(13Z,13'Z,3R,3'R,6'R)-Lutein has been isolated and purified from extracts of marigold flowers, fresh raw kale (Brassica oleracea var. Acephala), and human plasma and fully characterized by (1)H and (13)C NMR, UV/vis, and MS. While the concentration of (13Z, 13'Z)-lutein in kale and human plasma compared to (all-E,3R,3'R, 6'R)-lutein was found to be quite low, this compound was readily isolated by fractional crystallization of lutein from marigold extracts. Thus, the mother liquors from two consecutive crystallizations of lutein from a saponified extract of marigold flowers were enriched in (13Z,13'Z)-lutein (8.7% of total carotenoids) and employed for the isolation of this compound by HPLC. The identity of the di-Z-lutein in kale and human plasma has been established by comparison of the HPLC-UV/vis-MS profiles of the purified compounds with those of a fully characterized sample, isolated from marigolds. 3-Hydroxy-beta,epsilon-caroten-3'-one and 3'-epilutein have also been identified in extracts from marigolds. 相似文献

The effect of fat content on the release of volatile aroma compounds from frankfurters has been investigated. Although the release of most n-alkanals, alcohols, ketones, and furans was little affected by changes in the fat content of frankfurters, that of monoterpene hydrocarbons, sesquiterpene hydrocarbons, terpenes containing oxygen, cyclopentenones, phenyl propanoids, and phenols was greatly increased when the fat content was decreased. Some odors were also detected more frequently in the low-fat than in the full-fat sausages. These included smoky odors, due to phenols, spicy, synthetic, and floral odors due to terpenes, and meaty, roasted odors caused by sulfur-containing heterocyclic compounds. The release of aroma compounds from frankfurters appears to be closely related to the solvation of these compounds in the lipid phase. 相似文献

Length of sample storage can become significant in sensory studies due to panel fatigue limitations and samples needed for a reasonable expectation of finding significant differences. In roasted peanut sensory studies samples are stored between -10 and -23 degrees C to prevent or retard changes. Studies of up to 13 months' duration have examined stability and slow-rate sensory changes. Sweet taste was relatively stable, whereas bitter and tongue burn attributes increased slightly. Stale taste increased, suggesting lipid oxidation was taking place even at -23 degrees C. Painty attribute did not increase until stale was >3. An increase in fruity attribute was unexpected. With increases in fruity and stale attributes a decrease in roasted peanut was expected. However, storage at -23 degrees C seems to stabilize the roasted peanut lability when compared to storage at -10 degrees C. Fruity and stale interactions with roasted peanut and lability of roasted peanut were shown to be three separate and identifiable effects on roasted peanut. 相似文献

OBJECTIVE: To determine changes in results of selected serum biochemical tests after a single percutaneous liver biopsy and changes in results of hematologic and selected serum biochemical tests after multiple percutaneous liver biopsies in alpacas and llamas. ANIMALS: 5 llamas and 10 alpacas. PROCEDURE: A single percutaneous liver biopsy was performed in 5 llamas and 6 alpacas. Blood samples were collected before and 1, 2, and 3 days after liver biopsy, and submitted for serum biochemical analysis. In the other 4 alpacas, liver biopsy was performed on day 0 and then weekly for 4 weeks. Blood samples were collected at the time of each biopsy, and CBC and serum biochemical analyses were performed. Attitude and appetite of all animals were evaluated daily. RESULTS: None of the animals developed clinically apparent adverse effects. A mild decrease in sorbitol dehydrogenase activity was detected in animals that underwent a single liver biopsy, and mild decreases in plasma protein and albumin concentrations were detected in animals that underwent multiple biopsies. Other significant changes were not detected. CONCLUSIONS: Results of this study suggest that liver biopsy is safe in healthy llamas and alpacas and that, if necessary, multiple weekly liver biopsies can be performed safely in healthy alpacas. 相似文献

OBJECTIVE: To determine the diagnostic value of aerobic microbial culture and cytologic evaluation of corneal specimens in the diagnosis of infectious ulcerative keratitis (IUK). DESIGN: Prospective study. ANIMALS: 48 animals (26 dogs, 13 horses, 7 cats, 1 bird, and 1 llama) with corneal ulcers. PROCEDURE: Scrapings from corneal ulcers were examined cytologically. Corneal swab specimens were submitted for microbial culture. Animals were grouped according to whether they had been receiving antimicrobials at the time of admission. RESULTS: Of the 38 animals receiving antimicrobials, 19 had positive results for IUK on cytologic evaluation, 20 on microbial culture, and 26 on cytologic evaluation, microbial culture, or both. Of the 10 animals not receiving antimicrobials at the time of admission, 7 had positive results for IUK on cytologic evaluation, and 9 had positive results on microbial culture. In this group of 10 animals, additional animals with IUK were not identified on the basis of cytologic evaluation alone. When all 48 animals were considered irrespective of antimicrobial treatment, 26 and 29 had positive results for IUK on cytologic evaluation and microbial culture, respectively, whereas IUK was confirmed in 35 animals on the basis of cytologic evaluation, microbial culture results, or both. CONCLUSIONS AND CLINICAL RELEVANCE: Microbial culture and cytologic evaluation of corneal specimens maximizes identification of IUK, especially in animals receiving antimicrobial treatment. Because of serious consequences of untreated IUK, we recommend that both diagnostic tests be used to tailor treatment and reduce risk of vision impairment in animals. 相似文献

OBJECTIVE: To investigate the development of immune responses in calves experimentally and naturally infected with Mycobacterium paratuberculosis and to evaluate the potential for diagnostic tests to detect infected calves. DESIGN: Sequential testing of four treatment groups of calves over a 2 year period. PROCEDURE: Twenty-nine calves were allocated to four groups. Group D calves were orally dosed with M paratuberculosis, group N calves naturally exposed to M paratuberculosis, group V calves vaccinated for M paratuberculosis, and group C were control calves (not infected or vaccinated). Blood and faecal specimens were collected from each calf at monthly intervals to 18 months of age and then every 2 months until they were slaughtered between the ages of 21 and 29 months. Specimens were tested using absorbed EIA, IFN-gamma EIA and faecal culture. The infection status of the calves was confirmed by extensive histopathological examination and tissue culture. RESULTS: M paratuberculosis infection was confirmed in 10 calves, comprising six of eight orally dosed calves, three of five naturally exposed calves and one of nine vaccinated calves. The six artificially infected calves and one naturally infected calf were detected shedding M paratuberculosis in their faeces. Results with positive absorbed EIA were obtained from one artificially infected calf, one naturally infected calf and three vaccinated calves. All calves including controls had positive results on at least one occasion using the IFN-gamma EIA. In addition, seven calves had positive bovine tuberculosis results using the IFN-gamma EIA, even though bovine tuberculosis has been eradicated from Australia. OBJECTIVE: To determine the changes in Ca, P, Mg, Na, K, S, Cu, Fe and Zn concentrations of milk during the lactation in pasture-fed Thoroughbred mares and then calculate the dietary mineral requirements of the sucking foal and the lactating mare. PROCEDURE: Milk was sampled on days 1, 3, 7, 14, 21, 28 and at various times between 55 to 65, 85 to 95 and 135 to 150 days after parturition from 21 pasture-fed mares. The concentrations of macro- and micro-elements in the milk were determined by inductively coupled plasma emission spectrometry. RESULTS: Concentrations (mg/L) of these elements were highest in colostrum (Mg 302, Na 561, K 955, S 1035, Cu 0.76, Fe 0.79 and Zn 5.5) except for Ca (1245) and P (895), which where highest on day 7. The mean milk mineral element concentrations (mg/L) over days 55 to 150 were Ca 843, P 543, Mg 47, Na 120, K 590, S 219, Cu 0.19, Fe 0.34 and Zn 2.1. The mean plasma element concentrations (mg/L) over the same period were Ca 120, P 77.1, Mg 17.0, Na 3110, K 168, S 983, Cu 1.1, Fe 1.5 and Zn 0.49. Concentration gradients between plasma and milk were observed and, in the case of Ca, P, Mg, K and Zn, their concentrations in milk were greater than those in plasma, while a reverse situation was observed for Na, S, Cu and Fe. CONCLUSION: With the exception of Ca and P, the highest concentrations of mineral elements were observed in the colostrum. The nursing foal should have access to good pasture or creep feed, because the intakes of Ca, P and Cu from milk may be less than optimum to meet the daily mineral element requirements. 相似文献

Tissue-specific accumulation of tocopherols and tocotrienols in turkey tissues during embryonic development and their susceptibility to lipid peroxidation were investigated. Fertile turkey eggs were incubated using standard commercial conditions. Embryonic tissues were collected at 16, 22, 25 d of incubation and from day-old poults (referred to as day 29) and alpha-; beta- + gamma- and delta-tocopherols and respective tocotrienols were analysed by HPLC. A turkey diet provided to the parent hens contained the complete range of tocopherols and tocotrienols. Between days 16 and 22 of embryo development, the alpha-tocopherol concentration in the liver remained constant and then increased significantly (P<0.01) reaching a maximum just after hatching. Similar changes were observed for the other tocopherols and tocotrienols. The accumulation of alpha-tocopherol in the yolk sac membrane (YSM) started after day 20 of development and at hatching the alpha-tocopherol concentration in the YSM was twice that of beta- + gamma-tocopherols and 15 times greater than that of alpha-tocotrienol. In the kidney, heart, lung, muscle and adipose tissues a gradual increase in tocopherol and tocotrienol concentrations took place between days 20 and 25 of development with a sharp increase in particular of alpha-tocopherol between days 25 and 29. There was a discrimination between tocopherols and tocotrienols during their assimilation from the diet by the parent hen and during metabolism by the developing turkey embryo. Tissue-specific features in the susceptibility to lipid peroxidation were found with the brain being the most susceptible to lipid peroxidation at day 25 and in day-old poults. 相似文献
