (GIFT, Oreochromis niloticus) and freshwater prawn (Macrobrachium rosenbergii) in periphyton‐based systems were studied in farmers' ponds at Mymensingh, Bangladesh. Fifteen ponds (200–300 m2 area and 1.0–1.5 m in depth) were used to compare five stocking ratios in triplicate: 100% GIFT, 75% GIFT plus 25% prawn, 50% GIFT plus 50% prawn, 25% GIFT plus 75% prawn and 100% prawn. Ponds were stocked at a total density of 20 000 GIFT and/or prawn ha?1. Bamboo poles (mean diameter 6.2 cm and 5.5 pole m?2) were posted in pond bottoms vertically as periphyton substrate. Periphyton biomass in terms of dry matter (DM), ash‐free DM and chlorophyll a were significantly higher in ponds stocked with prawn alone than in ponds with different combinations of GIFT and prawn. Survival of GIFT was significantly lower in ponds stocked with 100% GIFT (monoculture) whereas, that of prawn was significantly higher in its monoculture ponds indicating detrimental effects of GIFT on prawn's survival. Individual weight gains for both species were significantly higher in polyculture than in monoculture. The highest total fish and prawn yield (1623 kg GIFT and 30 kg prawn ha?1) over 125–140 days culture period was recorded in ponds with 75% GIFT and 25% prawn followed by 100% GIFT alone (1549 kg ha?1), 50% GIFT plus 50% prawn (1114 kg GIFT and 68 kg prawn ha?1), 25% GIFT plus 75% prawn (574 kg GIFT and 129 kg prawn ha?1) and 100% prawn alone (157 kg ha?1). This combination also gave the highest economic return. Therefore, a stocking ratio of 75% GIFT plus 25% prawn at a total density of 20 000 ha?1 appeared to be the best stocking ratio in terms of fish production as well as economics for a periphyton‐based polyculture system.  相似文献
