Antioxidants are released during the NaOH extraction of soils, and this also releases phenolic compounds from plant and soil material. We hypothesized that phenolics would be important contributors to the antioxidant capacity (AOC) of soils. The concentrations of 12 phenolic compounds and the AOCs in 4 m ‐NaOH extracts of a range of UK soil types were measured. Samples of both surface and subsurface horizons were taken from 24 sites representing the major soil types (brown soils, gleys, podzols, peats and lithomorphic soils). The land use/vegetation varied from deciduous woodland through heather moorland to agricultural crops. The internal standards method was used to quantify the phenolics, which were detected by gas chromatography. The AOCs of the extracts and of standards of the individual phenolic compounds were measured using the TEAC (Trolox Equivalent Antioxidant Capacity) method. There were differences in the phenolic distributions extracted from soils with different land uses/plant inputs, as well as differences between surface and subsurface samples. A statistically significant linear relationship was found between the AOCs of the extracts and the sum of the phenolic compounds, as both these variables were positively correlated with the organic matter content of the soils. The AOCs of the individual phenols were used to calculate their contribution to the overall AOC of the extracts, and this was found to be small (approximately 1% of the total AOC). We concluded that the measured phenolic compounds were not important contributors to the AOCs of the soil extracts, and therefore that other unidentified antioxidant compounds were probably present.  相似文献

To evaluate the prevalence of methicillin-resistant Staphylococcus aureus (MRSA) in the pig population in Ireland, nasal swabbing was employed in three abattoirs to screen 440 pigs from 41 geographically distributed farms. One hundred individuals involved in the pig industry were also nasally screened. No MRSA isolates were recovered from the pigs and only two of the humans tested were identified as MRSA carriers. Importantly, MRSA was not obtained from pig producers, veterinarians or abattoir employees, but was isolated from individuals working in the wider pig industry. Multi-locus sequence typing revealed that these isolates belonged to sequence types (ST) ST22 and ST1307; the latter is a previously unreported single locus variant of ST5. Five dust samples from each of the three slaughterhouses were culture-negative for MRSA. These results indicate that porcine colonisation by MRSA, and in particular the animal-related strain MRSA-ST398, was not common in Ireland during the period of study.  相似文献

These guidelines have been designed to assist in the planning, operation and interpretation of studies which would serve to assess the efficacy of drugs against internal parasites of horses. Although the term anthelmintic is used in the title and text, these guidelines include studies on drug efficacy against larvae of horse bot flies, Gasterophilus spp., which are non-helminth parasites commonly occurring in the stomach of horses. The advantages, disadvantages and application of critical and controlled tests are presented. Information is also provided on selection of animals, housing, feed, dose titration, confirmatory and clinical trials, record keeping and necropsy procedures. These guidelines should assist both investigators and registration authorities in the evaluation of compounds using comparable and standard procedures with the minimum number of animals.  相似文献

Recently, indirect evidence was obtained for inhibition of soil net N mineralization by sterols in soil organic matter, which could have been caused by their antioxidant or antimicrobial properties. The objective of this study was to test the effect of potential inhibitors (i.e., individual compounds with known antioxidant and/or antimicrobial properties) on soil microbial mineralization processes during incubation for 7 and 14 d. A sandy agricultural soil was amended with four substances: two phenolic acids differing in their antioxidant capacity (AOC) (acetovanillone with no AOC, ferulic acid with large AOC), Trolox, an analogue of vitamin E (large AOC), and β‐sitosterol (no AOC, but potential antimicrobial properties). The two compounds with large AOC (ferulic acid and Trolox) showed no significant inhibition of C and net N mineralization; and the Trolox amendment actually caused a significant increase in C and net N mineralization after 7 d of incubation. Acetovanillone with no measurable AOC caused a significant increase in C mineralization (109% of substance C added), indicating degradation of the substance, and a very pronounced negative net N mineralization within 7 d (–356%), which was interpreted as N immobilization. Only β‐sitosterol showed strong inhibition of net N mineralization after 7 and 14 d (–59% and –26%, respectively) which was not interpreted as N immobilization, since there was no concomitant increase in C mineralization. Thus, an antimicrobial effect of β‐sitosterol specificly on microorganisms of the N cycle was suggested, but there was no clear inhibitory effect caused by the antioxidant compounds.  相似文献

Background

Sagittal ratio values (SRVs) of cervical vertebrae are used for ante‐mortem diagnosis of cervical vertebral stenotic myelopathy, but intraobserver and interobserver variability in measurement may influence radiographic interpretation of vertebral stenosis in horses with neurological disease.

Objectives

To determine intraobserver repeatability in SRVs, intra‐ and interobserver agreement in SRVs and whether or not agreement was influenced by animal age.

Animals

Forty‐two horses (>1 year old) with neurological disease from which laterolateral computed radiographic images of C2–C7 were obtained.

Methods

Four observers made measurements from C2 to C7 for each horse and interobserver agreement for intra‐ and intervertebral SRVs was determined using Bland–Altman analysis (acceptable agreement: limits of agreement [LOA] ≤ 0.05) on all horses and those ≤3 (n = 25) and >3 (n = 17) years old. Each observer also made repeated measurements for 10 horses and intraobserver repeatability and agreement were determined.

Results

Adequate intraobserver repeatability was achieved for 6 sites. Within observers, paired measurements had a median difference ≤5.7%, but a large range in differences often occurred, most frequently at intervertebral sites. For C5, C6, C7, and C3–4, LOA ≤ 0.05 were achieved by at least 1 observer. With the exception of C5 for 1 pair, LOA were >0.05 for interobserver agreement, regardless of animal age. LOA were largest at intervertebral sites.

Conclusions and Clinical Importance

Within and between observers, measurement error may limit the diagnostic accuracy of SRVs and result in discrepancies of diagnosis and treatment and warrants consideration when used clinically in horses with neurological disease.  相似文献

An automated reticulocyte counting method that used a flow cytometer and the nucleic acid staining dye, thiazole orange, was developed. Anticoagulated (EDTA) blood specimens were suitable for flow cytometric reticulocyte counting when stored at 4 C for 96 hours after collection. Thiazole orange-stained samples were stable for 5.5 hours after staining when stored capped at 20 C and protected from light. Flow cytometric and manual microscopic reticulocyte counts were compared for counts in the 0.27 to 5.32% range (as determined by flow cytometry) and 0.10 to 4.90% range (as determined by 1 technician). Although the results of flow cytometric analysis generally correlated well (r = 0.821) with manual counts, there was poor correlation between the procedures for counts less than or equal to 2.0% (r less than or equal to 0.272). Linearity of flow cytometric counts over the range 0.27 to 14.46% was excellent (r = 0.999). Within-run precision of flow cytometric counts (% coefficient of variation [cv] = 3 to 5) was superior to manual microscopic counts obtained by one technician (% cv = 19 to 23) and to manual microscopic counts, which were an average of counts done by 3 technicians (% cv = 8 to 18). Comparable flow cytometric counts were obtained by counting 50,000 or 100,000 blood cells in the flow cytometer.  相似文献

OBJECTIVE: To determine if the clinical classification of intermediate footrot (IFR) is changed to virulent footrot (VFR) by a transfer of the infected flock to a region where climatic conditions are more favourable for the transmission of the disease. DESIGN: Clinical examination of two groups of Merino wethers infected with IFR; one group of 309 in a region considered less favourable for footrot and another group of 343 at a second site considered more favourable. PROCEDURES: After characterising the form of footrot at the first site, infection was established at the second site by mixing 142 wethers from the first site with 201 unrelated wethers considered to be free of IFR and VFR. Observations of clinical characteristics were made over a 16 month period during which an outbreak of footrot occurred. Clinical assessments were made by inspecting every foot of every sheep at regular intervals and allocating a footscore. Evidence that the same clonal lines of D. nodosus were responsible for the footrot at both sites was provided by serotyping of isolates and using omp gene RFLP as a molecular epidemiological tool. RESULTS: The disease at the first site was classified as IFR because 7% of the sheep developed a maximum footscore (MFS) of 4, the most severe category, despite relatively low rates of transmission. When the outbreak occurred at the second site, which was more suitable for footrot transmission, the maximum proportion of the flock that developed a MFS of 4 was 3.6%, confirming the initial classification of IFR. CONCLUSIONS: When a flock infected with IFR was moved to a region where climatic conditions were more favourable for footrot transmission, the clinical classification of the disease remained the same in both the original flock and in sheep exposed to the infection for the first time.  相似文献
