分别以传染性法氏囊病病毒 ( IBDV) X毒株细胞适应的第 5代、第 1 1代和第 1 7代毒感染 38日龄非免疫雏鸡进行致病性试验 ,确定 X毒株的毒力 ,并比较 3个代次毒的致病性差异 ,进而利用 RT-PCR扩增其VP2 基因可变区 ,测定其核苷酸序列 ,从分子生态学角度研究了非鸡胚源细胞传代培养对 IBDV X毒株毒力和抗原性的影响。结果表明 ,IBDV X毒株表现为中等毒力 ,在 Vero细胞上传代后 ,其毒力有所减弱。VP2 可变区氨基酸序列有 3个位置发生了替换 ,即第 5代毒氨基酸残基 2 62位为半胱氨酸 ,而第 1 1代毒和第 1 7代毒则变为酪氨酸 ( Cys→ Tyr) ;第 5代毒的 2 90位为缬氨酸 ,而第 1 1代和第 1 7代毒则变为蛋氨酸 ( Val→Met) ;第 5代毒和第 1 1代毒的 31 6位为赖氨酸 ,而第 1 7代毒则变为精氨酸 ( Lys→Arg)。 2 90位氨基酸替换导致该区域二级结构的改变 ,并对 IBDV的抗原性有显著影响。上述结果提示 ,这些氨基酸残基可能对维持IBDV的毒力和抗原性是必要的。通过毒株间 VP2 可变区序列的比较和系统发生树的分析 ,证明 IBDV X毒株与 Bursin-2疫苗毒关系很近。  相似文献

为了解IBDV流行强毒HQ-b株囊毒与其细胞适应毒间生物学特性差异及2毒株毒力变化与VP2、VP5基因变异的关系,对2毒株的细胞适应性、致病性等进行比较,同时对其VP2、VP5基因序列进行分析。结果表明,HQ-b株囊毒对CEF、CEK、CELi、DF-1和Vero均不适应,而细胞适应毒HQ株仅不能适应Vero细胞、且批内及批间毒价稳定。致病性结果显示HQ-b株对4周龄SPF鸡致死率高达80%,是真正的超强毒,而细胞适应毒致死率已降为0%。对VP2基因高变区研究表明,HQ-b株具备IBDV超强毒株的分子特征,即222A、256I、294I和299S;其细胞适应毒HQ株除222A→P、256I→V、294I→L和299S→N外,在VP2公认的毒力位点253(Q→H)、279(D→N)、284(A→T)位氨基酸也发生改变,导致细胞适应毒具备经典弱毒株的分子特征,即222P、256V、279N、284T、294L和299N。对VP5基因研究表明:流行强毒HQ-b株也具有IBDV超强毒株的分子特征;其细胞适应毒VP5基因有12个位点碱基突变并导致9处氨基酸变异,尤其是ORF区第2个碱基由"T"突变为"C"后,导致细胞适应毒VP5的N端丢失了4个氨基酸,这种突变与现有的疫苗毒完全一致。本研究提供了超强毒HQ-b培育、驯化后致病性和细胞适应性转变的分子机理,也丰富了IBDV分子流行病学的理论。  相似文献

将两株鸡传染性法氏囊病病毒(IBDV)弱毒株在SPF鸡体内连续传代至第5、第6代时,出现明显的法氏囊萎缩和B:B指数下降,表明IBDV弱毒株在鸡体内连续传代后毒力增强。为进一步阐释哪些基因位点导致了上述毒力的变化,本试验测定了基础弱毒株及其在鸡体内传代后各个代次毒的基因组序列,比对分析后发现VP2蛋白253位氨基酸发生了由H到Q或N的变异,表明VP2蛋白253位氨基酸的替换可能会增强传染性法氏囊病病毒在鸡体内的致病性。  相似文献

试验旨在研究一株传染性法氏囊病病毒(IBDV)河南分离株的毒力特征及其与VP2氨基酸序列特征的关系。通过提取IBDV C4株RNA,利用RT-PCR扩增其VP2基因,与其他不同毒力IBDV毒株进行核苷酸及推导的氨基酸序列比对分析,同时使用pET-32a(+)原核表达载体表达VP2基因,用SDS-PAGE和Western blotting检测重组VP2蛋白的表达。结果显示,扩增的IBDV C4株的VP2基因序列在进化关系上属于超强毒力IBDV(vvIBDV)分类,与选取的vvIBDV毒株代表毒株核苷酸序列同源性在98.1%~98.7%之间,其七肽区为S-W-S-AS-G-S(第326—332位氨基酸)符合超强毒株特征,且222(A)、256(I)、294(I)和299(S)位氨基酸与超强毒力毒株的4个特征性氨基酸一致;但IBDV C4毒株的VP2蛋白氨基酸序列与超强毒力毒株代表毒株UK661相比,201(D/G)、281(G/R)、313(V/A)位氨基酸不同,其中281位氨基酸的改变处于279—290的小亲水区内,与病毒抗原性有关;构建的pET-32a(+)-VP2原核表达载体在大肠杆菌BL21感受态细胞上表达出分子质量约67ku的重组VP2蛋白,为进一步比较201(G)、281(R)、313(A)位氨基酸差异导致的抗原特性改变提供了研究基础。本试验结果表明,IBDV C4株VP2基因与vvIBDV毒株VP2基因的主要特性一致,但也有3处氨基酸与代表毒株UK661存在差异,这些改变可能与中国IBDV毒株毒力的进化有关。  相似文献

本研究成功将鸡传染性法氏囊病超强毒vvIBDVGx株通过SPF鸡胚的快速培育及鸡胚成纤维细胞传代致弱,揭示了vvIBDV从超强毒力向弱毒力转化过程中,其主要结构蛋白VP2基因核苷酸及推导的氨基酸序列的变化规律。对不同代次细胞毒进行了序列分析。发现细胞毒在第7代以前,VP2基因序列没有改变。与欧洲标准超强毒氨基酸同源性达100%;细胞毒第8代,有个别核苷酸发生了改变。但没有影响氨基酸序列;细胞毒第9代是变化复杂的过渡代;10代毒VP2基因与欧洲标准弱毒Cu-1氨基酸序列同源性达97%;以后的细胞适应毒至20代。其VP2基因序列不再改变。致病性实验表明原代毒对4周龄SPF鸡致死率为64%。细胞毒第5代的致死率为60%,而20代毒对鸡无致病性。在鸡体内连续传代6代不返强。  相似文献

以传染性法氏囊病病毒(IBDV)浙江分离株(ZJ2000)基因组RNA为模板,采用Long-accurate RT-PCR(LA-PCR)一步法扩增并克隆了IBDV ZJ2000株基因组A节段全长cDNA。序列测定结果表明,克隆的A节段全长共3259个核苷酸,包括5'、3'端的非编码区(NCRs)和2个部分重叠的开放阅读框(ORF1和ORF2),与参比的血清1型毒株核苷酸序列的同源性高达95.2%-99.2%。二级结构预测表明,在5'-NCRs存在1个大型的茎环发夹结构。ORF2编码145个氨基酸的VP5,与参比毒株的同源性高达98.6%-100%。ORF1编码1012个氨基酸的VP2/VP4/VP3,在氨基酸水平上VP2、VP3、VP4与参比毒株的同源性分别达到94.9%-98.8、96.1%-98.5%、97.1%-99.2%。ZJ2000共有14-38氨基酸的替代,其中特有氨基酸6个,变异大多数集中在VP2高变区,突变率达2.9%-11%。第2个小亲水区内280位氨基酸由S替代了N、290位M替代了L。这2个突变可能与IBDV的抗原性有关。VP2-VP4剪切位点附近511和540位氨基酸的变异可能使ZJ2000株的毒力增强。分子系统进化树分析表明,ZJ2000与欧洲Cu-1株、P2株、CEF94株和中国Harbin株的有关最近,而与欧洲、香港、日本的超强毒株和美国的变异株相对较远。  相似文献

本试验在江苏省鸡场分离获得3株鸡传染性法氏囊病病毒(IBDV),采用RT—PCR法扩增VP2基因,将产物克隆入pMD18T载体,经测序,并与IBDV代表株VP2基因的高变区序列进行分析比较。结果显示,3个分离株与超强毒株、强毒株、突变株及弱毒株的核苷酸同源性在89.5%~98.9%之间,与弱毒株Cu-1和疫苗株PBG-98同源性最高,为98.9%;推导出的氨基酸序列与代表性毒株的同源性在98.2%~99.5%之间。其中,七肽区的第三个丝氨酸残基突变为精氨酸或苏氨酸,279和284位氨基酸残基突变为天冬氨酸和苏氨酸,222、294和299位氨基酸残基分别突变为脯氨酸、亮氨酸和天冬氨酸。上述试验表明3株分离株均为临床弱毒株。  相似文献

传染性法氏囊病病毒(Infectious Bursal Diseas Virus,IBDV)的超强毒株GX8/99经在鸡胚成纤维细胞(CEF)上连续盲传23代后,细胞适应毒株GXC23时鸡的致病性显著减弱,对4~6周龄SPF鸡的致死率从85%~90%减为0~5%.GXC23在96孔细胞培养板上经2次无限稀释法后得到3个克隆病毒.经SPF鸡回传20代后,相应的毒株GX-2820、GX-4820、GX-5820对SPF鸡的致死率仍维持在0~5%.序列比较表明,GX8/99在传代过程中,其细胞适应毒GXC23有8个碱基突变,其中5个碱基突变导致了氨基酸改变.bp#2 T→C突变,导致传代毒ORF中的起始编码子后移了4个氨基酸.而3个克隆化病毒回鸡20代后致病性不变,这几个位点也保持不变,且与3个疫苗株完全一致.进一步分析表明,有4个超强毒株和4个强毒株的VP5基因的5个位点与GX8/99原始毒相同,而另3个超强毒及6个强毒株与细胞适应毒GXC23一致.这些结果表明,超强毒GX8/99在细胞传代过程中VP5基因上5个氨基酸的变异主要与适应细胞培养相关而与致病性无关.  相似文献

为了解传染性法氏囊病病毒(IBDV)HLJ-0504株的分子生物学特征,本研究利用融合PCR技术获得HLJ-0504株全基因组序列。核苷酸和推导氨基酸序列遗传演化分析发现,HLJ-0504A节段位于IBDV超强毒的分枝上,而B节段则介于超强毒株和减毒株之间,形成一个独立分枝,属于一株新的自然重组病毒。VP2抗原性预测分析表明,HLJ-0504株VP2第Ⅰ亲水区中的氨基酸发生突变(D212N),该突变可能导致了HLJ-0504株抗原性发生漂变。本实验结果为深入研究IBDV的分子特征奠定了基础。  相似文献

REASONS FOR PERFORMING STUDY: Centesis of the bicipital bursa using an 8.9 cm long spinal needle has been reported but the alternative of employing a 3.8 cm long hypodermic needle requires validation. OBJECTIVE: To compare the efficacy of 2 different methods of centesis of the bicipital bursa and to evaluate the usefulness of ultrasonographic imaging to determine the location of solution administered when centesis of the bursa is attempted. METHODS: For Trial 1, 6 clinicians, who had no previous experience of centesis of the bicipital bursa, attempted to inject a solution composed of an aqueous radiopaque contrast medium and physiological saline solution (PSS) into the bicipital bursae of 2/12 horses using the previously described distal approach to inject one bursa and a proximal approach to inject the contralateral bursa. The bicipital tendon and bursa were examined ultrasonographically before and after injection; and both shoulders were examined radiographically to identify the location of the medium. In Trial 2, another 6 clinicians, also with no previous experience of centesis, repeated Trial 1, using 6 horses, but the radiopaque contrast medium was mixed with air instead of PSS. RESULTS: Accuracy of centesis using the proximal approach was 39% and that of the distal approach 28%. Ultrasonographic examination of the shoulder allowed the location of solution and air to be accurately predicted in all 12 shoulders examined. CONCLUSIONS: Clinicians who have had no previous experience performing centesis of the bicipital bursa are unlikely to be successful in centesis using either approach. Radiographic examination after injecting a radiopaque contrast medium may be necessary to assess the success of centesis especially if bursal fluid is not obtained during centesis. Injecting air along with the radiopaque contrast medium provides more accurate ultrasonographic confirmation of centesis and better radiographic definition than does injection without air.  相似文献

Ingestively masticated fragments were collected and sized via sieving. Different sizes of esophageal masticate and ruminal digesta fragments, and ground fragments of larger masticated pieces were incubated in vitro, and undigested NDF remaining at intervals of up to 168 h of incubation was determined. The ruminal age-dependent time delay (tau) for onset of digestion of NDF was positively correlated (P < 0.004) with the mean sieve aperture estimated to retain 50% of the fragments between successive sieve apertures (MRA). Degradation rate of potentially degradable NDF (PDF) and level of indigestible NDF were not related (P > 0.10) to MRA of masticated and ground fragments. Estimates of tau were positively related to MRA, with slopes of bermudagrass < corn silage < ruminal fragments of corn silage. It was concluded that fragment size-, and consequently, ruminal age-dependent onset of PDF degradation of a mixture of different fragment sizes results in an age-dependent rate of degradation of the more rapidly degrading of two subentities of PDF. Models are proposed that assume a tau before onset of simultaneous degradation of PDF from two pools characterized as having gamma-modeled age-dependency and age-constant rates. The ruminal age-dependent pool seems to be associated with the faster-degrading pool, and its rate parameter increases with range in MRA in the population of fragments. Conceptually, the ruminal age-dependent rate parameter for PDF degradation seems to represent a composite of several effects: 1) effects of the size-dependent tau; 2) range in MRA of the population of ingestively masticated fragments; and 3) subentities of PDF that degrade via more rapid age-dependent rates compared with subentities of PDF that degrade via age-constant rates. The estimated fractional rates of ruminative comminution of ingestively masticated fragments (0.060 to 0.075/h) were of a magnitude similar to the mean fractional rates of PDF digestion (0.030 to 0.085/h), which implies that ruminative comminution may be first-limiting to fractional rate of PDF digestion. The in vivo roles of ingestive and ruminative mastication of fragments on PDF degradation must be considered in any kinetic system for estimating PDF digestion in the rumen. These results and others in the literature suggest that the rate of surface area exposure rather than intrinsic chemical attributes of PDF may be first-limiting to degradation rate of PDF in vivo.  相似文献

Fractures of the anconeal process of 5 pigs ranging in age from 4 to 8 months were studied radiographically and histologically. Clinically, animals with a fracture of the anconeal process had a "tight," restricted gait. In pigs at 4.5 months of age, a radiolucent line through the base of the anconeal process was composed of fibrocartilage, fibrous connective tissue, and hyaline cartilage. Subperiosteal proliferation of woven bone was located along the cranial surface of the olecranon, adjacent to the base of the anconeal process. In older animals, the radiolucent line through the anconeal process contained variable amounts of fibrous connective tissue and fibrocartilage. The proliferation of subperiosteal bone at the base of the anconeal process formed a "buttress callus" which retained a radiolucent area between the callus and the proximal surface of the anconeal process. The latter region of radiolucency was continuous with the transversely oriented line that traversed the base of the anconeal process.  相似文献

以国际标准强毒R株人工感染非免疫产蛋鸡,定时扑杀,分别从鼻窦、眶下孔、气管、肺、气囊、卵巢和输卵管分离MG,并收集感染鸡所产蛋分离MG。结果表明,人工感染48小时后上、下呼吸道及肺已被全面感染,96小时气囊已被感染,120小时输卵管已能分离到MG,卵巢始终分离不到MG。人工感染鸡自144小时便能在其所产蛋中分离出MG。药物治疗能在72小时内消除感染,油乳剂苗则需24天后逐渐降低蛋内MG分离率,药物卵内注射、种蛋药浴、高温处理均能杀死卵内MG,但以研制的种蛋浸泡剂药浴效果为最好。  相似文献

乳酸杆菌作为益生菌广泛用于人和动物。本文综述了乳酸杆菌改善宿主健康的机制。乳酸杆菌可通过产生抗菌物质如乳酸、过氧化氢、细菌素,或者通过竞争营养或肠道黏附位点来抑制致病菌;通过诱导黏附素的分泌或阻止细胞凋亡而增强肠道的屏障功能,从而保护肠道。文章重点讨论了乳酸杆菌表面成分(表面蛋白、脂磷壁酸和肽聚糖)与肠道受体(C型凝集素受体、Toll样受体和 Nod样受体),阐述了他们结合后启动免疫调节信号,调控肠道免疫功能以发挥改善健康作用的机制。  相似文献
