The 5‐lipoxygenase (5‐LOX) inhibitor tepoxalin has been shown to slow canine osteosarcoma (OSA) tumour xenografts growth, yet the mechanisms are poorly elucidated. Further examination of tepoxalin in canine OSA cell lines shows that tepoxalin treated cells undergo apoptosis through caspase‐3 activation and annexin staining. Interestingly, apoptosis is superseded by an increase in reactive oxygen species (ROS), as measured by activation of dihydrorhodamine 123 and mitosox. This increase in ROS appears to be related to the 5‐LOX inhibitor regardless of cellular 5‐LOX status, and was not observed after treatment with the tepoxalin metabolite RWJ20142. Additionally, 5‐LOX inhibition by tepoxalin appears to increase phosphatase and tensin (PTEN) homolog activity by preventing its alkylation or oxidation. PTEN modification or inhibition allows phosphoinositide‐3 (PI3) kinase activity thereby heightening activation of protein kinase B (AKT) phosphorylation. Our data suggest that off target oxidation and LOX inhibition play roles in the apoptotic response.  相似文献

Five fast-neutron-derived mutants were isolated from the wheat line Hobbit 'sib' that show enhanced field resistance towards Puccinia striiformis f.sp. tritici , the causal agent of yellow rust. Subsequent testing showed the yellow rust resistance phenotypes to differ between mutants, to be expressed at different growth stages and, in some cases, to show an isolate interaction. Three mutants, I3-48, I3-49 and I3-54, exhibited an enhanced yellow rust resistance phenotype from the third seedling leaf onwards, while mutants I3-27 and I3-30 did not show an altered yellow rust phenotype until later growth stages. Additional resistance for brown rust (causal agent Puccinia triticina ) was identified in mutants I3-27, I3-30, I3-48 and I3-49, and for powdery mildew caused by Blumeria graminis f.sp. tritici in mutants I3-27, I3-30, I3-48 and I3-54, although in some cases the resistance was isolate-specific.  相似文献

It has been proposed that susceptibility of potato to Phytophthora infestans would be a consequence of suppression and /or delaying of defense reactions by a soluble glucan which is released by compatible races of the fungus. In this report, the reaction of potato tuber slices (Solanum tuberosum cv. Huinkul) infected with either race I (1,4,7,8,10,11) or C (1,4,10,11) of Phytophthora infestans was studied. Race C grew better on slices than race I. Glucans from both races were isolated and their effect on the accumulation of phytoalexins and glucanases in tuber slices was studied. The glucans from the less virulent race (I) did not affect the accumulation of phytoalexins and glucanases in tuber slices infected or elicited with eicosapentaenoic acid, whereas the glucans from race C produced 70% inhibition of phytoalexin accumulation and reduced by 50% the induction of glucanase activities. Purified glucanases from potato degraded the glucans from race C but not from race I. The results reported here show that, at least on this cultivar, glucans from both races affected defense responses in a different manner, which could reflect structural differences between these glucans.  相似文献

根据已发表ScYLV-P0基因系列设计特异性引物,应用RT-PCR技术从甘蔗病叶的mRNA扩增得到目的DNA片段.以pET32a(+)为原核表达载体,构建重组表达质粒pET32a-P0.经过双酶切鉴定和DNA测序后,将重组表达质粒转入大肠杆菌BL21 (DE3)pLySs,在30℃培养条件下IPTG诱导表达.通过SDS-PAGE电泳检测融合蛋白表达情况.表达结果显示,在该表达系统中,融合表达蛋白P0是以包涵体形式的蛋白存在;P0融合蛋白大小约45kDa,与P0开放阅读框的理论推算值29.991 kDa加上载体自身蛋白约18 kDa相符,用Ni2+-NTA琼脂糖亲和层析纯化融合蛋白,免疫家兔制备出抗血清,通过酶联法(ID-ELISA)测定本实验制备的ScYLY-P0抗血清工作浓度为1:25000.  相似文献

A cell-based high-throughput screen that assessed the cellular stability of a tumor suppressor protein PDCD4 (Programmed cell death 4) was used to identify a new guanidine-containing marine alkaloid mirabilin K (3), as well as the known compounds mirabilin G (1) and netamine M (2). The structures of these tricyclic guanidine alkaloids were established from extensive spectroscopic analyses. Compounds 1 and 2 inhibited cellular degradation of PDCD4 with EC50 values of 1.8 μg/mL and 2.8 μg/mL, respectively. Mirabilin G (1) and netamine M (2) are the first marine natural products reported to stabilize PDCD4 under tumor promoting conditions.  相似文献

Eleven progenies, ten bi-parental and one selfed, of tetraploid potato were assessed for phenotypic segregation ratios of resistance to susceptible individuals against two isolates of P. infestans, one avirulent to all 11 known potato R genes (avirulent pathotype) and one virulent to eight R genes (virulent pathotype). Assuming Mendelian inheritance of independent R genes, the numbers of individuals segregating for resistance to both races could not be explained sufficiently for all progenies. However, the assumed presence of dominant, specific suppressors of the R gene function that were elicited by specific isolates of the fungus and that segregated in the host independently from R genes explained all frequencies of resistant and susceptible individuals in each of the progenies studied. Summary For RFLP mapping of R-genes, determining resistance to specific races of Phytophthora infestans in tetraploid potato, it is necessary to develop well segregating populations at the 2x level. During mapping studies, evidence was obtained that more genetic factor(s) are involved in the expression of R-genes than conventionally believed. Two experiments are described in which such an additional genetic factor was suppressing or enhancing the expression of unknown R nand R ifactors. R nand R iappeared to be present in the investigated plant material, containing R4 and R10, or in one of the susceptible crossing parents. In a third experiment, the expression and the segregation of the well known R1 gene was influenced by an additional genetic factor. In that case there were indications for a dominant suppressor. This was established by the selection of susceptible plants carrying a RFLP allele of probe GP21 closely linked to R1. In three of the four F1 populations, resulting from crosses between such susceptible plants and susceptible tester plants, resistnat progenies were found. The resistance appeared to be R1-specific. This clearly indicates that in three of the four investigated susceptible plants, the R1 gene was still present but not expressed.  相似文献

AIM:To explore the molecular effects of Astragalus polysaccharide(AP) on improving nervous functions and preventing neuronal apoptosis in rat cerebral cortex with cerebral ischemia and reperfusion. METHODS:One hundred and twenty male Wister rats were randomly divided into sham operation group(SOG), model groups(MG-1 d, 3 d and 7 d), low-dose AP treatment groups(L-APTG-1 d, 3 d and 7 d), and high-dose AP treatment groups(H-APTG-1 d, 3 d and 7 d). The right middle cerebral artery of the rats in MG and AGTG was intercepted by operation to induce ischemic brain injury. The rats in L-APTG and H-APTG were treated with AP at the doses of 5 mg/kg and 15 mg/kg by intraperitoneal injection, respectively. On the 1st day, 3rd day and 7th day after operation, those animals were sacrificed to collect the brain specimens for the study after cerebral blood flow reperfusion and determination of neurological deficit scores. The structural changes of the neurons were observed under electron microscope. Apoptosis was analyzed by flow cytometry. The protein levels of heat-shock protein 70(HSP70), protein kinase B(PKB) and P53 in cerebral corical neurons were determined by immunohistochemical staining and Western blotting. RESULTS:The neurological deficit scores and the apoptotic rate of cerebral cortical neurons in H-APTG were significantly lower than those in MG and L-APTG(P<0.05). The structures of the neurons in H-APTG, such as ribosome endoplasmic reticulum, nucleolus, Golgi complex, mitochondria, etc, were better than those in MG and L-APTG. On the 1st day, 3rd day and 7th day, the protein levels of HSP70 and PKB in cerebral cortical neurons in H-APTG were significantly higher than those in L-APTG, which were significantly higher than those in MG(P<0.05). However, the P53 protein level in H-APTG was significantly lower than that in L-APTG, which was significantly lower than that in MG(P<0.05). CONCLUSION:AP improves nervous functions and inhibits neuronal apoptosis during ischemia and reperfusion. The molecular mechanisms are associated with variations of protein expression in cerebral cortical neurons, such as promotion of HSP70 and PKB and inhibition of P53.  相似文献

目的:探讨肿瘤抑制基因p27在宫颈癌组织中的表达及其临床意义。方法:应用免疫组织化学方法(SP法)检测p27基因在62例官颈癌组织和20例正常官颈组织中的表达。结果:62例宫颈癌组织中,p27的阳性表达率为38.7%(24/62);p27的阳性表达与宫颈病灶大小、临床分期、细胞分化程度、转移及预后有密切关系(P〈0.01或〈0.05);与宫颈癌的病理类型及年龄无关(P〉0.05)。结论:p27基因的表达与宫颈癌的生长、浸润及淋巴结转移有关,其表达水平的高低可作为判断宫颈癌预后的重要指标之一。  相似文献

为筛选SOCS6基因启动子区域单核苷酸多态性(Single nucleotide polymorphisms,SNP)并研究其对启动子功能元件的影响,选择品种差异较大的贵州荷斯坦奶牛和务川黑牛构建DNA池,直接测序筛选SNP位点.结果表明,SOCS6基因5'调控区存在4个SNP位点,分别为T-513G、C-401T、A-332G和T-180G.生物信息学软件预测得到SOCS6基因核心启动子区和CpG岛,SNP位点导致附近部分转录因子结合位点消失和新位点产生.多态性位点对转录因子结合位点有显著影响,但并不改变CpG岛大小.  相似文献
