This study investigated the effects of low potassium diets with different levels of Ca compared to two diets low in dietary cation–anion difference (DCAD) fed prepartum as a strategy to prevent hypocalcemia on sorting behaviour, total tract digestibility, oxidative status and energy and protein metabolism of transition cows. Forty-eight pregnant dairy cows were assigned to 4 treatment groups: Low Ca, low K (LCLK), High Ca, low K (HCLK), Supplementation with anionic mineral mixture (AMS) supplementation with SoyChlor (CAS). After parturition, all animals were fed a standard postpartum diet. Data were collected until 21 DIM. Prepartum urinary pH was significantly reduced by the low DCAD diets, while postpartum Ca homeostasis was affected by the HCLK ration. Feeding AMS induced sorting against particles <1.18 mm in favour of particles >19 mm prepartum. In contrast, cows fed CAS showed an increase in selective consumption of fine particles and sorted against longer particles similar to the HCLK and LCLK groups. Postpartum sorting activity was not affected by the dietary treatments. After calving, apparent digestibility of NDF was significantly reduced in the HCLK group. Prepartum, we observed effects on serum concentrations of non-esterified fatty acids were higher and insulin sensitivity was lower in the AMS group. Blood urea nitrogen (BUN) was decreased in cows fed the CAS ration. Postpartum, we found serum protein to be decreased with the low DCAD diets while BUN was decreased in the CAS group. The low DCAD rations increased prepartum serum malondialdehyde concentrations, while postpartum total antioxidant capacity was lower in the HCLK and the AMS group. From these data, we conclude that AMS decreased prepartum intake due to compromised palatability. Intermediate protein metabolism was affected by the low DCAD diets, while parameters of oxidative stress were probably affected by acid–base balance and Ca homeostasis.  相似文献

Theileria equi (T. equi) is an obligate intra- and extra-erythrocytic parasite that causes equine theileriosis (ET) in equids. Equine theileriosis is considered a notifiable disease of global significance, a major constraint to the international movement of horses, and endemic in many countries. This disease may be difficult to diagnose, as it can produce variable and nonspecific clinical signs. A cross-sectional study was designed for the molecular characterisation of T. equi and to investigate the associated risk factors of ET accompanied by its consequences on haematological and sero-biochemical parameters. A convenience sampling of 500 blood samples were collected from ET suspect horses from January to December 2017. PCR was performed on all blood samples targeting the 18S rRNA gene of T. equi followed by sequencing; 9% animals tested positive with confirmed sequences. The isolates of this study showed high homology with Cuban, Russian and Brazilian isolates of T. equi (accession numbers KY111762.2 , MG551915.1 and KY952237.1 , respectively). Based on multivariate analysis, the principal risk factors consisted of absence of dogs on the premises and presence of tick infestation. The haemato-biochemical parameters showed a decrease in granulocytes and erythrocytes, and an increase in lymphocytes, monocytes, mean corpuscular volume, mean corpuscular haemoglobin, mean platelet volume, glucose, phosphorus and aspartate aminotransferase in positive horses. This is the first study which identified ET in Punjab (Pakistan) using molecular techniques and risk factors together with the haemato-biochemical variations in horses.  相似文献

The objective of this study was to evaluate the effect of collagenase and two final dimethylformamide (DMF) concentrations (4% and 7%) on alpaca frozen-thawed sperm quality. A total of 25 ejaculates from 5 alpaca were obtained using electroejaculation. Each individual ejaculate was evaluated and then diluted 4:1 in a solution of 1 mg/ml collagenase in HEPES-TALP medium and incubated for 4 min at 37°C. Subsequently, samples were diluted in TRIS-fructose-citric acid-egg yolk and cooled to 5°C. Then, each sample was divided in two aliquots and DMF at final concentration of 4% or 7% was added, equilibrated for 1 hr at 5°C and frozen over liquid nitrogen vapours. A Kruskal–Wallis test was used to evaluate the sperm morphometry, and Completely Random Block designs were used to analyse sperm motility, viability, membrane function and acrosome status. After collagenase incubation, none of the samples showed thread formation, and sperm parameters were preserved. Non-progressive motile sperm were higher (p < .05) in equilibrated samples (4% DMF: 31.8 ± 8.3% and 7% DMF: 36.3 ± 11.8%) compared to raw (10.1 ± 4.3%) and frozen-thawed semen (4% DMF: 9.7 ± 1.8% and 7% DMF: 7.5 ± 3.2%). Sperm membrane function, membrane integrity and intact acrosomes were higher (p < .05) in raw semen (40.1 ± 12.2%, 94.6 ± 3.2% and 91.3 ± 8.1%) compared to frozen-thawed samples (4% DMF: 19.8 ± 4.7%, 53.2 ± 2.7%, 65.7 ± 8.7% and 7% DMF: 20.4 ± 4.5%, 54.1 ± 1.4%, 64.6 ± 9.1%). Length of the sperm head was lower in frozen-thawed samples, being statistically different with 4% DMF compared to pre-freezing samples. The ratio between acrosome and head areas was greater (p < .05) in frozen-thawed samples. Incubation of raw alpaca semen with collagenase decreased the thread formation without affecting sperm quality. Frozen of collagenase treated alpaca semen with 4% or 7% DMF did not preserve the sperm parameters in thawed samples.  相似文献

In this study the starch digestion rates in broiler chickens from 18 samples of 5 commonly used feed grains (sorghum, wheat, maize, barley, triticale) were determined. The methodology to determine starch digestion rates in poultry is detailed herein. Starch digestion rates were not significantly different (P = 0.128) across the 18 feed grains, which reflects the wide variations that were observed within a given feedstuff. Nevertheless, starch digestion rates in broiler chickens offered wheat-based diets were significantly more rapid by 56.0% (0.117 versus 0.075 min−1; P = 0.012) than their sorghum-based counterparts on the basis of a pair-wise comparison. In descending order, the following starch digestion rates were observed: wheat (0.117 min−1), barley (0.104 min−1), triticale (0.093 min−1), maize (0.086 min−1), sorghum (0.075 min−1). The implications of these findings are discussed as they almost certainly have implications for poultry nutrition and the development of reduced crude protein diets for broiler chickens.  相似文献

Accurate and cost‐effective mapping of karst rocky desertification (KRD) is still a challenge at the regional and national scale. Visual interpretation has been utilised in the majority of studies, while an automated method based on pixel data has been investigated repeatedly. An object‐based method coupling with support vector machine (SVM) was developed and tested using Enhanced Thematic Mapper Plus (ETM+) images from three selected counties (Liujiang, Changshun and Zhenyuan) with different karst landscapes in SW China. The method supports a strategy of defining a mapping unit. It combined ETM+ images and ancillary data including elevation, slope and Normalized Difference Vegetation Index images. A sequence of scale parameters estimation, image segmentation, training data sampling, SVM parameters tuning and object classification was performed to achieve the mapping. A quantitative and semi‐automated approach was used to estimate scale parameters for segmenting an object at an optimal scale. We calculated the sum of area‐weighted standard deviation (WS), rate of change for WS, local variance (LV) and rate of change for LV at each scale level, and the threshold of the aforementioned index that indicated the optimal segment level and merge level. The KRD classification results had overall accuracies of 85·50, 84·00 and 84·86 per cent for Liujiang, Changshun and Zhenyuan, respectively, and kappa coefficients are up to 0·8062, 0·7917 and 0·8083, respectively. This approach mapped six classes of KRD and offered a visually appealing presentation. Moreover, it proposed a conceptual and size‐variable object from the classification standard of KRD. The feasibility of hyperspectral imaging (HSI) to detect deoxynivalenol (DON) content and Fusarium damage in single oat kernels was investigated. Hyperspectral images of oat kernels from a Fusarium‐inoculated nursery were used after visual classification as asymptomatic, mildly damaged, and severely damaged. Uninoculated kernels were included as controls. The average spectrum from each kernel was paired with the reference DON value for the same kernel, and a calibration model was fitted by partial least squares regression (PLSR). To correct for the skewed distribution of DON values and avoid nonlinearities in the model, the DON values were transformed as DON* = [log(DON)]3. The model was optimized by cross‐validation, and its prediction performance was validated by predicting DON* values for a separate set of validation kernels. The PLSR model and linear discriminant analysis classification were further used on single‐pixel spectra to investigate the spatial distribution of infection in the kernels. There were clear differences between the kernel classes. The first component separated the uninoculated and asymptomatic from the severely damaged kernels. Infected kernels showed higher intensities at 1,925, 2,070, and 2,140 nm, whereas noninfected kernels were dominated by signals at 1,400, 1,626, and 1,850 nm. The DON* values of the validation kernels were estimated by using their average spectra, and the correlation (R) between predicted and measured DON* was 0.8. Our results show that HSI has great potential in detecting Fusarium damage and predicting DON in oats, but it needs more work to develop a model for routine application.  相似文献

All forms of baking and processing cause a loss of nutrients, including vitamin E, but little is known about these occurrences or if they could be avoided. The objective of this research was to study the incorporation of palm oil and the stability of vitamin E in palm oil during breadmaking. Wheat meal and rye breads were baked with and without the addition of 0, 2, 5, or 8% palm oil. The eight E group vitamers (tocopherols and tocotrienols) were extracted by using accelerated solvent extraction, freeze dried, and then analyzed with normal‐phase HPLC. Compared with the controls, the inclusion of palm oil was found to increase the quantity of all forms of vitamin E in the final baked products. It is concluded that palm oil is effective in increasing the vitamin E content of whole grain bread.  相似文献
