A new chamber method and a stripping method were developed for field measurements of the rate of N2O emission from the water surface and for determinations of dissolved N2O in water from agricultural land. These methods were used for the measurement of drainage canal water and flooded water of rice fields during the period of June 1982 to January 1983. Grain compositional components impacting barley (Hordeum vulgare L.) use in food, feed and fuel products, must be combined with improved agronomic traits to produce a commercially viable barley cultivar. Little current information is available on grain composition and variability among winter barley genotypes. This study was conducted to determine the variability among modern hulled and hulless winter barley genotypes in grain composition. Barley types varied significantly in grain and kernel weight, starch, beta-glucan, oil and ash content, but not in protein concentration. Hulless barley had significantly higher grain test weight and starch concentration than hulled and malting types, and significantly higher beta-glucan than malting barley. Hulless barley had significantly lower kernel weights, oil, and ash concentrations than hulled and malting types. Higher starch and lower fiber and ash in grain of hulless barley versus hulled feed or malting type barley are characteristics that increase hulless barley desirability. Selection for high starch concentration among all barley types is feasible and will facilitate development of barley cultivars better suited for use in feed, malt, and ethanol production. 相似文献

The diffusive and nonspecific nature of nonpoint source contaminants such as sediment makes their management and mitigation challenging. Conventional source‐based tracing techniques for sediment simply apportion downstream sediment load to diffuse upstream sources classified by a limited number of source types including underlying rock type, land cover, and/or erosion process. Here, we develop a grid‐based sediment tracing technique that improves the precision of source contribution estimates and enhances the granularity of sediment source maps. We test the proposed technique using source and suspended sediment samples collected from the Emu Creek Catchment (911 km2), south‐east Queensland, Australia. Geochemical tracers were employed to distinguish sediments derived from the heterogenous and complex underlying rock types. Importantly, the proposed technique provided a greater spatial resolution of the sediment sources by assigning sediment contributions into grid sources rather than the area‐specific source types. 相似文献

Four sets of durum samples were used in this study to further understand the interrelationships among hard vitreous kernels (HVK), protein content, and pigment concentration, with a focus on the interaction and synergistic effects of protein content and vitreousness on durum quality. HVK level increases with higher protein content in the range of 9.5–12.5%, but this relationship is less evident in durum samples with high protein content (12.5–14.5%). Both protein content and kernel vitreousness can significantly affect durum milling quality. White starchy kernels (WSK) in low protein durum have a very detrimental impact on milling and pasta processing quality, but high protein content can mitigate the adverse impact of WSK on durum quality. Although protein content plays a dominant role, higher HVK might contribute positively to pasta firmness. There was no significant difference in yellow pigment content between HVK and WSK. However, pigment loss from semolina to dough was higher for WSK than HVK. Despite the difference in protein content, HVK and WSK have little difference in gluten strength. The monomeric protein was preferentially accumulated in HVK. The glutenin proteins of HVK and WSK were similar in the ratios of 1Bx/1By and HMW/LMW-GS. 相似文献

Despite great genetic diversity, sorghum grain consistently suffers from poor protein digestibility. The physicochemical packaging of protein bodies which consist of protease-resistant β- and γ-kafirin is considered a major obstacle. A synthetic β-kafirin gene, which shares the endosperm-specific promoter and signal peptide with the native β-kafirin gene (Sobic.009G001600.1), was transformed into sorghum inbred line Tx430. The gene was modified with ten additional proteolytic sites. These sites were designed to be amenable to cleavage by pepsin and/or chymotrypsin proteinases. Five independent transgenic lines were regenerated by microprojectile transformation. Notably, considerably more protein was observed in the peripheral endosperm of transgenic lines under scanning electron microscopy. Microscopy revealed invaginated or irregularly shaped protein bodies in the endosperm of transgenic lines. Grains of transgenic lines contained 11–37% more protein, which was 11–21% more pepsin digestible and 7–25% more chymotrypsin digestible than Tx430. Additionally, the abundant synthetic β-kafirin protein (5.6% of total protein) was detected by mass spectrometry data analysis in the transgenic line 9-1. Field-grown homozygous transgenics retained higher protein content, larger seed size and no reduction in grain number per plant. The study of food-grade isolated kafirin proteins has increased recently due to the interest of their potential applications in gluten-free products. However, functionality of extracted proteins and fractions have been poorly characterized. In this investigation, kafirins were extracted from whole or decorticated sorghum genotypes (white-regular, white-waxy, red-regular and high-tannin) with 70% aqueous ethanol with metabisulfite. Chemical composition, color parameters, functionality and molecular characterization of kafirin extracts were determined. When kafirin extracts were obtained from decorticated sorghums, higher protein purities (81.5–93.3%), lower lipid contents (1.9–12.6%), better color parameters, and higher yields were obtained compared to counterparts extracted from whole caryopses. Functional properties of the extracted kafirins showed low water solubility, free amino nitrogen, and emulsifying activity, but adequate water holding and fat absorption capacities and in vitro protein digestibility. Differences in functionality were attributed to sorghum genotype. SDS-PAGE showed extraction of α1-, α2-, β-, and γ-kafirins with no contamination of other protein fractions. FTIR indicated a reduction in α-helix:β-sheet after extraction, especially in proteins obtained from decorticated sorghums. High correlations found from Pearson analysis could be used to optimize protein yield and the functionality (emulsifying activity and solubility) of the extracted proteins to be applied in gluten-free food systems. 相似文献
