Aeromonas salmonicida was isolated from ulcerations in common dab (Limanda limanda). An experiment was performed to pinpoint its role in ulceration development, considering the importance of the skin barrier and the pigmented and non-pigmented sides. The skin of dab was treated in three zones, one where scales and epidermis were removed, one where mucus was discarded and one non-treated zone. Fish were tagged to allow individual identification and challenged with A. salmonicida. Mortality and severity of the developing lesions were recorded for 21 days post-inoculation. Starting 12 days post-inoculation, mortality occurred gradually in challenged fish; however, no direct cause could be established. Both control fish and challenged fish developed ulcerations containing A. salmonicida. Sequencing of vapA gene revealed that isolates retrieved from both groups were distinct, suggesting the presence of A. salmonicida prior to the trial. Most ulcerations developed in zones where skin was removed, suggesting that abrasion might be a predisposing factor in ulceration development. Ulcerations were also observed at the insertion site of the tag, where exposed muscle tissue might have favoured the development of ulcerations. In conclusion, A. salmonicida seems to be involved in the development of skin ulcerations in dab, although the exact pathogenesis needs to be elucidated.  相似文献

Mechanical weed control in low competitive, organic vegetable production systems is challenging, particularly in fields with large populations of Galinsoga spp. (Asteraceae). Various false seedbed techniques are used prior to crop planting or sowing to prevent weed emergence, albeit with variable success. This study investigated the impact of machinery type (flamer, hoe and harrow), number of passes (2 and 4), tillage depth (1–4 cm) and intensity (double and single hoeing, and hoeing with or without additional harrowing) on weed emergence and seedbank density in 0–5 cm topsoil of organic vegetable fields. False seedbed machinery that did not or minimally disturb the soil was most appropriate for preventive control of Galinsoga quadriradiata (Hairy galinsoga) and total weed seeds, with reductions in seedling emergence up to 99% and 73%, respectively, for flaming, and 74% and 67%, respectively, for 1 cm deep hoeing, 1 month after false seedbed creation. Compared with 1 cm deep hoeing, 1 cm deep harrowing was 16% less effective in the control of emerged seedlings, while flaming was highly effective in preventing weed seedling emergence, even after a low number of passes. Tillage intensity was less important than tillage depth for the reduction in weed emergence and seedbank density. Overall, tillage was more effective for seedbank reduction than flaming.  相似文献

Context

Mapping landscape connectivity across large spatial extents is an important component of ecological reserve network designs and species recovery plans. It can, however, be limited by computational power. One way to overcome this problem is to split the study area into smaller tiles, map landscape connectivity within each of those tiles, and then merge tiles back together to form composite connectivity maps.

Objectives

We tested the effects of landscape structure on the accuracy of composite landscape connectivity maps created from tiles and tested two methods to increase this accuracy.

Methods

We correlated replicate, composite current density maps with untiled maps. We tested whether our findings depended on the composition of the landscape by testing maps with corridors, barriers, different mixtures of high- and low-cost habitat, and road networks.

Results

We found that composite current density maps underestimated large-scale connectivity and overestimated the contribution of small habitat patches to overall connectivity. These biases became more pronounced as the tiles became relatively smaller. Landscapes with corridors or barriers were particularly sensitive. We increased the accuracy of tiled maps by increasing pixel size or by averaging several maps created using a “moving window” approach.

Conclusions

There is a trade-off between tile size and pixel size when modelling connectivity across large spatial extents. We suggest using the largest tile size possible when tiling is necessary, in conjunction with increased pixel size and a moving window method to increase accuracy of the composite current density maps.

Abstract

The aim of this study was to investigate the potential of simple sequence repeats as genetic markers in five species of the Solanum nigrum complex found in South Africa as well as their progeny. Some of the primers developed for sorghum, tomato, pepper and potato were used. Twenty-nine random sets of primers were selected for screening and seven were used to amplify each of the parents and their progeny. The optimum conditions were not the same for the various primer pairs with annealing temperatures of 45, 50 and 55 °C for different primers. The genetic-distance value amongst the species and their progeny varied from 0.33 to 0.55, showing the close relationship between entries and confirming the fact that the Solanum nigrum complex is a group of plants very closely related, yet the parents and their progeny could be clearly distinguished. This marker system therefore was very effective in distinguishing closely related accessions arising from a similar pedigree and could be used for molecular breeding in this species.  相似文献

The relative response of poppy (Papaver somniferum L.) and eight crop and vegetable species to excess manganese was investigated in a glasshouse, solution culture experiment. Plant yields and manganese concentrations were measured after two and six weeks growth at five levels of manganese (10–800 μM).

Poppies were highly sensitive to manganese toxicity in solution culture and reductions in shoot yield occurred at lower manganese levels in solution and at lower shoot manganese concentrations than that for the following sensitive species, ranked in order of increasing tolerance : brussels sprout, barley, green beans, lucerne and grean pea. In contrast lupins, oats and sugar beet were relatively tolerant producing about 80% or more of maximum shoot yield at the highest solution manganese level (800 μM Mn).

In this study the sensitivity of poppy, and brussels sprout, to manganese excess was attributed to their low shoot manganese “toxicity threshold values”; and their capacity to partition a high proportion of total plant manganese and dry matter to the shoot at solution manganese levels ≥ 100 μM.

The application of these results to field grown poppy is discussed in relation to interactions between manganese and other elements which modify plant tolerance to manganese excess.  相似文献

The objective of this study is to investigate the architecture to feature physical functionality of filling fats. This means an investigation of the different structure levels (crystallization, microstructure, macrostructure, etc.) that lead to good technological functionality. The isothermal crystallization behavior of two filling fats (one trans-containing and one trans-free) was examined by differential scanning calorimetry and microscopy. Furthermore, the hardness of the samples was examined after cooling in a water bath at two different temperatures and at three different storage times. The trans-containing filling fat crystallized faster and in smaller crystals as compared to the trans-free filling fat. The crystallization behavior of the trans-free filling fat was more complex, with the formation of different polymorphic forms. The hardness of the fillings was not only governed by the amount of solid fat present in the network but also by the structure of this network. The filling matrix components seem to have a pronounced influence on the microstructure and thus on the macroscopic properties.  相似文献

The charge properties of melanoidins in high molecular weight (HMw) coffee brew fractions, isolated by diafiltration and membrane dialysis, were studied. Ion exchange chromatography experiments with the HMw fractions showed that coffee brew melanoidins were negatively charged whereas these molecules did not expose any positive charge at the pH of coffee brew. Fractions with different ionic charges were isolated and subsequently characterized by means of the specific extinction coefficient (K(mix 405nm)), sugar composition, phenolic group content, nitrogen content, and the arabinogalactan protein (AGP) specific Yariv gel-diffusion assay. The isolated fractions were different in composition and AGP was found to be present in one of the HMw fractions. The AGP accounted for 6% of the coffee brew dry matter and had a moderate negative charge, probably caused by the presence of uronic acids. As the fraction that precipitated with Yariv was brown (K(mix 405nm) = 1.2), compared to a white color in the green bean, it was concluded that these AGPs had undergone Maillard reaction resulting in an AGP-melanoidin complex. The presence of mannose (presumably from galactomannan) indicates the incorporation of galactomannans in the AGP-melanoidin complex. As the uronic acid content in the more negatively charged melanoidin-rich, AGP-poor HMw fractions decreased, it was hypothesized that acidic groups are formed or incorporated during melanoidin formation.  相似文献

This case report describes the clinical presentation, the necropsy findings, and genetic results of a 13-year-old Warmblood mare presented with colic and a bilaterally loud, holosystolic murmur. Echocardiographic examination revealed the presence of a thoracic aortic aneurysm, an aortic pseudoaneurysm, a periaortic hematoma (circumferential cuffing by perivascular hemorrhage), and aortopulmonary fistulation. A supravalvular aortic stenosis (SVAS) was visible during echocardiography. Necropsy confirmed that the thoracic aortic aneurysm had ruptured and connected to the pseudoaneurysm, which fistulated into the pulmonary artery. Histologically, the aneurysm wall revealed chronic lesions such as fibrosis, mucin depositions, mineralizations, and elastin fragmentation. The mid abdominal aorta showed lesions suggestive of a systemic elastin arteriopathy. Molecular analysis, however, could not attribute this disease to a variant in the elastin gene, the most common causative gene for SVAS. To the authors' knowledge, this case report describes a case of aortopulmonary fistulation in a Warmblood horse associated with the presence of SVAS and an aortic aneurysm.  相似文献

Plants mainly rely on a mixture of Fe complexes with different organic ligands, like carboxylates and soluble fractions of water-extractable humic substances (WEHSs), to sustain the supply of this micronutrient. It has been demonstrated that the Fe-WEHS complex is more efficiently acquired by plant roots as it enhances functionality of the mechanisms involved in Fe acquisition at the root and leaf levels, allowing a faster recovery of the Fe-deficiency symptoms. The aim of this work is to verify whether this recovery involves also the allocation and accumulation of nutrients other than Fe to and within the leaf tissues. Iron-deficient plants treated with Fe-WEHS recovered more quickly the functionality both to uptake nitrate at the root level and to fixate CO2 in the leaves than those supplied with Fe-citrate. Concomitantly, Fe-WEHS-treated plants also accumulated other cationic nutrients faster and at a higher extent. Synchrotron 2D-scanning μ-X-ray fluorescence analyses of the leaves revealed that the recovery promotes a change in the allocation of these nutrients from the vascular system (K, Cu, and Zn) or trichomes (Ca and Mn) to the entire leaf blade. Fe-WEHS treatment efficiently promotes the recovery from Fe-deficiency-induced chlorosis with an enhanced allocation of other nutrients into the leaves and promoting their distribution into the entire leaf blade.  相似文献
