目的合成氨基脲人工抗原并制备其多克隆抗体。方法以氨基脲(SEM)和对醛基苯甲酸(CP)为原料合成半抗原(CP-SEM),并采用碳化二亚胺法和混合酸酐法将其分别与牛血清白蛋白(BSA)和鸡卵清蛋白(OVA)偶联,合成氨基脲人工抗原。以氨基脲人工抗原(CPSEM-BSA)为免疫原免疫Balb/c小鼠,利用间接ELISA法测定其抗体效价。结果经薄层层析、红外光谱和元素分析等方法鉴定合成半抗原即为CP-SEM。紫外扫描、聚丙烯酰胺凝胶电泳结果表明人工抗原合成成功。免疫获得抗氨基脲的多克隆抗体,其抗体效价为1:64000。结论氨基脲人工抗原合成成功,并获得抗氨基脲的多克隆抗体。 相似文献

Helicobacter‐like organisms are frequently found in canine stomachs, but the relationship between such organisms and gastric pathology has not been established. However, some such organisms have zoonotic importance. The aims of this study were to evaluate the morphological and biochemical characteristics of cultivable canine gastric Helicobacter‐like organisms (GHLOs) in pets and stray dogs and their prevalence in these two groups of dogs. Specimens were taken by gastroscopy from 30 clinically healthy stray dogs and 30 pet dogs. Cultures were positive from biopsies of 11/30 of stray and 6/30 of pet dogs. The isolated Helicobacters were observed by light microscopy and studied by biochemical, physiological and PCR analysis. Some of the isolated GHLO's displayed atypical shapes that were similar to Helicobacter pylori or Helicobacter acinonychis in stray dogs’ cytological examinations. They had 2–3 helices and were smaller than other canine GHLOs. One of these atypical Helicobacter strains was cultured. It was not possible to distinguish such strains by routine PCR and biochemical evaluations. Electron microscopy showed a smaller Helicobacter (2 μm in length) with 2 or 3 helixes. This study demonstrates that not all canine gastric Helicobacters are 5–15 μm in length, as has been previously proposed, and portrays the need for further investigation of canine GHLOs. 相似文献

试验旨在研究氨基酸副产物(amino acid by-products,ABP)对全株红高粱和全株甜玉米发酵品质及消化率的影响,并通过扫描电镜(SEM)探讨ABP对青贮饲料发酵品质及消化率的作用机制。试验设无添加剂的对照组和添加2.0% ABP的试验组Ⅰ、2.0% ABP与饲用菌混合添加的试验组Ⅱ,分别进行红高粱和甜玉米的发酵试验,检测其发酵品质及消化率。结果表明:①试验组Ⅰ红高粱和甜玉米青贮pH降至3.90和3.28,感官评定均属于优质青贮饲料。②对于红高粱青贮,与对照组相比,试验组干物质(DM)含量略有提高,但差异不显著(P>0.05),粗蛋白质(CP)含量显著高于对照组(P<0.05),试验组Ⅱ纤维成分略低于对照组,但差异不显著(P>0.05);各试验组乳酸和乙酸含量显著高于对照组(P<0.05),试验组Ⅱ丁酸含量显著低于其余组(P<0.05);试验组干物质和中性洗涤纤维(NDF)消化率均高于对照组,但差异不显著(P>0.05),酸性洗涤纤维(ADF)消化率显著高于对照组(P<0.05)。③对于甜玉米青贮,各试验组干物质和纤维含量均低于对照组,但差异不显著(P>0.05),粗蛋白质含量显著高于对照组(P<0.05);试验组Ⅰ乳酸含量显著高于其余组(P<0.05),试验组Ⅱ乙酸含量显著高于其余组(P<0.05),而各试验组丁酸含量显著高于对照组(P<0.05);试验组各养分消化率均低于对照组但高于未发酵原料,其中干物质和酸性洗涤纤维消化率差异显著(P<0.05)。④SEM结果表明,青贮过程中ABP通过破坏红高粱表面蜡质层促进饲用菌的黏附,并降解其细胞壁纤维成分而改善了青贮发酵品质并提高消化率;但ABP对青贮甜玉米蜡质层及细胞壁纤维结构破坏作用不明显。综上,添加ABP可提高红高粱青贮的发酵品质和养分消化率,但对甜玉米青贮的作用不大。 相似文献

应用扫描电镜和组织切片法研究球孢白僵菌侵染光肩星天牛幼虫体壁的过程以及寄主中肠发生的病理变化.结果表明:接菌后48h,芽管平均长度达到3.08μm时即可侵入幼虫体表.随着侵染时间延长,芽管伸长形成菌丝,逐渐萌发生成新的分生孢子,在后期菌丝间出现丝状物和网状结构,这2种现象在国内外未见报道.分生孢子多数以芽管方式侵入表皮,也可以产生附着胞再侵染.组织切片显示:侵染3天的幼虫腹部体腔中脂肪体组织变得模糊,细胞核不明显,马氏管出现空泡并变形.中肠围食膜被分解消失,肠壁肌肉层向内皱褶,排列紊乱,出现破裂,肠腔内变浑浊. 相似文献

Sesbania fiber is a fast-growing wood species that has been investigated for medium-density fiberboard (MDF) production. To assess the possibility of applying the local industrial defibration parameters of sugar-cane bagasse (SCB) on defibration of sesbania, the chemical constituents of unfibrated and defibrated sesbania, as well as their thermal stability and scanning electron micrographs, were estimated. Different preparation variables of MDF, such as density, level of urea-formaldehyde (UF) resin (with 0.19% free-formaldehyde [HCHO]), and pressing time were studied, in comparison with that produced by using SCB fibers. The results showed that most of the tested sesbania-based MDFs have mechanical properties that fulfill the minimum requirements of MDF ANSI standard. Additionally, applying 12% UF and pressing for 240 sec provided sesbania-based MDF with optimum reduction in thickness swelling (reached ~7%). It is important to note that the sesbania-based MDF produced under these conditions is characterized by a lower TS property, than that obtained from SCB, or that reported in standards. The preliminary feasibility study revealed that using sesbania fibers will be an added economical potential for MDF production. 相似文献
