In this paper, seed morphology and effects of pre-sowing treatments were studied. Matured seeds of the species were collected from healthy trees in the National Botanical Garden, Bangladesh, and treated with five pre-sowing treatments. The average length, breadth and thickness were found to be 0.56±0.03 cm, 0.44±0.009 cm and 0.26±0.008 cm in Albizia richardiana King and Prain, and 1.32±0.02 cm, 0.55±0.04 cm and 0.11±0.002 cm in Lagerstroemia speciosa L., respectively. Germination was carried out in polybags with a mixture of topsoil, coconut husk compost, coarse sand and fine sand in a ratio of 3:4:1:1. Results revealed that the germination rates of seeds in different pre-sowing treatments were significantly increased compared to those in cold-water treatment in both species. The highest germination rate was found to be 96% in hot-water treatment followed by 87%, 83% and 49% in treatments with scarification, H2SO4 and control in A. richardiana, respectively. However, the highest germination rate (79%) was found in H2SO4 treatment followed by 64%, 62% and 25% in treatments with hot water, scarification and control in L. speciosa, respectively. The lowest germination (35% in A. richardiana and 20% in L. speciosa) was found in cold-water treatment in both cases. Analysis of variance (ANOVA) showed the significant difference among the treatments for both species. It is concluded that hot-water treatment is recommended for seed germination of both species in rural Bangladesh.  相似文献

The H5 gene of avian influenza virus (AIV) strain A/chicken/Malaysia/5744/2004(H5N1) was cloned into pcDNA3.1 vector, and Esat-6 gene of Mycobacterium tuberculosis was fused into downstream of the H5 gene as a genetic adjuvant for DNA vaccine candidates. The antibody level against AIV was measured using enzyme-linked immunosorbent assay (ELISA) and haemagglutination inhibition (HI) test. Sera obtained from specific-pathogen-free chickens immunized with pcDNA3.1/H5 and pcDNA3.1/H5/Esat-6 demonstrated antibody responses as early as 2 weeks after the first immunization. Furthermore, the overall HI antibody titer in chickens immunized with pcDNA3.1/H5/Esat-6 was higher compared to the chickens immunized with pcDNA3.1/H5 (p < 0.05). The results suggested that Esat-6 gene of M. tuberculosis is a potential genetic adjuvant for the development of effective H5 DNA vaccine in chickens.  相似文献

We investigated the effect of prohexadione-calcium (Pro-Ca) on growth characteristics and endogenous gibberellins content of Chinese cabbage grown in the alpine region of South Korea. Pro-Ca was applied at the rates of 200 ppm and 400 ppm, after 10, 15 and 20 days of transplanting seedlings in to the field. Application of Pro-Ca through foliage improved quality and quantity of yield by promoting Chinese cabbage head yield, number of head leaves, total soluble sugar content and compactness of head. The leaf size was reduced, while the chlorophyll content increased under the influence of elevated Pro-Ca application, when measured after 40 days of transplantation. The endogenous bioactive GA1 and GA4 contents of Chinese cabbage drastically decreased with elevated Pro-Ca, indicating that gibberellins (GAs) biosynthesis was blocked by this chemical. Current study suggests that both of GAs biosynthesis pathways are operational in Chinese cabbage, although non-C13-hydroxylation pathway was found to be the major pathway. A local isolate of Metarhizium anisopliae (Hypocreales: Clavicipitaceae), Bacillus thuringiensis subsp. kurstaki and chlorantraniliprole were assessed against six field populations of tomato fruitworm Helicoverpa armigera Hübner (Lepidoptera: Noctuidae) in a series of laboratory bioassays. Two dose rates of B. thuringiensis (0.5, 1 μg g?1), one of both M. anisopliae (1.3?×?106 conidia ml?1) and chlorantraniliprole (0.01 ppm) were applied alone and in combination with each other against 2nd, 3rd, 4th and 5th larval instars. The mortality was observed every 24 h until pupation. The bioassays were carried out at 25°C and 75% r.h. The highest mortality was observed in Rawalpindi with the lowest pupation rate by applying the combined concentrations of B. thuringiensis and chlorantraniliprole. The lowest mortality was observed in population from Gujranwala among all the tested populations. The antagonistic interaction was noted where the high dose rate of B. thuringiensis was combined with M. anisopliae; however, the remaining interactions enhanced the mortality and reduced the percent pupation. The overall results demonstrated that all the treatments gave significant control of the larval instars of H. armigera. The population from Gujranwala proved least susceptible whereas the one from Rawalpindi was highly susceptible.  相似文献

Summary

Gerbera (Gerbera jamesonii) is an attractive ornamental flower of high economic importance. The present investigation was aimed at generating novel flower colour variants in the gerbera cultivar ‘Harley’ through physical and chemical mutagenesis. In vitro-raised shoot cultures of gerbera, established from petiole explants, were exposed to different doses of γ-rays (1.5, 2.0, 2.5, 5.0, 10.0, 15.0, 20.0, or 30.0 Gy) using a Cobalt-60 source emitting 2.51 kGy h–1. To induce mutations through chemical mutagenesis, different concentrations of ethylmethane sulphonate [EMS; 0.1, 0.2, 0.5, 0.8, or 1.0% (v/v)] were administered for 10 min or for 20 min. The LD50 values calculated for shoot survival and the induction of mutations were approx. 6.5 Gy for γ-rays and 0.65% (v/v) EMS for 10 min, or ≤ 0.1% (v/v) EMS for 20 min. Investigations revealed a negative correlation between mutagen dose and plant survival, both in vitro and after acclimatisation. Morphological variants showing changes in leaf shape, leaf size, scape length, flower diameter, and flower colour were obtained. Significantly, early flowering was induced in all mutated plants compared to non-mutated plants.The high frequencies of colour variants obtained using Bγ-rays, or the application of EMS to in vitro-raised shoot cultures could be an effective way to improve gerbera cultivars.  相似文献

The study examined the influence of compost and mineral fertilizer application on the content and stability of soil organic carbon (SOC). Soil samples collected from a long-term field experiment were separated into macroaggregate, microaggregate, and silt + clay fractions by wet-sieving. The experiment involved seven treatments: compost, half-compost N plus half-fertilizer N, fertilizer NPK, fertilizer NP, fertilizer NK, fertilizer PK, and control. The 18-year application of compost increased SOC by 70.7–121.7%, and mineral fertilizer increased by 5.4–25.5%, with no significant difference between control soil and initial soil. The C mineralization rate (rate per unit dry mass) in microaggregates was 1.52–2.87 mg C kg−1 day−1, significantly lower than in macroaggregate and silt + clay fractions (P < 0.05). Specific C mineralization rate (rate per unit SOC) in silt + clay fraction amounted to 0.48–0.87 mg C g−1 SOC day−1 and was higher than in macroaggregates and microaggregates. Our data indicate that SOC in microaggregates is more stable than in macroaggregate and silt + clay fractions. Compost and mineral fertilizer application increased C mineralization rate in all aggregates compared with control. However, compost application significantly decreased specific C mineralization rate in microaggregate and silt + clay fractions by 2.6–28.2% and 21.9–25.0%, respectively (P < 0.05). By contrast, fertilizer NPK application did not affect specific C mineralization rate in microaggregates but significantly increased that in silt + clay fractions. Carbon sequestration in compost-amended soil was therefore due to improving SOC stability in microaggregate and silt + clay fractions. In contrast, fertilizer NPK application enhanced SOC with low stability in macroaggregate and silt + clay fractions.  相似文献

The prolonged irrigation with marginal quality water can cause secondary salinization of soils, which necessitates for better understanding of water management alternatives. Relative performance of sulfuric acid and gypsum is still controversial to counter sodium hazards in soil/water system. As an alternative, sulfurous acid generators (SAG) are also being marketed. But up-till-now, there is not even a single field study published in scientific journals about their efficiency and economical viability for the treatment of saline-sodic water. Therefore, a field study was carried out to compare the effectiveness of SAG and alternate amendments applied on an equivalent basis to grow rice crop. SAG treatment of saline-sodic tube well water decreased only residual sodium carbonate (RSC) from 5.4 to 3.6 mmolc l−1, and had no beneficial effect on its sodium adsorption ratio (SAR) or electrical conductivity (EC). All the treatments kept soil EC and SAR around their respective threshold levels. For paddy yield, SAG, sulfuric acid, and gypsum treatments depicted nonsignificant differences. SAG and sulfuric acid treatments of water were about six times expensive than that of gypsum. It was concluded that soil-applied gypsum, to counter sodic hazards of irrigation water, is economical to sustain irrigated rice in dry regions.  相似文献

The development of anthelmintic resistance has made the search for alternatives to control gastrointestinal nematodes of small ruminants imperative. Among these alternatives are several medicinal plants traditionally used as anthelmintics. This present work evaluated the efficacy of ethyl acetate, acetone, and methanol dried leaf and seed extracts of five medicinal plants were tested in vitro ovicidal and larvicidal activities on Haemonchus contortus. The in vitro assay was based on egg hatch assay (EHA) and larval development assay (LDA), all plant extracts were evaluated at five concentrations 50, 25, 12.5, 6.25 and 3.13 mg/ml. The leaf and seed ethyl acetate, acetone and methanol of Annona squamosa, Eclipta prostrata, Solanum torvum, Terminalia chebula, and Catharanthus roseus extracts were showed complete inhibition (100%) at the maximum concentration tested (50 mg/ml). The overall findings of the present study have shown that our experimental plant extracts contain possible anthelmintic compounds.  相似文献

Cr3+ sorption on strong acid exchanger Amberlyst-15(H+) is studied as a function of time and temperature using CrCl3.6H2O and [Cr4(SO4)5(OH)2] solutions. The rate is found to be governed by a mixed diffusion for both the solutions and faster for Cl1? solution than SO4 2?. The exchange capacities are found to be higher for Cl1? system than SO4 2?. From the rate constant values, the energies of activation are calculated using the well-known Arrhenius equation. Equilibrium data is explained with the help of the Langmuir equation. The Langmuir parameters are also found to be higher for exchange from the chloride solutions. Various thermodynamic parameters (??Ho, ??So, and ??Go) for Cr3+ exchange on the resin are calculated. The ??Go values are found to be negative while ??Ho and ??So are positive for both the Cr3+/Cl1? and Cr3+/SO4 2? systems. It is suggested that in case of Cl1? solutions, the metal is exchanged as Cr3+, while in case of SO4 2? solutions, the metal exchanging specie is CrSO4 +.  相似文献
