In recent years, interest has grown in cultivating Allium species with enhanced health benefits and/or distinct flavor. Concentrations of phytochemicals determining these desired characteristics may be influenced by nitrogen forms (ammonium or nitrate) and arbuscular mycorrhizal (AM) fungi. We examined these relations with the test plant bunching onion (Allium fistulosum L.). Three different ammonium‐to‐nitrate (NH : NO ) ratios were supplied in combination with or without inoculation with an AM fungus (Glomus mosseae). The plants were evaluated for dry weight, leaf number, and content of nutrients (N, NO , P, S), sugars (glucose, fructose, and sucrose), and organosulfur compounds (measured as pyruvic acid). The experiment was carried out under controlled conditions in a greenhouse. Plants were grown on perlite amended twice a day with nutrient solution. In nonmycorrhizal plants, the application of nutrient solution with predominant NO or NH4NO3 as N source supported adequate growth of Allium fistulosum while predominant NH supply resulted in decreased growth and occurrence of wilting symptoms. Mycorrhizal inoculation significantly increased dry weight and leaf number of predominantly NH ‐fed or NH4NO3‐fed plants. While shoot P concentration increased with higher NH supply, shoot N concentration increased in predominantly NH ‐fed plants only. Nitrogen form and AM colonization had little effect on shoot S or sugar concentrations. The total content in organosulfur compounds was significantly affected by both, N form and AM colonization. The optimal growth condition for a high formation of organosulfur compounds in this experiment was a nutrient solution with predominant NO supply, but when supported by AM fungi, Allium fistulosum produced similar amounts of pyruvic acid in NH4NO3‐fed plants.  相似文献

Within recent years, a slight but significant increase of European eel Anguilla anguilla (L.) recruitment has been documented, but it remains questionable whether or not the increased recruitment levels resulted in higher eel numbers at the regional scale. To detect the changes in yellow eel numbers, logbook data covering a 15-year time series of catch per unit effort (CPUE) data from the German Baltic Sea were analysed. Monthly mean catch rates were calculated for two different size classes for two passive gears: fyke and stationary trap nets. Change-point analysis was applied to discover changes in the catch data. After a period of decreasing or constant catch rates, the fyke net data indicated that yellow eel numbers increased slightly in recent years in the Baltic Sea. Besides increasing numbers of immigrating juvenile eels, other population dynamics or conservation efforts might have added to the observed positive stock trend.  相似文献

Sequence diversity in the two barley (Hordeum vulgare L.) genes encoding sucrose synthase I (SSI) and sucrose phosphate synthase II (SPSII), both of which are involved in sucrose accumulation and grain filling, was studied by partial resequencing of eight reference genotypes and SNP analysis by pyrosequencing in a panel of 94 spring and 96 winter European barley varieties. The resequencing was based on two adjacent SSI fragments of size 880 and 820 bp, and a 2,322 bp SPSII fragment. In the SSI gene, 26 SNPs were present in the larger fragment, and 25 in the smaller one, and 11 of these were exploited to develop high-throughput SNP assays used for haplotype analysis. An association analysis based on either a general or a mixed linear model suggested that the predominant three haplotypes influenced certain components of both kernel and malting quality. However, the level of phenotype/haplotype association shown with the SPSII gene was rather low. SNP variation of SSI was used to map the locus to chromosome 7H.  相似文献

Cellular membrane fusion is thought to proceed through intermediates including docking of apposed lipid bilayers, merging of proximal leaflets to form a hemifusion diaphragm, and fusion pore opening. A membrane-bridging four-helix complex of soluble N-ethylmaleimide-sensitive factor attachment protein receptors (SNAREs) mediates fusion. However, how assembly of the SNARE complex generates docking and other fusion intermediates is unknown. Using a cell-free reaction, we identified intermediates visually and then arrested the SNARE fusion machinery when fusion was about to begin. Partial and directional assembly of SNAREs tightly docked bilayers, but efficient fusion and an extended form of hemifusion required assembly beyond the core complex to the membrane-connecting linkers. We propose that straining of lipids at the edges of an extended docking zone initiates fusion.  相似文献

In central neurons, information flows from the dendritic surface toward the axon terminals. We found that during in vitro gamma oscillations, ectopic action potentials are generated at high frequency in the distal axon of pyramidal cells (PCs) but do not invade the soma. At the same time, axo-axonic cells (AACs) discharged at a high rate and tonically inhibited the axon initial segment, which can be instrumental in preventing ectopic action potential back-propagation. We found that activation of a single AAC substantially lowered soma invasion by antidromic action potential in postsynaptic PCs. In contrast, activation of soma-inhibiting basket cells had no significant impact. These results demonstrate that AACs can separate axonal from somatic activity and maintain the functional polarization of cortical PCs during network oscillations.  相似文献

A method for the identification and quantification of phenolic compounds from artichoke (Cynara scolymus L.) heads, juice, and pomace by HPLC with diode array and mass spectrometric detection was developed. Among the 22 major compounds, 11 caffeoylquinic acids and 8 flavonoids were detected. Quantification of individual compounds was carried out by external calibration. Apigenin 7-O-glucuronide was found to be the major flavonoid in all samples investigated. 1,5-Di-O-caffeoylquinic acid represented the major hydroxycinnamic acid, with 3890 mg/kg in artichoke heads and 3269 mg/kg in the pomace, whereas in the juice 1,3-di-O-caffeoylquinic acid (cynarin) was predominant, due to the isomerization during processing. Total phenolic contents of approximately 12 g/kg on a dry matter basis revealed that artichoke pomace is a promising source of phenolic compounds that might be recovered and used as natural antioxidants or functional food ingredients.  相似文献

Assessment of critical values of phosphorus, potassium, and magnesium contents in arable soils Long-term fertilizer field experiments were carried out in order to calibrate soil test values for P, K, and Mg. In several years treatments gradually resulted in differences of soil nutrient contents. This made it possible to investigate the effect of fertilizer application on the relation between yield of plants and soil nutrient content. Methods were suggested to evaluate these relationships and to calculate critical soil nutrient contents. Emphasis was mainly directed towards the definition of level 3 of nutrient supply because it represents the aim of economic fertilizer application.  相似文献

Phenolic compounds of 14 pomace samples originating from red and white winemaking were characterized by HPLC-MS. Up to 13 anthocyanins, 11 hydroxybenzoic and hydroxycinnamic acids, and 13 catechins and flavonols as well as 2 stilbenes were identified and quantified in the skins and seeds by HPLC-DAD. Large variabilities comprising all individual phenolic compounds were observed, depending on cultivar and vintage. Grape skins proved to be rich sources of anthocyanins, hydroxycinnamic acids, flavanols, and flavonol glycosides, whereas flavanols were mainly present in the seeds. However, besides the lack of anthocyanins in white grape pomace, no principal differences between red and white grape varieties were observed. This is the first study presenting comprehensive data on the contents of individual phenolic compounds comprising all polyphenolic subclasses of grapes including a comparison of several red and white pomaces from nine cultivars. The results obtained in the present study confirm that both skins and seeds of most grape cultivars constitute a promising source of polyphenolics.  相似文献

The adsorption of the toxin from Bacillus thuringiensis (Bt‐toxin), which is synthesized in genetically modified maize, on sterilized Na‐montmorillonite and on H2O2‐treated and untreated clay fractions of three soils from different sites were studied. All adsorption isotherms can be described by a linear isotherm. Although all clay fractions from the different soils show nearly the same mineralogical composition, we found different affinities ranging from k = 47.7 to k = 366.7 of the adsorbates for the Bt‐toxin. The H2O2‐treated clay fractions show no correlation between the adsorption affinity and the amount of soil organic matter. On the other hand, there is a correlation between the content of organic carbon and the adsorption affinity of the untreated clay fractions. This can be explained by the fact that due to the coatings of soil organic matter on aggregates, the Bt‐toxin polymers are not able to adsorb within the clay aggregates.  相似文献

The molecular machinery mediating the fusion of synaptic vesicles (SVs) at presynaptic active zone (AZ) membranes has been studied in detail, and several essential components have been identified. AZ-associated protein scaffolds are viewed as only modulatory for transmission. We discovered that Drosophila Rab3-interacting molecule (RIM)-binding protein (DRBP) is essential not only for the integrity of the AZ scaffold but also for exocytotic neurotransmitter release. Two-color stimulated emission depletion microscopy showed that DRBP surrounds the central Ca(2+) channel field. In drbp mutants, Ca(2+) channel clustering and Ca(2+) influx were impaired, and synaptic release probability was drastically reduced. Our data identify RBP family proteins as prime effectors of the AZ scaffold that are essential for the coupling of SVs, Ca(2+) channels, and the SV fusion machinery.  相似文献
