Mutant-prevention concentrations of enrofloxacin for Escherichia coli isolates from chickens

OBJECTIVE: To investigate the development of enrofloxacin resistance among Escherichia coli isolates obtained from chickens by determining mutant-prevention concentrations (MPCs) and sequence the quinolone resistance-determining regions (QRDRs) of gyrA and parC genes in selected isolates. SAMPLE POPULATION: 15 chicken-derived E coli isolates. PROCEDURES: For all isolates, MPC and minimal inhibition concentration (MIC) of enrofloxacin were determined. The MPCs and maximum serum drug concentrations attained with enrofloxacin doses recommended for treatment of E coli infections in chickens were compared. Mutation frequencies and QRDR sequence changes in gyrA and parC were also determined. RESULTS: In 2 of 15 E coli strains, MPCs were low (0.016 and 0.062 microg/mL), MPC:MIC ratios were 2 and 4, and the GyrA and ParC proteins had no mutations. In 9 susceptible isolates with a GyrA point mutation, MPCs ranged from 2 to 16 microg/mL. For isolates with double mutations in GyrA and a single mutation in ParC, MPCs were > 32 microg/mL (several fold greater than the maximal plasma concentration of enrofloxacin in chickens); mutation frequencies were also much lower, compared with frequencies for single-mutation isolates. CONCLUSIONS AND CLINICAL RELEVANCE: For E coli infections of chickens, MPC appears to be useful for determining enrofloxacin-dosing strategies. The high MPC:MIC ratio may result in enrofloxacin-treatment failure in chickens infected with some wild-type gyrA E coli isolates despite the isolates' enrofloxacin susceptibility (MICs 0.125 to 1 microg/mL). For infections involving isolates with high MPCs, especially those containing mutations in gyrA and parC genes, treatment with combinations of antimicrobials should be adopted.     

Effect of diet with thiram on liver antioxidant capacity and tibial dyschondroplasia in broilers

1. An experiment was conducted to study the effect of thiram on liver antioxidant capacity and incidence of tibial dyschondroplasia in broilers. 2. One hundred and twenty Avian commercial broilers were allotted at random to three treatments: control group, low thiram group (50 mg/kg) and high thiram group (100 mg/kg). 3. Blood samples were collected to determine the activity of AST (aspartate aminotransferase). At the end of the trial, broilers were killed and liver samples were collected to determine the activity of SOD (superoxide dismutase), GSH-Px (glutathione peroxidase) and MDA (malondialdehyde) content, while the right proximal tibiotarsi were dissected in longitudinal section for assessment of tibial dyschondroplasia (TD) incidence and TD score. 4. The results showed that thiram increased the incidence of TD and TD scores, increased serum AST activity and MDA content of liver, and decreased the activity of SOD and GSH-Px in the liver. 5. They suggest that thiram causes TD in broilers by reducing liver antioxidation capability and damaging liver function; this may be one of the mechanisms by which thiram causes TD in broilers.     

Potential use of a transposon Tn916-generated mutant of Aeromonas hydrophila J-1 defective in some exoproducts as a live attenuated vaccine

Aeromonas hydrophila is a Gram-negative opportunistic pathogen that causes disease in a wide range of hosts due to its multifactorial virulence. Here we describe the application of transposon insertion mutagenesis approach to obtain an exoenzyme mutant of A. hydrophila strain J-1. Immunization of swordtail fish (Xiphophorus helleri Heckel) with the highly attenuated mutant provided protection (survival of 27 out of 35 fish, compared to survival of only 13 out of 35 control fish) in the fish given the highest immunization dose (10(7) CFU) against intraperitoneal challenge with the wild J-1 strain. Immunization with doses of 10(5) or 10(3) did not provide significant protection.

Expert System for Artificial Intelligence Blast Furnace Smelting Process

In order to upgrade traditional industry and realize the automation of furnace condition judgement and standardization of operation, an expert system for various furnace conditions and breakdown judgement and operation guidance on blast furnace are developed based on the theory of artificial intelligence and knowledge engineering. The system is fulfilled through C++Builder and combined with the actual production picture. The system has efficient and convenient man-machine interface by using menu operation method. The testing and operation of actual production data in simulation system indicate that the system is exact and reliable and can provide both the warning of various furnace breakdown and corresponding operation guidance.

Parsimonious Covering Probability Model for Turbogenerator Vibration Faulty Diagnosis

The turbogenerator vibration faults have the character of variety. Many faults often occur synchronously. This paper introduces a diagnosing model based on parsimonious covering theory and probability. A model for turtogenerator's fault diagnosis is proposed. The availability of this method is proved by two fault diagnosis examples of turbogenerator. The results show that the model proposed can be used for multi_fault diagnosis together. It may make up shortage for some of expert systems and neural networks in some aspect. From the practice,this model has higher reliability and practicability.
