One hundred serum samples collected from free-ranging white rhinoceros (Ceratotherium simum) in Kruger National Park (KNP) during the 2007 capture season were selected for measurement of antibody levels to several different vector-borne viral agents. These infectious diseases were chosen to compare with an earlier serosurvey that had been conducted in KNP in rhinos during 1987-1997. Positive antibody titers were found against epizootic hemorrhagic disease (EHD) of deer (8%), Bluetongue (BT) (1%), and Rift Valley fever (RVF) (49%). However, none of the 100 animals tested had detected antibody levels to African horse sickness (AHS). These values were in sharp contrast to those measured in the 1987-1997 survey in KNP white rhinos (AHS 60%, EHD 30%, BT 37%, RVF 0%). Vector-borne viral infection prevalence in white rhinos in the same geographical location appears to vary over time and may be important for monitoring presence of pathogens in an ecosystem.  相似文献

Leopards Panthera pardus are classified in the IUCN Red List as Vulnerable, primarily due to habitat loss, natural prey base depletion and exploitation caused by various anthropogenic activities. Although protected areas are important for leopard conservation, the majority of suitable leopard habitat lies beyond protected area boundaries exposing individuals to different environmental, physiological and psychosocial stressors. This study aimed to examine the suitability of five different enzyme immunoassays (EIAs) for monitoring adrenocortical function in the leopard based on faecal glucocorticoid metabolite (fGCM) analysis. After performing an adrenocorticotrophic hormone (ACTH) stimulation test and investigating the stability of fGCM post-defaecation, faeces from free-ranging leopards in a peri-urban and a conservation area were collected to investigate the potential impact of habitat variability on glucocorticoid output. An EIA measuring fGCM with a 5α-3β-11β-diol structure performed best, demonstrating a ~200%–330% increase in fGCM concentrations approximately 40 h post-ACTH administration. Concentrations of fGCM remained quite stable for up to six days post-defaecation and showed a maximum increase of 8% and a maximum decrease of 9%. Although not significantly different, overall median fGCM concentrations were 68% higher in individuals utilising the peri-urban area compared with leopards utilising the conservation area. The ranges of fGCM concentrations between sites, however, were similar. Individual median fGCM concentrations differed distinctly between free-ranging males and females, possibly linked to female reproductive status. The established method can now assist in addressing some of the issues facing local wildlife managers, conservationists and researchers tackling various aspects related to leopard conservation and management under different land-use practices.  相似文献

A 46-yr-old male white rhinoceros (Ceratotherium simum) died during anesthesia following agonal excitation. On postmortem, a well-demarcated 2.5-cm tan mass was identified in the right adrenal gland. Histopathology confirmed the presence of a pheochromocytoma, and elevated levels of epinephrine in serum collected shortly prior to the animal's death, as compared with sera from healthy controls, demonstrated the functional nature of the tumor. Although rare, pheochromocytoma should be considered as a differential diagnosis in cases of suspected hypertension and acute death in rhinos.  相似文献

本研究测定了石林龙晖野生动物科研中心饲养的非洲白犀牛的红细胞数、血红蛋白、红细胞比积、平均红细胞体积、平均血红蛋白含量、平均血红蛋白浓度、红细胞宽度变异系数、红细胞宽度标准差、血小板数、平均血小板体积、白细胞数、淋巴细胞绝对值、中间细胞绝对值、粒细胞绝对值、淋巴细胞百分比、中间细胞百分比、粒细胞百分比、大血小板百分比、血小板比积、血小板分布宽度。通过比较分析,雌、雄犀牛在红细胞数、大血小板百分比差异性显著(O.01〈P〈0.05)。其他雌、雄犀牛各项参数差异性不显著(P〉0.05)。雄性白犀牛与公乌蒙马相比,二者的血红蛋白差异性不显著。雌性白犀牛血液中的红细胞数与母乌蒙马的红细胞数差异性不显著。雌、雄白犀牛与安西牛的红细胞数和血红蛋白差异性不显著。因此,在诊断白犀牛血液常规指标时可参考乌蒙马和安西牛相关血液常规指标。  相似文献

Behaviour, plasma cortisol and heart rate were measured in 4-6-week-old calves during and after dehorning with and without the use of sedation and analgesia. Six groups of eight Friesian male and female calves were studied; four groups were dehorned using an electrical cauterizing dehorner, heated to approximately 600 degrees C. In group 1, a cornual nerve block was performed and the animals were sham-dehorned using a cold dehorner. Group 2 was treated similarly but dehorned with the heated dehorner. Group 3 received a mixture of xylazine and butorphanol intramuscularly, and were hot dehorned 20 min after the injection. Group 4 received the same sedatives-analgesics as group 3, and after 5 min also had a cornual nerve block, followed by hot dehorning 15 min later. Group 5 was hot dehorned without any form of sedation or analgesia. A sixth group of calves without any treatment or handling was used as controls for the behavioural observations.Head and leg movements during dehorning were significantly reduced when the cornual nerve was blocked. During the 4 h after dehorning, the behaviour of calves having a cornual block continued to differ from those in group 5. The cornual block prevented short-term increases in plasma cortisol concentrations and the long-term increases in heart rate seen in group 5. It was concluded that routine field use of local analgesia using a cornual nerve block improved the welfare of young calves subjected to dehorning with a hot iron.  相似文献

Objective To determine the effect of a long-acting local anaesthetic on the plasma cortisol response in calves dehorned using a scoop.
Design A physiological study with controls.
Procedure Seventy female calves, 3 to 4 months old, were divided into seven groups and dehorned by scoop with or without a long acting local anaesthetic (bupivacaine) which provides local anaesthesia for 3 to 4 h. In some treatment groups the local anaesthetic was given a second time, after 4 h, to give 8 h of anaesthesia. Blood samples were taken before and for 9 h after treatment and plasma cortisol concentrations were measured.
Results Scoop dehorning caused a marked rise in plasma cortisol concentration, which returned to pre-dehorning values after 7 h. The cortisol concentrations of calves given bupiva-caine were similar to those of control animals for 4 h after dehorning, but then a significant and protracted rise occurred, with concentrations returning to pretreatment values at about 9 h after dehorning. In calves given a second treatment of bupivacaine 4 h after dehorning, the plasma cortisol concentration remained similar to control values for 8 h after dehorning but then increased sharply.
Conclusions During its period of activity long-acting local anaesthetic prevented the rise usually seen after scoop dehorning in plasma cortisol concentrations; by inference it also alleviated the pain-induced distress caused by this procedure. However, when its effects had worn off the plasma cortisol values rose sharply indicating that animals still experienced pain-induced distress despite local anaesthetic being administered.  相似文献

Excess dietary glucose may be a factor in several captive wildlife diseases and reproductive abnormalities. The first step in understanding the health consequences of diets high in glucose is to characterize how dietary glucose concentrations change circulating glucose profiles. We adapted the glycemic index approach to detect differences in blood glucose concentrations in white rhinos in response to different meals. Six white rhinos were fasted overnight then randomly assigned to be fed 5 kg of grass hay and one of five meals varying in digestible energy (DE) availability and source (10% DE glucose, 5% DE glucose, 10% DE pelleted horse feed, 10% DE lucerne hay, 10% DE grass hay). After eating, the blood glucose response peaked 45–90 min later and remained elevated up to 180 min. Area under the curve results demonstrated that the blood glucose response was not different between diets. However, at 90 min, serum glucose levels in rhinos eating the 10% lucerne hay diet were significantly lower than the 5% glucose and 10% glucose diets but not the 10% pellet nor 10% grass hay diets. The changes in blood glucose responses to different diets were similar in magnitude to reported domestic horse profiles but are higher than predicted by allometric scaling. We conclude that the grass hay, lucerne hay and low glycemic index horse pellets fed in this study resulted in similar blood glucose responses in white rhinos. The validation of the methodology used in this study is a first step towards elucidating the relationship between glucose, obesity, health and reproduction in rhinos.  相似文献

Measuring physiological stress reactions through the quantification of plasma cortisol often involves physical restraint, which acts as a stressor itself. Here, we present the validation of a non-invasive method for assessing adrenocortical activity as an indicator of stress in the bat-eared fox (Otocyon megalotis). By conducting an adrenocorticotropic hormone (ACTH) challenge, we examined the suitability of three enzyme immunoassays (EIAs) detecting 11,17 dioxoandrostanes (11,17-DOA) as well as faecal glucocorticoid metabolites (fGCM) with a 5β-3α-ol-11-one (3α,11oxo-CM), or 11,17,21-trihydroxy-4-ene-20-one structure (cortisol), respectively, for monitoring stress-related physiological responses in male and female bat-eared foxes. Our results suggest that the cortisol EIA seems most suitable for measuring fGCMs in this myrmecophageous mammal. Using the cortisol EIA, we compared fGCM concentrations of three populations of foxes. Only one population experienced a sudden change in social environment and the assay appeared to effectively detect the expected resulting increase in stress hormone levels. Therefore, the identified EIA is effective at detecting intraspecific variation in fGCM levels and hence is a useful tool to evaluate physiological stress responses in this species.  相似文献

OBJECTIVE: To reassess whether local anaesthesia before dehorning and wound cauterisation after dehorning eliminates the cortisol response to dehorning. METHODS: A control group was compared with three dehorned groups (dehorned alone, dehorned after local anaesthesia, dehorned after local anaesthesia then cautery of the amputation wounds). The local anaesthetic regimen used produced nerve blockade for about 5 h. Blood samples were taken before and after dehorning and plasma cortisol concentrations were determined. RESULTS: Dehorning caused a marked cortisol response lasting for about 7 h. Local anaesthetic reduced the plasma cortisol concentrations to control values for 5 h, after which they increased for a further 5 h. Subsequently in both groups, cortisol concentrations remained at control values until at least 24 h after treatment. Local anaesthetic combined with wound cautery virtually abolished the cortisol response until at least 24 h after dehorning. CONCLUSION: Combining local anaesthesia with cautery of the amputation wounds substantially reduced the cortisol response until at least 24 h after dehorning.  相似文献

Dehorning adult cattle is a surgical procedure causing distress of varying intensities that can be reflected in behavioural changes and alterations in plasma cortisol levels. Stress responses during the dehorning process were evaluated in 18 Red Pied cows. The cows were divided into 3 groups of six and kept in tie-stall housing. Those in the first group were dehorned under general anaesthesia (GA) induced by intravenous administration of xylazine and ketamine. The second group was dehorned under sedation and local anaesthesia (SLA) induced by intramuscular administration of xylazine and local anaesthesia with lidocaine. The third group was dehorned under local anaesthesia (LA) with lidocaine. Dehorning was performed with a foetotomy wire. Blood samples were taken 0.5 h before dehorning to determine cortisol levels, and, by means of a central venous catheter inserted into the jugular vein, during surgery at 0, 0.5, 1, 1.5, 2, 2.5, 3, 3.5, 4, 5, 6, 7 and 8 h post-surgery. Concurrently, occurrence of stress behaviours was assessed. Cortisol levels were measured by enzyme immunoassay (EIA). The lowest mean peak levels of plasma cortisol (82.53 +/- 6.04 nmol l(-1)), the most rapid return of plasma cortisol levels to baseline values (1.92 +/- 1.11 h), and the lowest occurrence of stress behaviours (2.38 +/- 5.83%) were noted in the SLA group. The highest mean peak levels plasma cortisol (113.86 +/- 25.65 nmol l(-1)), the slowest return of plasma cortisol levels to baseline values (3.83 +/- 2.18 h) and the most frequent occurrence of stress behaviours (65.48 +/- 28.72%) were observed in the LA group. There were significant differences between the SLA and LA groups in peak plasma cortisol levels (p = 0.011) and in occurrence of stress behaviours (p = 0.003). Sedation induced by intramuscular administration of xylazine in conjunction with local anaesthesia with lidocaine is considered the most suitable method of anaesthesia when dehorning adult cattle. Local anaesthesia with lidocaine alone was least suitable.  相似文献

In a time of increasing environmental change caused by anthropogenic disturbance, there is a greater need to understand animal adaptations to manmade environments. In this regard, the measurement of stress‐related endocrine markers provides a useful tool to examine the impact of environmental challenges and the physiological consequences for wildlife occupying such space. The aims of the present study were to validate fecal glucocorticoid metabolite (fGCM) concentrations as a measure of stress using samples from a male African clawless otter (Aonyx capensis; n = 1) and to compare fGCM concentrations of otters occurring in a transformed and in 2 natural areas in South Africa. From the 5 different enzyme‐immunoassays (EIA) tested, a cortisol and oxoetiocholanolone (measuring 11,17 dioxoandrostanes) EIA revealed the highest response (74% and 48% increase, respectively) 30 and 24 hours after a stress event (translocation of a captive individual as part of its rehabilitation prior to release), respectively. For both EIAs, fGCM concentrations were comparable for samples collected up to 3 h post‐defecation. Using the cortisol EIA for subsequent analyses, fGCM concentrations of animals from the transformed area (n = 20; mean [± SD]: 0.468 [± 0.539] µg/g dry weight [DW]) were significantly higher (P = 0.013) than those from otters in the natural areas (n = 17; 0.242 [± 0.226] µg/g DW). These preliminary results suggest that African clawless otters may have increased adrenocortical activity that could be due to conditions linked to living in a transformed environment.  相似文献

AIMS: The aims of this study were (a) to evaluate the effect of xylazine and tolazoline, with and without lignocaine, on the cortisol response of calves following amputation dehorning and (b) to assess the effect of a non-steroidal anti-inflammatory drug (ketoprofen) and local anaesthesia on the cortisol response of calves to amputation dehorning. METHODS: Plasma cortisol concentrations were measured in 100 dehorned or non-dehorned 3-month-old calves over an 8-h period following five different sedative/analgesic or control treatments. Sedative/analgesic treatments were: control (no anaesthesia); local anaesthesia and ketoprofen; local anaesthesia and xylazine; local anaesthesia, xylazine and tolazoline; and xylazine only. Within each sedative/analgesic treatment group, half the calves (n=10 per group) were amputation dehorned and half were not dehorned. RESULTS: The change in plasma cortisol concentrations in calves dehorned after being given ketoprofen and local anaesthesia did not differ significantly from that of non-dehorned control calves for at least 8 h. In contrast, the cortisol response of dehorned calves not given analgesic drugs peaked 30 min after dehorning and lasted >4 h. Xylazine injected before dehorning significantly reduced but did not eliminate the peak of the cortisol response. When both xylazine and local anaesthesia were administered before dehorning the peak in the cortisol response was virtually eliminated. In the dehorned calves that received xylazine with or without local anaesthesia, cortisol concentration increased significantly 3 h after dehorning and did not return to baseline until at least 5 h later. When tolazoline was administered shortly after xylazine, it caused a marked cortisol response, higher than the response to any other treatment. CONCLUSIONS: Combining ketoprofen and local anaesthesia minimised the cortisol response, and by inference the pain-induced distress, following amputation dehorning in calves. Xylazine reduced the initial cortisol response to dehorning but not as much as when local anaesthesia was also given. The increase in cortisol concentration from 3-8 h after dehorning in calves given xylazine alone or in combination with local anaesthesia suggests that calves experienced pain-induced distress during this time and that xylazine had no long-term analgesic effect. Tolazoline, used to reverse the sedative effects of xylazine, caused a marked cortisol response in calves via a mechanism which remains unclear.  相似文献

Although observed in other Perrisodactyla, urine overmarking in white rhinos has not been described. Using a single opportunistic sighting, we were able to collect two dung samples from one oestrous female white rhino; one unmarked and one overmarked with male urine. Because of the behaviour of both the male and the female, we suggest that the female was in pro-oestrus. We hypothesised that the function of overmarking by the male was for oestrus concealment (i.e. odour masking), as observed in horses. Because dung from oestrous female white rhinos emits a higher proportion of alkanes than non-oestrous dung, we expected the proportion of alkanes emitted from oestrous dung to decrease after overmarking. In contrast, we found that after overmarking the proportion of alkanes emitted increased. We suggest that the function of urine overmarking in white rhinos could be to conceal all signals of reproductive condition, so that neither oestrous nor non-oestrous signals are recognisable, or that a signal is added to indicate that the female has been mated.  相似文献

White rhinoceroses (Ceratotherium simum) anesthetized with etorphine combinations develop severe pathophysiologic changes, including hypoventilation, hypoxemia and metabolic acidosis. The aim of this study was to evaluate the addition of butorphanol to the immobilizing mixture on the cardiopulmonary effects in free-ranging white rhinoceroses darted from the helicopter. In the control group (n=15), the rhinoceroses were anesthetized with etorphine, azaperone, detomidine, and hyaluronidase administered intramuscularly. In the treatment group (n=16), 10-20 mg of butorphanol was added to the combination. Within 10 min of becoming immobile, vital parameters (heart rate, respiratory rate, and temperature) and blood gas analyses were taken, and measurements were repeated after 10 (treatment group) and 20 min (control group). Both groups showed respiratory and metabolic acidosis, hypoxemia, and hypercapnia. In the control group, the arterial partial pressure of oxygen was significantly higher and the alveolar-to-arterial oxygen pressure gradients were significantly lower in all body positions compared with the butorphanol group. Oxygen hemoglobin saturation in the control group was higher than in the butorphanol group only in the lateral position. Improvements in arterial oxygen levels were observed in all animals when placed in sternal recumbency. There were no significant differences in the mean induction times between groups, but the distance the butorphanol group ran was significantly less after darting than in the control group. By adding butorphanol to the immobilizing mixture, no benefits in ventilation were seen; although, size differences make comparisons difficult. Running for a shorter distance during induction could be beneficial in the prevention of severe acid-base imbalances and capture myopathy.  相似文献

A 16-day-old white rhinoceros calf died suddenly while in excellent condition and showing no obvious previous clinical signs. It was the 9th calf of a mature female kept free-ranging with 11 other rhinoceros as well as various other game species on a 2000 hectare game breeding centre adjacent to the town of Lichtenburg and outlying cattle farmlands in the Northern Province. At post mortem examination, death was ascribed to heart failure. There was marked multifocal to coalescing subacute parasitic myocarditis with numerous protozoan bradyzoite cysts and free tachyzoites present amongst the predominantly round cell inflammatory infiltrate. The coccidian was positively identified as Neospora sp. using both polyclonal and murine monoclonal Neospora caninum antibody immunohistochemistry in the avidin-biotin technique. The parasites stained poorly with Toxoplasma gondii-specific immunoperoxidase staining. Ultrastructurally, a section of a bradyzoite-containing cyst, as well as tachyzoites, were largely but not totally consistent with those described for Neospora caninum. The dam showed no sign of illness. Neosporosis affecting white rhinoceros (Ceratotherium simum) has not previously been reported. Objective To measure plasma cortisol responses in calves dehorned using a scoop after administration of local anaesthesia and/or cautery of the wounds.
Design A physiological study with controls.
Procedure There were six treatments: control handling with and without local anaesthesia, dehorning, dehorning after local anaesthesia, dehorning followed by wound cautery, and dehorning after local anaesthesia followed by wound cautery. Blood samples were taken before and after dehorning.
Results Dehorning caused an increase in plasma cortisol concentrations, which decreased a little to plateau values and then declined to pretreatment values 3 to 4 h after dehorning. The peak was smaller after local anaesthesia was administered but when its effects wore off, cortisol concentrations increased and thereafter were similar to those in the dehorned animals. The combination of local anaesthesia and cautery resulted in a plasma cortisol response similar to those in control calves with or without local anaesthesia.
Conclusions If plasma cortisol concentrations reflect the distress being experienced by the calves, then local anaesthesia reduces the acute distress for about 3 h after dehorning but not during the subsequent 3 to 4 h. Combining local anaesthetic and cautery prevented the significant increase in plasma cortisol following dehorning and may eliminate the acute distress caused by scoop dehorning.  相似文献

Dehorning and disbudding are routine painful procedures carried out on cattle to facilitate management. The pain caused by these procedures and its alleviation may be evaluated by monitoring behaviour and physiological responses, and by measuring their effects on weight gain. The cortisol response to cautery disbudding is significantly smaller than that to amputation dehorning which infers that the latter is more painful. Amputation dehorning stimulates a defined cortisol response with a rapid rise to a peak value within 30 min followed by a decline to a plateau which then declines to pre-treatment values after about 8 h. A cornual nerve blockade using lignocaine virtually eliminates the escape behaviour seen during disbudding and dehorning and reduces the plasma cortisol response to dehorning for about 2 h. Thereafter there is an increase in the plasma cortisol concentration, a delayed response, which lasts for about 6 h. A cornual nerve blockade, using lignocaine combined with cauterizing the wound caused by amputation dehorning, virtually eliminates the cortisol response as does combining a lignocaine blockade with the non-steroidal anti-inflammatory drug (NSAID) ketoprofen. When xylazine is combined with a cornual nerve blockade using lignocaine before dehorning, the cortisol response is virtually eliminated for about 3 h. When this regime is used before cautery disbudding and includes a NSAID given before and after disbudding the behaviour of calves so treated suggests that pain may be alleviated for 24 h. Cautery disbudding is preferable to amputation dehorning, but for optimal pain relief xylazine sedation, local anaesthesia and a NSAID should be used with both procedures.  相似文献

In this study, an attempt was made to use vaginal electrical impedance to predict calving in a female white rhinoceros (Ceratotherium simum simum) and to determine the relationship between vaginal electrical impedance and hormonal profiles during pregnancy. The principle behind vaginal electrical impedance is that a change in the ionic balance of vaginal and cervical mucus occurs in response to changes in reproductive hormones. Three times weekly vaginal electrical impedance readings and fecal samples were collected from midgestation to calving (a 6-mo period). The extracted fecal samples were analyzed for immunoreactive estrogens, progestagens, and corticoids by RIA. Vaginal electrical impedance readings did not decrease before calving but remained consistent throughout the last 140 days of pregnancy. Fecal progestagens in the white rhinoceros decreased between day 17 and day 1 before calving, whereas estrogens increased between 4 and 2 mo before calving, with an additional increase occurring 1 mo before calving. Fecal corticoids increased 5 mo before calving, slowly declined, and increased again within 3 wk before calving. A decline in vaginal electrical impedance was noted 168 days before calving and remained at low levels for 4 wk. At the time of this decrease, the female became aggressive toward the male and began lactating. Fecal progestagens and estrogens did not change during this time; however, fecal corticoids increased as vaginal electrical impedance readings returned to normal along with her behavior and cessation of lactation. In summary, the use of vaginal electrical impedance could not predict parturition in the white rhinoceros. However, an anomaly occurred during pregnancy that was supported by vaginal electrical impedance readings, a change in female behavior, premature lactation, and a subsequent increase in fecal corticoids. The etiology of this physiological anomaly is unknown, yet it did not compromise pregnancy.  相似文献

Xylazine reduced the initial cortisol response to dehorning but not as much as when local anaesthesia was also given. The increase in cortisol concentration from 3–8 h after dehorning in calves given xylazine alone or in combination with local anaesthesia suggests that calves experienced pain-induced distress during this time and that xylazine had no long-term analgesic effect. Tolazoline, used to reverse the sedative effects of xylazine, caused a marked cortisol response in calves via a mechanism which remains unclear.  相似文献

Caustic paste is frequently used for disbudding young female dairy calves. Nerve blocking may not be completely effective after such chemical tissue damage. Regional anaesthesia, together with a non-steroidal-anti-inflammatory drug (NSAID), was shown to reduce plasma cortisol in calves disbudded using caustic paste. To find out whether pre-emptive NSAID alone could control pain or whether NSAID reduces cortisol response by a mechanism other than by pain control, we compared cortisol levels and behaviour of 10 chemically disbudded calves treated with IV flunixin-meglumine, five of which were injected at 5 min (F0) and five injected at 60 min before dehorning (F1), with 5 sham-dehorned (ND) and 5 non-treated chemically disbudded animals (CD). There was a higher (p < 0.001) cortisol level in both NSAID-treated groups compared with ND at 1 h after disbudding, but no differences from control animals (CD). Non-treated disbudded animals showed higher cortisol at + 3 h compared with ND. A higher incidence of pain-related behaviours was shown in disbudded animals up to 3 h post-disbudding.We conclude that pre-emptive analgesia treatment by itself is not effective in controlling pain and does not prevent blood cortisol increase after disbudding of calves with caustic paste.  相似文献
