The arrangement of a plant's roots in the soil determines the ability of the plant to resist uprooting. We have investigated the influence of root morphology on anchorage using simple patterns of root systems and numerical simulation. The form and mechanical properties of roots were derived from results found in the literature. Major parameters determining soil characteristics, root patterns and strength were varied so that their influence could be evaluated. The design of the experimental method we used generated an optimal number of configurations of different root systems, the tensile resistances of which were calculated by two‐dimensional finite element analysis. We could quantify the influence of specific parameters, e.g. branching angle, number of lateral roots and soil cohesion, as well as global parameters such as total contact area, basal diameter and volume of the whole root system. We found that the number of roots and the diameter of roots were major components affecting the resistance to uprooting. The combination of topology and biomass explained 70% of the variation of tensile resistance. 相似文献

Cyclosporine is a powerful immunosuppressive drug that is being used with increasing frequency to treat a wide range of immune-mediated diseases in the dog. To date, ideal dosing protocols that will achieve immunosuppression with cyclosporine in dogs remain unclear, and standard methods that can measure effectiveness of immunosuppression have not been established. The aim of our study was to evaluate the effects of in vitro cyclosporine exposure on a panel of molecules expressed by activated T cells to ascertain their potential as biomarkers of immunosuppression in dogs. Blood was drawn from six healthy dogs, and peripheral blood mononuclear cells (PBMC) were isolated and activated. Half of the cells were incubated with 200 ng/mL cyclosporine prior to activation, and the other half were not exposed to cyclosporine. Samples were analyzed using flow cytometry, and the expression of intracellular cytokines IL-2, IL-4, and IFN-γ was evaluated after 6, 12, and 24h of drug exposure. Each cytokine exhibited a time-dependent suppression profile, and all but two samples activated in the presence of cyclosporine showed lower cytokine expression than untreated controls. We also evaluated the expression of the surface T cell activation molecules CD25 and CD95 by flow cytometry after 36 h of drug exposure. Expression of these surface molecules decreased significantly when activated in the presence of cyclosporine. Our results suggest that suppressed expression of the markers related to T cell activation could potentially be utilized as an indicator of the efficacy of cyclosporine therapy in dogs. 相似文献

The molecular motion of water was studied in glucono-δ-lactone-acidified skim milk powder (SMP) solutions with various pH values and dry matter contents. NMR relaxometry measurements revealed that lowering the pH in SMP solutions affected 17O and 1H T2 relaxation rates almost identically. Consequently, the present study indicates that the proteins present in the samples do not affect the 1H relaxation behavior markedly, even at relatively high SMP concentrations (15-25%). Comparison of rheological measurements and NMR measurements suggested that the collapse of κ-casein during acidification could contribute to the initial decrease in 17O and 1H relaxation rate in the pH range between 6.6 and 5.5 for 15% SMP and in the pH range between 6.6 and 5.9 for 25% SMP. However, below pH 5.5 the viscosity and 17O and 1H NMR relaxation rates did not correlate, revealing that the aggregation of casein micelles, which increases viscosity below pH 5.5, does not involve major repartitioning of water. 相似文献

Soil water erosion and shallow landslides depend on aggregate stability and soil shear strength. We investigated the effect of vegetation on both soil aggregate stability and shear strength (through direct shear tests) in former croplands converted to vegetated erosion protection areas within the context of China's sloping land conversion programme. Four treatments were analysed in plots comprised of (i) 4 year old crop trees, Vernicia fordii, where understory vegetation was removed; (ii) V. fordii and the dominant understory species Artemisia codonocephala; (iii) only A. codonocephala and (iv) no vegetation. Soil samples were taken at depths of 0–5 cm and 45–50 cm. Root length density (RLD) in five diameter classes was measured, soil organic carbon (SOC), hot water extractable carbon (HWEC), texture and Fe and Al oxides were also measured. We found that mean weight diameter after slow wetting (MWDSW) in the A-horizon, was significantly greater (0.94–1.01 mm) when A. codonocephala was present compared to plots without A. codonocephala (0.57–0.59 mm). SOC and RLD in the smallest diameter class (< 0.5 mm), were the variables which best explained variability in MWDSW. A significant positive linear relationship existed between MWDSW and soil cohesion but not with internal angle of friction. As herbaceous vegetation was more efficient than trees in improving aggregate stability, this result suggests that the mechanisms involved include modifications of the cohesive forces between soil particles adjacent to plant roots and located in the enriched in SOC rhizosphere, thus also affecting shear strength of the corresponding soil volume. Thus, vegetation stabilised soil under different hierarchical levels of aggregate organisation, i.e. intra- and inter-aggregate. Our results have implications for the efficacy of techniques used in land conversion programs dedicated to control of soil erosion and shallow landslides. We suggest that mixtures of different plant functional types would improve soil conservation on slopes, by reducing both surface water erosion and shallow substrate mass movement. Planting trees for cropping or logging, and removing understory vegetation is most likely detrimental to soil conservation. 相似文献

Intake and digestion in cattle fed warm- or cool-season grass hay with or without low-level grain supplementation were studied with a 2 x 2 factorial arrangement of treatments in two 4 x 4 Latin square experiments. In Exp. 1, four cannulated beef cows (396 kg) were given Bermuda grass (B) or orchard grass (OG) hay at 1.5% body weight (BW) with 0 or .3% BW of ground corn (C; dry matter). Bermuda grass contained 12.1% crude protein, 79.3% neutral detergent fiber (NDF) and 5.5% acid detergent lignin (ADL); OG contained 10.6% crude protein, 82.4% NDF and 8.1% ADL. An interaction (P less than .07) between forage type and C supplementation was noted for microbial N entering the duodenum; C supplementation had a positive effect with B (30% increase) and little effect with OG. Corn supplementation did not affect ruminal NDF digestion with B, but it elicited an increase with OG (interaction, P less than .05; means were 60.7, 60.1, 61.5 and 66.3%). In the second experiment, growing dairy steers (196 kg) were given ad libitum access to similar B or OG hays and were fed 0 or .5% BW of C. Dry matter (DM) intake was lower for OG than for B (P less than .05) and was lower with than without C (P less than .06; means were 2.76, 2.56, 2.53 and 2.30% BW for B, BC, OG and OGC, respectively). Total tract organic matter digestion (%) was higher for OG than for B (P less than .10) and was higher with than without C (means were 54.7, 61.5, 60.4 and 65.3%). In conclusion, chemical constituents such as NDF may govern differences in intake between warm- and cool-season grasses, but physical attributes of the forages appear more important to digestion. 相似文献

A direct enzyme-linked antiglobulin test (DELAT) was used to measure the levels of red blood cell (RBC) bound IgG, IgM, IgA and C3 in dogs with autoimmune haemolytic anaemia (AIHA). At presentation, one or more DELAT parameters was raised in each AIHA case, and the RBC were typically coated with immunoglobulin of more than one class, together with C3. There was no relationship between the levels of RBC-bound IgG, IgM or IgA and the severity of the anaemia, although a significant negative correlation (rs = -0.66, P < 0.02) was found between bound C3 and blood haemoglobin concentration. These results indicate that the level of sensitisation of erythrocytes with IgG alone is not a reliable predictor of the severity of haemolysis in different cases, and that the pathogenesis of AIHA can be complex, involving multiple immunoglobulin classes and complement in the destruction of RBC. A significant relationship (rs = 0.63, P < 0.02) was found between serum IgG concentration and haemoglobin levels, and it is suggested that this may be due to free IgG inhibiting the interaction of IgG-sensitised RBC with macrophages. Serial measurements from individual AIHA cases during treatment revealed that the levels of RBC-bound immunoglobulins fell simultaneously with improvements in anaemia. In one dog, a relapse was associated with increases in bound IgG and IgM. Transient relative reticulocytopenia at presentation was common, but was not related to the severity of the anaemia. However, in other cases there was a persistent failure to increase RBC production, which was associated with slower recovery. 相似文献

Leukocyte populations present in the discrete Peyer's patches (PP) of the pig were characterized from birth (Day 0) to day 35 after birth by immunohistochemistry and image analysis. Immediately after birth, cell membrane expression of CD2 and CD3, major histocompatibilty complex (MHC) class 11 (both SLA (swine leukocyte antigen) -DQ+ and SLA-DR+), CD21, 74-22-15 and surface immunoglobulin (sIg) were all demonstrable. Computer assisted morphometric techniques were used to confirm the significant expansion of these cell populations from birth onwards. The distribution of the cell types was not random but suggested a preferential retention of cells at specific sites. This implies a degree of organization of immunological cells within the discrete PP, enhancing the potential to mount immune responses in the most efficient manner. 相似文献
