To identify genes of the necrotrophic pathogenic fungus Botrytiscinerea that are expressed during infection of tomato leaves, a differential screening of a genomic library with radioactively labelled cDNA was performed. This resulted in the identification of a B. cinerea gene, denominated Bcubi4, which encodes a precursor protein consisting of four identical head-to-tail repeats of a 76aa ubiquitin unit. Subsequently a gene denominated Bcubi1CEP79, encoding a single ubiquitin unit joined to a Carboxyl Extension Protein of 79 amino acids, was isolated. The expression of the two ubiquitin genes was studied during pathogenesis of B. cinerea on tomato. Bcubi1CEP79, but not Bcubi4, mRNA was transiently induced at 16h after inoculation. The increased expression of the Bcubi1CEP79 gene at this stage of pathogenesis might be required for enhanced ribosomal biogenesis. 相似文献
Nonhuman primates can be naturally infected with a plethora of viruses with zoonotic potential, including retroviruses. These simian viruses present risks to both captive nonhuman primate populations and persons exposed to nonhuman primates. Simian retroviruses, including simian immunodeficiency virus, simian type D retrovirus, simian T-lymphotropic virus, and gibbon ape leukemia virus, have been shown to cause clinical disease in nonhuman primates. In contrast, simian foamy virus, a retrovirus that is highly prevalent in most nonhuman primates, has not been associated with clinical disease in naturally infected primates. Although it has been shown that human retrovirus infections with human T-lymphotropic virus and human immunodeficiency virus originated through multiple independent introductions of simian retroviruses into human populations that then spread globally, little is known about the frequency of such zoonotic events. In this article, exogenous simian retroviruses are reviewed as a concern for zoo and wildlife veterinarians, primate handlers, other persons in direct contact with nonhuman primates, and other nonhuman primates in a collection. The health implications for individual animals as well as managed populations in zoos and research institutions are discussed, the cross-species transmission and zoonotic disease potential of simian retroviruses are described, and suggestions for working safely with nonhuman primates are provided. 相似文献
A high intake of easily fermentable carbohydrates and a low intake of fiber material are generally regarded as major factors affecting the health of captive langurs. The effect on fecal consistency of excluding fruits and vegetables from the diet was evaluated in Javan langurs (Trachypithecus auratus auratus). Cross-over trials were carried out at Rotterdam Zoo and at the Apenheul Zoo, The Netherlands. During the first and third dietary period, the langurs were fed their usual diet, which contained fruits, vegetables, langur pellets, and browse. During the second period, the vegetables and fruits were excluded from the diet and the diet essentially consisted of pellets and browse. Feces consistency was scored using a fecal score chart developed for langurs. During the second feeding period the feces consistency improved significantly in animals at both zoos. Across all trials, a firmer feces consistency was correlated with an increase in dietary cell wall (measured as neutral detergent fiber) and a decrease in dietary water. It is suggested that the combined decrease in the intake of soluble sugars, the increase of fiber intake, and a lower amount of dietary water in the diet resulted in more solid stools. The results indicate that a dietary neutral detergent fiber content of approximately 46% in dry matter will result in a feces consistency indicative of undisturbed gut function. 相似文献
In recent years the fate of human and animal pathogen microorganisms as a potential pollutant of the environment has been paid increased attention. Substantial quantities of these compounds and their metabolites are excreted, flushed down the drain, discarded as waste, or left over in animal feedlots. After passing to the sewer, several of these compounds are not adequately eliminated by the methods that are currently used in sewage treatment. Substantial quantities of biosolids and livestock manure end up on agricultural land. Effective sanitation of the environment, particularly of some of its special parts, which can be a source of spreading of diseases, plays an important role in prevention of infectious diseases. In this respect special attention should be paid to the disinfection of infected farm animal excrements. Sanitation of excrements should, on the one hand, ensure effective inhibition of infectious agents and, on the other hand, comply with the requirement of preserving the composition of the manure so it can be used in agricultural production. 相似文献
Papaya ringspot virus (PRSV) HA 5-1, a nitrous acid-induced mild mutant of severe strain HA, widely applied for control of PRSV by cross-protection,
was used to study the genetic basis of attenuation. Using infectious clones, a series of recombinants was generated between
HA 5-1 and HA and their infectivity was analyzed on the systemic host papaya and the local lesion host Chenopodium quinoa. The recombinants that contained mutations in P1 and HC-Pro genes caused attenuated infection on papaya without conspicuous
symptoms, similar to HA 5-1. The recombination and sequence analyses strongly implicated two amino acid changes in the C-terminal
region of P1 and two in HC-Pro of HA 5-1 involved in the attenuated infection on papaya. The recombinants that infected C. quinoa plants without local lesions contained the same mutations in the C-terminal region of HC-Pro for attenuated infection on
papaya. We conclude that both P1 and HC-Pro bear important pathogenicity determinants for the infection on the systemic host
papaya and that the mutations in HC-Pro affecting pathogenicity on papaya are also responsible for the inability to induce
hypersensitive reaction on C. quinoa. 相似文献
The aims of this follow-up study were: (a) to evaluate the role of ECT technology as a risk factor for several diseases; and (b) to determine if the effects of these diseases on cows' reproductive performance and as risk factors for culling are influenced by exposure to ECTs. Diseases considered were retained placenta, metritis, ketosis, cystic ovaries, silent heat, milk fever, clinical mastitis, and foot and leg problems. We used historical and contemporary controls (with control herds selected to match the experimental herds for size and location). Data consisted of 10264 Swedish Red and White (SRB) and 5461 Swedish Friesian (SLB) lactation records in 150 herds of which 33 used cow-trainers. Logistic regression was used to estimate the effects of parity and exposure to electric cow-trainers on the risks of diseases and the effects of diseases and exposure to electric cow-trainers on risk of culling. The least-squares procedure was used to estimate the effects of diseases on reproductive performance.
The dominant effects associated with use of electric cow-trainers were an increased risk for silent heat, clinical mastitis, ketosis and culling relative to cows in herds not using cow-trainers. Diseases had negative effects on reproductive performance and the effects were larger for cows in herds using cow-trainers. In herds using electric cow-trainers, the largest increase in the interval from first service to conception (58 days) was caused by the occurrences of silent heat, cystic ovaries and the combination of two or more diseases. Retained placenta, metritis, cystic ovaries, clinical mastitis and a combination of two or more diseases increased the risk of culling about two times relative to healthy primiparous cows with the increase being greater for cows in herds using cow-trainers. Silent heat did not increase risk of culling in control groups, but was the largest risk factor for culling in the exposed group. We concluded that exposure to electric cow-trainers increased the incidence risk of silent heat, clinical mastitis, and ketosis and changed silent heat from a neutral disease with respect to culling to a major risk factor. Finally, exposure to cow-trainers increased the general negative effect of diseases on the cows' reproductive performance and risk for culling. 相似文献
The set of twenty long‐term field nutrition trials starting from 1957 at five sites. The sites differed by their altitude (from 180 m to 620 m) with average daily temperatures (from 6,8°C m to 9°C m) and soil type (from chernozem to brown podsolic soil) Provided mainly the following results: ? The geonomic division to the fertile sugarbeet region and less fertile potato region did not strictly differentiate the influence of fertilizer and climatic effects to the crop yields. ? The effect of fertilizing was dependent on the nutrient content and on the conditions of releasing and binding of the nutrients in soil. ? The precipitation regime has a strong influence on the effect of the nitrogen escalated doses. ? The facts concerning the nutrient intake are very precious knowledge. These facts have not been examined yet during the long‐term trials. ? The site effect, particularly its climatic conditions, on the nutrient intake is often more obvious compared to the fertilizing effect. ? Quantitative multi-elemental analysis by inductively coupled plasma (ICP) spectrometry depends on a complete digestion of
solid samples. However, fast and thorough sample digestion is a challenging analytical task which constitutes a bottleneck
in modern multi-elemental analysis. Additional obstacles may be that sample quantities are limited and elemental concentrations
low. In such cases, digestion in small volumes with minimum dilution and contamination is required in order to obtain high
