To investigate the value of the BP26 protein in the serological diagnosis of ovine brucellosis caused by Brucella ovis, recombinant BP26 protein was produced in Echerichia coli and purified for use in an indirect enzyme-linked immunosorbent assay (I-ELISA). The majority of the recombinant protein was recovered from the supernatant of sonicated recombinant E. coli cells in a soluble form. This facilitated the purification of the recombinant BP26 protein which was achieved by using ion-exchange chromatography. After one step of purification, the purity of the recombinant BP26 protein was analyzed by using SDS-PAGE, Coomassie blue staining, and Western blot with a monoclonal antibody (MAb) directed against the BP26 protein. The degree of purity appeared satisfactory so that it could be directly used in I-ELISA. Although the recombinant BP26-ELISA appeared less useful than I-ELISA using the B. ovis hot saline (HS) extract as antigen, the high number of sera from B. ovis infected rams found positive (90%) in the recombinant BP26-I-ELISA indicated that the BP26 protein may be an additional suitable antigen for serological diagnosis of B. ovis infection in rams.  相似文献

ABSTRACT The host range of a pathogen can have special consequences on its evolution and the evolution of its virulence. For generalists, adaptation to different hosts may be conditioned by different trade-offs in the pathogen's life history and be affected by evolutionary processes that shape pathogen populations. We have examined adaptation of Cucumber mosaic virus (CMV) to different hosts, and analyzed the relationship between host adaptation and virulence. For this, six CMV isolates from central Spain from three different hosts were compared for the ability to multiply and to affect host growth. These analyses were done before and after an experimental evolution process consisting of 10 serial passages in the original host of the isolate. The differential capacity to infect different hosts was compatible with host adaptation. However, the capacity to multiply in different hosts did not provide evidence of host adaptation and was not improved after 10 passages, suggesting that fitness of the natural population of CMV was at, or near to, its maximum. No relationship was found between capacity of multiplication and virulence in any of the three different hosts. These results suggest that the "trade-off" model for the evolution of virulence may not apply to CMV.  相似文献

The side-effects of pesticides (insecticides, fungicides, herbicides and plant growth regulators) used on cotton were tested on adults and pupae of Trichogramma pretiosum Riley reared in the laboratory on two different hosts, the Angoumois grain moth (Sitotroga cerealella Olivier) and the Mediterranean flour moth (Ephestia kuehniella (Zeller)). The eggs of the host enclosing the parasitoid pupae received direct pesticide sprays, while the adults of the parasitoid were exposed to the pesticides through contact with residues on sprayed eggs offered to parasitism. Alpha-cypermethrin, carbosulfan, deltamethrin, endosulfan, profenofos and zeta-cypermethrin were highly noxious to the parasitoid, significantly reducing the percentage of emergence and parasitism of T. pretiosum developing in E. kuehniella or S. cerealella eggs. However, the pupal stage of the parasitoid developing in S. cerealella eggs was less susceptible to alpha-cypermethrin and deltamethrin. Lufenuron and metamidophos greatly reduced the percentage of adult emergence from eggs of both hosts, while novaluron only interfered on this variable when the wasps were developing in E. kuehniella eggs. However, lufenuron and monocrotophos had no effect on the parasitoid pupae of T. pretiosum developing in E. kuehniella eggs. Chlorfluazuron, diafenthiuron, diflubenzuron, fentin hydroxide, mepiquat chloride, novaluron, thiacloprid and triflumuron did not affect T. pretiosum emergence when eggs of S. cerealella enclosing pupae of the wasps were surface treated. The pesticides azoxystrobin, carbendazin + thiram, mepiquat chloride and novaluron had no effect on the ability of the wasps to parasitise E. kuehniella eggs. However, only mepiquat chloride did not affect the percentage of F1 wasps emerging from E. kuehniella eggs. The remaining pesticides moderately reduced the percentage of emergence and parasitism of the wasps when they had contact with the chemicals during their pupal or adult stage. Thus there were differences in pesticide toxicity according to the host used for parasitoid development. ● Grasslands in many regions of the world have been impacted by atmospheric nitrogen deposition. ● Nitrogen deposition commonly leads to reductions in species richness. ● Increases in biomass production is a common response to increased N deposition. ● In some parts of the world there has been limited research into the impacts of nitrogen deposition. Grasslands are globally-important ecosystems providing critical ecosystem services. The species composition and characteristics of grasslands vary considerably across the planet with a wide variety of different grasslands found. However, in many regions grasslands have been impacted by atmospheric nitrogen deposition originating from anthropogenic activities with effects on productivity, species composition and diversity widely reported. Impacts vary across grassland habitats but many show declines in species richness and increases in biomass production related to soil eutrophication and acidification. At a continental level there is considerable variation in the research effort that has been put into understanding the impacts of nitrogen deposition. In Europe, North America and parts of Asia, although there are unanswered research questions, there is a good understanding of N deposition impacts in most grassland habitats. This is not the case in other regions with large knowledge gaps in some parts of the world. This paper reviews the impacts of N deposition on grasslands around the world, highlighting recent advances and areas where research is still needed.  相似文献
