A new test design for the non-axenic submergent aquatic macrophytes Elodea canadensis Michx. and Myriophyllum spicatum L. has been developed for potential use in herbicide toxicity testing. For the non-axenic cultures, the best growth conditions were observed in the Elendt-M4 medium in which no growth of algae or bacteria was observed. Cuttings were placed in beakers containing only the artificial M4 medium or were planted in small beakers containing OECD (Organisation for Economic Cooperation and Development) sediment (5% peat, 75% sand, 20% kaolinite), which were then placed in larger vessels with the M4 medium. The plants were observed for main and secondary shoot length, biomass and root formation within 2-3 weeks of planting. Growth rates were calculated for total plant length and biomass. The variance between the replicates was low throughout the experiment [coefficient of variation (CV) < 26% for total plant length, and between 16 and 40% for biomass]. Relative growth rates based on total plant length were determined as 0.028 and 0.050 per day for M. spicatum in the systems containing M4 medium only and medium plus sediment respectively. Similar results were observed for E. canadensis, with relative growth rates of 0.26 and 0.073 per day in the two test systems. The root-shoot ratio at harvest was greater by a factor of 2-3 for E. canadensis in the M4 medium than in the system containing sediment. However, comparable ratios were observed for M. spicatum in the two test systems. Both growth in total plant length and growth in biomass of the two species have potential as measures of toxicity.  相似文献

Traditional homegardens (HGs) are considered to harbor high levels of plant diversity and have been therefore characterized as sustainable agro-ecosystems suitable for on-farm (incl. circa situm) conservation of plant genetic resources. While the functional structure of traditional HGs is poorly understood specifically for semi-arid and arid regions, their plant species richness and diversity is increasingly threatened by recent and fast evolving agricultural transformation processes. This has been particularly claimed for traditional jubraka HG systems of Sudan. Therefore, sixty-one HGs in four villages of the Nuba Mountains, South-Kordofan Province, Sudan, were randomly selected, geographically recorded and plant richness and abundance determined and plant diversity parameters calculated. In addition, socio-economic household data were assessed by interviews and soil samples taken to allow a comprehensive analysis of putative factors affecting HG plant diversity across different villages, levels of commercialization and plant species composition based clusters. A total of 110 species from 35 plant families were grown in the HGs along with 71 ornamentals. Perennial species accounted for 57 % including 12 indigenous fruit tree (IFT) species and six exotic fruit tree species. Mean species richness of useful plant species (excluding ornamentals) per HG was 23 (range 6–46). On average, 41 % of the 23 species per HG were of exotic origin, however, with a large range (21–83 %) among locations. Mean diversity and evenness indices were 1.46 (range 0.49–2.42) and 0.48 (0.15–0.87), respectively. The level of commercialization of HGs only marginally affected species diversity measures although the species richness was significantly higher for commercial than subsistence HGs. Species richness was higher on lower (6.6–7.2) pH soils. IFT richness was highly variable, but non-significantly different across the four locations. Plant species richness and diversity was high in comparison with other HG systems in semi-arid regions. Cluster analysis was found to be a valuable tool to classify HGs and to extract homogeneous HG types with low, intermediate and high richness and diversity. In addition, the share of exotic and ornamental species in HGs indicated a trend towards the loss of traditional farming practices, particularly in areas with good market access. The data did not indicate the hypothesized loss of inter-specific diversity due to commercialization and species richness was numerically even higher for market-oriented HGs compared to subsistence ones.  相似文献

This study evaluated outcomes of surgical treatment for carpal valgus in New World camelids and correlated successful outcome (absence of carpal valgus determined by a veterinarian) with patient characteristics and radiographic features. Univariable and multivariable analyses of retrospective case data in 19 camelids (33 limbs) treated for carpal valgus between 1987 and 2010 revealed that procedures incorporating a distal radial transphyseal bridge were more likely (P = 0.03) to result in success after a single surgical procedure. A greater degree of angulation (> 19°, P = 0.02) and younger age at surgery (< 4 months, P = 0.03) were associated with unsuccessful outcome. Overall, 74% of limbs straightened, 15% overcorrected, and 11% had persistent valgus following surgical intervention. To straighten, 22% of limbs required multiple procedures, not including implant removal. According to owners, valgus returned following implant removal in 4 limbs that had straightened after surgery.  相似文献

Tissue engineering is a promising field of study toward curing the meniscal deficient stifle; however the ideal cell type for this task is not known. We describe here the extraction of synoviocytes and meniscal fibrochondrocytes from arthroscopic debris from six dogs, which were cultured as tensioned bioscaffolds to synthesize meniscal-like fibrocartilage sheets. Despite the diseased status of the original tissues, synoviocytes and meniscal fibrochondrocytes had high viability at the time of removal from the joint. Glycosaminoglycan and collagen content of bioscaffolds did not differ. Meniscal fibrochondrocyte bioscaffolds contained more type II collagen, but collagen deposition was disorganized, with only 30–40% of cells viable. The collagen of synoviocyte bioscaffolds was organized into sheets and bands and 80–90% of cells were viable. Autologous, diseased meniscal fibrochondrocytes and synoviocytes are plausible cell sources for future meniscal tissue engineering research, however cell viability of meniscal fibrochondrocytes in the tensioned bioscaffolds was low.  相似文献

Meniscal injury is a common cause of canine lameness. Tissue engineered bioscaffolds may be a treatment option for dogs suffering from meniscal damage. The aim of this study was to compare in vitro meniscal-like matrix formation and biomechanical properties of porcine intestinal submucosa sheets (SIS), used in canine meniscal regenerative medicine, to synoviocyte-seeded SIS bioscaffold (SSB), cultured with fetal bovine serum (SSBfbs) or chondrogenic growth factors (SSBgf). Synoviocytes from nine dogs were seeded on SIS and cultured for 30 days with 17.7% fetal bovine serum or recombinant chondrogenic growth factors (IGF-1, TGFβ1 and bFGF). The effect on fibrochondrogenesis was determined by comparing mRNA expression of collagen types Iα and IIα, aggrecan, and Sry-type homeobox protein-9 (SOX9) as well as protein expression of collagens I and II, glycosaminoglycan (GAG), and hydroxyproline.The effect of synoviocyte seeding and culture conditions on biochemical properties was determined by measuring peak load, tensile stiffness, resilience, and toughness of bioscaffolds. Pre-culture SIS contained 13.6% collagen and 2.9% double-stranded DNA. Chondrogenic growth factor treatment significantly increased SOX9, collagens I and IIα, aggrecan gene expression (P < 0.05), and histological deposition of fibrocartilage extracellular matrix (GAG and collagen II). Culture with synoviocytes increased SIS tensile peak load at failure, resilience, and toughness of bioscaffolds (P < 0.05). In conclusion, culturing SIS with synoviocytes prior to implantation might provide biomechanical benefits, and chondrogenic growth factor treatment of cultured synoviocytes improves in vitro axial meniscal matrix formation.  相似文献

On the basis of a carbon isotopic record of both marine carbonates and organic matter from the Triassic-Jurassic boundary to the present, we modeled oxygen concentrations over the past 205 million years. Our analysis indicates that atmospheric oxygen approximately doubled over this period, with relatively rapid increases in the early Jurassic and the Eocene. We suggest that the overall increase in oxygen, mediated by the formation of passive continental margins along the Atlantic Ocean during the opening phase of the current Wilson cycle, was a critical factor in the evolution, radiation, and subsequent increase in average size of placental mammals.  相似文献

Massive microbial mats covering up to 4-meter-high carbonate buildups prosper at methane seeps in anoxic waters of the northwestern Black Sea shelf. Strong 13C depletions indicate an incorporation of methane carbon into carbonates, bulk biomass, and specific lipids. The mats mainly consist of densely aggregated archaea (phylogenetic ANME-1 cluster) and sulfate-reducing bacteria (Desulfosarcina/Desulfococcus group). If incubated in vitro, these mats perform anaerobic oxidation of methane coupled to sulfate reduction. Obviously, anaerobic microbial consortia can generate both carbonate precipitation and substantial biomass accumulation, which has implications for our understanding of carbon cycling during earlier periods of Earth's history.  相似文献
