The permeability of the upper mantle controls melt segregation beneath spreading centers. Reconciling contradictory geochemical and geophysical observations at ocean ridges requires a better understanding of transport properties in partially molten rocks. Using x-ray synchrotron microtomography, we obtained three-dimensional data on melt distribution for mantle peridotite with various melt fractions. At melt fractions as low as 0.02, triple junctions along grain edges dominated the melt network; there was no evidence of an abrupt change in the fundamental character of melt extraction as melt fraction increased to 0.2. The porosity of the partially molten region beneath ocean ridges is therefore controlled by a balance between viscous compaction and melting rate, not by a change in melt topology.  相似文献

There is a relation between stress and alcohol drinking. We show that the corticotropin-releasing hormone (CRH) system that mediates endocrine and behavioral responses to stress plays a role in the control of long-term alcohol drinking. In mice lacking a functional CRH1 receptor, stress leads to enhanced and progressively increasing alcohol intake. The effect of repeated stress on alcohol drinking behavior appeared with a delay and persisted throughout life. It was associated with an up-regulation of the N-methyl-d-aspartate receptor subunit NR2B. Alterations in the CRH1 receptor gene and adaptional changes in NR2B subunits may constitute a genetic risk factor for stress-induced alcohol drinking and alcoholism.  相似文献

Double-stranded ribonucleic acid (dsRNA) serves as a danger signal associated with viral infection and leads to stimulation of innate immune cells. In contrast, the immunostimulatory potential of single-stranded RNA (ssRNA) is poorly understood and innate immune receptors for ssRNA are unknown. We report that guanosine (G)- and uridine (U)-rich ssRNA oligonucleotides derived from human immunodeficiency virus-1 (HIV-1) stimulate dendritic cells (DC) and macrophages to secrete interferon-alpha and proinflammatory, as well as regulatory, cytokines. By using Toll-like receptor (TLR)-deficient mice and genetic complementation, we show that murine TLR7 and human TLR8 mediate species-specific recognition of GU-rich ssRNA. These data suggest that ssRNA represents a physiological ligand for TLR7 and TLR8.  相似文献

Transforming growth factor-β (TGFβ) signaling drives aneurysm progression in multiple disorders, including Marfan syndrome (MFS), and therapies that inhibit this signaling cascade are in clinical trials. TGFβ can stimulate multiple intracellular signaling pathways, but it is unclear which of these pathways drives aortic disease and, when inhibited, which result in disease amelioration. Here we show that extracellular signal-regulated kinase (ERK) 1 and 2 and Smad2 are activated in a mouse model of MFS, and both are inhibited by therapies directed against TGFβ. Whereas selective inhibition of ERK1/2 activation ameliorated aortic growth, Smad4 deficiency exacerbated aortic disease and caused premature death in MFS mice. Smad4-deficient MFS mice uniquely showed activation of Jun N-terminal kinase-1 (JNK1), and a JNK antagonist ameliorated aortic growth in MFS mice that lacked or retained full Smad4 expression. Thus, noncanonical (Smad-independent) TGFβ signaling is a prominent driver of aortic disease in MFS mice, and inhibition of the ERK1/2 or JNK1 pathways is a potential therapeutic strategy for the disease.  相似文献

The mountainous environment of the Ethiopian highlands has a great variety of ecotopes and thus demands great flexibility in land management. Different climatic conditions and landscape positions induce different soil forming processes, leading to various soil types with specific risks and potentials. The present study portrays a soil sequence of the central section of the Main Ethiopian Rift Valley, from the footslopes of the eastern escarpment to the marginal volcano structures. Six profiles under natural vegetation are described for classification according to the USDA Soil Taxonomy and World Reference Base for Soil Resources classification systems, and special site characteristics are discussed. The Acacia savannah of the footslopes (1900 m a. s. l.) is characterized by Vertisols (Mazic Vertisols/Aridic Haplusterts), with often pronounced effects of seasonal waterlogging. All other soils are well-drained and reflect the general increase in rainfall with elevation and slope, causing a decline in topsoil pH and a change from cation-rich clay soils of the Podocarpus-dominated forest at 2300 m a. s. l. (Mollic Nitisols/Typic Palehumults) to strongly-leached Humic Umbrisols/Humic Dystrudepts of the Hagenia-dominated forest around 2600 m a. s. l. The highland savannah plain (2700 m a. s. l.) with a drier and cooler environment has typically Mollic Cambisols/Dystric Haplustepts, which are less leached and have a rather brownish colour. At 2900 m a. s. l., Niti-umbric Alisols/Andic Hapludalfs are found in the Hypericum forest at the midslopes of the marginal volcanoes receiving high rainfall, whereas soil development is at a more initial state in the Erica-dominated forest at 3200 m a. s. l. (Umbric Andosols/Alic Hapludands). Clay mineral composition is kaolinite-dominated for the upper five profiles, with a high proportion of poorly crystalline components in the upper savannah and the volcano upslopes. The lowermost profile probably has a polygenetic origin indicated by an abrupt change from a smectitic to a kaolinitic composition in the subsoil. Soil development on quite homogeneous bedrock appears essentially controlled by relief and climate, underlining the suitability of the region as a model area for in-depth gradient studies on ecosystem processes and land use.  相似文献

Total phenolics, ascorbic acid, and betalain contents of differently colored cactus pear clones (nine Opuntia ficus-indica [L.] Mill. clones and one O. robusta Wendl. clone) were investigated and related to their respective antioxidant potential assessed by Trolox-equivalent antioxidant capacity (TEAC) and oxygen radical absorbance capacity (ORAC) assays. TEAC and ORAC values were very highly correlated with each other and also with values for total phenolics, betalain contents, and ascorbic acid concentrations. Total phenolics had the greatest contribution to ORAC and TEAC values. High-performance liquid chromatography (HPLC)-diode array detector (DAD)-tandem mass spectrometry (MS/MS) measurements of cactus pear juices permitted the differentiation of the clones based on variations in pigment patterns and betalain concentrations. The red and yellow betalains were absent in lime green colored cactus fruits. The ratio and concentration of these pigments were responsible for the yellow, orange, red, and purple colors in the other clones. Progeny of purple and lime green colored parents were characterized by 12% and 88% of plants bearing lime green and purple fruit, respectively. This implies that the genes for betalain production were lacking in the lime green fruits but could be provided by a parent with a complete set of genes, that is, purple fruits. Besides known pigments typical of Cactaceae, two unexpected betalains were identified. Whereas gomphrenin I was found for the first time in tissues of cactus plants, methionine-betaxanthin has never been described before as a genuine betalain. In addition to their alleged health-promoting properties, various combinations of yellow betaxanthins and red-purple betacyanins may allow the development of new food products without using artificial colorants.  相似文献
