Sheep lacrimal glands are mixed glands, consisting of tubulo-acinar units succeeded by ducts of simple morphology. The secretory portions consist of three cell types: mucous, seromucous and serous, which may be intermingled in the same acinus or may form acini wholly made of only serous or mucous cells. Mucous cells show a rough endoplasmic reticulum that is reduced to a few cisternae located near the cell base and among the interstices of the secretory droplets. Mucous granules appear uniformly electron-lucent. Serous cells display a typical structure; serous granules can be uniformly electron-dense or composed of dense inclusions dispersed in an electron-lucent matrix. The seromucous granules have a bizonal substructure: a dense core is embedded in a highter matrix. Secretory acini are succeeded by intercalated ducts; the epithelium of these ducts gradually increases in height to form a kind of excretory duct, without the intervention of striated ducts.  相似文献

Intraocular choristomas are rare anomalies in domestic animals and are often associated with multiple ocular malformations. A Thoroughbred foal presented for ocular abnormalities and was diagnosed with microphthalmia, corneal dermoids, severe anterior segment dysgenesis (including glandular choristomas), aphakia, retinal dysplasia, and optic nerve hypoplasia. Morphological, histochemical, and immunohistochemical comparisons were made between ocular choristomatous tissues from this foal and lacrimal gland, third eyelid gland, nasopharynx, trachea, and lacrimal sac/nasolacrimal duct from normal horses. Morphologically the choristomatous tissues (glands and epithelium lining the anterior segment) were most similar to the lacrimal sac. Histochemistry of glandular components found the glands associated with the lacrimal sac/nasolacrimal duct to be serous, as was the glandular intraocular choristomas. Our findings suggest that the origin of intraocular glandular choristomas in this case is from the lacrimal sac.  相似文献

In the present study, we firstly demonstrated immunohistochemical expressions of secretory carbonic anhydrase (CA-VI) isozyme in bovine forestomach, large intestine and major salivary glands. CA-VI was detected in basal layer epithelial cells of esophageal and forestomach stratified epithelium, in mucous cells of upper glandular region of large intestine, in serous acinar cells of the parotid gland, in serous demilune cells and some ductal liner cells of mandibular, monostomatic sublingual and esophageal glands. These immunohistolocalizations suggested that bovine CA-VI plays various roles in pH regulation, maintenance of ion and fluid balance, and cell proliferation.  相似文献

The functional properties of sialic acids appear to be manifold. Therefore, they are considered as essential components of saliva. In this study, the localization of sialoglycoconjugates in the submandibular glands of Japanese miniature (Shiba) goat was examined by light and electron microscopic histochemical methods. The submandibular glands exhibited a large amount of sialic acids. Additionally, sialic acids with O-acetyl substitutions were detectable in the mucous acinar cells and serous demilunar cells. According to lectin histochemical methods, the mucous and serous cells mainly contained the Siaα2-6Gal/GalNAc sequence. These sialoglycoconjugates generated by the submandibular glands may specifically participate in the maintenance of the viscoelastic properties of saliva, protection of oral tissues and prevention of pathogenic microbial attacks. Therefore, our results suggest that they are essential components of saliva to maintain oral health.  相似文献

This study aimed to investigate the major salivary glands in the southern white-breasted hedgehog (Erinaceus concolor) histologically and histochemically. Five adult males were included in this study. The results showed that anatomically the shape of the parotid, submandibular and sublingual glands of the Erinaceus concolor was respectively almost pear, elliptical to pyramidal and oval. Histologically, the parotid gland had serous acini and secretory cells showed negative reaction to alcian blue (AB) (pH = 2.5) and negative response to aldehyde fuchsin (AF), methylene blue (MB) and PAS stains. The submandibular gland was a mixed gland of mucous and serous acini. The mucous acini were strongly positive for PAS, AB, MB and AF. However, serous acini were week for PAS and negative against AB, MB and AF stains. The sublingual gland was purely composed of mucous acini. The mucous acini of the sublingual gland were strongly positives for PAS, AB and MB methods. While their reaction to the AF staining was negative. In conclusion, the histological and histochemical observations of the major salivary glands of the southern white-breasted hedgehog (E. concolor) indicated that these glands shown similarities and some special different histochemical features as compared to other mammalian species.  相似文献

Complex carbohydrates were examined in glandular cells of the horse duodenal gland by using lectin histochemical techniques. In the horse, the duodenal gland was distributed in the area from the uppermost part of the small intestine to a point about 6m caudal to the pylorus. It consisted of two types of cells, mucous and serous cells. The former was found in glands distributed almost all over this part, but the latter was present in glands distributed restrictedly to the uppermost part of the small intestine at a point about 10 cm caudal to the pylorus. The cytoplasm of the mucous cell contained neutral glycoproteins with different saccharide residues as alpha-D-mannose, N-acetyl-beta (1----4)-D-glucosamine, galactose, alpha-galactose, alpha-N-acetylglucosamine, beta-D-Gal (1----3)-D-GalNAc, alpha-L-fucose and sialic acid. On the other hand, the serous cell contained neutral and acid glycoproteins with different residues such as alpha-D-mannose, alpha-D-glucose, beta-D-galactose(1----3)-D-N-acetylgalactose, alpha-L-fucose, N-acetyl-alpha-D-galactosamine, N-acetyl-beta (1----4)-D-glucosamine, galactose, alpha-galactose, alpha-N-acetylglucosamine and sialic acid. It is also elucidated in the present study that lipase, an enzyme for digestion, is contained in the serous cell of the equine duodenal gland.  相似文献

Previously, the distribution of myoepithelial cells (mecs) in the salivary glands was studied by both immunohistochemistry, and transmission electron microscopy; however, little was elucidated concerning their morphological features, especially in goats. This study was performed to investigate the correlation between the cytoarchitecture of the mecs in goat major salivary glands (parotid, mandibular, and sublingual glands) and the nature of the saliva secretion. The cytoarchitectural features of the mecs were examined by scanning and transmission electron microscopy as well as immunohistochemically. The secretory endpieces in the parotid gland are of the pure serous type, but in both the mandibular and sublingual glands they are of the mixed type. In all studied glands, the intercalated ducts were covered by mecs which, unlike the large stellate cells that surrounded the secretory endpieces, were spindle-shaped with few cytoplasmic processes. Interestingly, the mecs were found to bulge on the basal surfaces of the serous acini and intercalated ducts in all glands and to be in close contact to the seromucous tubules surface in the mandibular and sublingual glands forming a continuous network around it. In conclusion, the differences in the degree of development of the mecs as well as the number of their cytoplasmic processes may be correlated with the nature of the secretion and the number of the secretory granules. Thus these observations may have some relevance in the diagnosis of atrophy and pathogenic conditions of these glands.  相似文献

The acini of the bovine mandibular gland consist of mucous and demilunar seromucous cells. The mucous cells contain electron-lucid mucigen droplets or mucoid masses, only rarely delimited by membranes. The cisternae of the RER communicate freely. Mitochondria are scarce. The seromucous demilunar cells contain considerably more granules, and perigranular membranes are far more common. Many granules have characteristic alternating electron opaque and less electron dense areas. The prominent RER appears to be associated with a peculiar labyrinth of interconnecting tubules. Mitochondria are numerous. Numerous myoepithelial cells are observed. The cuboidal cells of the intercalated ducts lack signs of secretory activity. The columnar cells of the striated ducts have extensive infoldings of the basal plasma membrane, associated with mitochondria. The cell apex contains many granulated vesicles. Endogenous lactoperoxidase activity could be demonstrated only in the demilunar cells, within the secretory granules, the RER, including the perinuclear cisternae, as well as the labyrinth mentioned above. A positive reaction could be observed over acinar lumina and in intercellular canaliculi. The possible role of peroxidase as a component of a bactericidal system of the bovine saliva is discussed.  相似文献

An ultrastructural study has been made of gastric mucosa of a teleostean fish, Tilapia spp. The cytological features of the surface mucous cells, mucous neck cells, glandular cells and endocrine cells are described. The surface mucous cells, identified by their superficial localization, are characterized by apical granules. The mucous neck cells are distinguished by the appearance of their mucous granules and their localization between surface mucous cells and glandular cells. The gastric glands contain only one form of cell whose fine structure is similar to cells that secrete hydrochloric acid. Physiological implications of some ultrastructural features are also discussed.  相似文献

The ultrastructural differentiation and maturation of the neck cells and the zymogenic cells during physiological cell renewal were investigated in the abomasal oxyntic-gland region of cattle. Immature neck cells of the distal isthmus and proximal neck exhibit transitional morphology to the predominantly mucous isthmus cells. Neck cells confined to the glandular neck are characterized by bipartite peptic-cored mucous secretory granules. In a proximal-distal gradient along the neck, a progressive increase in the peptic granular component and concomitant reduction in mucous components paralleled by proliferation of the rough endoplasmic reticulum creates pre-zymogenic cells in the proximal glandular base. These, in turn, give rise to mature zymogenic cells with pure peptic secretory granules and typical zymogenic cell morphology. In the depth of the gland, older degenerative zymogenic cells are found. Variations in size and number of the zymogenic granules point to different secretory activities of the mature zymogenic-cell population of the glandular base. These results favour the conception of a zymogenic-cell lineage arising within the isthmus and passing through different developmental stages, including neck cells, during their migration down the gland.  相似文献

The morphology of the gland cells in the glandular sac area of the Bactrian camel and the composition of secretory substances were examined by histochemical methods. It was found that the gland cells of the glandular sac area were of the same type and size as those of the cardiac glands. The composition of secretory substances from the glandular sac area was the same as that of secretory substances from the cardiac glands. Moreover, secretory substances from the gland cells of the glandular sac area contained a great deal of acid glycoconjugates, such as sialic acid, in addition to neutral saccharides (fucose, mannose, glucose, N-acetyl-D-glucosamin, galactose and N-acetyl-galactosamin). Furthermore, immunohistochemical examination showed that progastricsin was present in the gland cells of the glandular sac area and the cardiac gland. In this study, histological analysis suggested that the stomach of the Bactrian camel is a single cavity stomach, formed as a result of multiple differentiation and growth of cardiac glands through the process of evolution.  相似文献

Morphometric, histological and histochemical studies were carried out on the sublingual salivary glands of the Arabian camel (Camelus dromedarius). The glands are of the tubulo-acinar type and consist of many lobules that are composed of two types of cells, mucoserous and seromucous. The mucoserous cells form the main secretory units of the gland but seromucous cells are much more seldom and form associated acini. The former cells secrete and elaborate large quantities of neutral mucosubstances, sialomucins and little sulphomucins while only the apical portion of the latter cells shows weak to moderate activity for neutral and acid mucosubstances. The histoenzymological tests employed here detected a considerable activity of alkaline phosphatase, succinic dehydrogenase, aminopeptidase and non-specific esterases, but weak activities of cytochrome oxidase, peroxidase and no activities of triacylglycerol lipase, beta-glucoronidase and amylase. The functional significance of these findings is discussed.  相似文献

The distribution of postganglionic autonomic nerve fibers in the lacrimal gland and gland of the third eyelid of dogs was studied by use of histochemical techniques. Adrenergic nerve distribution was identified by use of the sucrose-potassium phosphate-glyoxylic acid technique. A loose network of adrenergic nerves was found throughout the interstitium around acini and blood vessels and in vessel walls. Acetylcholinesterase staining was used to identify cholinergic nerve fibers. A cholinergic distribution pattern was found, although the cholinergic innervation appeared more dense than the adrenergic. In the gland of the third eyelid, mucus-secreting lobules and lipid-secreting lobules appeared to be equally innervated by parasympathetic fibers. These lobules could not be differentiated when the sucrose-potassium phosphate-glyoxylic acid technique was used. The techniques used in this study could not demonstrate whether direct contact was made by either cholinergic or adrenergic nerve fibers with secretory or myoepithelial cells. The presence of both nerve fiber types around acini suggests an interrelationship between the sympathetic and parasympathetic nervous system in lacrimal gland secretion in dogs.  相似文献

The histomorphological aspects as well as the histochemical content and distribution of glycoproteins (GPs) in the mucosa of the digestive tract of the white croaker Micropogonias furnieri were studied. The buccopharyngeal cavity and the esophagous showed a squamous stratified epithelium with mucous cells. The stomach presented three portions: cardias, fundus and pylorus. Tubular glands formed by a single type of gland cell were located along the cardias and fundus. Histochemical tests showed that the buccopharyngeal cavity and the esophagous presented the largest amount of the different types of mucosubstances. Both organs showed abundant secretory mucous cells that synthesize large quantities of neutral, sulphated and sialylated GPs. The surface epithelium in the cardias and fundus synthesized and secreted scarce sialylated and neutral GPs whereas the secretions of the apical surface were abundant. The pylorus secreted large amounts of neutral as well as sulphated and sialylated GPs. Gland cells secreted neutral GPs. The ultrastructural features of the gut cells were quite similar to those of other teleosts. The buccopharyngeal cavity and the esophagous surface epithelial cells, identified by their superficial localization, were characterized by cytoplasmic vesicles of different size. Abundant goblet cells with secretory mucous granules were also present. Gastric glands in the stomach contained just one form of cell with a fine structure similar to cells that secrete pepsinogen.  相似文献

Objective The third eyelid of domestic animals is important for the production and distribution of tears, in removing ocular debris and in protection of the globe, and has significant immunologic functions. Although it is known that tears contain antibodies of the immunoglobulin A (IgA) isotype which are produced mainly by plasma cells of the lacrimal gland, very little is known about the antibody repertoires in the third eyelid of domestic animals. To assess whether IgA is derived from local synthesis, we analyzed the location of IgA‐producing cells and the cellular distribution of secretory component (SC) in the third eyelid of domestic animals in a comparative study. Animal studied A total of 83 third eyelids of dogs, cats, pigs, cows, sheep, goats and horses were investigated in the course of this study. Procedures Third eyelids were obtained immediately after death, cut length‐wise, fixed overnight and processed for immunohistochemical detection of IgA and SC by the ABC technique. Results The results show that IgA‐producing plasma cells are densely populated in subepithelial spaces of the surface epithelium as well as in the nictitating gland in a species‐specific manner. In contrast, the SC could be demonstrated exclusively in glandular acinar and ductal epithelial cells and in different cell types of the surface epithelium, preferentially located on the bulbar side of the nictitating membrane. Conclusion It is suggested that most of the SC is locally produced by resident plasma cells and subsequently transferred through the surface epithelium and glandular duct cells by transcytosis. This indicates that the third eyelid is an important member of the secretory immune system in domestic animals.  相似文献

In this study, we aimed to observe the localization and expression of peptide hormones—leptin, ghrelin and obestatin—in the sheep tongue by immunohistochemistry. For that purpose, tongues of ten adult sheep were used. Leptin expression of moderate intensity was observed in the basal and parabasal epithelial cells of the luminal epithelium, and leptin was strongly expressed in the taste buds of the circumvallate and fungiform papillae and in von Ebner's glands. Ghrelin was primarily expressed in some of the skeletal muscle cells and the smooth muscle cells of the middle layer of blood vessels. A strong expression was observed in the epithelial cells lining the base of the groove surrounding the circumvallate papillae. Obestatin expression was particularly strong in the epithelial cells of the salivary ducts. It was also stronger in the von Ebner's glands than in the seromucous glands. Leptin, ghrelin and obestatin were shown to be produced at varying levels in different cell types, including epithelial, stromal and skeletal muscle cells, as well as in ganglion neurons, neural plexuses and blood vessels in the sheep tongue. Cellular localization and expression of these peptide hormones have not been investigated in many species including sheep.  相似文献

The histology of the pharyngeal cavity and oesophagus of the freshwater 'silverside'Odontesthes bonariensis (Cuvier and Valenciennes) and the characteristics of their mucous cells were investigated. The histological characterization of its digestive wall revealed that the mucosa is thrown with longitudinal folds. The epithelium covering the folds was stratified with abundant mucous cells and gustative corpuscles, which are lacking in the oesophagus. The muscularis mucosa was absent. The submucosa presented the compactum stratum. The muscularis was organized in longitudinal and circular layers of muscular striated fibres. The serosa with a flat epithelium was located only in the oesophagus. Using histochemical procedures including methods for localization and characterization of glycoproteins (GPs), no differences were detected between the mucous cells contents of the pharyngeal cavity and those of the oesophagus. The mucous cells showed a weak periodic acid-Schiff (PAS) positive reaction in their content. The reactions for the differential analysis of sialic acids from GPs are feeble for periodic acid-Schiff at low temperature and low pH (PA*S) and KOH/PA*S and strong for periodic acid/borohydride/KOH/PAS (PA/Bh/KOH/PAS) and KOH/PA*/Bh/PAS revealing the scarce presence of C7 or C9 substituted and non-substituted sialic acids and the abundance of C7, C8 substituted sialic acids, O-acyl sugars and neutral sugars respectively. The results suggest that the pharyngeal cavity with the gustative corpuscles would induce the gastric secretion whereas the oesophagus is mainly involved in the transport of the food bolus to the stomach with the aid of abundant secretion of mucus. GPs secreted on the surface of the mucous cells, likely related to environmental conditions, would be involved in the lubrication, protection against abrasion and inhibition of microorganism proliferation.  相似文献
