Objectives

1) To determine which peripheral artery commonly used for invasive arterial blood pressure (IBP) monitoring yields the least bias when compared with noninvasive blood pressure (NIBP) values obtained at the antebrachium of the dog, and 2) to identify and describe differences in systolic (SAP), mean (MAP) and diastolic arterial pressures (DAP) among different anatomical locations.

Study design

Prospective experimental study.

Animals

Twenty adult hound dogs weighing 24.5 ± 1.1 kg (mean ± standard deviation).

Methods

Four peripheral arteries—dorsal pedal, median caudal, intermediate auricular and superficial palmar arteries—were catheterized with 20 gauge, 3.8 cm catheters. One NIBP cuff was placed in the middle third of the antebrachium. Four sets of IBP and NIBP measurements were simultaneously collected every 2 minutes. A linear mixed model was performed to analyze the collected data.

Results

IBP values varied depending on the arterial catheterization site. The difference was greater for SAP. NIBP measured at the antebrachium had the best agreement with IBP measured at the median caudal artery.

Conclusion and clinical relevance

IBP varies among anatomical locations. The smallest bias and narrowest limits of agreement were obtained at the median caudal artery, providing the best overall agreement with the equipment studied. The median caudal artery may be the preferable anatomical location for clinical comparison studies between IBP and NIBP in dogs when the cuff is on the antebrachium.  相似文献

Abiotic stress limits crop productivity worldwide, making stress‐resistant crops a major goal for improving food security. Stress resistance is often assessed by comparing genotypes under a single control and stressed level of an abiotic factor, then ranking the genotypes’ overall performance, using a metric like geometric mean productivity (GMP). However, abiotic stress is rarely a presence/absence phenomenon, but often varies continuously. Here, we tested whether the common practice of assessing performance using a single control and stress level is predictive of performance across a range of stress levels. Twelve sunflower genotypes were grown in the greenhouse under one of eight nutrient levels (from non‐limiting to severely limiting) and assessed for performance (shoot dry mass at the budding stage) and related traits at both seedling and budding stages. Contrary to expectations, genotype rankings for GMP were consistent regardless of the two nutrient levels used to calculate GMP. This suggests that, for nutrient stress in sunflower, studies using an arbitrary control and stress level are broadly representative: genotypes with high GMP exhibit high GMP regardless of the nutrient levels used. In addition, GMP values were highly correlated with performance in the respective nutrient levels from which they were calculated; thus, higher GMP for a given genotype indicates that it performed well (relative to other genotypes) under both stressed and non‐stressed conditions. Together, these findings strongly support the current paradigm of comparing genotypes at a single control and stress level as a valid strategy for screening for nutrient stress resistance in sunflower. However, few traits were consistently associated with performance (shoot dry mass) at either high‐ or low‐nutrient availability, and seedling traits were rarely predictive of budding‐stage traits and performance, suggesting further studies are needed to determine the mechanisms underlying nutrient stress resistance in this important oilseed crop.  相似文献

Pasture legumes that persist under challenging agroecological conditions are crucial to ensure high lamb growth rates in dryland pastures. Pasture and lamb production from binary and diverse mixtures (Mix) of tall fescue (TF) with white clover (Whc), balansa clover (Bc), subterranean clover (Sc) and birdsfoot trefoil (Bft) were compared in a summer‐dry hill site in Corvallis, Oregon over a two‐year period. In 2018, all pasture combinations provided similar lamb liveweight gains (LWG, mean 177 g/day) in the first half of spring. Lambs in TF‐Bc and TF‐Mix pastures grew 31 to 41 g/day faster than those grazing TF‐Sc, TF‐Whc and TF‐Bft in the second half of spring (p < .05). Overall, TF‐Bc and TF‐Mix had higher (p < .05) legume contents (32% and 37% respectively) compared to other pasture combinations. In spring 2019, lambs that grazed the TF‐Mix and TF‐Whc pastures had higher LWG than those on other pastures (p < .05). The superior lamb growth rates were associated with the higher legume content and pasture quality maintained into the late spring period. Overall, the legume content of all pastures decreased over the course of the two‐year trial, with the decline being substantial for balansa clover. The present study confirmed that a high legume content of pastures leads to greater lamb growth rates. Total annual yields of pastures that had greater legume contents were superior to others (p < .05). Thus, a combination of self‐regenerating annual clovers with perennial legumes in pasture mixtures may ensure a higher legume content and longer persistence in dryland hill pastures.  相似文献

Purpose

Gensburg Markham Prairie (GMP), a high-quality tallgrass prairie and wetland in the Chicago area, is surrounded by high-traffic highways and residential communities. Anthropogenic inputs are a concern given observed ecosystem degradation around the periphery of the prairie. To understand soil quality and the potential for habitat degradation in GMP, we evaluated concentrations of metals and phosphorus in soil profiles, identified possible sources of contamination, and assessed the likelihood that contamination would impact the prairie ecosystem.

Materials and methods

We collected 15 intact soil cores (to a depth of 120 cm) and 22 surface soil samples (0–10-cm depth), determined the soil type, and measured pH, organic matter, and concentrations of Pb, Cu, Zn, Mg, Fe, K, Ca, and P using ICP-OES. To quantify soil pollution, we calculated the element enrichment factor (EF) and single element pollution index (SEPI) for each sample. We also used principal component analysis (PCA) to interpret relationships between site variables, elemental concentrations, and sources of metals.

Results and discussion

Heavy metals (Pb, Cu, and Zn) and P were found to accumulate in surface samples. The mean EF values for Pb, Cu, Zn, and P revealed significant enrichment of these elements in the soil profiles. However, SEPI analyses indicate that GMP soils have a low level of contamination. PCA revealed that concentrations are highest for samples with high organic matter content near the ground surface and that Pb, Cu, and Zn have common sources of pollution.

Conclusions

We observed enrichment of Pb, Cu, Zn, and P because of deposition from the urban built environment, but the enrichment levels are low enough that they are not expected to negatively impact the ecosystem of GMP. The detailed soil chemistry data constitute a spatial contamination map that can be used to assess potential long-term impacts on the ecosystem, such as reduction of plant growth and species diversity, and inform site management and biodiversity conservation efforts.

Streptococcus agalactiae is a major bacterial pathogen that is the cause of serious economic losses in many species of freshwater, marine and estuarine fish worldwide. A highly efficacious S. agalactiae vaccine was developed using extracellular products (ECP) and formalin-killed whole cells of S. agalactiae. The vaccine efficacy following storage of S. agalactiae ECP and formalin-killed S. agalactiae cells at 4 degrees C for 1 year was determined. The stored ECP containing S. agalactiae formalin-killed cells failed to prevent morbidity and mortality among the vaccinated fish, and the relative percentage survival was 29. Serum antibody responses of the stored ECP and freshly prepared ECP against soluble whole cell extract of S. agalactiae indicated that significantly less antibody was produced in fish immunized with stored ECP and S. agalactiae cells than in those fish immunized with freshly prepared ECP and S. agalactiae cells at day 31 post-vaccination. Silver staining of sodium dodecyl sulphate-polyacrylamide gels and immunostaining of Western blots with tilapia antiserum to S. agalactiae revealed that predominant 54 and 55 kDa bands were present in the freshly prepared ECP fraction. The 55 kDa band was absent from the stored ECP and new bands below 54 kDa appeared on the Western blot. The results of this study on S. agalactiae ECP provide evidence for a correlation between protection and antibody production to ECP and for the importance of the 55 kDa ECP antigen for vaccine efficacy.  相似文献

Hybrid striped bass (HSB) were immunized with bovine serum albumin (BSA) and the specific anti-BSA immunoglobulin (Ig) was affinity purified from the resulting serum by means of an agarose gel-BSA column. The native Ig had an apparent molecular size of 893 KDD, by size exclusion chromatography, and when examined by polyacrylamide gel electrophoresis (PAGE) under denaturing conditions, resolved to heavy (H) and light (L) chains of 76 and 27 KDD, respectively. Affinity purified native HSB Ig was used to immunize a goat which produced specific anti-HSB Ig antibody (Ab). Purified native HSB Ig was also used to produce two murine monoclonal antibodies (mAbs) with specific affinities for H and L chain moieties of the HSB Ig molecule. Both polyclonal and monoclonal antibodies could be used individually in an indirect enzyme-linked immunosorbent assay (ELISA) to measure specific anti-BSA Ig in HSB serum. These antibodies could also be used in combination to measure total Ig in a capture ELISA format. Using both assays, the kinetics of the humoral immune response of HSB was measured for 98 days following two injections of BSA.  相似文献

A 15‐wk study was conducted to evaluate the effect of supplemental menhaden fish oil levels and feeding duration on growth performance and tissue proximate and fatty acid (FA) compositions of juvenile channel catfish, Ictalurus punctatus. Dietary fish oil levels had no effect on final weight gain, feed efficiency, and survival of channel catfish. Tissue lipid contents were directly correlated to dietary lipid levels, while moisture contents were inversely related to dietary lipid levels. Fillet moisture contents progressively decreased, whereas fillet lipid increased with increasing feeding duration. Significant increase in saturated and total n‐3 FAs and decrease in monoenoic and total n‐6 FA in whole body and fillet were observed at each incremental level of dietary fish oil. Percentages of n‐3 and n‐3 highly unsaturated fatty acids in fillet of fish fed the control and 3% fish oil diets decreased with increasing feeding periods, whereas those of fish fed 6 or 9% added fish oil diets remained stable or increased. Ratios of n‐3/n‐6 were statistically comparable throughout the 15‐wk feeding. When expressed in terms of mg/g of fillet, the highest concentration of n‐3 was obtained in fillets of fish fed the 9% added fish oil diet for 15 wk.  相似文献

We present a draft sequence of the genome of Aedes aegypti, the primary vector for yellow fever and dengue fever, which at approximately 1376 million base pairs is about 5 times the size of the genome of the malaria vector Anopheles gambiae. Nearly 50% of the Ae. aegypti genome consists of transposable elements. These contribute to a factor of approximately 4 to 6 increase in average gene length and in sizes of intergenic regions relative to An. gambiae and Drosophila melanogaster. Nonetheless, chromosomal synteny is generally maintained among all three insects, although conservation of orthologous gene order is higher (by a factor of approximately 2) between the mosquito species than between either of them and the fruit fly. An increase in genes encoding odorant binding, cytochrome P450, and cuticle domains relative to An. gambiae suggests that members of these protein families underpin some of the biological differences between the two mosquito species.  相似文献
