Lymphocyte survival during immune responses is controlled by the relative expression of pro- and anti-apoptotic molecules, regulating the magnitude, quality, and duration of the response. We investigated the consequences of deleting genes encoding the anti-apoptotic molecules Mcl1 and Bcl2l1 (Bcl-x(L)) from B cells using an inducible system synchronized with expression of activation-induced cytidine deaminase (Aicda) after immunization. This revealed Mcl1 and not Bcl2l1 to be indispensable for the formation and persistence of germinal centers (GCs). Limiting Mcl1 expression reduced the magnitude of the GC response with an equivalent, but not greater, effect on memory B cell formation and no effect on persistence. Our results identify Mcl1 as the main anti-apoptotic regulator of activated B cell survival and suggest distinct mechanisms controlling survival of GC and memory B cells.  相似文献

The substrate availability for microbial biomass (MB) in soil is crucial for microbial biomass activity. Due to the fast microbial decomposition and the permanent production of easily available substrates in the rooted top soil mainly by plants during photosynthesis, easily available substrates make a very important contribution to many soil processes including soil organic matter turnover, microbial growth and maintenance, aggregate stabilization, CO2 efflux, etc. Naturally occurring concentrations of easily available substances are low, ranging from 0.1 μM in soils free of roots and plant residues to 80 mM in root cells. We investigated the effect of adding 14C-labelled glucose at concentrations spanning the 6 orders of magnitude naturally occurring concentrations on glucose uptake and mineralization by microbial biomass. A positive correlation between the amount of added glucose and its portion mineralized to CO2 was observed: After 22 days, from 26% to 44% of the added 0.0009 to 257 μg glucose C g?1 soil was mineralized. The dependence of glucose mineralization on its amount can be described with two functions. Up to 2.6 μg glucose C g?1 soil (corresponds to 0.78% of initial microbial biomass C), glucose mineralization increased with the slope of 1.8% more mineralized glucose C per 1 μg C added, accompanied by an increasing incorporation of glucose C into MB. An increased spatial contact between micro-organisms and glucose molecules with increasing concentration may be responsible for this fast increase in mineralization rates (at glucose additions <2.6 μg C g?1). At glucose additions higher than 2.6 μg C g?1 soil, however, the increase of the glucose mineralization per 1 μg added glucose was much smaller as at additions below 2.6 μg C g?1 soil and was accompanied by decreasing portions of glucose 14C incorporated into microbial biomass. This supports the hypothesis of decreasing efficiency of glucose utilization by MB in response to increased substrate availability in the range 2.6–257 μg C g?1 (=0.78–78% of microbial biomass C). At low glucose amounts, it was mainly stored in a chloroform-labile microbial pool, but not readily mineralized to CO2. The addition of 257 μg glucose C g?1 soil (0.78 μg C glucose μg?1 C micro-organisms) caused a lag phase in mineralization of 19 h, indicating that glucose mineralization was not limited by the substrate availability but by the amount of MB which is typical for 2nd order kinetics.  相似文献

Nutrient-poor, sandy soils form the prevailing substrate at post-mining sites of the Lusatian region(Brandenburg, Germany) and present a challenge for vegetation development. We studied the organic acid quantity and composition of three commonly occurring pioneer plant species, the legumes Lotus corniculatus L. and Trifolium arvense L. and the grass Calamagrostis epigeios(L.) Roth, to determine if plant growth and exudation differed with(non-sterilized soil) and without(sterilized soil) an indigenous soil microbial community. We investigated whether organic acids were found in the rhizosphere and surrounding soil and whether this influenced nutrient mobilization. This study consists of linked field investigations and a greenhouse experiment. Plants were grown in the greenhouse in either sterilized or non-sterilized sandy soil from a reclamation site in the Lusatian mining landscape(Welzow Su¨d, East Germany). After seven months, the plant biomass, root morphology, organic acids, and water-soluble nutrients and root colonization with arbuscular mycorrhizal fungi(AMF) and dark septate endophytes(DSE) were analyzed. Roots of all three plants in the field and greenhouse experiments were highly colonized with AMF. Calamagrostis epigeios and T. arvense had a significantly higher colonization frequency with DSE than L. corniculatus. The quantity and composition of organic acids strongly differed among plant species, with the highest number of organic acids found for L. corniculatus and lowest for C. epigeios. The quantity of organic acids was greatly reduced in all plants under sterilized soil conditions. However, the composition of organic acids and plant growth in sterilized soil were reduced for both legumes, but not for C. epigeios, which had a higher biomass under sterilized conditions. Changes in nutrient concentrations in the field rhizosphere soil relative to those in the control were measurable after seven months. While the spectrum of organic acids and the growth of legumes seemed to be dependent on a highly diverse soil microbial community and a symbiotic partner, the grass C. epigeios appeared capable of mobilizing enough nutrients without an indigenous microbial community, and might be more competitive on sites where soil microbial diversity and activity are low.  相似文献

A balsam poplar clone (Populus trichocarpa cv. Weser 6) was inoculated by two ectomycorrhizal strains (Laccaria bicolor MW 158 and Paxillus involutus 1444) in Kick‐Brauckmann‐pots. The substrates were two arable sandy soils (Cambisols) with different organic matter content and nutrient supply. One soil (WIL) was rich in organic matter (Corg = 1.6%) and total nitrogen (Nt = 0.14%), whereas the other soil (RIE) had low contents of Corg (0.8%) and Nt (0.08%). Leaf nutrient concentrations, shoot lengths, root and shoot biomass production and nitrogen accumulation in the biomass were determined to discover possible inoculation effects. Mycorrhization indices (% colonized fine roots) of 36% with Laccaria bicolor and 40% with Paxillus involutus were observed on the Corg rich soil (WIL) in contrast to 16% and 14% on the Corg poor soil (RIE), respectively. Inoculation of poplar on the soil WIL increased shoot length, biomass production, shoot:root ratio and total N uptake of the cuttings, whereas on the soil RIE only the shoot:root ratio increased and the N nutrition was improved. We conclude that interactions between soil and fungus should be tested when choosing ectomycorrhizal strains for inoculation.  相似文献

Recruitment dynamics are challenging to assess or predict because of the many underlying drivers that vary in their relevance over time and space. Stock size, demographic and trait composition, condition and distribution of spawning fish and the spatio‐temporal dynamics of trophic and environmental interactions all influence recruitment processes. Exploring common patterns among stocks and linking them to potential drivers may therefore provide insights into key mechanisms of recruitment dynamics. Here, we analysed stock‐recruitment data of 64 stocks from the north‐east Atlantic Ocean for common trends in variation and synchrony among stocks using correlation, cluster and dynamic factor analyses. We tested common trends in recruitment success for relationships with large‐scale environmental processes as well as stock state indicators, and we explored links between recruitment success and demographic, environmental and ecological variables for a subset of individual stocks. The results revealed few statistically significant correlations between stocks but showed that underlying common trends in recruitment success are linked to environmental indices and management indicators. Statistical analyses confirmed previously suggested relationships of environmental–ecological factors such as the subpolar gyre and Norwegian coastal current with specific stocks, and indicated a large relevance of spawning stock biomass and demographics, as well as predation, whereas other suggested relationships were not supported by the data. Our study shows that despite persistent challenges in determining drivers of recruitment due to poor data quality and unclear mechanisms, combining different data analysis techniques can improve our understanding of recruitment dynamics in fish stocks.  相似文献

To evaluate the manner in which a cow's claws make contact with the ground at the walk, the gait, and in particular the claw-ground contact pattern, were studied in 12 healthy, lactating dairy cows, using high-speed cinematography (500frames/s) while the animals were walking on a treadmill. The results showed that the limbs were advanced around the contralateral limbs in a sigmoid curve. The feet contacted the ground with the foot axis and the tips of the claws rotated slightly outwards. In all cows the lateral claws contacted the ground before the medial claws in the hindlimbs, and in 10/12 cows in the forelimbs. The heel of the lateral claws was the region of initial contact with the ground in the hindlimbs of all cows and in the forelimbs in 9/12 cows. Lateral 'heel first' contact in the fore and hindlimbs appeared to be the normal gait pattern in these animals. Compared with a previous study of heifers, lactating cows had a larger step width in the hindlimbs and a smaller step width in the forelimbs. These ground contact patterns offer an explanation for the predisposition to claw disorders of the lateral claw of the hindlimb. The results of this study reinforce the suggestion that soft floor surfaces should be provided for cattle to prevent mechanical injury to the claws.  相似文献

The so-called "yellow pigment" content of durum wheat has been used for a long time as an indicator of the color quality of durum wheat and pasta products. For decades the chemical nature of these pigments has been assigned to carotenoids, mainly to the xanthophyll lutein and its fatty acid esters. The chemical composition of the yellow pigments of eight German durum wheat cultivars was studied. Grains were milled on a laboratory mill. Pigment extraction of millstream fractions was performed according to the optimized ICC standard method 152 procedure, and the chemical composition of the extract was analyzed by isocratic reversed phase high-performance liquid chromatography. all-trans-Lutein ranged from 1.5 to 4 mg kg(-1), and zeaxanthin was found in traces. No lutein esters and carotenes were detected. Surprisingly, the fraction of carotenoids of the complete yellow pigment content amounted to only 30-50% of the yellow pigment quantities, so there are still compounds in durum wheat not yet identified that contribute considerably to the yellow color of the grain extracts. The isolation and chemical identification of those pigments are under investigation.  相似文献

Postmortem proteome changes of porcine muscle related to tenderness   总被引:12,自引:0,他引:12  
Proteome analysis was used to investigate the relation between changes in postmortem proteome of porcine muscle and tenderness development. Muscle samples were taken at slaughter and 72 h postmortem, and the registered changes in the proteome were related to Warner-Bratzler shear force. One hundred and three protein spots were found to change significantly (P < 0.01) over time, and of these the 27 most pronounced changes were identified. Eleven out of the 27 changes were fragments of actin. Other identified myofibril proteins or fragments included myosin heavy chain, titin, myosin light chain I, myosin light II, CapZ, and cofilin. Correlation analysis revealed significant correlations between three of the identified actin fragments and the myosin heavy chain fragment to shear force. Moreover, myosin light chain II and triose phosphate isomerase I were also found to correlate significantly to shear force. The results clearly demonstrate that postmortem degradation of actin and myosin heavy chain is related to meat tenderness.  相似文献
