Impacts of Bolbophorus damnificus (Digenea: Bolbophoridae) on Production Characteristics of Channel Catfish,Ictalurus punctatus,Raised in Experimental Ponds

Bolbophorus damnificus was recognized as a threat to channel catfish, Ictalurus punctatus, production in Louisiana and Mississippi in 1997 and 1999, respectively. Severe to moderate outbreaks (≥34% prevalence) are correlated to heavy losses, but impact of mild outbreaks with low prevalence (<33%) was unknown. Laboratory and field studies were conducted to evaluate sublethal acute (early stage) and chronic (late stage) trematode infection impacts on catfish production. Fish raised in ponds seeded with ram's horn snails from ponds harboring active B. damnificus outbreaks resulted in infection prevalence ranging from 1.6 to 20.4%. Compared to fish raised in ponds not seeded with infected snails (0% prevalence), fish biomass, number harvested, and total feed fed were significantly lower. These data agree with previous studies where mild infections reduced feed consumption, fish production, and economic returns. Infected fish were collected from ponds and used in a laboratory growth study. Once removed from the infection source, weight gain, growth rate, feed consumption, and feed efficiency were equal to or greater than trematode‐free fish. This suggests that although early B. damnificus infection (premetacercarial development) can severely impact production, infection has little impact on performance once metacercarial cysts are fully developed, and the infection source is removed.     

Radiosurgery using a stereotactic headframe system for irradiation of brain tumors in dogs.

Radiation therapy of brain tumors in dogs typically involves administration of multiple fractions over several weeks. Fractionation is used to minimize damage to normal tissue. Radiosurgery uses multiple non-coplanar stereotactically focused beams of radiation in a series of arcs to deliver a single dose to the target with extreme accuracy. The large number of beams facilitates a high degree of conformation between the treatment area and the target tumor and allows for a steep dose gradient; the use of nonintersecting arcs minimizes exposure of normal tissue. Computed tomography with a stereotactic localizer secured to the skull allows generation of a 3-dimensional image of the target and provides accurate spatial coordinates for computerized treatment planning and delivery. Three dogs were treated with radiosurgery, using 1,000 to 1,500 cGy. A linear accelerator mounted on a rotating gantry was used to generate and deliver the radiation. Two dogs with meningiomas survived 227 and 56 weeks after radiosurgery. A dog with an oligodendroglioma survived 66 weeks. No complications were observed following the use of this technique.     

Monoclonal antibodies to Leptospira interrogans serovar pomona.

Three monoclonal antibodies produced against Leptospira interrogans serovar pomona have been studied for their diagnostic usefulness. All three monoclonals reacted strongly in the enzyme-linked immunosorbent assay and indirect fluorescent antibody test with serovar pomona and did not react with serovars grippotyphosa, canicola, icterohaemorrhagiae and hardjo.

Summary The potential of isozymes for distinguishing asparagus varieties was carried out by a survey on 21 varieties using 10 enzyme systems: GOT, SkDH, DIA, PGM, MDH, IDH, PGD, ACP, PGI, MR and ADH. Only 3 enzymes, SkDH, GOT and PGM, showed useful polymorphisms. The varieties were found heterogeneous according to their genetic structure: open pollinated varieties were more heterogeneous than clonal hybrids; the F1 hybrid and the vitroclones were homogeneous. As expected from the narrow genetic basis of the varieties, only a few alleles per isozyme locus were present. Moreover, for each enzyme, one allele or type was predominant so that the discriminating power of the method was low. Pre-extraction preparation (fresh, frozen, freeze-dried, or acetone powdered) and long-term storage of fruit and vegetable tissues: effects on antioxidant enzyme activity

Activities of the antioxidant enzymes ascorbate peroxidase, catalase, dehydroascorbate reductase, glutathione reductase, guaiacol peroxidase, monodehydroascorbate reductase, and superoxide dismutase were assayed in honeydew (Cucumis melo L.) fruit and spinach (Spinacia oleracea L.) leaves either as fresh, frozen to -80 degrees C, frozen in liquid nitrogen, freeze-dried, or acetone powder, representing the various ways tissues are treated prior to enzyme extraction. Treated tissues were analyzed following treatment or stored for up to 8 weeks at -80 degrees C. Enzyme activities in fruit frozen with or without liquid nitrogen and leaves frozen with or without liquid nitrogen or freeze-dried were equal to those of fresh tissue. Enzyme activities in freeze-dried or acetone-powdered fruit and leaves and in acetone-powdered tissues were significantly higher or lower than those in fresh tissue. Enzyme activities in both tissues frozen with or without liquid nitrogen and stored for 8 weeks at -80 degrees C changed little; those in freeze-dried and acetone-powdered tissues, however, significantly increased/decreased over the same period. Fresh tissue should be used in antioxidant enzyme assays, but if storage is necessary, tissues should be placed directly into a -80 degrees C freezer.     

Heterologous expression of excitability proteins: route to more specific drugs?

Many clinically important drugs act on the intrinsic membrane proteins (ion channels, receptors, and ion pumps) that control cell excitability. A major goal of pharmacology has been to develop drugs that are more specific for a particular subtype of excitability molecule. DNA cloning has revealed that many excitability proteins are encoded by multigene families and that the diversity of previously recognized pharmacological subtypes is matched, and probably surpassed, by the diversity of messenger RNAs that encode excitability molecules. In general, the diverse subtypes retain their properties when the excitability proteins are expressed in foreign cells such as oocytes and mammalian cell lines. Such heterologous expression may therefore become a tool for testing drugs against specific subtypes. In a systematic research program to exploit this possibility, major considerations include alternative processing of messenger RNA for excitability proteins, coupling to second-messenger systems, and expression of enough protein to provide material for structural studies.     

Ethanol inhibition of vitamin A metabolism in the testes: possible mechanism for sterility in alcoholics

Vitanin A (retinol) is essential for spermatogenesis. Alcohol dehydrogenase, the enzyme responsible for ethanol metabolism, is also required for the conversion of retinol to bioactive retinal at the end organ site. Ethanol inhibits the oxidation of retinol by testicular homogenates containing alcohol dehydrogenase. Thus, a possible biochemnical mechanism for the sterility of chronic alcoholics is identified.     

Calicivirus isolation and persistence in a pygmy chimpanzee (Pan paniscus)

What may be the first calicivirus isolate from any primate species, including man, was recovered from a herpesvirus-like lip lesion on a pygmy chimpanzee and then, 6 months later, from the throat of the same animal. The infected individual and its cage mates had circulating antibodies that were type-specific for this calicivirus. The agent was antigenically different from 30 other calicivirus serotypes and is tentatively designated primate calicivirus Pan paniscus type 1 (PCV-Pan 1).     

Oroville earthquakes: normal faulting in the sierra nevada foothills

Aftershocks of the Oroville, California, earthquake of 1 August 1975 define a 16- by 12-kilometer fault plane striking north-south and dipping 60 degrees to the west to a depth of 10 kilometers. Focal mechanisms from P-wave first motions indicate normal faulting with the western, Great Valley side downdropped relative to the Sierra Nevada block. The northward projection of the fault plane passes beneath Oroville Dam and crops out under the reservoir.
