Fatty acids have been used in marine biogeochemistry as food chain biomarkers, but in freshwater these studies are rare. In order to evaluate the fatty acid potential as biomarkers in freshwater, their profile was analyzed during vitellogenesis in two fish species, in both waterfall and reservoir environments of the Paraíba do Sul River Basin. Detrivorous Hypostomus affinis and omnivorous Geophagus brasiliensis seem to elongate and desaturate polyunsaturated fatty acids (PUFA) and transfer them to the ovaries’ phospholipids. Waterfall Geophagus brasiliensis have more highly unsaturated fatty acids in the liver, but in the reservoir, accumulation mainly occurs in muscle and ovary triglycerides, suggesting trophic opportunism and a plasticity during vitellogenesis. In Hypostomus affinis, PUFA alteration occurs only in the reservoir, suggesting a high phytoplankton occurrence. Eutrophication and water speed is reflected in Hypostomus affinis ovaries by higher PUFAn3 and bacterial fatty acids. As in marine environments, analysis of mono- and polyunsaturated fatty acids during vitellogenesis can be used as a tool in food chain studies in freshwater.  相似文献

An IgM capture ELISA using heterologous antibodies was developed to evaluate the kinetics of the humoral immune response in dogs experimentally infected with Toxoplasma gondii RH strain. Detection of parasite in tissues from inoculated dogs was evaluated by mouse bioassay and immunohistochemical techniques. Serum samples were obtained at regular intervals up to 62 days post-inoculation (p.i.), when the animals were necropsied and their tissues examined. Antibody levels were measured by IgM capture ELISA (McELISA), indirect hemagglutination (IHA), indirect fluorescent antibody test (IgG-IFAT) and indirect immunoenzymatic assay (IgG-ELISA). All dogs seroconverted but only one exhibited severe clinical signs of infection. IgM antibodies were detected by McELISA from the seventh day on, with decreasing IgM levels around the 27th day. Similar results were obtained from IHA, although McELISA showed earlier and longer detection of IgM antibodies. IgG antibodies were detected from the seventh day on, and throughout the period of observation. Immunohistochemical findings and mouse bioassay revealed the presence of free tachyzoites in tissues of the clinically affected dog only. These results suggest that T. gondii acute infection in dogs shows a remarkably transient IgM synthesis, and this feature may constitute an important marker of active infection. Furthermore, McELISA was shown to be a potential tool to diagnose canine toxoplasmosis.  相似文献

The demand for sustainable and environmentally friendly food sources and food ingredients is increasing, and microalgae are promoted as a sustainable source of essential and bioactive lipids, with high levels of omega-3 fatty acids (ω-3 FA), comparable to those of fish. However, most FA screening studies on algae are scattered or use different methodologies, preventing a true comparison of its content between microalgae. In this work, we used gas-chromatography mass-spectrometry (GC-MS) to characterize the FA profile of seven different commercial microalgae with biotechnological applications (Chlorella vulgaris, Chlorococcum amblystomatis, Scenedesmus obliquus, Tetraselmis chui, Phaeodactylum tricornutum, Spirulina sp., and Nannochloropsis oceanica). Screening for antioxidant activity was also performed to understand the relationship between FA profile and bioactivity. Microalgae exhibited specific FA profiles with a different composition, namely in the ω-3 FA profile, but with species of the same phylum showing similar tendencies. The different lipid extracts showed similar antioxidant activities, but with a low activity of the extracts of Nannochloropsis oceanica. Overall, this study provides a direct comparison of FA profiles between microalgae species, supporting the role of these species as alternative, sustainable, and healthy sources of essential lipids.  相似文献

Mercury bioaccumulation kinetics of two important macrobenthic species, the polychaete Hediste diversicolor and the bivalve Scrobicularia plana, were evaluated following a dietary pathway (i.e. contaminated algae), through a mesocosm laboratory experiment. Both studied species presented a similar model of Hg bioaccumulation kinetics, a linear pattern of accumulation through time being the mercury accumulation in the organisms proportional to the mercury concentration in the food. Mercury bioaccumulation rates were higher in the polychaete H. diversicolor (reaching approximately 0.15 μg g−1 at the end of the experiment) than in the bivalve S. plana (≈0.07 μg g−1), which could be related to their feeding strategies, ingestion rates and assimilation efficiencies. Moreover, the mercury bioaccumulation revealed to be quite a fast process especially for the polychaete, and despite the fact that this species is not an edible organism, it is an important prey item, which can greatly contribute to the transport of contaminants to higher trophic levels. Therefore, the bioaccumulation of mercury by these important macrobenthic species, especially the bivalves, represents a non-negligible risk for humans.  相似文献

Alpha-synuclein is implicated in several neurodegenerative disorders, such as Parkinson's disease and multiple system atrophy, yet its functions remain obscure. When expressed in yeast, alpha-synuclein associated with the plasma membrane in a highly selective manner, before forming cytoplasmic inclusions through a concentration-dependent, nucleated process. Alpha-synuclein inhibited phospholipase D, induced lipid droplet accumulation, and affected vesicle trafficking. This readily manipulable system provides an opportunity to dissect the molecular pathways underlying normal alpha-synuclein biology and the pathogenic consequences of its misfolding.  相似文献

Marine invertebrates contain nutritionally important elements that make them suitable to be included in the human diet, and they are considered an outstanding source of secondary metabolites with multiple biotechnological uses. The sea hare Bursatella leachii is an invasive species in Mar Menor that can be managed through the control of its reproduction and/or by massive biomass removal. In this context, this work evaluated the possible use of biomass from B. leachii in the food and cosmetics industries. Biomass was appraised for nutritional properties, and extracts were prepared and evaluated in vitro for antioxidant and anti-inflammatory capacity, cholinesterase and tyrosinase inhibition, and for total contents in different phenolic groups. It had a moderate moisture level, low ash and fat levels, and an adequate protein amount and protein/lipid ratio. The fatty acids (FA) profile was mainly saturated, dominated by palmitic acid. The methanol extract had the highest acetylcholinesterase inhibition. The acetone extract had the strongest capacity to scavenge 1,1-diphenyl-2 picrylhydrazyl (DPPH) radical, reduce iron, inhibit tyrosinase, decrease nitric oxide production in lipopolysaccharide (LPS)-stimulated microglia cells, and the upmost levels of total phenolics, flavonoids, and tannins. Our results suggest multiple biotechnological applications for B. leachii biomass in different economic areas.  相似文献

Seaweeds are considered healthy and sustainable food. Although their consumption is modest in Western countries, the demand for seaweed in food markets is increasing in Europe. Each seaweed species has unique nutritional and functional features. The preparation of blends, obtained by mixing several seaweeds species, allows the obtaining of maximum benefits and ingredients with single characteristics. In this work, five seaweed blends, commercially available and produced under organic conditions in Europe, were characterized. The proximal composition included contents of ash (20.28–28.68% DW), proteins (17.79–26.61% DW), lipids (0.55–1.50% DW), and total carbohydrates (39.47–47.37% DW). Fatty acid profiles were determined by gas chromatography–mass spectrometry (GC–MS), allowing quantification of healthy fatty acids, namely n-3 and n-6 polyunsaturated fatty acids (PUFA), and calculation of lipid quality indices. Each blend showed a characteristic PUFA content in the lipid pool (35.77–49.43% of total fatty acids) and the content in essential and healthy n-3 PUFA is highlighted. The atherogenicity (0.54–0.72) and thrombogenicity (0.23–0.45) indices evidenced a good nutritional value of lipid fractions. As nutritional and environmentally attractive products, the consumption of the studied seaweed blends can contribute to a healthy lifestyle.  相似文献
