Changes in VP2 gene during the attenuation of very virulent infectious bursal disease virus strain Gx isolated in China

Very virulent (vv) infectious bursal disease virus (IBDV) Gx strain with high pathogenicity was attenuated through replication in specific-pathogen-free (SPF) chicken embryos and in chicken embryo fibroblast (CEF) cell cultures. The changes in VP2 nucleotide and the deduced amino acid sequences were obtained during attenuation of vvIBDV in CEF culture. Sequence analysis of selected passages from numbers 0 to 20 in CEFs (designated here Gx to CEF-20) showed that no changes were detectable in the VP2 gene before CEF-7. There were a few changes in the nucleotide sequence of the VP2 gene but no amino acid substitutions at CEF-8. The virus of CEF-9 was an intermediate with some amino acid changes that possibly were related to virulence. CEF-10 virus had become similar to CU-1 strain. The VP2 gene sequence remained the same from CEF-10 to CEF-20. The results of pathogenicity tests showed that the mortalities of Gx, CEF-5, CEF-8, and CEF-9 in 4-wk-old SPF chickens were 64%, 60%, 60%, and 32%, respectively; whereas CEF-10, CEF-15, and CEF-20 were nonpathogenic. Virus neutralization tests with Gx strain showed that the antigenicities are similar from Gx to CEF-20.     

Effects of elevated temperature in vivo on the maturational and developmental competence of porcine germinal vesicle stage oocytes

Postslaughter processing of sow carcasses results in the ovaries being exposed to temperatures of 41.3 to 42.1 degrees C within a 30-min time frame. This study investigated whether the maturational and developmental competence of the recovered germinal vesicle stage oocytes could be compromised by post-slaughter processing. The results showed that the in vitro maturation rates of GV stage oocytes exposed to elevated temperature did not significantly differ from the corresponding controls (74.1 vs. 75.8%). Immunocytochemical staining revealed that elevated temperature did not adversely affect metaphase II spindle formation but resulted in extensive disruption of oocyte cytoskeletal organization. This, in turn, had a detrimental effect on parthenogenetic development compared with the corresponding nonheat-treated controls (cleavage rate = 27.7 vs. 65.3%, P < 0.01; blastulation rate = 6.7 vs. 20.6%, P < 0.01). Hence, transient exposure to elevated temperature during slaughter did not have any detrimental effects on nuclear maturation per se, but it did result in extensive cytoskeletal damage, which in turn drastically decreased the developmental competence.     

几种药剂防治水稻飞虱的田间试验

比较了几种药剂对水稻稻飞虱的防治效果:采用10%吡虫啉WP可以最经济、有效的控制飞虱的扩展和发生,对严重发生的田块可适当添加50%敌敌畏EC提高效果。     

胶孢炭疽菌的血清学研究

以4种胶孢炭疽菌的分生孢子为免疫原免疫家兔制备抗血清,采用琼脂双扩散和对流免疫电泳的方法对不同来源的20株炭疽菌进行了免疫学对比研究。结果表明:4种抗血清可分别与15个同种胶孢炭疽菌抗原中的大多数发生阳性反应,而只能与少部分不同种炭疽菌的抗原发生阳性反应。这说明同种的胶孢炭疽菌不同菌株间的亲缘关系较近,而不同种的炭疽菌间的亲缘关系较远。     

紫叶稠李育苗方法研究

紫叶稠李是新引进沈阳地区的具有较高观赏与应用价值的彩叶树种.为准确指导育苗生产,我们在林职院林盛基地和沈阳东陵区君宝彩叶苗圃进行了芽接、枝接、扦插等方面的试验,结果表明,芽接方法以T芽接、嵌芽接成活率最高;T芽接以6月效果最好,嵌芽接以4～5月及9月上旬效果最好;枝接以劈接、插皮接成活率最高;枝接以涂保湿剂效果最好;硬枝和软枝扦插均不适宜.     

High-resolution three-dimensional imaging of dislocations

Dislocations and their interactions govern the properties of many materials, ranging from work hardening in metals to device pathology in semiconductor laser diodes. However, conventional electron micrographs are simply two-dimensional projections of three-dimensional (3D) structures, and even stereo microscopy cannot reveal the true 3D complexity of defect structures. Here, we describe an electron tomographic method that yields 3D reconstructions of dislocation networks with a spatial resolution three orders of magnitude better than previous work. We illustrate the method's success with a study of dislocations in a GaN epilayer, where dislocation densities of 1010 per square centimeter are common.     

小菜蛾为害不同生育期秋甘蓝对产量的影响及经济阈值研究

采用笼罩法测定了小菜蛾在秋甘蓝不同生育期为害造成的产量损失,建立了产量损失率与小菜蛾虫口密度的回归关系式。结果表明,在秋甘蓝苗期、莲座期、结球始期和结球中后期秋甘蓝损失率(y)与小菜蛾虫口密度(x)关系式分别为:y=9.480 6x-2.586 2,y=3.621 8x-0.790 2,y=-0.210 7x2+6.698 1x-1.906 5和y=0.081 1x2-0.147 9x+0.405 4。根据经济阈值模型,秋甘蓝不同生育阶段的小菜蛾幼虫经济阈值分别为苗期0.54头/株,莲座期0.91头/株,结球始期0.67头/株,结球中后期6.08头/株。该研究为秋甘蓝小菜蛾的防治策略和经济阈值的确定提供了科学依据。     

PEG介导的玉米弯孢叶斑病菌遗传转化

为了探索不同酶系组成对玉米弯孢叶斑病菌原生质体制备的影响,建立该病菌原生质体遗传转化系统,采用酶系混合物裂解菌丝体制备原生质体,采用PEG介导方法进行原生质遗传转化,通过PCR和Southern blotting技术对转化子进行验证。结果表明:1%溶壁酶+1%蜗牛酶+1%纤维素酶混合酶系为玉米弯孢叶斑病菌原生质体制备的最佳酶系,可以产生原生质体6.78×106 cfu/mL。用PEG介导方法转化共获得16个稳定的转化子,从中随机挑取5个转化子发现质粒pV2已被成功整合到基因组中。本研究获得了制备玉米弯孢叶斑病菌原生质体的最佳酶系,建立了PEG介导的原生质体遗传转化体系,为开展该菌致病相关基因克隆和基因功能研究提供了一种手段。     

古尔班通古特沙漠南部梭梭鼠害特征及防治生态阈值研究

鼠类的"有害"或"无害"与其生境密切相关,文中以位于古尔班通古特沙漠南部的梭梭鼠害防治示范区和非示范区为调查样地,对比分析了两种生境中梭梭林的鼠害特征,进而探讨了沙漠区梭梭鼠害防治的生态阈值。研究发现:(1)古尔班通古特沙漠南部梭梭鼠害整体较轻。由鼠类引起的梭梭死亡率,非示范区平均为4.29%,示范区平均为2.12%。(2)梭梭的生长状况是吸引鼠类前来取食的因素。梭梭的基径、高度、冠幅与其冠下有效鼠洞数和鼠洞总数都成极显著的线性正相关关系,其生长状况的差异性决定了鼠类危害的程度。(3)鼠类对梭梭基部的取食较轻,而对梭梭各级侧枝的取食不仅具有选择性,还受生境和自身种群数量的影响。梭梭基部的轻度危害比率大于中度危害和重度危害比率,而梭梭侧枝、一级侧枝、二级侧枝的危害程度与鼠洞总数呈显著相关关系,有效鼠洞数只与梭梭二级侧枝的危害程度呈显著相关关系。(4)单株梭梭整株危害程度与其冠下鼠洞总数呈指数函数关系,当鼠洞总数>5个/株时,梭梭整株危害程度开始加剧,故该研究认为梭梭鼠害防治的生态阈值为鼠洞总数为5个/株。     

辽宁口岸进境原木疫情分析及检疫监管建议

本文对2001～2010年间辽宁口岸进境原木疫情情况进行分析,探讨了进境原木截获有害生物种类特点、疫情年发生规律特点并对其形成原因进行了深入分析。同时,对今后的检疫监管工作提出了相关建议。
