潜叶蝇在大庆市日光温室蔬菜上的发生与为害

对潜叶蝇在北方节能日光温室蔬菜上的发生和为害进行调查研究。结果表明,3～11月潜叶蝇在日光温室蔬菜上均有发生,其中5月中旬～6月中旬、9月下旬～10月中旬是为害高峰期;一日中9:00～11:00、13:00～15:00是潜叶蝇成虫为害高峰期;3月下旬、6月上中旬潜叶蝇/明显升高。     

Increased expression of CD40 ligand by peripheral blood mononuclear cells in patients with systemic lupus erythematosus

AIM:To investigate expression and function of CD40 ligand by peripheral blood mononuclear cells (PBMCs) in patients with systemic lupus erythematosus (SLE).METHODS:Expression of CD40 ligand by PBMCs in patients with SLE and control were examined by flow cytometric analysis before and after stimulated by phytohemagglutinin(PHA)and depressed by Dexamethasone(Dex). The correlation between expression of CD40 ligand and SLE activity index(SLEDAI) was analysed in patients with SLE.RESULTS:The expression of CD40 ligand by PBMCs in patients with active SLE was higher than that in patients with inactive SLE and control. Though the expression of CD40 ligang by PBMCs could be stimulated by PHA in three groups, it was the highest in patients with active SLE. Dex depressed the expression of CD40 ligand by PBMCs significantly in patients with SLE, but not in control. There was high positive correlation between expression of CD40 ligand and SLEDAI in patients with active and inactive SLE.CONCLUSION:Increased expression of CD40L by PBMCs in patients with SLE may play an important role in pathogenesis of SLE.     

Association of polymorphism of Fc receptor γ chain gene at position-29 in promoter with the susceptibility to systemic lupus erythematosus in southern Chinese

AIM:To detect the association between the polymorphism of Fc receptor γ chain gene at position-29 in promoter and systemic lupus erythematosus(SLE).METHODS:The genotypes at position -29 in promoter of Fc receptor γ chain gene were determined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method in 180 patients with SLE and 140 ethnically matched controls in southern China.RESULTS:The frequencies of TT genotype(33.3%) and T allele (54.4%) at position -29 in patients with SLE were significantly higher than those in controls (17.2% and 42.9%, respectively), whereas, the frequencies of GG genotype (24.4%) and G allele (45.6%) in patients with SLE were remarkably lower than those in controls (31.4% and 57.1%, respectively) (P＜0.05). The TT genotype and T allele at position -29 were not associated with lupus nephritis in SLE patients (P＞0.05).CONCLUSION:Our results indicate that the T allele at position -29 in promoter of Fc receptor gene probably contributes to the susceptibility to SLE, but does not play a role in the occurrence of lupus nephritis.     

Using antisense nucleic acid technology to study the influence of POLκ on genetic stability

AIM:To establish cell line FL-POLκ^- and to study the role ofPOLκ(polymerase kappa) on genetic stability.METHODS:A mammalian expression vector expressing antisense POLκ gene fragment pMAMneo -amp^--POLκ was constructed by cloning the 1 690-1 918 fragment of POLκ gene into the mammalian expression vector pMAMneo-amp^- in antisense orientation. FL cells were fransfected with this antisense RNA expressing vector and selected by G418. Based on the shuttle-plasmid pZ189, the mutation assay was made.RESULTS:The spontaneous mutation frequency of supF tRNA gene in the plasmid replicated in the FL- POLκ^- was 11.2×10^-4, while it was 4.9×10^-4 and 3.7×10^-4 in the control cells FL and FL-M, respectively.CONCLUSION: POLκ playes an important role in maintenance of genetic stability.     

‘三棱榄''橄榄果实香气成分分析

1 材料与方法选取广东优良鲜食橄榄品种‘三棱榄’,2001年12月6日采样,采用固相微萃取法(SPME)富集香气成分(鲜橄榄果肉于15℃下捣碎后取样1.0 g放入4 mL聚四氟乙烯硅橡胶垫密封螺口玻璃瓶中,插入100μm聚二甲基硅氧烷纤维头于室温25-30℃顶空取样2 h),用美国Finnigan TRACE GC-MS气相色谱-质谱联用仪进行分析。气相色谱柱为DB-1弹性毛细管柱30 m×0.25 mm,载气为He(99.99％),流速1.0mL/min。程序升温从40℃开始先保持10 min,后以2℃/min的升温速率升至150℃保持10 min。质谱条件:电子能量70 eV,离子源温度250℃,质量范围35-450 aum,不分流进样。2002年12月18日采样重复分析。     

苏云金杆菌小试生产发酵影响因子的研究

利用 1 0L全自动发酵罐对苏云金杆菌小试生产的发酵条件进行了研究。结果显示 ,不同的温度、pH值、培养基成分及接种方式对发酵产量和发酵周期均有影响 ;在培养基配方中存在葡萄糖时 ,葡萄糖单独灭菌对提高苏云金杆菌发酵产量是重要的。初步生物测定显示 ,发酵上清中存在一些杀虫增效因子 ,建议配制苏云金杆菌制剂时尽可能加以利用。     

水稻白叶枯病菌单细胞系毒力分化研究

 以稀释倒平板法从0型菌086和IV型菌967-4和9620中分离到59个单细胞系;在12个近等基因系品种上,086和其单细胞系表现为弱毒力,2个IV型菌及其单细胞系能克服抗病基因Xa-1、2、3、8、10、11、14的抗性,不能克服Xa-21、4、5、7、13的抗性;带主效抗病基因的品种Asominori、XM5、M41、XM6和丰锦能把3个母株的59个单细胞系区分为12种数量差异或质量差异的不同致病型;将此5品种与近等基因系配合,适合作为病菌致病基因变异频度监测的寄主;采用"段叶沙培,切口取菌胶"法分离病菌,在中国致病型鉴定品种上划分的致病型,是田间病菌群体毒力结构的表型反应。     

离体条件下水稻种衣剂对串珠镰刀菌生长及形态毒理影响

 检测了黑龙江水稻主产区4个当地主栽品种的种子内部镰刀菌寄藏情况,测定了20%克福甲和20%克多甲种衣剂对种子带菌消毒处理效果及对水稻串珠镰刀菌(Fusarium moniliforme)的抑菌作用和联合毒力,并借助扫描电镜观察了上述2种混配种衣剂对串珠镰刀菌的形态毒理影响。结果表明,种子内部镰刀菌的分离频率高达56.7%~96.0%,其中串珠镰刀菌的分离频率为32.6%~48.2%,2种种衣剂对带菌种子具有显著的消毒处理效果。20%克福甲和20%克多甲种衣剂对镰刀菌F.moniliforme的毒力指数分别为457.11和802.04,增效倍数分别为6.53和0.13。20%克多甲种衣剂(多菌灵:甲基立枯磷为5:5,W/W)对串珠镰刀菌的抑菌作用优于20%克福甲种衣剂(福美双:甲基立枯磷为8:6,W/W),增效作用低于20%克福甲种衣剂。电镜观察表明,种衣剂低浓度至高浓度处理下均可引起串珠镰刀菌菌丝不同程度的异常生长,表现为主菌丝局部膨大或形成菌丝束,菌丝顶端异常膨大、缢缩或形成穗状和花絮状分枝。     

棉铃虫性诱剂诱捕器的改进及其应用研究

利用废弃的食用油油壶制成棉铃虫诱捕器,可提高诱蛾的效果。挡板距诱捕器器口的距离以3cm的效果最佳。与常规水钵、水盆诱捕器相比较,全年诱蛾总量分别增加116.3％和减少79.8％,但诱蛾灵敏度均高出18.72％。
