Summary Orobanche species are commonly identified using morphological characteristics. In many cases, the distinction of closely related species is difficult, and a molecular tool is more suitable to differentiate them. In this study, genomic polymorphism between morphologically distinct species was investigated through amplification by polymerase chain reaction (PCR) of intersimple sequence repeat (ISSR) regions. Five primers were used to study genetic variation in the morphologically distinct species O. hederae and O. amethystea, as well as the closely related species O. cernua and O. cumana. For the first two species, all the primers detected genetic polymorphism. Anchored primers allowed the identification of more specific molecular markers than non‐anchored tri‐ and tetranucleotide primers. Genetic polymorphism was investigated among three O. hederae populations using the two types of primer. One non‐anchored and two anchored primers detected intraspecific variation, which was not correlated with the geographical location of those populations. The primer (GATA)4 detected polymorphism between five specimens each of O. cernua and O. cumana species collected from different countries, permitting these two closely related species to be clearly differentiated. This study demonstrated that ISSR markers can be highly reliable for precise identification of Orobanche species.  相似文献

The cumulative cardiotoxicity that occurs as a result of doxorubicin chemotherapy is irreversible and can affect both quality and quantity of life for the cancer patient. Cardiac troponin I (cTnI) is a sensitive and specific marker of cardiomyocyte death. The purpose of this retrospective study was to evaluate serum concentrations of cTnI in dogs with lymphoma or osteosarcoma given doxorubicin chemotherapy, and with known cardiac outcome, based on a minimum assessment by physical examination and thoracic radiography. Serum samples were also available for cTnI measurement from seven healthy dogs given intracoronary doxorubicin. Serial serum samples obtained before, during and after doxorubicin chemotherapy showed increased cTnI concentrations in some clinical patients following chemotherapy (P = 0.0083 compared to baseline), but this did not correlate with clinical signs of cardiomyopathy. In dogs that subsequently developed cardiomyopathy however, serum cTnI concentrations were elevated before clinical signs became evident (confirmed with echocardiography).  相似文献

Fusarium culmorum is a phytopathogenic, toxigenic fungus causing seedling diseases, foot rot and head blight of cereals. For estimating competition effects in mixtures of two single-spore isolates, two winter rye single crosses were tested with either four isolates individually or four 1 : 1 mixtures of the same isolates in six field environments. Two isolates (FC46, FC64) were highly aggressive deoxynivalenol (DON) and 3-acetyl DON-producers, the other two (FC30, FC71) were medium aggressive nivalenol-producers. Rye heads were inoculated during flowering with conidia of pairs of isolates expressing similar (FC46 + FC64, FC30 + FC71) or contrary (FC46 + FC71, FC30 + FC64) levels of aggressiveness and similar or different concentrations and chemotypes of mycotoxins, respectively. Head blight rating and yield components relative to the non-inoculated plots were recorded as aggressiveness traits. Additionally, mycotoxin concentrations were measured in the rye grain. Random pathogen samples were re-isolated from heads at the onset of symptom development and analysed by molecular markers (RAPD–PCR) in one environment. Aggressiveness of the isolate mixtures was significantly lower than that of the isolates applied individually on both rye genotypes. Similarly, mycotoxin concentrations were significantly lower in the mixtures in seven out of eleven comparisons. Among the re-isolates, the component genotypes of a mixture significantly deviated from the inoculated 1 : 1 ratio when a particular isolate (FC46) was present in the mixture. This isolate displayed a superior competitive ability irrespective of the aggressiveness or mycotoxin profile of the mixing partner illustrating that pathogenic fitness is caused by additional factors that have not, as yet, been identified.  相似文献

本研究利用MISA软件挖掘长江刀鲚(Coilia ectenes)肌肉和肝脏转录组中的微卫星标记,为刀鲚选育群体的种质资源评估和分子标记辅助育种奠定基础。结果显示,从71869条Unigenes中共获得33896条重复单元长度为1~6碱基的微卫星序列;刀鲚转录组中不同类型微卫星的重复基序具有不同的分布特征,其中,单核苷酸重复、二核苷酸重复和三核苷酸重复为主要的微卫星重复类型,分别占总微卫星数目的34.94%、49.47%和13.34%;不同微卫星重复类型的优势重复基序亦有所不同,其中,A/T为单核苷酸重复基序的优势重复基序占86.25%,AC/GT为二核苷酸重复基序的为优势重复基序占75.25%,AGG/CCT为三核苷酸重复基序的优势重复基序占28.57%;不同微卫星重复基序核苷酸的数量和重复次数亦有所不同,重复次数伴随着重复单元中核苷酸数量的增加而呈现降低的趋势;从100对四核苷酸重复的SSR引物中筛选获得了16对多态性微卫星标记,并以此为基础,对长江刀鲚选育群体(F3)的遗传学特征进行了初步评估,结果显示,长江刀鲚选育群体F3的平均有效等位基因数(Ne)、平均观测杂合度(Ho)、平均期望杂合度(He)和Shannon多样性指数I分别为1.7580、0.3414、0.3977和0.6278。以上结果表明,基于刀鲚转录组数据批量开发微卫星是切实可行的,所开发的多态性微卫星标记能够应用于长江刀鲚选育群体的遗传背景评估和进一步的遗传育种研究。  相似文献

Maize rough dwarf disease caused by Rice black-streaked dwarf virus (RBSDV) is transmitted by planthopper in China. Identification and development of resistant hybrids are complicated because of the inconsistencies in viral disease pressure every year. Marker-assisted selection can provide means for main-taining virus resistance alleles even in the absence of disease. In this paper a F2 segregation population was constructed to identity the molecular markers linked to the resistance gene using a cross between a resistant and a susceptible parents (Qi319×Ye107). Fifteen-day-old seedlings of F2 population were exposed to small brown planthoppers carrying RBSDV for 3 days in specific inoculation chamber. The inoculated plants were transplanted to screenhouse after removing the insects completely. In plant maturity stage the disease resistance of all the individuals were visually assessed. The results showed that 17, 8, 11, 51 and 122 plants were scaled from 0-4 respectively, in which 0 means no symptoms and 4 represents highly susceptible. Chi-square test demonstrated that the segregation ratio of phenotype was 1∶15 (resistant: susceptible) or 1∶6∶9 (resistant∶moderate∶susceptible) in the F2 population, indicating RBSDV resistance of maize was controlled by two recessive genes. The F2 individuals DNA were extracted and 261 SSR (simple sequence repeat) primers derived from maize genome ten chromosomes were selected from maize GDB database to construct genetic linkage map. The linkage map consisted of 71 polymorphic SSR markers, spanning a genetic distance of 996.6 cM with an average interval of 14.0 cM between adjacent markers. The resistant and susceptible gene pools were set up for BSA (bulked segregant analysis) and 6 polymorphism markers were obtained with BSA-SSR method between the two pools. The F2individuals were further analyzed with 6 polymorphism markers. Chi-square test showed that phi 051, umc1407 and umc1432, mapped on chromosome 7 and 10, exhibited segregation distortion significantly and very significantly in susceptible individuals. These three SSR markers were identified as potential markers linked to the resistant loci.  相似文献

以2006年辽宁、吉林、黑龙江三省水稻区域试验品种(系)为试材,采用SSR标记分析了东北三省35个水稻品种(系)的遗传多样性和亲缘关系,结果共检测出237个等位基因,分子量变异范围在95~320 bp。品种(系)间不同位点等位基因数目不等(2~8个),平均3.3857个,平均Nei基因多样性指数为0.4935,变幅为0.0328(PSM116)~0.8577(PSM363)。多态性位点的比率变化范围为71.25%~90.04%,多样性指数(H)介于0.3869~0.4903,信息指数(I)在0.6399~0.8489。吉林和黑龙江供试品种的SSR多样性低于辽宁水稻品系,而吉林和黑龙江基本相同。各稻区内品种的多样性均低于总体水平,显示了东北三省2006年水稻育种遗传基础的狭窄性。  相似文献

【目的】小麦条锈病由条形柄锈菌小麦专化型(Puccinia striiformis. f. sp. tritici,Pst)引起的气传性真菌病害,是世界范围内最具破坏性的小麦病害之一。发掘抗病资源和培育抗病品种是防治条锈病最经济有效的措施。通过鉴定和评价云南小麦地方品种对中国当前流行的条锈菌生理小种的抗性水平,综合分析可能携带的抗条锈病基因,为小麦抗病育种提供理论依据。【方法】利用当前中国小麦生产上毒性强、流行频率高的条锈菌生理小种CYR32和CYR34对243份云南小麦地方品种进行苗期抗病性鉴定,成株期使用CYR32、CYR33、CYR34和贵农致病类群等混合生理小种进行抗性鉴定,并利用小麦生产上重要抗条锈病基因Yr5、Yr10、Yr15、Yr18、Yr26、Yr28、Yr29、Yr30、Yr36、Yr39、Yr41、Yr48、Yr65、Yr67、Yr80和Yr81的紧密连锁标记对供试材料进行分子检测。【结果】在243份小麦材料中,18份种质对CYR32表现苗期抗性,32份种质对CYR34表现苗期抗性,对CYR32和CYR34均表现苗期抗病的有8份,占3.29%。结合苗期和成株期抗性...  相似文献

北粳南移在江西省已推行11年,为进一步明确适合江西晚粳常规稻的稻瘟病抗性情况及粳渗籼骨干亲本的选择,本研究利用Pi37/Pi35/Pish、Pi5、Pi2、Pi9、Pia、Pi23、Pigm、Pi33/Pi42、Pi56、Pik/kg(t)/ks/kp/km/kh/43、Pikh共11个抗瘟基因的标记结合井冈山抗性谱的鉴定,对28份骨干粳稻和54份骨干籼稻进行分子标记与抗性分析。结果表明,粳稻中有1份携带其中的7个抗性基因,有5份携带6个抗性基因,有7份携带5个抗性基因,有12份携带4个抗性基因,有3份携带3个抗性基因,抗性频率最高的Pia为0.786,其次为Pikh和Pi9,频率均为0.643,Pigm和Pi37/Pish在这批粳稻材料中的抗性频率为0;籼稻中有8份携带其中的7个抗性基因,有10份携带6个抗性基因,有13份携带5个抗性基因,有17份携带4个抗性基因,有4份携带3个抗性基因,有2份携带2个抗性基因,抗性频率最高的Pi9为0.981,其次Pik/Pikh/Piks频率为0.833,Pigm、Pi37/Pish、Pikh、Pia、Pi5、Pi2、Pi23和Pi33/Pi42...  相似文献

为了解和掌握青海省引进和收集藜麦种质资源的遗传背景和多样性,本研究选择11个表型性状和221对SSR引物对114份藜麦种质资源进行遗传多样性分析。结果表明,114份藜麦种质资源的11个表型性状差异极显著(P<0.01),变异系数范围为4.57%~87.79%,主穗直径的变异最大(87.79%),籽粒直径的变异最小(4.57%);遗传多样性指数范围为0.47~2.29,单株产量的遗传多样性指数最大(2.29),茎秆长最小(0.47)。46对多态性较好的SSR引物扩增出165条多态性条带,每对引物的平均等位变异数为3.59,观测等位基因(Na)平均值为1.65,有效等位基因(Ne)平均值为1.50,Shannon’s信息指数(I)和Nei’s基因多样性指数(He)平均值分别为0.39和0.27,平均多态性比率为64.35%。通过聚类分析发现,114份藜麦种质基于表型性状和SSR分子标记的聚类结果存在一定的相似和差异,来源地相同的种质被分到了不同的类群,来源地不同的种质被分到了同一类或单独聚成一类,说明参试藜麦种质资源的表型性状和分子遗传信息多样性较丰富。  相似文献
