We demonstrated that Demodex canis was transferred to skin xenografts of a dog and a hamster onto severe combined immunodeficiency mice. After the transfer of mites, the number of eggs, larvae, nymphs and adult mites per gram of canine and hamster xenografts increased, whereas no live mites were detected on murine allograft. These results indicate that D. canis proliferates in hair follicles of dog and hamster skins but not in murine allograft. Therefore, D. canis may have host preference but not strict host-specificity. 相似文献

To assess the permeability of mouse oocytes and embryos, matured oocytes and embryos at various stages of development were placed in five cryoprotectant solutions at 25 C for 25 min. From the cross-sectional areas of the oocytes/embryos, the relative change in volume was analyzed. In oocytes, shrinkage was least extensive and recovery was quickest in the propylene glycol solution, showing that propylene glycol permeates the oocytes most rapidly. Dimethyl sulfoxide, acetamide, and ethylene glycol permeated the oocytes slightly more slowly than propylene glycol. The oocytes in glycerol shrunk extensively and then expanded marginally, indicating slow permeation. The volume changes of 1-cell and 2-cell embryos were similar to those of oocytes, showing little change in permeability. In 8-cell embryos, the volume recovered much faster than in the earlier stages especially in glycerol and acetamide. In morulae, the volume recovery was much faster in glycerol and in ethylene glycol; in ethylene glycol, the extent of shrinkage was small and the recovery was fast, indicating an extremely rapid permeation. Although the permeability of oocytes/embryos generally increased as embryo development proceeded, the degree of increase varied greatly among the cryoprotectants. Interestingly, the volume change in propylene glycol was virtually unaffected by the stage of development. Such information will be valuable for determining a suitable protocol for the cryopreservation of oocytes/embryos at different stages of development. 相似文献

Blastocystis infection in amphibians was surveyed in three species of anuran and one species of urodele amphibians captured at two distinct locations in Japan. All three species of frogs were highly infected with Blastocystis, while 69 individual urodele newts, Cynopus pyrrhogaster, were negative for infection. Eleven Blastocystis isolates (47.8%) were recovered from 23 Rana nigromaculata leopard frogs. Twenty-three (92%) of 25 Rana catesbeiana bullfrogs and all (100%) of 24 Bufo japonicus japonicus toads were positive for Blastocystis. Two distinct populations of the toad and bullfrog showed a high prevalence (100 or 84.6%) of Blastocystis infection, while in two populations of the leopard frog only one population was positive for Blastocystis (84.6%). Three Blastocystis isolates from different species of the frogs were established. Since none of the three isolates could survive at 37 degrees C, a temperature tolerance assay was performed to assess the optimal growth temperature and to determine the range of non-lethal temperatures. During the exponential growth phase of 3- or 4-day cultures at 25 degrees C, three isolates were exposed to 4, 28, 31, or 34 degrees C for 3 days and then returned to 25 degrees C to monitor the cell growth. Based on the optimal growth temperatures and different ranges of temperature tolerance among the three new isolates from frogs and two known species, Blastocystis hominis and Blastocystis lapemi, it was established that the three isolates recovered from different species of frogs had different physiological features from B. hominis and B. lapemi. 相似文献

C/O specific pathogen-free White Leghorn chickens were intracerebrally inoculated at one day of age with a brain homogenate of Japanese bantams (Gallus gallus domesticus) affected with fowl glioma. Histologically, six of eight inoculated chickens developed nonsuppurative meningoencephalitis in cerebrum and two of them had the characteristic lesions of fowl glioma. Hyperplastic lymphoid foci concomitantly developed in many organs of these birds, especially in the heart. Apart from these lymphoid foci, lymphocytic myocarditis was observed in all inoculated birds. Matrix inclusions were also noted in myocardial cells. Immunohistochemically, avian leukosis virus antigens were detected in reticular cells in the lymphoid foci, mesangial cells of the kidney, smooth muscle cells of the blood vessels, and myocardial cells. Of these tissues, the myocardium of all inoculated birds consistently showed strong reactivity for this antigens. The matrix inclusions were also positive for the antigens. These results suggest that the causal virus of fowl glioma has a high propensity to replicate, especially in myocardium and nonsuppurative myocarditis occurs associated with so-called fowl glioma. 相似文献

In 2001, a bacterial rot of onion (Allium cepa L.) bulbs was observed in Japan. The causal agent was identified as Bukholderia cepacia (Palleroni & Holmes 1981 ex Burhkolder 1950) Yabuuchi, Kosako, Oyaizu, Yano, Hotta, Ezaki, and Arakawa 1993. The identified bacteria were divided into two groups (Y and W) based on colony colors, and several phenotypic and genetic characteristics. Based on recA polymerase chain reaction assays, the strains of the Y and W groups belong to genomovar I (B. cepacia sensu stricto) and genomovar III (B. cenocepacia), respectively.The nucleotide sequence data reported are available in the DDBJ/EMBL/GenBank databases under the accession numbers AB162427 and AB162428 相似文献

We investigated the effect of myostatin on the differentiation of bovine preadipocyte. Stromal-vascular cells containing preadipocytes were prepared from perirenal adipose tissue of approximately 30-month-old Japanese Black steers. After confluence, the differentiation was induced by 1-methyl-3-isobutyl-xanthine, dexamethasone, insulin, and troglitasone for 2 days, and then subsequently cultured for 6 days. The cells were treated with myostatin during the induction of differentiation (the early phase of differentiation) or throughout the differentiation period. We measured the terminal differentiation markers such as glycerol-3-phosphate dehydrogenase activity, lipid accumulation, and the expression of adipocyte fatty acid-binding protein mRNA at the end of cultures. The treatment with myostatin throughout the differentiation period severely suppressed the induction of all differentiation markers. The treatment with myostatin in the early phase of differentiation also suppressed the induction of terminal differentiation markers but three-fold higher dose of myostatin was required for the suppression compared with its treatment throughout the differentiation period. Myostatin treatment reduced the expression of peroxisome proliferator-activated receptor (PPAR) gamma mRNA and interfered with the induction of CCAAT/enhancer binding protein (C/EBP) alpha mRNA. We also observed that follistatin stimulates preadipocyte differentiation in the presence of myostatin. These results suggest that myostatin inhibits bovine preadiopocyte differentiation through suppressing PPARgamma and C/EBPalpha mRNA expressions and that follistatin counteracts the suppressive effect of myostatin. 相似文献

Zinc (Zn) is an essential micronutrient with numerous cellular functions in plants, and its deficiency represents one of the most serious problems in human nutrition worldwide. Zn deficiency causes a decrease in plant growth and yield. On the other hand, Zn could be toxic if excess amounts are accumulated. Therefore, the uptake and transport of Zn must be strictly regulated. In this review, the dominant fluxes of Zn in soil?Croot?Cshoot translocation in rice plants (Oryza sativa) are described, including Zn uptake from soils in the form of Zn2+ and Zn-DMA at the root surface, and Zn translocation to shoots. Knowledge of these fluxes could be helpful to formulate genetic and physiologic strategies to address the widespread problem of Zn-limited crop growth. 相似文献
