Both Bos indicus (zebu) and Bos javanicus (banteng) contribute to the Indonesian indigenous livestock, which is supposedly of a mixed species origin, not by direct breeding but by secondary cross-breeding. Here, the analysis of mitochondrial, Y-chromosomal and microsatellite DNA showed banteng introgression of 10-16% in Indonesian zebu breeds with East-Javanese Madura and Galekan cattle having higher levels of autosomal banteng introgression (20-30%) and combine a zebu paternal lineage with a predominant (Madura) or even complete (Galekan) maternal banteng origin. Two Madura bulls carried taurine Y-chromosomal haplotypes, presumably of French Limousin origin. There was no evidence for zebu introgression in five populations of the Bali cattle, a domestic form of the banteng.  相似文献

In the current study, milk protein variation was examined in cattle (Bos indicus), mithun (Bos frontalis), yak (Bos grunniens) and their hybrid populations in Bhutan to estimate genetic variability, conduct genetic characterization and assess the possibility of gene flow between mithun and cattle. Isoelectric focusing of 372 milk samples from 11 populations detected four molecular types of β‐lactoglobulin (A, B, E and M), five molecular types of αS1‐casein (A, B, C, E and X) and three molecular types of k‐casein (A, B and X). Mithun and yak shared alleles but were found to exhibit different allele frequencies for the proteins studied. The degree of genetic variability within populations was measured by average heterozygosity and ranged from 24–40% in cattle, 26% for yak and 33% for mithun. We also resolved the traditional mithun and cattle hybridization system via principal component analysis. Our results suggested secondary introgression of mithun genes to the village Thrabum population, and a close genetic relationship between Bhutanese indigenous cattle and Indian cattle.  相似文献

Biochemical polymorphisms and major mtDNA haplotypes in Bergschaf breeds and Waldschaf as contribution to descent of domestic sheep Morphological similarities between domestic sheep and certain subspecies of Ovis ammon L. 1758 are not necessarily proof of the domestication of a specific subspecies, as crossbreeding of foreign wild sheep populations of different genetic origin with domestic sheep could have led to observed similarities. The maternal inherited mitochondrial genome offers the opportunity to trace modern domestic sheep breeds across numerous generations to presumptive wild ancestors regardless of domestication induced morphological changes and crossbreeding, etc. An investigation based on haplotype specific HinfI – mtDNA fragments and protein polymorphisms in different mountain sheep breeds thought to represent maternal descendants of the Torfschaf as well as in the Waldschaf breed, which belongs to the Zaupel group, were used to help clarify if the major European mtDNA haplotype B was associated with the Torfschaf (Ovis aries palustris RÜTIMEYER 1862), the oldest domesticated sheep in Europe. Samples derived from Steinschaf of the Slovenian and Bavarian type, white and brown varieties of Bergschaf, Brillenschafe from Bavaria and Villnösserschafe showed haplotype B. A single investigated ram of the Bergamasker breed was of the same haplotype. mtDNA haplotype A was detected in more than 50% of the Waldschaf animals. This demonstrates that not all sheep breeds derived from Zaupelschaf are descendants of the same wild ancestor. Within haemoglobin (HBB) the variants HBBA and HBBB occur in a well balanced pattern in all breeds investigated, while the frequency of HBBA was high (0.857) in the brown Bergschaf. Steinschafe of Bavarian type were polymorphic at the CA2‐locus and Waldschafe at the GC‐locus. At the transferrin locus, differences in the occurrence and frequency of the different alleles could be demonstrated between populations. Relationships between Torfschaf, Kupferschaf, Steinschaf, Zaupelschaf, Bergamasker and European Mouflon are discussed. It is likely that the domesticated ancestors of most European sheep can be traced from the Torfschaf to an as yet unknown wild sheep (Ovis ammon) subspecies in Asia.  相似文献

Fecal DNA analysis is a useful tool for the investigation of endangered species. Tamaraw (Bubalus mindorensis) is endemic to the Philippine island of Mindoro but knowledge of its genetic and ecological information is limited. In this study, we developed a species identification method for tamaraw by fecal DNA analysis. Eighteen feces presumed to be from tamaraw were collected in Mount Iglit‐Baco National Park and species‐known feces from domestic buffaloes and cattle were obtained from a farm. Additionally, one species‐unknown fecal sample was obtained in Mount Aruyan Preserve, where the sighting of tamaraw has not been reported in recent years. Based on DNA sequence data previously reported, the genus Bubalus‐ and tamaraw‐specific primers for PCR of cytochrome b gene were newly designed. The Bubalus‐specific primer yielded a 976 bp fragment of cytochrome b for all fecal samples from tamaraw and domestic buffaloes, but not for cattle, whereas the tamaraw‐specific primer yielded a 582 bp fragment for all tamaraw fecal samples and for one of the four domestic buffalo samples. PCR‐RFLP (restriction fragment length polymorphism) analysis of the 976 bp PCR fragment with AvrII or BsaXI provided distinct differences between tamaraw and domestic buffalo. PCR‐RFLP analysis also showed that the species‐unknown sample obtained in Mount Aruyan Preserve, originates from tamaraw.  相似文献

State‐of‐the‐art health care includes genome sequencing of the patient to identify genetic variants that contribute to either the cause of their malady or variants that can be targeted to improve treatment. The goal was to introduce state‐of‐the‐art health care to cats using genomics and a precision medicine approach. To test the feasibility of a precision medicine approach in domestic cats, a single cat that presented to the University of Missouri, Veterinary Health Center with an undiagnosed neurologic disease was whole‐genome sequenced. The DNA variants from the cat were compared to the DNA variant database produced by the 99 Lives Cat Genome Sequencing Consortium. Approximately 25× genomic coverage was produced for the cat. A predicted p.H441P missense mutation was identified in NPC1, the gene causing Niemann‐Pick type C1 on cat chromosome D3.47456793 caused by an adenine‐to‐cytosine transversion, c.1322A>C. The cat was homozygous for the variant. The variant was not identified in any other 73 domestic and 9 wild felids in the sequence database or 190 additionally genotyped cats of various breeds. The successful effort suggested precision medicine is feasible for cats and other undiagnosed cats may benefit from a genomic analysis approach. The 99 Lives DNA variant database was sufficient but would benefit from additional cat sequences. Other cats with the mutation may be identified and could be introduced as a new biomedical model for NPC1. A genetic test could eliminate the disease variant from the population.  相似文献

Kazakhstan is the largest landlocked country and contains two important propagation routes for livestock from the Fertile Crescent to Asia. Therefore, genetic information about Kazakhstani cattle can be important for understanding the propagation history and the genetic admixture in Central Asian cattle. In the present study, we analyzed the complete mtDNA D‐loop sequence and SRY gene polymorphism in 122 Kazakhstani native cattle. The D‐loop sequences revealed 79 mitochondrial haplotypes, with the major haplogroups T and I. The Bos taurus subhaplogroups consisted of T (3.3%), T1 (2.5%), T2 (2.5%), and T4 (0.8%) in addition to the predominant subhaplogroup T3 (86.9%), and the Bos indicus subhaplogroup of I1 (4.1%). Subsequently, we investigated the paternal lineages of Bos taurus and Bos indicus, however, all Kazakhstani cattle were shown to have Y chromosome of Bos taurus origin. While highly divergent mtDNA subhaplogroups in Kazakhstani cattle could be due to the geographical proximity of Kazakhstan with the domestication center of the Fertile Crescent, the absence of Bos indicus Y chromosomes could be explained by a decoupling of the introgression dynamics of maternal and paternal lineages. This genetic information would contribute to understanding the genetic diversity and propagation history of cattle in Central Asia.  相似文献

Dwarf Lulu cattle, the only Bos Taurus type of cattle in Nepal, are raised under severe environments in the mountainous zone of that country. In the present study, the body measurement traits, cytogenetic and molecular genetic characteristics of the Lulu cattle are investigated. Blood samples were collected from 31 animals in four villages (altitudes 2590–3550 m) in the southern part of Mustang. The Lulu cattle had a normal karyotype with 2n = 60, XY or XX. Only one male examined had a large submetacentric X‐chromosome and a small submetacentric taurine type Y‐chromosome. The mitochodrial DNA (mtDNA) genotypes were analyzed by PCR mediated restriction fragment length polymorphisms, displacement (D)‐loop region PCR mediated single strand conformation polymorphisms, and D‐loop region sequences. Many base substitutions were found in the D‐loop region, suggesting that the Lulu cattle originated from at least 10 maternal lines. Three types of mtDNA from these cattle were found, the Bos taurus type (n = 23), the Bos indicus type (n = 6), and the Bos grunniens type (n = 2). In the village at the lowest altitude, four of the five cows were of the Bos indicus type. These results indicated that mtDNA types of the Lulu cattle mostly belong to Bos taurus, but have been hybridized with Bos indicus cattle in lower‐elevation regions in their maternal lineage.  相似文献

2′, 5′‐Oligoadenylate synthetases (OAS) are important components of an interferon‐mediated antiviral pathway. No polymorphisms in exonic regions of bovine OAS1 gene have been identified and associated with reproduction traits. The objective of the study was to detect and evaluate the effects of mutations in exonic region of bovine OAS1 gene with reproduction traits in cattle. DNA samples collected from 250 individual cows of two Indian dairy breeds (Sahiwal and Frieswal) of cattle were used in the study. The genetic variants of the OAS1 gene were identified with polymerase chain reaction–single‐strand conformation polymorphism (PCR‐SSCP) and sequence analysis using seven set of primer pairs. The PCR‐SSCP analysis revealed polymorphism in the fragments comprising of exon 2, exon 5 and first fragment of exon 6 while the fragments of exons 1, 3, 4 and second fragment of exon 6 were monomorphic in Sahiwal and Frieswal cattle. The mutations in the amplified region comprising of exon 2 were found to have significant association with age at first breeding and calving, service period, dry period and pregnancy rate. Significant associations were found between SNPs in the exon 5 and service and dry periods of the animal, whereas the genetic variants in the first fragment of the exon 6 showed significant association with age at first breeding and calving. To our knowledge, this study demonstrated for the first time that the polymorphisms in OAS1 gene were associated with reproductive traits and it can be chosen as a candidate gene for improvement of reproductive performance of cattle.  相似文献

Endogenous retroviruses present in the genomes take a specific role in placental formation in various vertebrates, including bovine and sheep. Fematrin‐1, which is the envelope (Env) protein of bovine endogenous retrovirus found in bovine placenta, is involved in the formation of fetomaternal hybrid cells in cattle placenta. This study was conducted to clarify whether fematrin‐1 possesses fusogenic activity in trophoblast cells. Another question is whether Env proteins only have species‐specific activity or not. For this, fematrin‐1 gene was transfected in ovine trophoblast cells, and we examined fusogenic activity with Cos‐7 cells. Although fematrin‐1 fusogenic activity was detected in both neutral and acidic pH conditions, acidic condition significantly enhanced it. These activities were rather weaker than those of vesicular stomatitis virus G protein as a positive control. However, the ratio of fematrin‐1 and vesicular stomatitis virus G protein fusion index was confirmed similar to those in the previous reports. Some fusion cells showed multinucleate cells. These results imply that fematrin‐1 is involved in the formation of trophoblast hybrid cells even in different species trophoblastic cells.  相似文献

Protein JY‐1 is an oocyte‐specific protein that plays an important regulatory role in the granulosa cell layer and during the early embryo development stages. It is the first specific protein of maternal origin discovered in a single‐ovulating species. In this study, the exon regions of the JY‐1 gene were characterized by sequencing in 20 unrelated cattle (Bos taurus indicus) and 20 unrelated buffaloes (Bubalus bubalis). Eighteen polymorphisms were detected in cattle and 10 polymorphisms in buffaloes. Some of the polymorphisms were identified in codifying regions and caused amino acid changes. The insertion of a thymine was detected in the codifying region of exon 3 of the buffalo sequence when compared to the cattle one. This insertion causes a change in the codons frameshift from this point onwards, modifying the 19 terminal amino acids of the buffalo protein and creating a premature stop codon. This finding may explain reproductive differences between cattle and buffaloes in terms of follicle recruitment, embryo development and incidence of twin pregnancies.  相似文献

The objectives of this study were to detect effective genetic polymorphisms of bovine growth hormone (bGH) gene associated with calf weight in Japanese Black cattle. Fifty‐eight sires and 47 breeding cows were used to detect the polymorphisms in exons by single‐strand conformation polymorphism (SSCP). Four homozygous and six heterozygous SSCP genotypes were identified in exon 5. Although each single nucleotide polymorphism (SNP) had been reported, these genotypes were caused by three SNPs at the nucleotide positions 2141, 2277 and 2291. Four haplotypes C‐C‐A, G‐C‐A, C‐C‐C and G‐T‐A were newly identified. It was suggested that other haplotypes not detected in this study may not exist, considering the allele frequencies reported in Bos taurus and Bos indicus, and the migrating process of native Japanese cattle. Thereafter, we examined associations between the detected polymorphic sites in exon 5 by PCR – restriction fragment length polymorphism and calf weight using 53 breeding dams and 135 calves. The birth weights of calves with haplotype G‐C‐A are significantly lighter and calves' weights produced by cows with such haplotype are also lighter at 30 days old, using regression analysis. Although further research is necessary, these results may serve as a useful criterion to select breeding stocks, especially in maternal abilities.  相似文献

Antibiotic resistance mediated by bacterial production of extended‐spectrum beta‐lactamase (ESBL) is a global threat to public health. ESBL resistance is most commonly hospital‐acquired; however, infections acquired outside of hospital settings have raised concerns over the role of livestock and wildlife in the zoonotic spread of ESBL‐producing bacteria. Only limited data are available on the circulation of ESBL‐producing bacteria in animals. Here, we report ESBL‐producing Escherichia coli in wild common vampire bats Desmodus rotundus and livestock near Lima, Peru. Molecular analyses revealed that most of this resistance resulted from the expression of blaCTX‐M‐15 genes carried by plasmids, which are disseminating worldwide in hospital settings and have also been observed in healthy children of Peru. Multilocus sequence typing showed a diverse pool of E. coli strains carrying this resistance that were not always host species‐specific, suggesting sharing of strains between species or infection from a common source. This study shows widespread ESBL resistance in wild and domestic animals, supporting animal communities as a potential source of resistance. Future work is needed to elucidate the role of bats in the dissemination of antibiotic‐resistant strains of public health importance and to understand the origin of the observed resistance.  相似文献

Insulin‐like growth factor‐1 (IGF‐1) is one of the important factors for growth, milk production and reproductive functions and mainly released from the liver in response to growth hormone (GH) via GH receptor (GHR) in cattle. Recently, some single nucleotide polymorphisms (SNPs) were identified in the bovine GHR gene. Some GHR‐SNPs were shown to be related to plasma IGF‐1 concentration in cattle. Hence, the capacity to IGF‐1 production in the liver might be affected by GHR‐SNP and associated with performance in the future. This study examined whether GHR‐SNP is associated with IGF‐1 production in the liver of pre‐pubertal heifers. In 71 Holstein calves, blood samples for genomic DNA extraction were obtained immediately after birth. To genotype the GHR‐SNPs in the promoter region, polymerase chain reaction (PCR) products were digested with restriction enzyme NsiI (cutting sites: AA, AG and GG). All heifers at 4 months of age were intramuscularly injected with 0.4 mg oestradiol benzoate. Blood samples were obtained from the jugular vein just before (0 h) and 24 h after injection. The number of AA, AG and GG at the NsiI site was 0, 17 and 54 respectively. In AG and GG, plasma GH concentrations were higher pre‐injection than 24 h post‐injection (p < 0.01). Moreover, plasma GH concentrations in AG post‐injection were higher than in GG (p < 0.05). In contrast, the GG genotype exhibited higher plasma IGF‐1 concentrations in pre‐injection than post‐injection (p < 0.01), although oestradiol did not change IGF‐1 concentration in the AG genotype. We conclude that the GG polymorphism in the promoter region of GHR is associated with a higher potential capacity of IGF‐1 production in the liver of cattle.  相似文献

The main objective of the study was to determine the possible influence of host species on the maintenance of ticks in the field by determining the relative contribution of game animals compared to domestic animals. The study was carried out on a game ranch 32 km south-east of Nairobi. Tick counts were carried out on 30 Zebu cattle (Bos indicus) aged 1 to 3 years and 20 red Maasai sheep (Ovis aries) aged 6 months to 1 year grazing with game animals in a common area for a period of 2 years and these counts were compared with those on eland and Thomson's gazelle. Half-body counts were carried out on the cattle and sheep once every week. To avoid excessive stress, the animals were dipped in amitraz whenever the half body counts exceeded 50 fully engorged female ticks of any species. Tick counts on two wild animal species (eland (Taurotragus oryx) and Thomson's gazelle (Gazella thomsonii)) were carried out during the weekly culling of the herbivores. The results revealed that there was no significant difference in the number ticks per square metre between the wild ungulates and the domestic animals.  相似文献

A cytogenetical study using metaphase chromosomes from cultured lymphocytes, was made of 2 Banteng (Bibos banteng) steers and 218 bulls representing 13 purebreeds (Bos taurus type, Bos indicus type and Sanga) and 7 cross-breeds. Studies were made of photographic karyotypes of Giemsa stained and C-banded chromosomes of bulls of each breed and of B-banded chromosomes from 3 breeds of Bos indicus and one cross-breed Australian Friesian Sahiwal) cattle. The relative lengths of chromosomes of Bos taurus and Bos indicus bulls were compared and significant difference in relative lengths of the X chromosomes were noted between these two species. There was a differences in morphology of the Y chromosomes; Sanga, Banteng and Bos taurus type breeds had a small submetacentric Y chromosome, except for the Jersey which had a metacentric Y chromosome. All Bos indicus type bulls had an acrocentric Y chromosome but the Droughtmaster breed had two forms of the Y chromosome (submetacentric and acrocentric). The C-banding patterns of the autosomes and X chromosomes were similar for all breeds while those of the Y chromosomes of Bos indicus type cattle allowed their accurate identification. G-banding patterns of Bos indicus resembled those of Bos taurus and enabled pairing of homologous chromosomes. Centromeres of the autosomes were unstained but those of the sex chromosomes were darkly stained.  相似文献

Lulu is a dwarf cattle population bred in the Mustang district of western and central Nepal. This area is located around the habitat boundary between Bos taurus and Bos indicus. The peculiarities of Lulu are their small size (weight range in the adult female: 68–153 kg) and rearing in high mountain areas at 2800 m to 4000 m in altitude. There were 5770 head of Lulu cattle in the Mustang district in 1998, 4333 females and 1437 males. The morphological appearance of Lulu is Bos taurus. However, one of the five Lulu studied in Kagbeni, Mustang had a Bos indicus mitochondrial DNA type based on the D‐loop sequence, while the other four were Bos taurus. It is suggested that there are hybrids of Lulu with Bos indicus maternal lineage in a mostly taurine‐breed genetic background. Steps must be taken to preserve the unique Lulu.  相似文献

There are hump, humpless cattle and gayal distributed in Yunnan province, south‐west China, but their genetic background remains unclear. To determine the origin and genetic diversity of Yunnan gayal and cattle (Diqing, Nujiang and Wenshan cattle), we analysed mtDNA control region sequences of 71 samples and SRY gene sequences of 39 samples, together with the available sequences in GenBank. The neighbour‐joining phylogeny and the reduced median network analysis showed that Yunnan gayal originated from the hybridization between male Bos frontalis and female Bos taurus or Bos indicus, and that Yunnan cattle mostly originated from B. indicus, also containing some hybrids of male B. indicus and female B. taurus. The phylogenetic pattern of Yunnan cattle was consistent with the recently described cattle matrilineal pool from China and indicated more contribution to the Yunnan cattle from B. indicus than from B. taurus.  相似文献
