DNA vaccinations against fish viral diseases as IHNV at commercial level in Canada against VHSV at experimental level are both success stories. DNA vaccination strategies against many other viral diseases have, however, not yet yielded sufficient results in terms of protection. There is an obvious need to combat many other viral diseases within aquaculture where inactivated vaccines fail. There are many explanations to why DNA vaccine strategies against other viral diseases fail to induce protective immune responses in fish. These obstacles include: 1) too low immunogenicity of the transgene, 2) too low expression of the transgene that is supposed to induce protection, 3) suboptimal immune responses, and 4) too high degradation rate of the delivered plasmid DNA. There are also uncertainties with regard distribution and degradation of DNA vaccines that may have implications for safety and regulatory requirements that need to be clarified. By combining plasmid DNA with different kind of adjuvants one can increase the immunogenicity of the transgene antigen – and perhaps increase the vaccine efficacy. By using molecular adjuvants with or without in combination with targeting assemblies one may expect different responses compared with naked DNA. This includes targeting of DNA vaccines to antigen presenting cells as a central factor in improving their potencies and efficacies by means of encapsulating the DNA vaccine in certain carriers systems that may increase transgene and MHC expression. Specific contrast ultrasound is widely applied in diagnostic procedures on humans but remains underused in veterinary medicine. The objective of this study was to evaluate the use of microbubble-based contrast for rapid ultrasonographic diagnosis of thrombosis in small animals, using male New Zealand white rabbits (average weight about 3.5 kg) as a model. It was hypothesized that the use of microbubble-based contrast agents will result in a faster and more precise diagnosis in our model of thrombosis. A pro-coagulant environment had been previously established by combining endothelial denudation and external vessel wall damage. Visualization of thrombi was achieved by application of contrast microbubbles [sterically stabilized, phospholipid-based microbubbles filled with sulfur hexafluoride (SF6) gas] and ultrasonography. As a result, rapid and clear diagnosis of thrombi in aorta abdominalis was achieved within 10 to 30 s (mean: 17.3 s) by applying microbubbles as an ultrasound contrast medium. In the control group, diagnosis was not possible or took 90 to 180 s. Therefore, sterically stabilized microbubbles were found to be a suitable contrast agent for the rapid diagnosis of thrombi in an experimental model in rabbits. Background
The moose (Alces alces) is an intensively managed keystone species in Fennoscandia. Several aspects of reproduction in moose have not been fully elucidated, including puberty, timing of mating and oestrus, and the length of the oestrus period. These aspects are relevant for an adaptive management of moose with respect to harvest, population size, demography and environmental conditions. Therefore, an investigation of female moose reproduction was conducted during the moose-hunting period in southern Sweden from 2008 to 2011.Results
A total of 250 reproductive organs and information on carcass weight and age was collected from four different hunting areas (provinces of Öland, Småland, Södermanland, and Västergötland) in southern Sweden. The results showed that puberty in female moose varied with carcass weight, age, and time of season. The period for oestrous/mating lasted from about mid September to the beginning of November.Conclusions
The oestrus period (predominantly for heifers) is longer than previously reported and was not finished when the hunting period started. Sampling the uterine cervix to detect spermatozoa was a useful method to determine if mating had occurred. To avoid hunting of moose during oestrus, we suggest that the hunting period should be postponed by at least 14 days in southern Sweden. 相似文献74.

The aim of the present study was to determine the relationship of progesterone (P4),
bovine pregnancy-associated glycoprotein-1 (bPAG-1) and nitric oxide (NO) levels with late
embryonic (LEM; day 28 to day 42) and early fetal mortalities (EFM; > day 42 to day 56)
in dairy cows. Transrectal ultrasonography (6–8 MHz) was performed in 100
Holstein-Friesian cows at days 28, 42 and 56 after artificial insemination (AI; day 0) to
diagnose pregnancy and to monitor the fate of the embryo. After ultrasound scanning of
each cow, a milk sample was collected for assessment of P4 by an ELISA test and a blood
sample was collected for assessment of bPAG-1, by using a double-antibody
radioimmunoassay, and serum NO metabolites (nitrate + nitrite). Based on ultrasonographic
examinations and bPAG-1-RIA, 41 of 100 inseminated cows were confirmed pregnant at day 28
after AI. Nine cows suffered of LEM, and 6 cows suffered of EFM and the overall pregnancy
loss rate was 36.6% (15/41) between days 28 and 56 of pregnancy. By logistic regression
analysis, there were no significant relationships between the level of P4 and bPAG-1 at
day 28 after AI and the occurrence of LEM and EFM. Also, there were no significant
relationships between the levels of P4 and bPAG-1 at day 42 and the occurrence of EFM. On
the other hand, a significant relationship (P<0.05) was found between NO level at day
28 and the occurrence of LEM. In conclusion, measurement of the serum NO concentration at
day 28 of pregnancy might help to predict the outcome of pregnancy by day 42 in dairy cows

Estrus cycle is a well recognized cause of insulin resistance in bitches. The insulin receptor (IR) as well as the insulin-like
growth factor-I receptor belong to the same subfamily of tyrosine kinase (TK) receptors. The objective of this study was to
evaluate basal TK activity in muscle tissue of bitches during the estrus cycle. Twenty-four bitches were used in the study
(7 in anestrus, 7 in estrus, and 10 in diestrus). Muscle samples, taken after spaying surgery to determine TK activity, were
immediately frozen in liquid nitrogen and then stored at −80°C until the membranes were prepared by sequential centrifugation
after being homogenized. TK activity was determined by Poly (Glu 4:Tyr 1) phosphorylation and expressed in cpm/μg of protein.
TK activity was significantly lower (P < 0.001) in the animals in estrus (104.5 ± 11.9 cpm/μg of protein) and diestrus (94.5 ± 16.9 cpm/μg of protein) when compared
with bitches in anestrus (183.2 ± 39.2 cpm/μg of protein). These results demonstrate, for the first time, lower basal TK activity
in the muscle tissue of female dogs during estrus and diestrus, which may represent lower insulin signaling capacity, opening
a new field of investigation into the molecular mechanisms of insulin resistance in dogs. 相似文献

The detection of 4-hydroxycoumarin rodenticides in poisoned domestic animals requires a highly sensitive method as tissue and serum levels of anticoagulants may be very low owing to rapid elimination, metabolism or post-mortem degradation. Thin-layer chromatography (TLC) and reversed-phase high-performance liquid chromatography (RP-HPLC) with fluorescence detection were used to identify the anticoagulants in spiked tissues and in suspicious samples. The analysis of ten suspicious samples highlighted the limitations of both methods. Only the three samples of baits were found positive by TLC whereas one of the five anticoagulants was detected in eight samples by RP-HPLC with fluorescence detection. Therefore, RP-HPLC with fluorescence detection proved to be the more sensitive method for detecting low levels of 4-hydroxycoumarins in blood serum, liver and ingesta, whereas TLC is usually sufficient for analysing baits.Abbreviations RP-HPLC

Eight calves were fed from the fourth to the twentieth week of age a milk powder diet containing 40 mg lead acetate per kg dry substance. This is twice the lead concentration legally permitted. In average, each animal has daily taken up 0.834 mg lead per kg body weight. Eight calves were used as controls. The animals gained the usual fattening weights and did not show clinical symptoms. Beginning with week 14, increased levels of blood urea were encountered in the animals which received lead in their diet. Morphologically, severe renal lesions were found in these calves. The kidneys were increased in weight, pale and of firm consistency. Histologically, fibrosis and periglomerular interstitial non-purulent nephritis were found. Light- and electronmicroscopically, numerous intranuclear inclusion-bodies typical for lead poisoning were demonstrated in epithelial cells of proximal tubules. The results show, that daily uptake of less than 1 mg lead per kg body weight must be considered as a toxic dose for calves, and not 5-7 mg as stated in literature. 相似文献

Foot and mouth disease (FMD) is a highly transmissible viral infection of cloven hooved animals associated with severe economic losses when introduced into FMD-free countries. Information on the impact of the disease in FMDV-endemic countries is poorly characterised yet essential for the prioritisation of scarce resources for disease control programmes. A FMD (virus serotype SAT2) outbreak on a large-scale dairy farm in Nakuru County, Kenya provided an opportunity to evaluate the impact of FMD on clinical mastitis and culling rate. A cohort approach followed animals over a 12-month period after the commencement of the outbreak. For culling, all animals were included; for mastitis, those over 18 months of age. FMD was recorded in 400/644 cattle over a 29-day period. During the follow-up period 76 animals were culled or died whilst in the over 18 month old cohort 63 developed clinical mastitis. Hazard ratios (HR) were generated using Cox regression accounting for non-proportional hazards by inclusion of time-varying effects. Univariable analysis showed FMD cases were culled sooner but there was no effect on clinical mastitis. After adjusting for possible confounders and inclusion of time-varying effects there was weak evidence to support an effect of FMD on culling (HR = 1.7, 95% confidence intervals [CI] 0.88-3.1, P = 0.12). For mastitis, there was stronger evidence of an increased rate in the first month after the onset of the outbreak (HR = 2.9, 95%CI 0.97-8.9, P = 0.057).

本试验旨在研究外源精胺对断奶仔猪小肠黏膜成纤维细胞因子-2( FGF2)蛋白质表达量及小肠发育的影响。将9窝胎次和初生体重相近的7日龄“杜×长×大”哺乳仔猪随机分配到0、12和15 mg/kg外源精胺组,每组3窝。仔猪从7日龄起补饲相应的饲粮,21日龄断奶后继续补饲断奶前饲粮至28日龄结束。从每窝选2头接近平均体重的仔猪屠宰,采集小肠及黏膜样品,测定FGF2蛋白质表达量和小肠的发育状况。结果表明:1)12 mg/kg外源精胺组仔猪小肠黏膜FGF2蛋白质表达量分别高于0和15 mg/kg外源精胺组,但3个外源精胺组仔猪小肠黏膜FGF2蛋白质表达量之间无显著差异(P>0.05)。2)12和15 mg/kg外源精胺组仔猪小肠绒毛高度和宽度分别极显著大于0 mg/kg外源精胺组( P<0.01),而12 mg/kg外源精胺组仔猪十二指肠和空肠黏膜绒毛高度与隐窝深度比值( V/C )分别极显著高于0 mg/kg外源精胺组( P<0.01)。3)28日龄仔猪小肠黏膜蔗糖酶和麦芽糖酶活性随外源精胺添加量的增加而升高。由此可见,给7~28日龄仔猪连续补饲外源精胺能提高28日龄仔猪小肠黏膜FGF2蛋白质表达量、V/C和二糖酶活性。 相似文献
