In this study, the pathogenic behavior of PRRSV 13V091 and 13V117, isolated in 2013 from two different Belgian farms with enzootic respiratory problems shortly after weaning in the nursery, were compared with the Belgian strain 07V063 isolated in 2007. Full-length genome sequencing was performed to identify their origin. Twelve weeks-old pigs were inoculated intranasally (IN) with 13V091, 13V117 or 07V063 (9 pigs/group). At 10 days post inoculation (dpi), 4 animals from each group were euthanized and tissues were collected for pathology, virological and serological analysis. 13V091 infection resulted in the highest respiratory disease scores and longest period of fever. Gross lung lesions were more pronounced for 13V091 (13%), than for 13V117 (7%) and 07V063 (11%). The nasal shedding and viremia was also most extensive with 13V091. The 13V091 group showed the highest virus replication in conchae, tonsils and retropharyngeal lymph nodes. 13V117 infection resulted in the lowest virus replication in lymphoid tissues. 13V091 showed higher numbers of sialoadhesin infected cells/mm2 in conchae, tonsils and spleen than 13V117 and 07V063. Neutralizing antibody response with 07V063 was stronger than with 13V091 and 13V117. It can be concluded that (i) 13V091 is a highly pathogenic type 1 subtype 1 PRRSV strain that replicates better than 07V063 and 13V117 and has a strong tropism for sialoadhesin cells and (ii) despite the close genetic relationship between 13V117 and 07V063, 13V117 has an increased nasal replication and shedding, but a decreased replication in lymphoid tissues compared to 07V063.

The polyacetylene falcarinol, isolated from carrots, has been shown to be protective against chemically induced colon cancer development in rats, but the mechanisms are not fully understood. In this study CaCo-2 cells were exposed to falcarinol (0.5-100 microM) and the effects on proliferation, DNA damage, and apoptosis investigated. Low-dose falcarinol exposure (0.5-10 microM) decreased expression of the apoptosis indicator caspase-3 concomitantly with decreased basal DNA strand breakage. Cell proliferation was increased (1-10 microM), whereas cellular attachment was unaffected by <10 microM falcarinol. At concentrations above 20 microM falcarinol, proliferation of CaCo-2 cells decreased and the number of cells expressing active caspase-3 increased simultaneously with increased cell detachment. Furthermore, DNA single-strand breakage was significantly increased at concentrations above 10 microM falcarinol. Thus, the effects of falcarinol on CaCo-2 cells appear to be biphasic, inducing pro-proliferative and apoptotic characteristics at low and high concentrations of falcarinol, respectively.  相似文献

OBJECTIVE: To examine the relationship between mental distress, academic performance and regular breakfast consumption across gender and immigration status. DESIGN: Cross-sectional population-based study. Two four-page questionnaires were filled in during two school sessions. SETTING: All junior high schools in Oslo, Norway using the classroom as the setting for the study. SUBJECTS: All 10th grade students 15-16 years olds in 2000 and 2001. Of 8316 eligible students, 7343 (88.3%) participated in the study. RESULTS: All immigrant groups, except the Western countries group, are skipping breakfast more often than Norwegian students, and girls more often than boys (27 versus 19%). After adjustment for possible confounding factors, the odds ratio (OR) for being mentally distressed when eating breakfast seldom/never compared with every day was 3.0 (2.0-4.5) for boys, 1.6 (1.2-2.1) for girls and 1.6 (1.5-2.6) for the immigrant group. The comparable OR for having low school grades was similar for boys and girls, 2.0 (1.3-3.0), and 1.6 (1.5-2.6) for the immigrant groups. CONCLUSIONS: Skipping breakfast is a common feature among 10th grade students. The implications of skipping breakfast on mental distress and academic performance are stronger for boys than girls and stronger for Norwegians compared with immigrants.  相似文献

The goal of this paper is to propose and evaluate automated image analysis methods for describing muscle cutlets in rainbow trout. The proposed automated image analysis methods were tested on a total of 983 scanned images of trout cutlets, and included quality traits such as fat percentage, flesh colour and the size of morphologically distinguishable subparts of the cutlet. A sub-sample of 50 images was randomly selected for manual segmentation of the cutlet, the dorsal fat depot and the red muscle and regions. The identification of these regions by manual and automatic image analysis correlated strongly (r = 0.97, r = 0.95 and r = 0.91, respectively). The estimated fat percentage obtained from image analysis, based on the area of visible fat and the colour of the cutlet flesh, correlated well with chemical fat percentage measured by mid-infrared transmission spectroscopy (MIT) (r = 0.78). The automated image analysis methods are therefore a reliable means of predicting the fat percentage of trout cutlets. Principal component analysis (PCA) loading plots were used to identify subsets of variables from the image analysis of special significance for further studies; cutlet area, dorsal fat depot area, red muscle area, back height, cutlet width, and width of left and right abdomen wall were among the variables selected. PCA loading plots of different colour variables indicated that simple statistical coefficients such as percentiles and mean values can be used to quantify different aspects of flesh colour. In conclusion, the methods presented here provide a powerful toolbox for describing important morphological structures and quality traits of trout cutlets.  相似文献

Samples of hatchery-reared Atlantic salmon parr (age 0+ and 1+) from two different Norwegian river strains were investigated for connections between sexual maturity and prevalence of dorsal fin ray erosion. Mature males showed consistently lower prevalence as well as better recovery than immature males. Among the immature fish, no differences were detected between males and females. The results are discussed relative to morphological and physiological changes in maturing salmon parr.  相似文献

Lime and phosphorus (P) applications are common agricultural management practices. Our aim was to quantify the effects of long-term application practices on root growth and abundance of arbuscular mycorrhizal fungi (AMF) under field conditions. We assessed the effects of lime and P fertilizer applications on barley yield, root growth and AMF abundance in 2016. Treatments were no, low, medium and high liming rate corresponding to application of 0, 4, 8 and 12 Mg lime ha−1 every 5–9 years since 1942 combined with no or yearly application of 15.6 kg P ha−1 since 1944. At harvest, grain yield, root intensity (core-break) and AMF abundance at different soil depths were estimated. Root development was monitored during early growth with minirhizotrons in treatments receiving low, medium and high liming rates and P fertilization. A quadratic model relating grain yield to liming rate estimated yields to peak at 6.4 Mg lime ha−1 with yields of 4.2 and 3.2 Mg grain ha−1 with and without P fertilization, respectively. Low and medium liming rates resulted in greater AMF abundance, especially in the no P treatments. During early growth in P-fertilized treatments, 77% and 65% more roots developed in the soil profile when treated with medium and high liming rate, respectively, compared to low liming rate. We conclude that long-term application of lime in soils receiving yearly P fertilization improved conditions for root growth in soil layers below 30 cm, but at the high liming rate, this did not translate into higher yield.  相似文献

Hybrid aspen is an interesting tree species for wood production in northern Europe. In this study we examined growth dynamics over the whole rotation period. Height and diameter development, as well as annual growth of stem volume and stem biomass, were repeatedly recorded in 14 planted and 2 root sucker stands, aged up to 26 years, in southern Sweden. A main aim was to study the productivity level for hybrid aspen forestry with an expected rotation period of about 25 years. The study verified earlier prognoses, showing a mean annual increment (MAI) of 19.5 m3 of stem wood ha?1 yr?1 after 25 years. This corresponds to a dry weight of stem biomass of 7.2 tons dry matter (DM) ha?1 yr?1. Maximum MAI was still not reached after 25 years, although the growth curve was flattening out. If branch biomass is included, MAI is estimated to about 9 tons DM ha?1 yr?1 and further improvements in growth is expected by using the best genotypically selected clones available. Results from three different thinning regimens showed that thinning intensity provided significantly larger diameter growth, while no significant yield effects were seen among the thinning regimens.  相似文献

The amounts of three main polyacetylenes in carrots; falcarinol, falcarindiol, and falcarindiol-3-acetate, were determined by HPLC, during three seasons, in carrots harvested several times per season and at different locations in Sweden. The amounts of falcarindiol first decreased from a relatively high level and then increased later in the harvest season. The amounts of falcarindiol-3-acetate showed similar variations, whereas the amounts of falcarinol did not exhibit any significant variation during the harvest season. During storage the amount of polyacetylenes leveled off, increasing in samples initially low and decreasing in samples initially high in polyacetylenes. The amounts of all polyacetylenes varied significantly due to external factors and between stored and fresh samples. This variation opens up possibilities to achieve a chemical composition of polyacetylenes at harvest that minimizes the risk of bitter off-taste and maximizes the positive health effects reported in connection with polyacetylenes in carrots.  相似文献

The closed-chamber method is the most common approach to determine CH4 fluxes in peatlands. The concentration change in the chamber is monitored over time, and the flux is usually calculated by the slope of a linear regression function. Theoretically, the gas exchange cannot be constant over time but has to decrease, when the concentration gradient between chamber headspace and soil air decreases. In this study, we test whether we can detect this non-linearity in the concentration change during the chamber closure with six air samples. We expect generally a low concentration gradient on dry sites (hummocks) and thus the occurrence of exponential concentration changes in the chamber due to a quick equilibrium of gas concentrations between peat and chamber headspace. On wet (flarks) and sedge-covered sites (lawns), we expect a high gradient and near-linear concentration changes in the chamber. To evaluate these model assumptions, we calculate both linear and exponential regressions for a test data set (n = 597) from a Finnish mire. We use the Akaike Information Criterion with small sample second order bias correction to select the best-fitted model. 13.6%, 19.2% and 9.8% of measurements on hummocks, lawns and flarks, respectively, were best fitted with an exponential regression model. A flux estimation derived from the slope of the exponential function at the beginning of the chamber closure can be significantly higher than using the slope of the linear regression function. Non-linear concentration-over-time curves occurred mostly during periods of changing water table. This could be due to either natural processes or chamber artefacts, e.g. initial pressure fluctuations during chamber deployment. To be able to exclude either natural processes or artefacts as cause of non-linearity, further information, e.g. CH4 concentration profile measurements in the peat, would be needed. If this is not available, the range of uncertainty can be substantial. We suggest to use the range between the slopes of the exponential regression at the beginning and at the end of the closure time as an estimate of the overall uncertainty.  相似文献
