The influence of reaction conditions for oligosaccharide synthesis from stachyose using a commercial enzymatic preparation from Aspergillus aculeatus (Pectinex Ultra SP-L) was studied. Oligosaccharides were analyzed by gas chromatography with flame ionization detection (GC-FID) and matrix-assisted laser desorption/ionization-time-of-flight-mass spectrometry (MALDI-TOF-MS). Galactosyl-melibiose (DP(3)) was synthesized as a result of fructosidase activity, whereas fructosyl-stachyose (DP(5)) and difructosyl-stachyose (DP(6)) were formed as a consequence of the fructosyltransferase activity of Pectinex Ultra SP-L. The optimal reaction conditions for the synthesis of penta- and hexasaccharides were 60 °C, pH 5.5, 600 mg/mL stachyose, and 34 U/mL enzyme. Reaction time played an important role in oligosaccharide mixture composition constituted by 20% DP(5), 0.7% DP(6), 55% stachyose, 21% galactosyl-melibiose, and 1% monosaccharides after 1 h and 16% DP(5), 4% DP(6), 27% stachyose, 44% galactosyl-melibiose, and 2% monosaccharides after 3 h. In conclusion, stachyose could be used as a substrate for the enzymatic synthesis of new oligosaccharides that may open new opportunities in the development of future prebiotics.  相似文献

The influence of pond management on water quality for drip-irrigated crops was studied throughout a field survey and a mesocosm experiment. Water sources were pooled into two groups: ground or surface water (GW/SW) and recycled wastewater. Pond covering, which was limited to about a quarter of them, improved water quality by reducing phytoplankton biomass. However, biocide applications and pond dredging were ineffective at improving in-pond water quality. Dredging did not reduce the concentrations of planktonic chlorophyll a or total suspended solids (TSS) in GW/SW fed ponds, whereas biocide applications increased both parameters. Field and experimental data proved that the two predominant taxa of submerged aquatic vegetation (SAV) found in ponds (Potamogeton pectinatus and Chara spp.) improved water quality by increasing water oxygenation and decreasing chlorophyll a and TSS concentrations. Preserving SAV (especially Chara spp.) appears to be an environment-friendly, cost-effective and recommendable alternative strategy for irrigation pond management.  相似文献

Productive performances of triploid (3n) and diploid (2n) shi drum (Umbrina cirrosa L.) were evaluated over an experimental period of 76 days. Fish, selected according to body weight (196 specimens from each ploidy), were reared in tanks (two for each thesis) under the same environmental and dietary conditions. A practical extruded diet (crude protein: 43.4%; ether extract: 19.3%) was offered to fish according to a feeding rate that varied from 1% to 1.5% of live weight. Both at the beginning and at the end of the trials, samples of fish were submitted to morphometrics and chemical analysis. The final body weight and the specific growth rate of triploid fish were significantly lower than those of fish diploid. The feed efficiency of triploids showed a reduction in crude protein retention. Juvenile triploids evidenced a larger amount of coelomatic fat, and their gonads were atrophic. The whole‐body proximate composition of 3n fish was characterized by a higher ether extract and a lower crude protein content than 2n fish. Triploids had higher liver lipid content; there were no significant differences in viscera proximate composition. Sum, the results showed that chromosome set manipulation in this species can reduce productive performances at a juvenile stage, influencing some morphometric traits.  相似文献

Difficulties to develop an easy and reproducible protocol to get healthy and well formed plants from somatic embryos of papaya (Carica papaya L.) had included low germination, callus production at the base of the embryo radicle and the occurrence of hyperhydric plantlets among others, and by consequence unsuccessful transfer to the field. With the aim of improving a propagation method, the effects of light quality, gelling agent and phloridzin concentration on the germination of somatic embryos of hermaphrodite C. papaya L. var. Maradol were studied. Somatic embryos were grown on half strength MS medium, with the addition of Chen vitamins [Chen, M.H., Wang, P.J., Maeda, E., 1987. Somatic embryogenesis and plant regeneration in Carica papaya L. tissue culture derived from root explants. Plant Cell Rep. 6, 348–351], solidified with three distinct gelling agents: Sigma® Agar–Agar, Difco® Bacto agar and Phytagel®; supplemented with phloridzin and exposed to different light qualities: blue (54 μmol m−2 s−1), red (65 μmol m−2 s−1), gro-lux (68 μmol m−2 s−1), red + blue, white (32 μmol m−2 s−1) and wide spectrum (49 μmol m−2 s−1) during a period of 4 weeks. Results show that light quality and gelling agent had important effects on germination and plant growth, while 3.0 mg L−1 phloridzin had an important role on germination as well as in root development. Somatic embryos exposed to white light, culture medium solidified with 3.0 mg L−1 phytagel and 3.0 mg L−1 phloridzin showed longer roots. Meanwhile, germination and plant length were promoted on an improved culture medium solidified with 7.5 g L−1 Difco® Bacto agar, 3.0 mg L−1 phloridzin and exposed to gro-lux lamps. Under these conditions, 70% of somatic embryos germinated and developed normal roots without hyperhydricity. The regenerated plantlets with well developed roots and shoots were successfully transferred to a greenhouse with a survival rate of 95%.  相似文献

Two new series of nortopsentin analogues, in which the imidazole ring of the natural product was replaced by thiazole and indole units were both substituted by 7-azaindole moieties or one indole unit was replaced by a 6-azaindole portion, were efficiently synthesized. Compounds belonging to both series inhibited the growth of HCT-116 colorectal cancer cells at low micromolar concentrations, whereas they did not affect the viability of normal-like intestinal cells. A compound of the former series induced apoptosis, evident as externalization of plasma membrane phosphatidylserine (PS), and changes of mitochondrial trans-membrane potential, while blocking the cell cycle in G2/M phase. In contrast, a derivative of the latter series elicited distinct responses in accordance with the dose. Thus, low concentrations (GI30) induced morphological changes characteristic of autophagic death with massive formation of cytoplasmic acid vacuoles without apparent loss of nuclear material, and with arrest of cell cycle at the G1 phase, whereas higher concentrations (GI70) induced apoptosis with arrest of cell cycle at the G1 phase.  相似文献

Intraventricular ependymoma is a rare type of feline intracranial neoplasia and published information on magnetic resonance imaging (MRI) characteristics is currently lacking. The purpose of this retrospective case series study was to describe the clinical and MRI characteristics of histopathologically confirmed intraventricular ependymomas in a group of cats. Five cats met inclusion criteria. In relation to normal gray matter, ependymomas appeared hyperintense on T2W, T2W‐FLAIR, PD, and DW‐EPI images; isointense on ADC images; and had subtle to strong contrast enhancement. Some variability was seen on T2*GRE and on T1W images with masses being isointense to hyperintense. Four ependymomas were small and homogeneous, and one was centrally cavitated. All cats had obstructive hydrocephalus, transtentorial herniation, and foramen magnum herniation. Perilesional edema was identified in most cats but was questionable in one. Intraventricular ependymoma should be considered as a differential diagnosis for cats with this combination of MRI signs.  相似文献
