Differences in the bacterial communities of soils caused by disturbances and land management were identified in rRNA gene libraries prepared from conventional tilled (CT) and no tilled (NT) cropland, a successional forest after 30 y of regrowth (NF) and an old forest of >65 y (OF) at Horseshoe Bend, in the southern Piedmont of Georgia (USA). Libraries were also prepared from forests after 80 y of regrowth at the Coweeta Long Term Ecological Research site (CWT) in the Southern Appalachians of western North Carolina (USA). The composition of the bacterial communities in cropland soils differed from those of the Horseshoe Bend OF and CWT forest soils, and many of the most abundant OTUs were different. Likewise, the diversity of bacterial communities from forest was less than that from cropland. The lower diversity in forest soils was attributed to the presence of a few, very abundant taxa in forest soils that were of reduced abundance or absent in cropland soils. After 30 y of regrowth, the composition of the bacterial soil community of the NF was similar to that of the OF, but the diversity was greater. These results suggested that the bacterial community of soil changes slowly within the time scale of these studies. In contrast, the composition and diversity of the bacterial communities in the Horseshoe Bend OF and Coweeta soils were very similar. Thus, this forest soil bacterial community was widely distributed in spite of the differences in soil properties, vegetation, and climate as well as resilient to disturbances of the above ground vegetation.  相似文献

OBJECTIVE: To assess intestinal mucosal function by measuring permeability and absorptive capacity in dogs with chronic enteropathy (CE) before and after treatment and to determine whether those variables were correlated with clinical disease activity or histologic scoring of intestinal biopsy specimens. ANIMALS: 29 dogs with CE. PROCEDURE: Dogs were designated as having dietresponsive CE or CE requiring glucorticoid treatment. Severity of clinical signs was assessed by calculating the canine inflammatory bowel disease activity index (CIBDAI). Histologic severity of intestinal infiltration was assessed before and after 4 weeks of treatment in the diet-responsive group and before and after 10 weeks of treatment in the glucocorticoid group. Gastrointestinal permeability and mucosal absorptive capacity were assessed by use of intragastric administration of a solution containing lactulose, rhamnose, xylose, 3-O-methylglucose, and sucrose. Urine was collected 6 hours after administration of the sugar solution to determine urinary lactulose-to-rhamnose (L:R), xylose-to-methylglucose (X:M), and sucrose-to-methylglucose (S:M) ratios. RESULTS: Median CIBDAI scores decreased significantly in both groups of dogs after treatment. However, the median histologic grade of intestinal biopsy specimens did not change with treatment in either group. There were no significant differences in L:R, X:M, or S:M ratios after treatment in either group and no significant correlations between L:R, X:M, or S:M ratios and CIBDAI or histologic scores. CONCLUSIONS AND CLINICAL RELEVANCE: Results of tests for intestinal permeability and mucosal absorptive capacity were not useful indicators of clinical disease activity as assessed by the CIBDAI or the sever ity of infiltration as indicated by histologic evaluation.  相似文献

EMLA is a lidocaine/prilocaine cream used for topical analgesia in human pediatric patients. The purpose of this study was to establish the safety of EMLA in clinically ill cats, to measure systemic absorption and to determine whether EMLA reduced the need for sedation for the placement of jugular catheters. Thirty-one cats were randomized to either a placebo or EMLA cream group. Cream was applied to a 10 cm(2) area over the jugular vein, with 1h of occlusive dressing. Neither anesthetic was systemically absorbed in any cat, and no adverse clinical signs were observed. Struggling during catheter placement was less in the EMLA-treated cats compared to placebo, but did not reach significance (P = 0.06). Jugular catheters were successfully placed in 60% of EMLA-treated cats and 38% of placebo cats; this difference was not statistically significant and may not justify the added steps of EMLA cream administration for this purpose. However, EMLA does appear to be safe in clinically ill cats, and may be useful for other applications such as for skin mass removal or repeated venepuncture.  相似文献

Mast cell tumours (MCTs) are relatively common tumours of cats, and are the second most common cutaneous tumours in cats in the USA. While the primary splenic form of the disease is far less common, it is usually associated with more severe clinical signs. Signalment, clinical and survival characteristics of mast cell neoplasia were characterised in 41 cats. The most common tumour location was cutaneous/subcutaneous head and trunk. Stage 1a was the most common tumour stage at first diagnosis (n=20), followed by stage 4 (both stage 4a and stage 4b; n=10). Of 22 cats that underwent excisional biopsy, mast cell neoplasia recurred in four cats during the study period. Three of the 41 cats presented with simultaneous cutaneous and either splenic or lymph node tumours. A comparison between cats with only cutaneous tumours (n=30) and those with tumours involving the spleen or lymph nodes (n=11) showed longer survival times for the cutaneous-only group (P=0.031). Twelve of the 41 cats died of mast cell neoplasia during the study period. When a subgroup of cats with only cutaneous tumours (no lymph node or visceral involvement) were divided according to whether there were multiple (five or more) tumours (n=6) or a single tumour (n=19), cats with single tumours survived longer than those with multiple tumours (P=0.001). Solitary cutaneous feline MCTs without spread to the lymph nodes usually manifest as benign disease with a relatively protracted course. However, multiple cutaneous tumours, recurrent tumours and primary splenic disease should receive a guarded prognosis due to the relatively short median survival times associated with these forms of the disease.  相似文献

The United Nations sustainable development goals (SDGs) provide an ambitious and comprehensive framework for addressing the development needs on a global, regional, and national scale. In order for the SDG targets to be obtainable, diverse stakeholders need the technical, institutional, and organizational capacity to implement the wide variety of initiatives covered under these goals. Inspired by a 2016 workshop, this article synthesizes capacity-building themes and strategies for the following approaches to sustainable development, specifically related to forestry, restoration, agriculture, and other forms of land management. Using three case studies from the authors’ experience, we present approaches to building local capacity for sustainable land management (SLM) in the tropics. These projects have taken different steps in order to build technical and leadership capacity as well as develop management or financial skills, and offer insight into various approaches that may be used in order to improve the effectiveness and long-term impact of SLM efforts. By presenting a range of tools and approaches to capacity building, the authors hope that this synthesis can serve as a valuable guide for the development of diverse capacity-building initiatives required to meet the SDGs.  相似文献

Replanting at appropriate times after harvesting a coniferous forest stand can help efforts to suppress seedling mortality caused by the pine weevil Hylobius abietis, but optimal times are uncertain. We hypothesized that planting in June rather than May in the third season after harvest would reduce feeding damage by the pine weevil and increase seedling survival rates in central Sweden, where new-generation weevils mainly fly away from their development sites in May/early June. An experimental test of the hypothesis in eight clear-cuts confirmed that planting in June instead of May reduced proportions of seedlings attacked by pine weevil, bark removal from seedlings’ stems, and proportions of seedlings killed by feeding damage. These differences between seedlings planted in May and June declined to some extent with time but still remained significant after two growing seasons. The total seedling mortality after two seasons did, however, not differ significantly between seedlings planted in May and June. Overall, 29% of all seedlings were killed by pine weevil, 4.0% by Hylastes bark beetles, and 2.3% by drought. The results indicate that replanting in spring during the third season after harvest can advantageously continue until mid-June with respect to damage and mortality.  相似文献

The aim of this work was to study the influence of process parameters and the starter culture on the characteristics of wheat sourdough by using response surface methodology. Influence of fermentation temperature (16–32°C), ash content of flour (0.6–1.8%), and fermentation time (6–20 hr) were considered as independent factors and their effects were studied in sourdough fermented with Lactobacillus plantarum, L. brevis, Saccharomyces cerevisiae, or with a combination of yeast and lactic acid bacteria. Formation of acidity, free amino acids, and volatile compounds were considered the main responses. A possibility to enhance formation of potential flavor compounds and precursors without excessive acidity formation in wheat sourdoughs was established. The total amount of amino acids increased by 25–50%, depending on the strain and fermentation conditions. The total amount of volatile compounds increased seven‐ to 100‐fold, depending on the strain and fermentation conditions. Sourdough started with S. cerevisiae was an effective way to optimize the amount of volatile compounds without excessive acidity formation in appropriate processing conditions. Ash content of flour and fermentation time were the most significant factors to modify metabolic activity of wheat sourdoughs. Frequent interactions between the studied factors were observed on the formation of acidity, amino acids, and volatile compounds with most of the strains studied. Possibility to improve current industrial fermentation processes and control flavor attributes of breads by using optimized sourdough was established.  相似文献

Interactions between microbial communities and organic matter were analyzed for soils from the project regions ’︁Ecosystem Research in the Agricultural Landscape/FAM, Munich’ in southern Germany and ’︁Ecosystem Research in the Bornhöved Lake district’ from northern Germany using ratios between microbial biomass content (Cmic), microbial metabolic quotient (qCO2) and organic carbon content (Corg). In the agricultural soils in southern Germany, the qCO2/Corg ratio differed significantly with respect to agricultural management in contrast to ecophysiological Cmic/Corg ratio. In addition, Cmic/Corg ratio decreased from 39 to 21 mg Cmic g—1 Corg and qCO2/Corg ratio increased from 72 to 180 mg CO2‐C g—1 Cmic h—1 (g Corg g—1 soil)—1 with increasing soil depth. For the upper soil horizons from the landscape in northern Germany the two quotients differed significantly with reference to land use showing highest microbial colonization under grassland and lowest under beech forest. In contrast, C use efficiency was lowest in arable field under maize monoculture and highest in a wet grassland having a high organic C content.  相似文献

In a two‐year field trial at the sites Ruthe (Germany, loess soil, Orthic Luvisol) and Schermer (The Netherlands, marine clay soil, Eutric Fluvisol) the cauliflower F1‐hybrids Marine, Lindurian and Linford were compared in their efficiency of N use from limiting and optimum supplies of N. Limiting N was Nmin at planting. Optimum N was 250 kg ha—1 as the sum of inorganic N content of the soil (Nmin) at planting and fertilizer‐N. Marine was the most efficient variety, producing the highest shoot dry‐matter and quality (% class 1 curds) at both limiting and optimum N supplies. The N supply did not affect the horizontal and vertical distribution of root length density per soil volume (RLD, cm cm—3) irrespective of variety. The RLD decreased exponentially with increasing soil depth. Varietal differences in RLD were not found at Ruthe, whereas at Schermer Marine had the highest RLD in all soil layers investigated (0 to 60 cm). No correlations were found between RLD and residual Nmin at harvest, except at limiting N supply in Schermer where a strong negative correlation was found between RLD in the 45 to 60 cm layer and Nmin at harvest. Thus, varietal differences in N efficiency are speculated to be rather due to different internal N‐use efficiency than to differences in N‐uptake efficiency.  相似文献

The aim was to study the effect of concentration and molecular weight of four different β‐glucan preparations on the perceived sensory quality of a beverage prototype. The correlations between sensory and instrumental measures were investigated. Two of the preparations were brantype containing high molecular weight β‐glucan, two were more‐processed low molecular weight β‐glucan preparations. Twelve beverage samples containing 0.25–2% β‐glucan and one reference sample thickened with carboxymethyl cellulose (CMC) were profiled by a sensory panel and analyzed by instrumental measurements (viscosity and molecular weight). Sensory profiles of the beverages varied at the same concentration of β‐glucan, depending on β‐glucan preparation. Beverages made with the bran‐type preparations were more viscous and had higher perceived thickness than beverages made with more‐processed, low molecular weight preparations. Moderate correlations were obtained between perceived thickness and sliminess and instrumental viscosity at all shear rates between 26 and 100/sec (r = 0.63–0.78; P ≤ 0.001). Technologically, more‐processed β‐glucan preparations are easier to add into a beverage in amounts sufficient for achieving a physiologically functional amount of β‐glucan in a product.  相似文献
