The phylogenetic relationships of five isolates of Pasteurella multocida serotype B:2 belonging to buffalo, cattle, pig, sheep and goat were investigated by comparative sequence analysis of 16S rRNA gene. The 1468bp fragment of 16S rRNA gene sequence comparison showed that the isolates of cattle (PM75), pig (PM49) and sheep (PM82) shared 99.9% homology with the buffalo isolate (vaccine strain P52) whereas, the goat isolate (PM86) shared 99.8% homology with the vaccine strain. The 16S rRNA gene sequences of these isolates were also found monophyletic with type B reference strain NCTC 10323 of P. multocida subsp. multocida. The present study indicated the close relationships of haemorrhagic septicaemia causing P. multocida serotype B:2 isolates of buffalo and cattle with other uncommon hosts (pig, sheep and goat).  相似文献

Cultivar ‘Thatcher’, and ‘Thatcher’ lines with Lr 21 and Lr 22 were studied against a number of races of Puccinia recondita for seedling and adult plant reaction. The study has established that Lr 21 and Lr 22 are genes effective against P. recondita at adult plant stage. It has also shown that these genes confer resistance against all races when plants are inoculated at boot leaf stage.  相似文献

Summary The resistance to cotton jassid in okra was found controlled by dominant genes. Both additive and dominance gene effects were significant but both additive gene effects and dominance x dominance type of interactions appear to be more important than other effects. Sorghum midge [Stenodiplosis sorghicola (Coquillett)] is an important pest of grain sorghum, and host plant resistance is one of the important components for the management of this pest. We studied the inheritance of resistance to this insect involving a diverse array of midge-resistant and midge-susceptible genotypes in India and Kenya. Testers IS 15107, TAM 2566, and DJ 6514, which were highly resistant to sorghum midge in India, showed a greater susceptibility to this insect in Kenya. The maintainer lines ICSB 88019 and ICSB 88020 were highly resistant to sorghum midge in India, but showed a susceptible reaction in Kenya; while ICSB 42 was susceptible at both the locations. General combining ability (GCA) effects for susceptibility to sorghum midge for ICSA 88019 and ICSA 88020 were significant and negative in India, but such effects were non-significant in Kenya. The GCA effects of ICSB 42 for susceptibility to sorghum midge were significant and positive at both the locations. The GCA effects were significant and positive for Swarna, and such effects for IS 15107 and TAM 2566 were negative at both the locations. GCA effect of DJ 6514 were significant and negative in India, but non-significant and positive in Kenya; while those of AF 28 were significant and positive during the 1994 season in India, but significant and negative in Kenya. Inheritance of resistance to sorghum midge is largely governed by additive type of gene action. Testers showing resistance to sorghum midge in India and/or Kenya did not combine with ICSA 88019 and ICSA 88020 to produce midge-resistant hybrids in Kenya. Therefore, it is essential to transfer location specific resistance into both parents to produce midge-resistant hybrids.  相似文献

We have constructed a linkage map of the rice brown planthopper (BPH)resistance gene, Bph1. RFLP and AFLP markers were selected by thebulked segregant analysis and used in the mapping study of 262 F2sthat were derived from a cross of `Tsukushibare', a susceptible japonica cultivar, and `Norin-PL3', an authentic japonicaBph1-introgression line. Twenty markers were mapped within a 28.9-cMregion containing the Bph1 locus on the long arm of rice chromosome12. Combining the result of segregation analysis of BPH resistance by themass seedling test and that of the markers, the Bph1 locus wasmapped within a 5.8-cM region between two flanking markers. The closestAFLP markers, em5814N and em2802N, was at 2.7 cM proximal to theBph1 locus. Together with the previously constructed high-resolutionmap of bph2 locating the locus at ca. 10 cM proximal to the Bph1 locus, this improved version of the linkage map would facilitatepyramiding these two important BPH resistance genes.  相似文献

Summary Soybean germplasm was screened for resistance to bacterial pustule disease. The etiological agent, Xanthomonas campestris pv. glycines, was isolated from the leaves of field grown soybean in Maharashtra, India. The screening of soybean stocks was carried out by excised leaf inoculation method. A differential susceptibility to the pathogen was observed in the tested stocks. Two stocks P-4-2 and P-169-3 were found to be completely resistant to the pathogen and displayed an incompatible reaction. Four cultivars, EC-34160, Bragg, Kalitur and PK-472 displayed moderate resistance and the remaining stocks were susceptible to the attack of the pathogen. The stocks P-4-2 and P-169-3 remained resistant even to a high concentration of 109 colony forming units (cfu)/ml of the pathogen.  相似文献

Summary Sorghum midge, Contarinia sorghicola Coq. (Diptera: Cecidomyiidae) is one of the most important pests of grain sorghum worldwide. We studied the reaction of midge-resistant and midge-susceptible genic-cytoplasmic male-sterile (A-lines) and their maintainers (B-lines), and the effect of resistant and susceptible restorers on sorghum midge. Midge damage and adult emergence were significantly lower on the B-lines of midge-resistant genotypes (PM 7061 and PM 7068) than their corresponding A-lines, while the reverse was true for the midge-susceptible genotypes (296A and ICSA 42). Differences in midge damage and the number of midges emerged were not significant between the midge-resistant and midge-susceptible A-lines when infested without pollination (except midge emergence on PM 7061A). Pollination with a midge-resistant restorer (DJ6541) reduced midge emergence significantly in one of two seasons. Source of pollen did not influence midge emergence on the highly-resistant A-line, PM 7061A. The implications of these observations in the development of midge-resistant hybrids were discussed.  相似文献

Summary The effect of direct vernalization of immature embryos on flowering was studied in six winter wheat genotypes. Fourteen-, 17-, and 20-day-old embryos were excised and vernalized for 0–6 weeks on synthetic medium during a conditioning period. Percent germination of embryos was high (overall 96.1%), and free from genotypic effects. Genotypes differed for flowering in response to cold treatment of excised embryos. Embryo vernalization was as effective as or more than conventional vernalization (control, seedling vernalization for 6 weeks). Seventeen-day-old embryos were the most responsive to vernalization. With a 5-week vernalization of 17-day-old embryos, the percentage of plants anthesed was higher than those from 14-and 20-day-old embryos. For 17-day-old embryos vernalized for 5 weeks, the mean number of days from culture to anthesis was less than that of 6 week vernalization, less than that of 14- and 20-day-old embryos, and less than controls.Purdue Univ., Agronomy Dept., W. Improving the level and stability of grain yield is the primary objective of wheat breeding programs in the Eastern Gangetic Plains (EGP) of South Asia. A regional wheat trial, the Eastern Gangetic Plains Yield Trial (EGPYT), was initiated by CIMMYT in collaboration with national wheat research programs in Bangladesh, Nepal, and India in 1999–2000 to identify wheat genotypes with high and stable grain yield, disease resistance, and superior agronomic traits for the EGP region. A set of 21 wheat experimental genotypes selected from a regional wheat screening nursery in South Asia, three improved widely grown cultivars (Kanchan, PBW343 and Bhrikuti), and one long-term cultivar (Sonalika) were tested at 9–11 sites in six wheat growing seasons (2000–2005) in the EGP. The 21 experimental genotypes were different in each year, whereas the four check cultivars were common. In each year, one or more of the experimental genotypes showed high and stable grain yield and acceptable maturity, plant height, and disease resistance compared to the check cultivars. Three improved cultivars have already been commercially released in the region through EGPYT and many germplasm lines have been used in the breeding programs as parents. Identification of wheat genotypes with high-grain yield in individual sites and high and stable yield across the EGP region underlines their value for regional wheat breeding programs attempting to improve grain yield and agronomic performance.  相似文献
