The influence of housing system on the initial bacterial contamination of the eggshell was studied. Two long-term experiments were performed. Bacterial eggshell contamination, as expressed by total count of aerobic and Gram-negative bacteria, was periodically analysed for eggs from a conventional cage, a furnished cage with nest boxes containing artificial turf or grids as nest-floor material and an aviary housing system. Results were log-transformed prior to statistical analyses. For both experiments no systematic differences were found between the conventional cage and furnished cage. The type of nest-floor material in the nest boxes of the furnished cages also did not systematically influence the bacterial contamination. A possible seasonal influence on contamination with a decrease in the winter period (up to > 0.5 log cfu/eggshell) of total count of aerobic and Gram-negative bacteria was observed in the first experiment. The contamination with total aerobic flora was higher (more than 1.0 log) on eggs from the aviary housing system compared to the conventional and the furnished cage systems. For Gram-negative bacteria this was not the case. During the entire period of both experiments, independent of housing system, shell contamination was not influenced by age of hens or period since placing the birds in the houses. For the total count of aerobic bacteria a restricted positive correlation (r2 = 0.66) was found between the concentration of total bacteria in the air of the poultry houses and initial shell contamination.  相似文献

1. The hygienic properties of eggs produced in two designs of furnished cage were investigated and compared to two standard cage designs. 2. At 28, 37, 47 and 58 weeks of age, the proportion of dirty eggs was higher in one of the furnished cage designs while, in the other, it was similar to standard cages. 3. At 27, 33 and 60 weeks, the bacterial load on the eggshell (total aerobic bacteria and enterococci) was higher in furnished cage designs. A seasonal effect was observed with lower counts at 60 weeks (winter) than at 27 weeks (summer). 4. More dirty eggs and a higher bacterial load were observed in eggs laid outside the nests, which suggests egg hygiene in furnished cages could be similar to standard cages if the equipment in furnished cages was improved to enhance nest laying.  相似文献

鸡蛋是人类膳食的重要组成部分,其产出与贮运销售过程中均存在沙门氏菌等致病菌污染隐患,直接影响鸡蛋的消费安全。作者对北京市场德青源、留民营等4种品牌包装鲜鸡蛋及散装鸡蛋在贮存过程中细菌总数、大肠菌群数及沙门氏菌数量变化进行了测定,研究了贮存过程中各品牌鸡蛋蛋壳、蛋清及蛋黄中的细菌总数,大肠菌群数随时间的变化情况及沙门氏菌的检出情况。研究结果发现,不同品牌的鸡蛋蛋壳表面在贮存初期细菌总数介于9.315×102~1.367×104 CFU/个,且随着贮存时间的延长呈上升趋势;品牌鸡蛋贮存初期蛋内容物均未检出污染细菌,而在贮存6 d后检出污染细菌,细菌总数与大肠菌群存在随着贮存时间的延长而增加的变化。除A品牌鸡蛋外,市售鲜蛋均存在沙门氏菌污染,蛋壳表面污染率为1.67%~8.3%,内容物中污染率为1.67%~5%。其中散装鸡蛋的细菌总数,大肠菌群数及沙门氏菌检出率均高于其它几种品牌的鸡蛋。  相似文献

1. The survival and penetration of Salmonella enterica serovar Enteritidis (SE) inoculated on the eggshell was examined upon storage for up to 20 d at real-life conditions (15 to 25 degrees C and 45 to 75% relative humidity (RH)). 2. Penetration was assessed by emptying the egg contents and filling the eggs with a selective medium that allowed visualising Salmonella growth on the inside of the shell and membrane complex. 3. The study of survival on the eggshells was based on viable counts and showed that numbers of surviving organisms decreased over time. Survival was inversely related to storage temperature and RH. Although the average counts decreased over time, a limited proportion of shells carried high numbers of SE at all storage conditions. 4. Penetration spots were observed earlier using an increased storage temperature due to increased growth rates of SE on the agar. After 20 d of storage a similar percentage (c. 44.7%) of eggshells became penetrated, irrespective of the storage conditions tested in this study. 5. The higher the Salmonella shell contamination at the end of storage, the higher the probability that the eggshell was penetrated. Salmonella shell counts exceeding 4 log cfu yielded more than a 90% probability of eggshell penetration occurring.  相似文献

The shell of the egg is essential in providing shape to the egg and ensuring safe packaging of the internal contents; however, shell defects have been shown to increase the risk of microbial contamination of eggs. Eggs were collected from commercial cage and free-range flocks at the ages of 25, 35, 45, 55, 65, and 75 wk. From each collection per flock, 30 eggs were processed for the eggshell and egg internal quality determination, 30 eggs for cuticle estimation, 30 eggs for mammillary layer ultrastructural features scoring, and 60 eggs for egg microbial enumeration. Translucency score and shell reflectivity were significantly higher in free-range eggs and increased with flock age in both production systems. Egg weight, shell weight, percentage shell, shell thickness, albumen height, Haugh unit, and yolk color were higher for cage eggs. The amount of cuticle was higher in cage eggs and fluctuated with flock age in both production systems. For the mammillary layer ultrastructural variables, a significant effect of production system and flock age was observed for early fusion, Type A bodies, and Type B bodies, whereas aragonite, depression, erosion, and hole were rarely observed. Variability of mammillary cap size, the incidence of poor mammillary cap quality, incidence of late fusion, alignment, Type A bodies, Type B bodies, and cubic cone formation were greater in the free-range versus cage system and increased with flock age in both production systems. The incidence of confluence and early fusion were greater in cage eggs and decreased with age in both production systems. Significantly lower total microbial load was observed for cage compared with free-range eggs, but the overall bacterial load recorded in this study was low. It can be concluded that cage eggs were better in overall quality when directly compared with free-range eggs.  相似文献

1. Effects of two housing systems (cages vs outdoor) on external and internal egg characteristics were investigated. 2. In total 785 eggs from three different lines in cages and 268 eggs from outdoor-housed layers were examined for egg weight, albumen, yolk and shell content, albumen height and pH, and albumen and yolk dry matter content. 3. Interactions between layer age and housing systems were found for egg weight, eggshell content, albumen height, albumen pH, and dry matter content of the albumen and yolk. This was mainly due to the greater variation with age in the outdoor layers, compared to the caged layers. 4. Irrespective of age eggs from outdoor layers were relatively broader than eggs from the caged layers. Yolk colour was considerably darker in the outdoor group (11.0 vs 9.3). 5. We concluded that it is more difficult to maintain constant external and internal egg quality in an outdoor housing system than in a battery cage system. Factors that determine the greater fluctuations in internal egg quality need to be investigated.  相似文献

This study was conducted to determine the relationship between bacteria destruction on poultry carcass skin and bacteria in raw ground poultry meat from the same carcasses. Immersion time in boiling water of broiler chicken whole carcasses required for maximum reduction of naturally occurring aerobic bacterial count on skin was measured. Treatments for chicken carcasses consisted of immersion in boiling water (approximately 95 degrees C) for 0, 0.5, 1, 1.5, 2, 2.5, 3, 3.5, and 4 min. Four skin samples taken following treatment and three taken from subsequently ground carcass meat were analyzed for total aerobic plate counts (APC). Analysis of the data indicated a linear increase in bacterial destruction on skin with increased boiling water immersion time from 0 to 4 min. Reduction of skin bacteria to less than 1 log10 occurred at 3 min carcass immersion or longer. The analysis also indicated that treatment with boiling water and removal of skin was effective in reducing bacterial counts in ground meat to similar levels at all treatment times from 0.5 to 4.0 min. Findings from this study indicated that a boiling water immersion intervention and removal of skin could reduce subsequent bacteria contamination of ground meat. This intervention could minimize the risk of pathogen-contaminated primary processed poultry carcasses used in further processing.  相似文献

The aim of the present study was to determine the elimination of Salmonella by different lactic acid concentrations in microbiological media and on turkey carcass elements. The average bacteria counts in the control samples without lactic acid were: 1.8 x 10(8), 1.1 x 10(8) and 2.3 x 10(8), for S. Enteritidis, S. Anatum and S. Typhimurium, respectively. The concentration of lactic acid of 0.1% in the agar media completely inhibited the growth of all Salmonella strains. At 0.05% lactic acid concentration, the bacteria count was 2 log cycles lower and at a 0.03% solution it was 1 log cycle lower than that in the respective control samples. However, the examined bacteria developed in the presence of 0.02% and 0.01% lactic acid concentrations and their counts fell into the same log brackets. An analysis of the experimental results obtained from turkey carcass elements immersed in the lactic acid solution showed that the Salmonella identification rate was determined by the bacteria inoculum spread over the turkey carcass surface. The contamination of 10(1) CFU of Salmonella spread onto the turkey carcass was completely eliminated by immersing the carcasses in 1% or 2% lactic acid solutions. The contamination of turkey carcass elements with 10(2) CFU of S. Enteritidis and their immersion in 2% lactic acid solution for 15 min resulted in the reduction of the number of samples with Salmonella compared to the number of control samples with Salmonella. At contaminations of 10(3) CFU on the carcass surfaces, the immersions in 1% and 2% lactic acid solutions did not reduce Salmonella counts.  相似文献

Phage therapy reduces Campylobacter jejuni colonization in broilers   总被引:1,自引:0,他引:1  
The effect of phage therapy in the control of Campylobacter jejuni colonization in young broilers, either as a preventive or a therapeutic measure, was tested. A prevention group was infected with C. jejuni at day 4 of a 10-day phage treatment. A therapeutic group was phage treated for 6 days, starting 5 days after C. jejuni colonization of the broilers had been established. Treatment was monitored by enumerating Campylobacter colony forming units (CFU) and phage plaque forming units (PFU) from caecal content. Counts were compared with control birds not receiving phage therapy. A clear 3 log decline in C. jejuni counts was initially observed in the therapeutic group, however, after 5 days bacterial counts stabilized at a level 1 log lower than that of the control group. Colonization of C. jejuni in the prevention group was delayed by the treatment and after an initial 2 log reduction, colonization stabilized within a week at levels comparable to the therapeutic group. The CFU and PFU counts displayed opposing highs and lows over time, indicative of alternating shifts in amplification of bacteria and phages. There were no adverse health effects from the phage treatment. Two different phages were combined as therapeutic treatment of Campylobacter positive chickens challenged at the age approaching broiler harvest. This again resulted in a significant decrease in Campylobacter colonization. We conclude that phage treatment is a promising alternative for reducing C. jejuni colonization in broilers.  相似文献

Egg weight, shell thickness, number of pores, cuticle deposition and ability of Salmonella enterica serovar Enteritidis (SE) to penetrate the shell were determined for eggs from one layer flock through the entire production period. 2. Penetration was assessed by filling the eggs with a selective medium that allowed visualising Salmonella growth on the inside of the shell and membrane complex. After inoculation of each shell with on average 2.59 log cfu, the eggs were stored for up to 20 d at 20 degrees C and 60% relative humidity (RH). 3. On average 38.7% of the eggshells became penetrated. Mostly penetration occurred on d 3. Although it affected all shell characteristics studied, hen age did not significantly influence eggshell penetration. 4. No correlations were observed between any of the shell characteristics studied and the ability of SE to penetrate the shell. The growth of SE on the shell is of major importance because shell contamination at 20 d of storage and SE penetration were highly correlated.  相似文献

1. The present study was conducted to investigate the effects of inulin on laying hens. A total of 360 Brown Nick laying hens were divided randomly into 6 groups of 60 with 6 replicates of 10 hens and fed on diets containing 0 (control), 0·1, 0·5, 1·0, 1·5 or 2·0% inulin during the 4-week trial. 2. Dietary supplementation of inulin reduced cholesterol concentration (mg/g yolk) and content (mg/egg) in eggs. Cholesterol content in eggs decreased linearly with increasing levels of dietary inulin level. 3. Supplementation of inulin in diets decreased coliform bacteria counts and pH in the caecum. The lowest coliform bacteria counts (6·30 ± 0·03 log10 cfu/g) and pH (6·47 ± 0·01) were obtained in the 2·0% inulin group, the two indices decreasing by 21·6% and 3·0% respectively, compared with the control group. Coliform bacteria count and pH were changed linearly in accordance with increasing levels of dietary inulin level. Caecal Bifidobacteria counts were increased in the 2·0%-inulin group. 4. Inulin supplementation of layer diets did not appear to have any adverse effects on laying rate, egg weight, feed intake, feed conversion efficiency, cracked-egg rate, eggshell thickness or Haugh unit compared with the control laying hens. 5. Therefore, dietary supplementation with inulin may lead to the development of low-cholesterol chicken eggs as demanded by health-conscious consumers.  相似文献

Concentration of airborne endotoxins, airborne aerobic bacteria and airborne aerobic gram-negative bacteria were measured in 3 rabbit houses. Further, the species composition of the airborne gram-negative bacterial flora was investigated. The total amount of airborne endotoxin ranged from 22 to 774 EU/m3 (Endotoxin Units/m3). The number of total airborne aerobic bacteria varied between 780 and 20100 CFU/m3, the number of airborne aerobic gram-negative bacteria between 39 and 1030 CFU/m3. Most gram-negative bacterial isolates belonged to the family Enterobacteriaceae with E. coli as primary species. In two rabbit houses also airborne Pasteurella multocida spp. multocida, the most common respiratory pathogen of rabbits, was isolated.  相似文献

OBJECTIVE: To evaluate the effects of footwear hygiene protocols on bacterial contamination of floor surfaces in an equine hospital. DESIGN: Field trial. PROCEDURES: Footwear hygiene protocols evaluated included use of rubber overboots with footbaths and footmats containing a quaternary ammonium disinfectant, rubber overboots with footbaths and footmats containing a peroxygen disinfectant, and no restrictions on footwear type but mandatory use of footbaths and footmats containing a peroxygen disinfectant. Nonspecific aerobic bacterial counts were determined via 2 procedures for sample collection and bacterial enumeration (contact plates vs swabbing combined with use of spread plates), and the effects of each footwear hygiene protocol were compared. RESULTS: There were no consistent findings suggesting that any of the protocols were associated with differences in numbers of bacteria recovered from floor surfaces. Although there were detectable differences in numbers of bacteria recovered in association with different footwear hygiene protocols, differences in least square mean bacterial counts did not appear to be clinically relevant (ie, were < 1 log10). CONCLUSIONS AND CLINICAL RELEVANCE: Although cleaning and disinfection of footwear are important aids in reducing the risk of nosocomial transmission of infectious agents in veterinary hospitals, the numbers of aerobic bacteria recovered from floor surfaces were not affected by use of rubber overboots or the types of disinfectant used in this study. Further study is warranted to evaluate the usefulness of footwear hygiene practices relative to their efficacy for reducing transmission of specific pathogens or decreasing nosocomial disease risk.  相似文献

蛋产品安全一直是人们重点关注的问题,也是研究的热点。随着福利养殖的推广,鸡蛋被微生物污染的机率增大。除了加强饲养管理之外,强化鸡蛋自身保护屏障从而抵抗细菌入侵也十分重要。胶护膜是蛋壳最外层覆盖的无色透明膜,鸡蛋产出前1.5~2.0 h于子宫部形成,是蛋抵御微生物污染的重要屏障,研究表明胶护膜较厚的蛋不易被细菌入侵。鸡蛋蛋壳胶护膜主要由糖蛋白(90%)、多糖(4%)、脂质(3%)和以羟基磷灰石晶体形式存在的无机磷(3%)组成。胶护膜中含有溶菌酶C、卵转铁蛋白、ovocleidin-32等多种抗菌蛋白,可有效地阻碍细菌跨壳污染。此外,胶护膜能终止蛋壳矿化、帮助蛋产出前在子宫中翻转、调节水气交换以延长鸡蛋的保存时间。本文对胶护膜的结构、生物学功能、胶护膜品质评价方法及胶护膜质量的影响因素等方面进行了阐述,并对尚待解决的问题展开了探讨。  相似文献

Haylage for horses is often harvested in late plant maturity, which could be associated with an increased risk of impaired hygienic quality in the forage and short aerobic storage stability after bale opening, but knowledge in this area is scant. An experiment was conducted in which the microbial composition was analyzed before and after conservation of primary growth haylage harvested early (June), middle (July), and late (August) in the season during 1 year. The counts of yeast, enterobacteria, and lactic acid bacteria (LAB) in preconserved herbage increased with the advancing harvest time (P ≤ .02). After conservation, the August haylage had increased counts of enterobacteria (log 4.3 colony-forming unit [CFU]/g) and LAB (log 6.5 CFU/g), compared with the June and July haylage (log ≤1.7 CFU/g for enterobacteria and ≤5.7 CFU/g for LAB, P < .001). The yeast counts were the lowest in the June haylage (log 5.0 CFU/g) compared with the July and August haylage (log ≥6.3 CFU/g, P < .001). After conservation, the mold counts were lower in the June haylage and greater in the August haylage (P = .01). In the preconserved herbage, Cladosporium cladosporioides was the most common mold species in June but Fusarium poae was in July, and Mucor fragilis in August. After conservation, Penicillium carneum was the only species found in the June haylage, with M. circinelloides most frequently found in the July haylage and M. hiemalis and M. circinelloides found at similar frequencies in the August haylage. An advanced harvest time resulted in greater counts of enterobacteria, yeast, and LAB and an increased number of mold species in the conserved haylage. The aerobic storage stability of the opened haylage bales measured by temperature was similar among the harvests.  相似文献

The aim of this study was to analyse the qualitative composition of Gram-negative microbes, mainly of the family Enterobacteriaceae, including pathogenic bacteria such as Salmonella, in the albumens and yolks and on the shells of hens' eggs, depending on their source and on the temperature and duration of their storage. A total of 375 table eggs were studied, from a large-scale poultry farm, a small-scale poultry farm and a supermarket. Each group was divided into 5 subgroups according to the temperature and duration of their storage during the study. Two serotypes of bacteria of the genus Salmonella were identified: S. Enteritidis and S. Arizonae. Strains of Salmonella spp. were also isolated. Apart from Salmonella and Escherichia coli, among the most frequently isolated bacteria of the family Enterobacteriaceae were Enterobacter spp., Klebsiella spp. and Citrobacter freundii. Qualitative analysis of the bacterial microflora of the eggs also showed the presence of other Gram negative bacteria, including Acinetobacter spp., Pseudomonas spp., Tatumella ptyseos, Providencia stuartii, Serratia liquefaciens, Flavimonas oryzihabitans, Vibrio metschnikovii, Leclercia adecarboxylata, Kluyvera spp., Rahnella aquatilis, Proteus mirabilis, and Achromobacter spp. The study demonstrated that the conditions applied, i.e., the temperature and duration of storage, did not significantly influence the prevalence of particular species of Gram-negative bacteria in the eggs. However, based on the analysis of contamination of eggs with Salmonella depending on their source, it can be concluded that the system in which the hens are housed affects the risk of contamination of eggs with these pathogens.  相似文献

1. The eggshell cuticle is the proteinaceous outermost layer of the eggshell which regulates water exchange and protects against entry of micro-organisms. In this study, we investigated the hypothesis that the cuticle may also reduce microbial contamination by providing a chemical defence. 2. Outer eggshell and cuticle protein was extracted from domestic chicken (Gallus gallus), duck (Anas platyrhynchos) and goose (Anser anser) eggs by HCl and urea treatment, respectively. Antimicrobial activity of the extracts against Gram-positive and Gram-negative bacteria was evaluated. 3. C-type lysozyme, ovotransferrin and ovocalyxin-32 were identified in all extracts by Western blotting. All extracts from all species demonstrated lysozyme enzymatic activity. Immobilised c-type lysozyme retained some enzymatic activity. Protein extracts demonstrated activity against Pseudomonas aeruginosa, Staphylococcus aureus and Bacillus subtilis suggesting the action of antimicrobial proteins in addition to lysozyme. 4. The results suggest that the antimicrobial outer eggshell and cuticle proteins present in a number of avian species may be a mechanism which enhances avian reproductive success.  相似文献

AIMS: This paper compares changes in visible and microbiological contamination on lamb carcasses dressed using one system which includes process factors previously identified as potential critical control points for HACCP-based approaches to hygienic slaughter and dressing, and another system which excludes those factors. METHODS: Longitudinal changes in microbiological and visible contamination of lamb carcasses were quantified in two slaughterhouses, one utilising clean, shorn, unwashed lambs in an inverted dressing system, the other utilising dirty, woolly, washed lambs in a traditional dressing system. Excision samples (5 cm2 each) for microbiological analyses were taken from two sites on 25 carcasses per treatment group immediately after pelting, at the completion of dressing, after overnight chilling and after boning and packaging. Visible contamination on the surface of 300 carcasses per treatment group was assessed after pelting and after overnight chilling. RESULTS: Mean aerobic plate counts and Escherichia coli counts on the leg and loin in the inverted dressing system were low after pelting (log10/cm2 1.86 and 1.71; log10/cm2 0.13 and 0.05 respectively) but generally showed statistically significant increases through to final packaging. In the conventional dressing system, there were much higher counts on the leg and loin after pelting (log10/cm2 4.66 and 2.71; log10/cm2 2.21 and 0.24 respectively), but subsequent handling of the carcass by slaughter line workers had no measurable deleterious effects. The inverted dressing system resulted in final mean aerobic plate counts on meat at packaging that were 1.38% (leg) and 48.98% (loin) of those derived from the conventional dressing system. Mean E. coli counts from the inverted system were 21.37% (leg) and 67.61% (loin) of those from the conventional system. There was an inverse relationship between the prevalence of carcasses with visible faecal contamination and their microbiological status. CONCLUSION: Significant reductions in microbiological contamination of sheep carcasses can be brought about by HACCP-based systems. Possible critical control points are pre-slaughter presentation status (including avoidance of pre-slaughter washing), inverted dressing, handling by slaughter line workers and meat inspectors, and chilling. Use of levels of visual faecal contamination as an on-line monitoring parameter for slaughter hygiene can give erroneous results. Interactions between different process steps may alter the effectiveness of the HACCP plan, and successful design and application depends on a detailed knowledge of the specific process utilised in each slaughterhouse.  相似文献

[目的]研究鸡蛋暗斑对蛋品质及其抗氧化能力的影响。[方法]从饲喂同种日粮、饲养环境相同、同日龄褐壳蛋鸡同一批次所产的褐壳蛋中,用灯光照射法挑选出24枚典型正常蛋、24枚典型暗斑蛋进行试验,并分别对正常蛋和暗斑蛋进行蛋品质、蛋黄抗氧化指标的测定及比较。[结果]蛋品质比较:暗斑蛋与正常蛋相比,其蛋形指数、蛋壳强度、蛋重、哈氏单位、蛋白高度、鸡蛋等级、蛋黄颜色、蛋黄重、蛋壳重、蛋清重、蛋壳厚度、蛋黄比重、蛋壳比重、蛋清比重均差异不显著(P>0.05);暗斑蛋蛋壳颜色极显著深于正常蛋(P<0.01)。抗氧化指标比较:暗斑蛋蛋黄的总抗氧化能力(T-AOC)显著低于正常蛋蛋黄的总抗氧化能力(P<0.05),暗斑蛋蛋黄的丙二醛(MDA)含量与正常蛋蛋黄的丙二醛含量相比差异不显著(P>0.05)。[结论]与正常蛋相比,暗斑蛋的蛋壳颜色较深,总抗氧化能力较差。  相似文献
