The antibody response of turkeys experimentally infected with Mycoplasma synoviae was determined by the serum plate agglutination (SPA), hemagglutination-inhibition (HI), and microagglutination (MA) tests and the enzyme-linked immunosorbent assay (ELISA). No antibody response was detected until 2 weeks postinfection (PI) with the MA test (17% positive), 3 weeks PI with the SPA test (11% positive), 4 weeks with the HI test (21% positive), and 5 weeks PI with the ELISA, and even then, only 16% of the birds were positive. Although at least 89% of the birds were positive by culture, only 58% of the turkeys developed a detectable antibody response.  相似文献

Of the non-ruminant wildlife species known to harbor Mycobacterium avium paratuberculosis (MAP), the rabbit (Oryctolagus cuniculus) is thought to pose the greatest risk of transmission to cattle. We analyzed 80 hunter-harvested wild rabbits from a core study area in southern Spain, and sera from 157 wild rabbits sampled opportunistically on seven additional sites. Gross lesions compatible with paratuberculosis were observed in two of 80 necropsied rabbits. Histopathology revealed focal to diffuse multibacillary MAP-compatible lesions in 8 of 10 rabbits examined. Presence of MAP was confirmed in one rabbit with gross lesions by positive amplification curves for both IS900 and ISMAP02. However, no isolate was obtained from 47 samples by culture. We adapted an indirect ELISA for the detection of MAP antibodies. At the established cut-off of 0.5, 6 of 237 wild rabbit sera (2.5%) yielded a positive ELISA result. Antibodies were detected in rabbits from 3 of 8 sampling sites. Considering the increasing relevance of MAP infection for animal health, these results open a challenging field for future research.  相似文献

Red deer (Cervus elaphus) have a pronounced seasonality in their physiology. The aim of this study was to assess the effect of the season on red deer responsiveness to skin testing with the phytohaemagglutinin (PHA) mitogen. Study subjects included 270 farmed adult red deer (19 stags and 251 hinds). The skin testing was carried out between January 2009 and August 2010. The animals were injected intradermally with a 0.1 ml volume containing 250 μg of PHA diluted in phosphate buffered saline. The skinfold thickness was measured immediately prior to injection and 72 h after administration, always by the same person and with three repeats per measurement. Single effects of sex and time on skin test responsiveness were significant (p < 0.001) as well as their interaction (p < 0.001). In winter (January), and considering the average of two years, the skinfold increase in response to the intradermal injection of 250 μg PHA was 2.1 times larger in stags and 1.4 times in hinds than in summer (August). While stags had 1.3 times larger responses than hinds in winter, the inverse occurred in summer, with 1.1 times larger responses in hinds. We also evidenced a limited inter-annual variation of skinfold increase in response to PHA in red deer. These findings have important consequences regarding the interpretation of skin test results in the ante-mortem diagnosis of tuberculosis and paratuberculosis, at least in deer.  相似文献

Photodegradation has been shown to play a role in plant litter degradation in some ecosystems, and therefore potentially in the global carbon cycle. To introduce photodegradation into models of carbon turnover we need an equation that relates mass loss to incident radiation. Based on experimental data from the literature we developed a linear equation for photodegradation as a function of light exposure. We also scaled up the results to global scale, based on global data on incident radiation and shading by plant cover. The results indicate that photodegradation plays a part in semi-arid environments and potentially some arctic and alpine environment. However, the percentage of global plant production decomposed by photodegradation is estimated to be less than 1% (range 0.5-1.6). Photodegradation is therefore important locally, but probably not very important for the global carbon budget.  相似文献

A protozoological survey was performed on waste stabilization lagoons to isolate, identify, and determine the incidence of zooflagellates present in these waters. A correlation between the species found and the physicochemical parameters determined was also done. Fifteen samplings were analyzed from June 1981 to April 1982. Free-living zooflagellates included: Bodo edax Klebs: Bodo caudatus Dujardin;Trepomonas agilis Dujardin;Pleuromonas jaculans Perty (all of them polisaprobic indicators);Bodo saltans Ehrenberg (a mesosaprobic indicator); and Bodomorpha minima Hollande. A human commensal species Enteromonas hominis Da — Fonseca, was also isolated. The performance of the lagoons was predominantly anaerobic. The temperature seemed to be the most influential factor on the incidence of the zooflagellates isolated, especially in winter time.  相似文献

Because of the small thermal capacity of ultrathin ( approximately 200 nanometers) metal single crystals, it is possible to explore the coupling of catalytic and thermal action at low pressures. We analyzed a chemothermomechanical instability in this regime, in which catalytic reaction kinetics interact with heat transfer and mechanical buckling to create oscillations. These interacting components are separated and explored through experimentation, mathematical modeling, and scientific computation, and an explanation of the phenomenon emerges from their synthesis.  相似文献

Purpose

The sensitivity of soil organic carbon to global change drivers, according to the depth profile, is receiving increasing attention because of its importance in the global carbon cycle and its potential feedback to climate change. A better knowledge of the vertical distribution of SOC and its controlling factors—the aim of this study—will help scientists predict the consequences of global change.

Materials and methods

The study area was the Murcia Province (S.E. Spain) under semiarid Mediterranean conditions. The database used consists of 312 soil profiles collected in a systematic grid, each 12 km2 covering a total area of 11,004 km2. Statistical analysis to study the relationships between SOC concentration and control factors in different soil use scenarios was conducted at fixed depths of 0–20, 20–40, 40–60, and 60–100 cm.

Results and discussion

SOC concentration in the top 40 cm ranged between 6.1 and 31.5 g?kg?1, with significant differences according to land use, soil type and lithology, while below this depth, no differences were observed (SOC concentration 2.1–6.8 g?kg?1). The ANOVA showed that land use was the most important factor controlling SOC concentration in the 0–40 cm depth. Significant differences were found in the relative importance of environmental and textural factors according to land use and soil depth. In forestland, mean annual precipitation and texture were the main predictors of SOC, while in cropland and shrubland, the main predictors were mean annual temperature and lithology. Total SOC stored in the top 1 m in the region was about 79 Tg with a low mean density of 7.18 kg?Cm?3. The vertical distribution of SOC was shallower in forestland and deeper in cropland. A reduction in rainfall would lead to SOC decrease in forestland and shrubland, and an increase of mean annual temperature would adversely affect SOC in croplands and shrubland. With increasing depth, the relative importance of climatic factors decreases and texture becomes more important in controlling SOC in all land uses.

Conclusions

Due to climate change, impacts will be much greater in surface SOC, the strategies for C sequestration should be focused on subsoil sequestration, which was hindered in forestland due to bedrock limitations to soil depth. In these conditions, sequestration in cropland through appropriate management practices is recommended.  相似文献

Potato (Solatium tuberosum) generally requires high amounts of phosphate fertilizer to reach economically acceptable yields, particularly in soils originating from volcanic ash. This is a consequence of the potato plants low root density and the slow soil diffusion rate of phosphorus (P) in these soils. Our objective was to evaluate the effect of P rates on tuber yield, biomass production, and distribution, biomass P accumulation and concentration, and P distribution in potato cv. Mexiquense. The experiment was carried out in an Andisol (7.8 μg g‐1 Olsen‐P) located at the east of Valle de México. Fertilization rates were 0, 18,41,46,69,78,90,106,113,135,150,163, and 207 kg ha‐1 P, from ordinary superphosphate. Top growth and root biomass, tuber yield, P percentage and P accumulation in different plant parts were measured at harvest. Minimum and maximum average tuber yields were 8.4 and 18.0 Mg ha‐1; the plants absorbed 5.8 and 11.8 kg ha‐1 P, corresponding to 0 (control) and 207 kg ha‐1 P, respectively. Phosphorus fertilization had little influence on plant P concentration, where average concentrations in tuber and top growth were 0.20 and 0.24 % P, respectively. By contrast, P accumulation increased with increasing P rates, but P distribution between tuber and top growth was dependent on the amount of P applied. The control treatment showed approximately 1:1 distribution of P between top growth and tuber, but as P rate increased, top growth P decreased and tuber P increased. When applying the highest P rate, 36% of P accumulated in the top growth and 64 % in the tuber. The information obtained will permit decisions on the correct use of phosphate fertilizer for potato in Andisols of the Valle de Mexico.  相似文献

This paper describes findings in the characterization of 300 accessions in a collection of common beans (Phaseolus vulgaris L.) in Honduras. The plant material was collected from 1990 to 1994 (4 years before the damaging Mitch Hurricane) covering most of the Departments of the country and kept at the Escuela Agrícola Panamericana Zamorano. Thirty-two morpho-agronomical characters were evaluated and the results were grouped by classes. The studied accessions showed a marked diversity with high predominance of red and small seeds. The accessions with purple flowers had black seeds and originate mainly from the western area of the country where this type of seed is preferred. Principal Component Analysis shows that clearly defined groups do not exist. The conservation of this diversity is recommended for future propagation, breeding and the investigation of the genetic relationships and other studies.  相似文献

Iranian wheat landrace accessions (IWAs) were collected from country‐wide farm fields and market places in 1935 by a professor at the University of Tehran and shared with University of California at Davis, California. IWAs were further submitted to the genebank of International Maize and Wheat Improvement Center (CIMMYT), Mexico. 2,403 IWAs from CIMMYT’s genebank were assayed by DArT‐seq technology to assess genetic diversity. No apparent ecogeographic patterns related to genetic diversity were detected, probably due to long‐term transport and frequent interchange of landraces among farmers. A multivariate clustering procedure combining genotypic and phenotypic information was used in selecting a core‐set, which represented 15% of the hexaploid wheat accessions included in this study. This subset captured an estimated 93% of rare (frequency <0.05) alleles. Multisite phenotypic data (India, Mexico) validated the ability of the core‐set in detecting useful variants. Potential donor accessions for multiple traits (disease resistance, zinc concentration) were identified from the core‐set for wheat‐breeding. This report illustrates a breeder friendly robust core‐set formulation strategy for harnessing the useful genetic variation stored in the genebanks.  相似文献
